Biodiesel Production and Nutrients Removal from Piggery Manure Using Microalgal Small Scale Raceway Pond (SSRP) (미세조류 옥외배양 시스템을 이용한 돈분 액체 비료의 영양염류 제거 및 바이오디젤 생산)

  • Due to the rapid energy consumption and fossil fuel abundance reduction, the world is progressively in need of alternative and renewable energy sources such as biodiesel. Biodiesel from microalgae offers high hopes to the scientific world for its potential as well as its non-competition with arable lands. Taking consideration to reduce the cost of production as well as to attain twin environmental goals of treatment and use of animal waste material the microalgal cultivation using piggery manure has been tested in this study. Unialgal strains such as Chlorella sp. JK2, Scenedesmus sp. JK10, and an indigenous mixed microalgal culture CSS were cultured for 20 days in diluted piggery manure using Small Scale Raceway Pond (SSRP). Biomass production and lipid productivity of CSS were $1.19{\pm}0.09gL^{-1}$, $12.44{\pm}0.38mgL^{-1}day^{-1}$, respectively and almost twice that of unialgal strains. Also, total nitrogen and total phosphorus removal efficiencies of CSS was 93.6% and 98.5% respectively and 30% higher removal efficiency compared to the use of unialgal strains. These results indicate that the piggery manure can provide microalgae necessary nitrogen and phosphorus for growth thereby effectively treating the manure. In addition, overall cost of microalgal cultivation and subsequently biodiesel production would be significantly reduced.

Monitoring of Microorganisms Added into Oil-Contaminated Microenvironments by Terminal-Restriction Fragment Length Polymorphism Analysis

  • Terminal-restriction fragment length polymorphism (T-RFLP) analysis was used to monitor inoculated oil-degrading microorganisms during bioremedial treatability tests. A pair of universal primers, fluorescently labeled 521F and 1392R, was employed to amplify small subunit rDNA in order to simultaneously detect two bacterial strains, Corynebacterium sp. IC10 and Sphingomonas sp. KH3-2, and a yeast strain, Yarrowia lipolytica 180. Digestion of the 5'-end fluorescence/labeled PCR products with HhaI produced specific terminal-restriction fragments (T-RFs) of 185 and 442 bases, corresponding to Corynebacterium sp. IC10 and Y. lipolytica 180, respectively. The enzyme NruI produced a specific T-RF of 338 bases for Sphingomonas sp. KH3-2. The detection limit for oildegrading microorganisms that were inoculated into natural environments was determined to be $0.01\%$ of the total microbial count, regardless of the background environment. When three oil-degrading microorganisms were released into oil-contaminated sand microenvironments, strains IC10 and 180 survived for 35 days after inoculation, whereas strain KH3-2 was detected at 8 days, but not at 35 days. This result implies that T-RFLP could be a useful tool for monitoring the survival and relative abundance of specific microbial strains inoculated into contaminated environments.

Studies on the Phage of Vibrio parahaemolyticus (Vibrio parahaemolyticus의 Phage에 관한 연구)

  • Authors have isolated phages of V. parahaemolyticus from shellfish and investigated some of their characteristics. The results obtained were as follows: Twenty-three phage strains(9.2%) out of 250 specimens were isolated. Plaques of phages were small, clear or turbid and $0.5{\sim}1.5mm$ in diameter. The electron micrographs of K3 phages showed two morphology; one was a hexagonal head about 105nm with a tail about 12nm, the other was a hexagonnal head about 60nm with a tail about 25nm. The host ranges of pahges were limited to V. parahaemolyticus strains and there appeared to be no relationship between the K serotypes of V. parahaemolyticus strains and the host ranges of the phage isolates. The adsorption rate of phages were more than 80% for $10{\sim}15$ minutes, the inactivation rate at $60^{\circ}C$ was more than 99% for $40{\sim}45$ minutes. The pH stability range was between 6.0 and 8.0. The inactivation rate of phages by UV irradiation was more than 99% for $45{\sim}75$ seconds.

Role of Dual Flagella in the Pathogenesis of Vibrio parahaemolyticus

  • Vibrio parahaemolyticus possesses two flagella systems: polar and lateral flagella for swimming in liquid and swarming on solid surfaces or in viscous environments. To elucidate the pathogenic role of these dual flagella systems, we constructed single- and double-deletion mutants of the lafA and flhAB flagellum genes and investigated their biofilm formation, cell adhesion, and colonization of the small intestine of suckling mice. The double-mutant strain was more impaired in biofilm formation than either of the single-mutant strains. In addition, the lafA, flhAB, and double-mutant strains showed 40%, 45%, and 60%, respectively, lower adherence to HeLa cells than the wild-type strain. Moreover, the lafA, flhAB, and double-mutant strains exhibited 49%, 5.6 and 6.7 times, respectively, lower colonization in a competition assay than the wild-type strain. These findings indicated that polar flagella were more important than lateral flagella for the pathogenesis of V. parahaemolyticus.

  • To gain insights into the phylogenetic relationships of genus Staurastrum and Staurodesmus, we analyzed nuclearencoded small subunit rDNA of 82 strains, and chloroplast atpB gene sequences of 44 strains belonging to three genera (Staurastrum, Staurodesmus, Cosmarium). Excluding the Staurastrum muticum and S. orbiculare, forty five strains of genus Staurastrum formed a well supported clade. It was shown that with no cell wall sculpture and processes, these two species have a strong phylogenetic relationship with genus Staurodesmus. Therefore, it is strongly recommended to transfer Staurastrum without processes and cell wall sculpture into Staurodesmus. S. obsoletus is a taxa that is transferred from Cosmarium. But, from this study, it has shown a phylogenetic relationship with Cosmarium. Therefore, this species is strongly recommended to transfer back to Cosmarium instead of Staurodesmus. As it was studied before, genus Staurastrum has shown monophyletic. Since the genus taurodesmus groups with Cosmarium, they were shown to be polyphyletic.

  • The movement of bone fluid from the region of the bone vasculature through the canaliculi and the lacunae of the surrounding mineralized tissue accomplishes three important tasks. First it transports nutrients to the osteocytes in the lacunae buried in the mineralized matrix. Second, it carries away the cell waste. Third, the bone fluid exerts a force on the cell process, a force that is large enough for the cell to sense. This is probably the basic mechanotrasduction mechanism in bone, the way in which bone senses the mechanical load to which it is subjected. The mechanism of bone fluid flow are described below with particular emphasis on mechanotransduction. Also described is the cell to cell communication by which higher frequency signals might be transferred, a potential mechanism in bone by which the small whole tissue strain is amplified so the bone cells can respond to it. One of the conclusions is that higher frequency low amplitude strains can maintain bone as effectively as low frequency low amplitude strains can maintain bone as effectively as low frequency high amplitude strains. This mechanism has many similarities with the mechanotransduction of acoustical signals in the ear. These conclusion leads to a paradigm shift in how to treat osteoporosis and how to cope with microgravity.

Involvement of Growth-Promoting Rhizobacterium Paenibacillus polymyxa in Root Rot of Stored Korean Ginseng

  • Paenibacillus polymyxa is a plant growth-promoting rhizobacterium (PGPR) which can be used for biological control of plant diseases. Several bacterial strains were isolated from rotten roots of Korean ginseng (Panax ginseng C. A. Meyer) that were in storage. These strains were identified as P. polymyxa, based on a RAPD analysis using a P. polymyxa-specific primer, cultural and physiological characteristics, an analysis utilizing the Biolog system, gas chromatography of fatty acid methyl esters (GC-FAME), and the 16S rDNA sequence analysis. These strains were found to cause the rot in stored ginseng roots. Twenty-six P. polymyxa strains, including twenty GBR strains, were phylogenetically classified into two groups according to the ERIC and BOX-PCR analyses and 16S rDNA sequencing, and the resulting groupings systematized to the degrees of virulence of each strain in causing root rot. In particular, highly virulent GBR strains clustered together, and this group may be considered as subspecies or biovar. The virulence of the strains seemed to be related to their starch hydrolysis enzyme activity, but not their cellulase or hemicellulase activity, since strains with reduced or no starch-hydrolytic activity showed little or no virulence. Artificial inoculation of the highly virulent strain GBR-1 onto the root surfaces of Korean ginseng resulted in small brown lesions which were sunken and confined to the outer portion of the root. Ginseng root discs inoculated in vitro or two-year-old roots grown in soil drenched with the inoculum developed significant rot only when the inoculum density was $10^{6}-10^{7}$ or more colony-forming units (CFU) per ml. These results suggest that P. polymyxa might induce ginseng root rot if their population levels are high. Based on these results, it is recommended that the concentration of P. polymyxa should be monitored, when it is used as a biocontrol agent of ginseng, especially in the treatment of stored roots.

  • The evolution of strain states and textures during rolling with various conditions was investigated by finite element method (FEM) simulations and measurements of rolling textures. Symmetrical rolling with a high friction gives rise to a strong variation of shear strains in rolled sample leading to the formation of texture gradients throughout the thickness layers. A small variation of shear strains during rolling with a well lubrication condition leads to the formation of a fairly homogeneous rolling texture throughout the sheet thickness. During asymmetrical rolling, a proper control of rolling parameters provides the evolution of a fairly homogeneous shear texture throughout the whole sheet thickness.

  • Ten terpenes (1-10), two sterols (11 and 12), and two unusual phenylpropanoids (13 and 14) were isolated from the brittle star Ophioplocus japonicus (Family Ophiuridae). Their structures were identified by analyses of the spectral data and by comparison with the literature data. The terpenes (1-10) and phenylpropanoids (13 and 14) are first encountered in brittle stars. These compounds were evaluated for cytotoxicity against a small panel of human solid tumor cell lines. Compounds 6-10 and 14 displayed moderate to significant cytotoxicity. The compounds were evaluated for antibacterial activity against 20 clinically isolated strains. Compound 5 exhibited antibacterial activities against three Streptococcus and three Staphylococcus strains.

  • In this paper, dynamic properties of fiber reinforced soils were investigated at shearing strains between $10^{-4}%\;and\;10^{-1}%$ using resonant column test. Resonant column test has been widely used as a primary laboratory testing technique in investigating dynamic soil properties expressed in term of shear modulus and material damping. At strains above elastic threshold, the variations of shear modulus(G) and damping ratio(D) were investigated. Based on test results, the small strain shear modulus($G_{max}$) and damping ratio($D_{min}$) were determined and the effects of confinement on $G_{max}$ and $D_{min}$ were characterized.
