Objectives : Hyaluronic acid(HA) is a mucopolysaccharide, occuring naturally in living organisms. It is one of the most hydrophilic molecules, so it has been known as being related to skin hydration and skin aging. The purpose of this study is to examine the effects of Angelica gigas(A. gigas) ethanol extract on hyaluronic acid synthesis. Methods : To determine cytotoxicity and hyaluronic acid synthase 2 gene expression, hyaluronic acid production in HaCaT cells, MTT assay and RT-PCR ELISA was used. Results : There were no cytotoxicity in $50{\mu}g/ml$ concentration A. gigas extract in MTT assay. Hyaluronic acid synthase 2(HAS2) gene expression was increased by all treated concentration A. gigas extract. Hyaluronic acid production was higher than control group in $50{\mu}g/ml$ & $100{\mu}g/ml$ concentration A. gigas extract. Conclusions : Hyaluronic acid production was increased by A. gigas extracts. Therefore, We suggest that A. gigas can make a contribution to the moisturizing effect on human skin.
본 논문에서는 치아 씨앗으로부터 분리한 단백다당체(proteoglycan)의 성분분석과 분자량 측정 및 피부보습 효과를 연구하였다. 분리된 단백다당체의 성분분석 결과 치아 씨앗 단백다당체를 이루고 있는 당 성분(%)은 galactose (46.8), glucuronic acid (27.1), rhamnose (8.7), xylose (7.6), glucose (4.9), fructose (2.3), mannose (1.8), arabinose (0.9)로 확인되었으며, 함유된 단백질의 함량은 31.3 mg/g으로 이를 구성하는 아미노산 성분(mg/g)은 Asp (1.9), Glu (3.6), Ser (0.9), Gly (3.6), Thr (0.8), Arg (1.0), Ala (2.0), Tyr (0.4), Cys (4.8), Val (1.1), Phe (0.5), Ile (0.6), Leu (0.9)로 확인 되었다. Gel permeation chromatography (GPC)에 의해 측정한 치아 씨앗 단백다당체의 분자량 범위는 100,000~250,000 Da로 분자량 분포에 의한 평균 분자량은 170,000 Da이었다. 또한, 사람을 이용한 피부수분 보유 효과 및 피부 수분 손실량을 평가한 결과, 피부의 수분함량을 유지하는 효과가 우수하였으며 표피 수분 손실량 또한 적어서 피부 보습제로서 매우 우수하였다. 결론적으로 치아 씨앗의 다당체로부터 분리한 단백다당체를 화장품에 응용하면 우수한 보습효과를 가지는 화장품을 개발 할 수 있을 것으로 사료된다.
알레르기성 접촉피부염은 T세포와 대식세포가 관여하는 세포매개성 면역반응으로 항원에 노출된 뒤 수 일 후에 증상이 나타나는 지연형 반응이다. 그 과정은 감작기와 유발기로 나뉘는데 감작기에는, 표피장벽을 통해 유입된 항원이 표피기저층에 있는 항원전달세포에 의해 처리된 후 림프절로 이동되어 T세포에 의해 인식되고 그 T세포는 항원특이 T세포로 활성화된다. 유발기는 동일 항원이 재감작될 때 항원특이 T세포의 반응을 활성화시키고 다양한 cytokine 분비를 통해 염증세포를 항원 유입부위로 이동시킨다. 본 연구에서는 DNCB로 알레르기성 접촉피부염을 유발한 개의 모델에 폴리감마글루탐산의 항염증 효과를 평가하였다. 폴리감마글루탐산은 12일간 적용하였고 실험기간 동안 이틀 간격으로 피부 생리학적 지표를 측정하였으며 적용 후 cytokine 측정과 조직병리학적 검사를 실시하였다. DNCB 적용후 피부 생리학적 지표의 변화로 표피경유수분손실, 피부 수화도, 피부 두께 그리고 홍반지수는 증가하였고 피부 산도는 감소하였다(p < 0.05). 조직병리학적 검사결과 염증세포 침윤과 부종성 변화에 의한 상피두께 증가 및 진피 결합조직의 감소가 특징적으로 나타났다. 또한 진피에서 pro-inflammatory cytokine인 TNF-${\alpha}$와 IFN-${\gamma}$ 수치 및 상피에서 apoptotic change의 지표인 caspase-3와 PARP 면역반응세포의 수치가 유의적으로 증가하였다(p < 0.01). 하지만 폴리감마글루탐산 적용으로 피부 생리학적 지표(p < 0.05) 및 조직병리학적 변화가(p < 0.01) 기본 수치로 회복되었다. 따라서 본 연구를 통해 개에서 DNCB에 의한 알레르기성 접촉피부염 유발 및 폴리감마글루탐산의 우수한 항염증 효과를 확인하였고 그 결과 폴리감마글루탐산은 향후 피부염에 대한 치료제로 사용할 수 있을 것으로 기대된다.
피부는 신체에서 가장 크고 육중한 기관 중 하나로 인간의 생리 및 병리 과정에 참여한다. 피부는 자기 유지 및 치유, 기계적 및 화학적 손상 방어, 자외선 과 외부 병원성 미생물로부터의 방어, 비타민 D 합성 그리고 사회 심리적 기능을 한다고 알려져 있다. 이 연구의 목적은 한국 여성의 부위와 연령에 따라 피부 생물학 인자와 연관된 생물리학 인자의 변화를 평가하는 데 있다. 20 ~ 49세의 약 70명의 건강한 성인 여성이 이 실험에 참여하였다. 측정부위는 하박 내측과 뺨으로 진행하였다. 인체 피부의 생물리학 인자를 측정하기 위하여 여러 가지 비침습적인 방법으로 진행하였다. 피부의 생물학 인자를 분석하기 위하여 코티졸, 파이브로넥틴, 케라틴-1, 10, 11, 인보루크린, 케라틴 6를 인체의 얼굴과 하박내측으로 비교하였다. 또한 비침습적 방법으로 피부 생물리학 인자는 피부 부위와 연령에 따른 차이를 측정하였다. 측정 부위에 따른 결과, 각질층 수분량, 경피수분손실량과 피부색(L과 a값)은 유의적인 차이가 나타났다. 연령에 따른 결과, 오직 피부색에서만 연령에 따른 차이가 유의적으로 나타났다. 코티졸, 케라틴-6, 파이브로넥틴, 케라틴-1, 10, 11 은 연령과 부위간 유의적 차이가 없지만 인볼루크린은 30 ~ 39세 연령대에서 다른 연령대보다 유의적으로 가장 높았다. 이러한 결과는 개인의 피부 환경에 대한 상세한 피부 상태 변화로 설명할 수 있을 것이다.
Transdermal drug delivery(TDS) offers many important advantages. For instance, it is easy and painless, it protects the active compound from gastric enzymes, and it avoids the hepatic first-pass effect. Also, it is simple to terminate the therapy if any adverse or undesired effect occurs. But skin is a natural barrier, and only a few drugs can penetrate the skin easily and in sufficient quantities to be effective. Therefore, in recent years, numerous studies have been conducted in the area of penetration enhancement. The most commonly used transdermal system is the skin patch using various types of technologies. Compared with other method of dosage, it is possible to use for a long term. It is also possible to stop the drug dosage are stopped if the drug dosage lead to side effect. Polysaccharide, such as xanthan gum and algin were selected as base materials of TDS. Also, these polymers were characterized in terms of enhancers and drug contents. Among these polysaccharide, the permeation rate of Paroxetine such as lipophilic drug was the fastest in xanthan gum matrix in vitro. We used glycerin, PEG400 and PEG800 as enhancers. Since dermis has more water content(hydration) than the stratum corneum, skin permeation rate at steady state was highly influenced when PEG400 was more effective for lipophilic drug. Proper selection of the polymeric materials which resemble and enhance properties of the delivering drug was found to be important in controlling the skin permeation rate.

Especially, this result suggests a possible use of polysaccharide gel ointment matrix as a transdermal delivery system of anti-hyperlipoproteinemic agent.
Chronic exposure to ultraviolet (UV) radiation, regarded as a major cause of extrinsic aging or photoaging characterized by wrinkle formation and skin dehydration, exerts adverse effects on skin by causing the overproduction of reactive oxygen species. Agastache rugosa Kuntze, known as Korean mint, possesses a wide spectrum of biological properties including anti-oxidation, anti-inflammation, and anti-atherosclerosis. Previous studies have reported that A. rugosa protected human keratinocytes against UVB irradiation by restoring the anti-oxidant defense system. However, the anti-photoaging effect of A. rugosa extract (ARE) in animal models has not yet been evaluated. ARE was orally administered to hairless mice at doses of 100 or 250 mg/kg/day along with UVB exposure for 12 weeks. ARE histologically improved UVB-induced wrinkle formation, epidermal thickening, erythema, and hyperpigmentation. In addition, ARE recovered skin moisture by improving skin hydration and transepidermal water loss (TEWL). Along with this, ARE increased hyaluronic acid levels by upregulating HA synthase genes. ARE markedly increased the density of collagen and the amounts of hydroxypoline via two pathways. First, ARE significantly downregulated the mRNA expression of matrix metalloproteinases responsible for collagen degradation by inactivating the mitogen-activated protein kinase/activator protein 1 pathway. Second, ARE stimulated the transforming growth factor beta/Smad signaling, consequently raising the mRNA levels of collagen-related genes. In addition, ARE not only increased the mRNA expression of anti-oxidant enzymes but also decreased inflammatory cytokines by blocking the protein expression of nuclear factor kappa B. Collectively, our findings suggest that A. rugosa may be a potential preventive and therapeutic agent for photoaging.
Ursolic acid (UA) and Oleanolic acid (ONA), known as urson, micromerol and malol, are pentacyclic triterpenoid compounds which naturally occur in a large number of vegetarian foods, medicinal herbs, and plants. They may occur in their free acid form or as aglycones for triterpenoid saponins, which are comprised of a triterpenoid aglycone, linked to one or more sugar moieties. Therefore UA and ONA are similar in pharmacological activity. Lately scientific research, which led to the identification of UA and ONA, revealed that several pharmacological effects, such as antitumor, hepato-protective, anti-inflammatory, anticarcinogenic, antimicrobial, and anti-hyperlipidemic could be attributed to UA and ONA. Here, we introduced the effect of UA and ONA on acutely barrier disrupted and normal hairless mouse skin. To evaluate the effects of UA and ONA on epidermal permeability barrier recovery, both flanks of 8-12 week-old hairless mice were topically treated with either 0.01-0.1 mg/ml UA or 0.1-1 mg/ml ONA after tape stripping, and TEWL (Transepidermal water loss) was measured . The recovery rate increased in those UA or ONA treated groups (0.1 mg/ml UA and 0.5 mg/ml ONA) at 6 h more than $20\%$ compared to vehicle treated group (p<0.05). Here, we introduced the effects of UA and ONA on acute barrier disruption and normal epidermal permeability barrier function. For verifying the effects of UA and ONA on normal epidermal barrier, hydration and TEWL were measured for 1 and 3 weeks after UA and ONA applications (2mg/ml per day). We also investigated the features of epidermis and dermis using electron microscopy (EM) and light microscopy (LM). Both samples increased hydration compared to vehicle group from f week without TEWL alteration (p<0.005). EM examination using RuO4 and OsO4 fixation revealed that secretion and numbers of lamellar bodies and complete formation of lipid bilayers were most prominent $(ONA{\geq}UA>Vehicle)$. LM finding showed that thickness of stratum corneum (SC) was slightly increased and especially epidermal thickening and flattening was observed (UA>ONA>Veh). We also observed that UA and ONA stimulate epidermal keratinocyte differentiation via $PPAR\;\alpha$. Protein expression of involucrin, loricrin, and filaggrin increased at least 2 and 3 fold in HaCaT cells treated with either $ONA\;(10{\mu}M)$ or UA $(10{\mu}M)$ for 24h respectively. This result suggested that the UA and ONA can improve epidermal permeability barrier function and induce the epidermal keratinocyte differentiation via $PPAR\;{\alpha}$. Using Masson-trichrome and elastic fiber staining, we observed collagen thickening and elastic fiber elongation by UA and ONA treatments. In vitro results of collagen and elastin synthesis and elastase inhibitory activity measurements were also confirmed in vivo findings. These data suggested that the effects of UA and ONA related to not only epidermal permeability barrier functions but also dermal collagen and elastic fiber synthesis. Taken together, UA and ONA can be relevant candidates to improve epidermal and dermal functions and pertinent agents for cosmeseutical applications.
생체수는 링거액, 인공관절액, 세포 배양 등 다양한 분야에서 연구되어 왔다. 이는 생체수가 체온을 조절하는데 결정적인 역할을 하고, 생체 내 다양한 대사 과정에서 용매로 사용되며, 삼투압이나 능동적 섭취를 통한 혈액 또는 림프액을 통해 세포에서 세포로 전달되는 미네랄, 에너지원, 호르몬, 시그널과 약물의 전달 물질로 이용되기 때문이다. 세포외 지질과 자연보습성분(natural moisturizing factor, NMF)을 함유하는 각질층은 외부의 수분을 끌어들여 내부 수화에 이용하며, 이러한 과정들은 피부 장벽 기능과의 연관성이 높다. 본 연구에서는 피부 장벽 기능을 강화하는 아미노산, 펩타이드, 단당류를 포함한 생체모사수(Cell Bio Fluid SyncTM)를 인체 피부에 처리하여 세포활성을 관찰하고, 생체 모사수를 함유한 제품을 피부에 도포하여 인체피부 개선 효과를 연구 하였다. 피부 세포 활성을 보기 위해 각질세포인 HaCaT 세포를 이용하였고, 에너지 고갈상태로 만들기 위해 3시간 동안 PBS에 전 처리한 후 이후 세포 배양액인 DMEM 및 등장액인 PBS, 생체 모사수(Cell Bio Fluid $Sync^{TM}$)에 각각 3시간 처리하였다. 이후 MTT assay와 이미지 분석을 수행하였다. 인체의 피부 개선 효능을 위한 임상 연구에 21명의 여성이 참여하였다. 생체 모사수를 함유한 제품을 1주일간 안면에 도포하였고, 수분량, 피부결, 밝기 그리고 피부 균일도를 측정하였다. 모든 측정은 피험자가 세안 후 항온항습조건($22{\pm}2^{\circ}C$, $50{\pm}5%$)에서 20분 간 대기하고 수행하였다. 모든 데이터는 SPSS ver. 21을 이용하여 분석하였다. 그 결과, 생체 모사수(Cell Bio Fluid $Sync^{TM}$)가 세포 활성을 높이고 인체의 피부에서 수분, 거칠기, 밝기, 균일도, 투명도 등의 개선 효과가 있음을 확인할 수 있었다.

1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
