Application of methylene blue color test for the detection of inherited susceptibility to hemolysis of Korean native cattle (한우(韓牛)의 선천성용혈감수성검사(先天性溶血感受性檢査)에 대한 methylene blue 청색소실시험법(靑色消失試驗法)의 적용(適用))

  • Blood samples were obtained from Korean native cattle and dairy cattle of Holstein species in the slaughter house and methylene blue color tests were performed for the detection of the inherited susceptibitity to hemolysis. Glucose-6-phosphate dehydrogenase activities expressed as the optical density obtained by methylene blue color test were the highest as 0.54 in male Korean cattle, 0.62 in female Korean cattle and 0.72 in dairy cattle of Holstein species. Percent hemolysis, packed cell volume and plasma protein contents were measured and compaired with relation to the results of methylene blue color test and no correlation were observed in each.

  • The incidences of liver abscess, fatty liver and fascioliasis were 11.9%, 23.2% and 36.0%, respectively in 211 Holstein cattle observed at the slaughter house. Blood chemical values of them were measured. The results were as follows : 1. Liver abscess group was significantly higher than normal group in the levels of SDH, AST, TP, globulin and fibrinogen, but it was significantly lower than normal group in the levels of albumin and A/G ratio (p<0.01). 2. Fatty liver group was significantly higher than normal group in the levels of SDH, AST, OCT, TP and globulin, but it was significantly lower than normal group in the levels of albumin and A/G ratio (p<0.01). 3. Fasciolisais group was significantly higher than normal group in the levels of AST. TP and globulin (p<0.01), but it was significantly lower than normal group in the levels of albumin and A/G ratio (p<0.01).

  • Muscular lesion was detected in a 23-month-old castrated bull encountered at Kyungsan slaughter house. The lesion appeared as fat intervening muscle fibers. The affected animal had no clinical signs. On microscopic examination, there was replacement of many muscle fibers by normal fat cells. Numerous fat cells were located between muscle fibers. Remaining skeletal muscle cells were in degenerative process, and thus abnormal skeletal muscle cells had loose fibers while normal had intact ones. The advent of inflammatory cells is not at the lesion, which is unique view in steatosis.

Nuclear Changes Occurring During Cannine Oocyte Maturation In Vitro (개 난자의 체외성숙중 핵변화)

  • Canine follicular oocytes were used to establish a reliable system for maturation and fertilization in vitro. Ovaries were obtained from either slaughter house or hormone-primed bitches of mixed breeds. The oocytes were recovered by mincing the ovaries in M2+BSA. Good quality of oocyte-cumulus complexes (OCCs) were selected and cultured in TCM 199 containing 15% fetal calf serum(FCS) for 24~56 h in an atmosphere of 5% CO2 at 39$^{\circ}C$. Maturation rate of follicular oocytes was >87% showing metaphase I. Unlike other domestic animals the cumulus expansion did not occur fully in canine OCCs although minimum expansion was found between the cumulus cells and corona radiata cells, the clear nuclear morphology was presented for the first time by rapid staining. The IVM system used in this study may be useful to obtain fully maturated metaphase I oocyte in dog.

Cutaneous Squamous Cell Carcinoma Occurred in Broiler Chickens (육용계의 피부에 발생한 편평세포암종)

  • Two 53-day-old broiler chickens were condemned due to multiple cutaneous nodular masses at slaughter house. Grossly, variable sized round masses were scattered in the abdomen and pelvis. Some of them were large and fused each other. Center of large fused nodules were ulcerated. Histopathologically, tumor masses consisted of multiple cords or masses of epidermal cells. Some of \ulcornerem contained central conoentric lamella of keratin (cancer pearl). From these results, these cases were diagnosed as cutaneous squamous cell carcinoma. This is the first report of cutaneous squamous cell carcinoma occurred in broiler chickens in Korea.

  • One cytopathogenic virus was isolated in alveolar macrophages from pig without any apparent respiratory clinical signs. Biophysical properties and electron microscopy of the isolate showed the characteristics of adenovirus. Intracytoplasmic inclusion bodies were seen in virus-inoculated cells. The genetic analysis indicated the presence of DNA with the size of >20Kb. In a serological survey of 40 serum samples collected from two different farms in slaughter house, 9 sera were positive for neutralizing antibody against the isolate. The potential implications of the isolate as the causative agent in respiratory disorder were discussed.

  • From April to May 1967 3,275 Korean native cattle slaughtered at Taegu public slaughter house were investigated for infection rate of faciola and 222 adult fasciola species taken from the bile duct, of the patients were examined for morphological study. The results were as follows: 1. Among 3,275 Cattle 1,099 Cattle (32.5%) were infected with fasciola species. 2. Seasonal changes of infection rate were 34.7%(Winter). 33.2%(Spring), 30.9%(fall) and 29.1%(Summer). 3. Three different species(large, small and intermediate type) of fasciola were detected from the patients. These large, small and intermediate types were identified morphologically with fasciola gigantica, fasciola hepatica and fasciold indica respectivelly. Distribution of these three kind, were 52.4%(intermediate type), 28.4%(Small type) and 19.0%(large type).

A Study on the Characteristics and Incidences of T. hyodysenteriae on Pigs. (돈적병균의 분포와 특성에 관한 연구)

  • Feces were collected from pigs of various ages on slaughter house and swinery in the southern area of the kangwon-do. Treponema hyodysenteriae were isolated from the feces by the method of Jenkinson. and the effects of temperature on the survival of these organism were investigated. The results abtained are summerized as follows ; 1. T. hyodysenteriae were isolated from 22 fecal samples of 1, 296 samples, and showed higher incidence at 4-8week age groups than other age groups. 2. The organisms were isolated from 17 samples of 329 diarrhea samples and 5 samples of 967 non-diarrhea samples. 3. T hyodysenteriae survived long at $0-10^{\circ}C$ but survived short at $37^{\circ}C$ or above.

  • A serological survey for antibodies to Leptospira (L.) interrogans in dairy and Korean native cattle in Gyeongbuk area was performed using 6 different living antigens, which were L. icterohaemorrhagiae(RGA), L. canicola(Hond Utrecht IV), L. autumnalis(Akiyami A), L. australis(Ballico), L. pomona(Pomona) and L. hebdomadis(Hebdomadis), by microscopic agglutination test. In the microscopic agglutination test, greater than partial agglutination at a serum dilution of 1:300 or over was recorded as positive. In the dairy cattle(Holstein), four(4.7%) of 86 sera from 13 dairy farms were positive for L. canicola and L. pomona antibodies. In the Korean native cattle, four(3.2%) of 124 sera from the slaughter house in Taegu city were positive for L. canicola, L. icterohaemorrhagiae and L. pomona antibodies. Among the positive sera, L, canicola was dominated in this experiment.

  • At the Seoul first slaughter house, 2,964 heads of swine were examined for the infection and distribution of Cysticercus tenuicollis. The results were summarized as follows. 1. The Cysticercus tenuicollis were detected from 106 heads of swine and the infection rate were 3.58 percent. 2. Incidence at each province were 5.7% in Jeon-ra-puk Do, 4.4% in Chung-cheong-nam Do, 3.83% in Kyong-sang-nam Do, 3.57% in Jeon-ra-nam Do, 2.52% in Kang-won Do, 1.75% in Kyong-sang-puk Do, and Kyong-gi Do and Seoul City were not recognized. 3. The monthly variation of the infection of Cysticercus tenuicollis were not found nearly through a year. 4. The usual habitat of Cysticercus tenuicollis were recognized the omentum and liver, but unusually one case was found around the rectum.
