The effects of Two Terpenoids, UA and ONA on Skin Barrier and Its Application

  • Ursolic acid (UA) and Oleanolic acid (ONA), known as urson, micromerol, prunol and malol, are pentacyclic triterpenoid compounds which naturally occur in a large number of vegetarian foods, medicinal herbs, and plants. They may occur in their free acid form or as aglycones for triterpenoid saponins, which are comprised of a triterpenoid aglycone, linked to one or more sugar moieties. Therefore UA and ON A are similar in pharmacological activity. Lately scientific research, which led to the identification of UA and ONA, revealed that several pharmacological effects, such as antitumor, hepatoprotective, anti-inflammatory, antimicrobial, and anti-hyperlipidemic could be attributed to UA and ONA. Here, we introduced the effects of UA and ONA on acute barrier disruption and normal epidermal permeability barrier function. To clarify the effects of UA and ONA on skin barrier recovery, both flank skin of 8-12 weeks hairless mice were topically treated with samples (2mg/ml) after tape stripping, then measured recovery rate using TEWL on hairless mice. The recovery rate increased in UA and ONA treated groups at 6h more than 20% compared to vehicle treated group (p <0.05). For verifying the effects of UA and ONA on normal epidermal barrier, hydration and TEWL were measured for 1 and 3 weeks after UA and ONA applications (2mg/ml per day). We also investigated the features of epidermis and dermis using electron microscopy (EM) and light microscopy (LM). Both samples increased hydration compared to Vehicle group from 1 week without TEWL alteration (p<0.005). EM examination using Ru04 and OsO4 fixation revealed that secretion and numbers of lamellar bodies and complete formation of lipid bilayers were most prominent (ONA$\geq$UA>Vehicle). LM finding showed that stratum corneum was slightly increased and especially epidermal thickening and flattening was observed (UA>ONA>Vehicle). Using Masson-trichrome and elastic fiber staining, we observed collagen thickening and elastic fiber increasing by UA and ONA treatments. In vitro results of collagen and elastin synthesis and elastase inhibitory experiments were also confirmed in vivo findings. This result suggested that the effects of UA and ONA related to not only skin barrier but also collagen and elastic fibers. Taken together, UA and ONA can be relevant candidates to improve barrier function and pertinent agents for cosmetic applications.

  • Neonatal lupus(NL) is characterized by typical clinical features and the presence of maternal autoantibodies. The principal serologic markers of NL are anti-Ro/SSA or anti-La/SSB maternal autoantibodies, which are transferred across the placenta and can be detected for the first few months of the affected child. The major clinical manifestations are cardiac disease, notably congenital heart block, and cutaneous lupus lesions. Hepatobiliary disease is relatively rare clinical manifestation of NL. We experienced a case of NL with abnormal liver function test and skin lesion.

The Improvement of skin barrier function and anti-obesity effect of Codonopsis lanceolata by supercritical carbon dioxide extraction

  • The root of Codonopsis lanceolata has been used in traditional medicine. This study was conducted to confirm the comparative effect of ethanol solvent extraction (CLE) and supercritical carbon dioxide extraction (CLS) of C. lanceolata roots. CLS had higher antioxidant than CLE. For supercritical co-solvent modified carbon dioxide extraction (CLS), it were extracted at 250 bar 50℃ 150 min at a flow rate of ethyl alcohol 3 mL/min for 90min. In addition, CLS inhibited the adipocyte differentiation of 3T3-L1 cells. When treated with the extract at a concentration of 100 ㎍/mL, the Wnt/β-catenin pathway reporter luciferase activity of HEK 293-TOP cells increased approximately by 3-folds compared to that of the untreated control group. Also, the treatment by CLS (50 ㎍/mL) showed a significant increase of involucrin expression. These results indicate that supercritical carbon dioxide extract of C. lanceolatamay serve as a cosmeceutical agent for improving skin barrier function and the treatment of obesity.

  • Recently, it has been developed for Eco-environment, Super light, Multi-functional nano materials. As needed mobile parts in Smart phone or TV, computer, information communication for high pass signal, multi-function, Magnetic thin film materials have been developed. As last, magnetic powder, sintered and sputtering parts were thick and low purity than electroplating layer, low pass signal and noise were resulted, vibrated TV screen. Because chemical complex temperature was high and ununiform surface layer, it has been very difficult for data pass in High Frequency (GHz) area. Large capacity data pass is used to GHz. Above GHz, signal pass velocity is dependent on Skin Effect of surface layer. If surface layer is thick or ununiform, attachment is poor, low pass signal and cross talk, noise are produced and leaked. It has been reported technical trend of Electrochemically plating and Surface treatment of Metal, Polymer, Ceramic etc. by dispersion/complex for Multi functional nano-magnetic material in this paper.

  • Modern society is prevailed a lot of allergies. So, the allergy test is very important. There are many kinds of allergy test. A doctor usually uses skin allergy test among many allergy tests. However, little standadization and objectivity of grading-standard has been established in the skin allergy test. A measurement of the reaction area has been a major objective to perform skin allergy test. Recently, a doctor's method is to measure the reaction area after drawing a line that represents the reaction area on the skin. But this method differs slightly from the real reaction area and individual doctor's measurement is different, because the edge of the reaction area is obscure. In this paper, we propose a algorithm which is able to detect vague edges using the fuzzy set. The algorithm that detects the line and curve is proposed first. Here, the maximum value is calculated by comparing the membership function of the line and curve seperately. We also encode the direction of the line and curve by using 8-direction code. Then, we calculate the reaction area by measuring the pixels which are inside the reaction area. And finally the Allergy grade is decided by grading-standard, and we accomplish faster, the 80re accurate and objective allergy grade decision.

Biocompatibility of the Prepared Biodegradable Film from Algin and Chitosan (Algin과 Chitosan으로부터 제조한 생분해성 Film의 생체적합성)

  • Biodegradable films were prepared by solution blending method in the weight ratio of Chitosan and Algin for the purpose of useful bioimplants. The possibility of bioimplants, which prepared from natural polymers as a skin substitute and food wrapping materials were evaluated by measuring the biodegradability. these biodegradable films were inserted in the back of rats and their biodegradability was investigated by hematological change as a function of time. Rats study showed that low-Chitosan induced increments of monocyte and basophil after 48 hours of implantation. And medium-Chitosan showed increase of lymphocyte and decreased neutrophil counts after 48 hours of implantation. Low, medium Chitosan showed high hemoglobin contents, medium and high Chitosan showed high hematocrit value after 48 hours of implantation. As a result, medium, high-Chitosan induced potential incompatibility in the tissue after 48 hours, but there was little effects to the skin inflammation. The values of biodegradable films, which prepared from natural polymers measured in this study were some satisfiable results at short period with those of ideal skin bioimplants and artificial skin.

The Localization of Cytokeratin 19 and Vimentin in Sprague Dawley Albino Rat Skin Tissue

  • Cytokeratin 19 (CK19) expressed in epidermis of skin, bulge region of hair follicle, outermost layer of outer root sheath and proximal and distal to bulge. Vimentin is a fibrous protein that localized in cytoplasm of fibroblast and forms cytoskeleton to maintain shape of cell and nucleus. In this study, CK19 and vimentin in skin were confirmed with light, fluorescence and transmission electron microscope. As a result, CK19 was localized epidermis, hair follicles, outer root sheath and nucleus of Merkel's cell. However, vimentin was localized some epidermis, dermis, hypodermis and nucleus of Merkel's cell. The role of CK19 is self-renewal and homeostasis in skin. Also, hair follicle regeneration and hair growth is known to be related. It is supposed that required of structural proteins that make up cytoskeleton is increased. Thereby, expression of CK19 is increased. It is considered that vimentin localized in order to stabilize structure of cell and cytoskeleton of fibroblasts. Also, CK19 and vimentin present in nuclei of Merkel's cell, and to act as a fibrous protein that make up end of a nerve fiber present in Merkel's cell and paracrine function of Merkel's cell.

IL-4 Suppresses UVB-induced Apoptosis in Skin

  • In this study, cutaneous role of IL-4 in UVB-induced apoptosis was investigated using transgenic mice with skin-specific expression of IL-4 (IL-4 Tg mice). The transgenic mice did not show any gross clinical abnormalities. However, epidermis was thickened and increased MHC class II positive cells were detected as well as enhanced expression of inflammatory cytokines such as IL-1 and TNF-$\alpha$ in skin. In addition, histological analysis revealed increased infiltration of lymphocytes, acanthosis, hyperkeratosis, and parakeratosis in skin of IL-4 Tg mice. The physiological effect of IL-4 overexpression in skin against environmental stimulus such as UVB was investigated by irradiating wild-type and IL-4 Tg mice with UVB followed by evaluation of apoptosis. The result demonstrated suppressed apoptosis in epidermis of IL-4 Tg mice compared with wild-type mice. To further assess anti-apoptotic function of IL-4 in keratinocytes, stable cell clones were made where IL-4 was constitutively overexpressed and examined for UVB-induced apoptosis. The results showed that apoptosis was remarkably decreased in IL-4 over-expressing cell clones compared with that in mock transfected cells. Collectively, data presented here shows that IL-4 has an inhibitory effect against UVB-induced apoptosis in keratinocytes, suggesting that IL-4 may be an important regulator in cutaneous immunity against UVB.

A many-objective evolutionary algorithm based on integrated strategy for skin cancer detection

  • Nowadays, artificial intelligence promotes the rapid development of skin cancer detection technology, and the federated skin cancer detection model (FSDM) and dual generative adversarial network model (DGANM) solves the fragmentation and privacy of data to a certain extent. To overcome the problem that the many-objective evolutionary algorithm (MaOEA) cannot guarantee the convergence and diversity of the population when solving the above models, a many-objective evolutionary algorithm based on integrated strategy (MaOEA-IS) is proposed. First, the idea of federated learning is introduced into population mutation, the new parents are generated through sub-populations employs different mating selection operators. Then, the distance between each solution to the ideal point (SID) and the Achievement Scalarizing Function (ASF) value of each solution are considered comprehensively for environment selection, meanwhile, the elimination mechanism is used to carry out the select offspring operation. Eventually, the FSDM and DGANM are solved through MaOEA-IS. The experimental results show that the MaOEA-IS has better convergence and diversity, and it has superior performance in solving the FSDM and DGANM. The proposed MaOEA-IS provides more reasonable solutions scheme for many scholars of skin cancer detection and promotes the progress of intelligent medicine.
