Direct Analysis in Real Time Mass Spectrometry (DART-MS) Analysis of Skin Metabolome Changes in the Ultraviolet B-Induced Mice

  • Ultraviolet (UV) radiation is a major environmental factor that leads to acute and chronic reactions in the human skin. UV exposure induces wrinkle formation, DNA damage, and generation of reactive oxygen species (ROS). Most mechanistic studies of skin physiology and pharmacology related with UV-irradiated skin have focused on proteins and their related gene expression or single-targeted small molecules. The present study identified and analyzed the alteration of skin metabolites following UVB irradiation and topical retinyl palmitate (RP, 5%) treatment in hairless mice using direct analysis in real time (DART) time-of-flight mass spectrometry (TOF-MS) with multivariate analysis. Under the negative ion mode, the DART ion source successfully ionized various fatty acids including palmitoleic and linolenic acid. From DART-TOF-MS fingerprints measured in positive mode, the prominent dehydrated ion peak (m/z: 369, M+H-$H_2O$) of cholesterol was characterized in all three groups. In positive mode, the discrimination among three groups was much clearer than that in negative mode by using multivariate analysis of orthogonal partial-least squares-discriminant analysis (OPLS-DA). DART-TOF-MS can ionize various small organic molecules in living tissues and is an efficient alternative analytical tool for acquiring full chemical fingerprints from living tissues without requiring sample preparation. DART-MS measurement of skin tissue with multivariate analysis proved to be a powerful method to discriminate between experimental groups and to find biomarkers for various experiment models in skin dermatological research.

복합전극을 이용한 정혈의 최대피부저항특성에 관한 연구 (A Study of Skin Resistance of Jeong-Acupuncture Points Using the Compound Electrode)

  • Objectives : The purpose of this study is to 1. measure the skin resistance of jeong-acupuncture points using compound electrode, and 2. develop a database of skin resistance based on age and gender. Methods : The skin resistance of jeong-acupuncture points on the hands and feet of 191 healthy volunteers was measured using a specially designed compound electrode. A constant current of $5{\mu\textrm{A}}$ was applied to the measurement points and the skin resistance was obtained using amplifier, A/D converter and Acknowledge software program, Results : 1. ANOVA statistical analysis showed no significant differences between jeong-acupuncture points on hands and feet. 2. Paired samples t-test showed no significant differences between Yin-Yang meridian points, 3. When the data were sorted by gender, factor analysis showed statistically significant differences between male and female groups in all measured points except for left foot. 4. Based on the factor analysis, a significant relationship between the skin resistance and age was found in feet. However, no such relationship was found in hands. Conclusions : There are differences in skin resistance according to sex and age, Further clinical research is necessary using compound electrode method.

Extracellular Superoxide Dismutase (EC-SOD) Transgenic Mice: Possible Animal Model for Various Skin Changes

  • We have generated transgenic mice that expressed mouse extracellular superoxide dismutase (EC-SOD) in their skin. In particular, the expression plasmid DNA containing human keratin K14 promoter was used to direct the keratinocyte-specific transcription of the transgene. To compare intron-dependent and intron-independent gene expression, we constructed two vectors. The vector B, which contains the rabbit -globin intron 2, was not effective for mouse EC-SOD overexpression. The EC-SOD transcript was detected in the skin, as determined by Northern blot analysis. Furthermore, EC-SOD protein was detected in the skin tissue, as demonstrated by Western blot analysis. To evaluate the expression levels of EC-SOD in various tissues, we purified EC-SOD from the skin, lungs, brain, kidneys, livers, and spleen of transgenic mice and measured its activities. EC-SOD activities in the transgenic mice skin were approximately 7 fold higher than in wild-type mice. These results suggest that the mouse overexpressing vector not only induces keratinocyte-specific expression of EC-SOD, but also expresses successfully functional EC-SOD. Thus, these transgenic mice appeared to be useful for the expression of the EC-SOD gene and subsequent analysis of various skin changes, such as erythema, inflamation, photoaging, and skin tumors.

실험동물에 대한 para-phenylenediamino을 함유하는 염모제의 피부 및 눈에 대한 시험 (A Study on the Skin and Eye Testing of para-phenylenediamine and commercial hairdyes containing paraphenylenediamine in animals)

  • Now para-phenylenediamine(PPDA) is generally used as component of most hairdyes because it can be used more conveniently and effectively than the others. But PPDA become known to cause side effects such as skin sensitization, contact dermatitis and eye irritation. So this study was done to restudy the safety of hairdyes containing PPDA. The results of experiment were as follows. 1. As a result of primary skin irritation test and eye irritation test in white rabbits, the solution containing 5% PPDA(in saline) were classified as weak irritant but four kinds of commercial hairdyes containing PPDA were proved not to have any irritation. 2. As a result of skin sensitization test in guinea pig, four kinds of commercial hairdyes were classified as Calss I(week irritant) but three kinds of commercial hairdyes induced skin sensitization for guinea pig to 5% in test groups. These results indicate that the actual toxicity of commercial hairdyes(four kinds) is negligible. But based on the results of experiment, it is thought that there is possibility of some commercial hairdyes to induce skin sensititization.

The Ratios of Pre-emulsified Duck Skin for Optimized Processing of Restructured Ham

  • The purpose of this study was to investigate the quality of duck ham formulated with duck skin through the pre-emulsification process. The experiments to investigate the quality characteristics of duck ham were carried out to measure proximate composition, cooking loss, emulsion stability, pH, color, texture profile analysis, apparent viscosity, and sensory characteristics. Duck ham was prepared with various ratios of duck skin in pre-emulsion as follows: Control (duct skin 30%), T1 (duck skin 20% + pre-emulsified duck skin 10%), T2 (duck skin 15% + pre-emulsified duck skin 15%), T3 (duck skin 10% + pre-emulsified duck skin 20%), and T4 (pre-emulsified duck skin 30%). As the ratio of duck skin to pre-emulsified skin changed, the quality of duck ham in terms of moisture content, fat content, cooking loss, emulsion stability, lightness, textural analysis, apparent viscosity, and overall acceptability changed. The moisture content of T2 was the highest (p<0.05) and that of the control and T4 was the lowest (p<0.05). The fat content of control was higher than all treatments (p<0.05). T2 had the lowest values in cooking loss, total expressible fluid, fat separation, hardness, springiness, and gumminess (p<0.05). The score of overall acceptability of all treatments with pre-emulsified skin was higher than control (p<0.05). Therefore, the pre-emulsification process can improve the quality characteristics of duck ham and 1:1 ratio of duck skin and pre-emulsified skin was the proper ratio to improve the quality characteristics of duck ham.

Development of big data based Skin Care Information System SCIS for skin condition diagnosis and management

  • 피부상태의 진단과 관리는 뷰티산업종사자와 화장품산업종사자에게 그 역할을 수행함에 있어서 매우 기초적이며 중요한 기능이다. 정확한 피부상태 진단과 관리를 위해서는 고객의 피부상태와 요구사항을 잘 파악하는 것이 필요하다. 본 논문에서는 피부상태 진단 및 관리를 위해 소셜미디어의 빅데이터를 사용하여 피부상태 진단 및 관리를 지원하는 빅데이터기반 피부관리정보시스템 SCIS를 개발하였다. 개발된 시스템을 사용하여 텍스트 정보 중심의 피부상태 진단과 관리를 위한 핵심 정보를 분석하고 추출할 수 있다. 본 논문에서 개발된 피부관리정보시스템 SCIS는 빅데이터 수집단계, 텍스트전처리단계, 이미지전처리단계, 텍스트단어분석단계로 구성되어 있다. SCIS는 피부진단 및 관리에 필요한 빅데이터를 수집하고, 텍스트 정보를 대상으로 핵심단어의 단순빈도분석, 상대빈도분석, 동시출현분석, 상관성분석을 통해 핵심단어 및 주제를 추출하였다. 또한 추출된 핵심단어 및 정보를 분석하고 산포도, NetworkX, t-SNE 및 클러스터링 등의 다양한 시각화 처리를 함으로써 피부상태 진단 및 관리에 있어 이를 효율적으로 사용할 수 있도록 하였다.

연조직 변형에 의한 해부학적 지표와 피부마커의 변위 상관성을 이용한 동작분석 오차 보정 방법의 적용 (Application of Compensation Method of Motion Analysis Error Using Displacement Dependency between Anatomical Landmarks and Skin Markers Due to Soft Tissue Artifact)

  • Of many approaches to reduce motion analysis errors, the compensation method of anatomical landmarks estimates the position of anatomical landmarks during motion. The method models the position of anatomical landmarks with joint angle or skin marker displacement using the data of the so-called dynamic calibration in which anatomical landmark positions are calibrated in ad hoc motions. Then the anatomical landmark positions are calibrated in target motions using the model. This study applies the compensation methods with joint angle and skin marker displacement to three lower extremity motions (walking, sit-to-stand/stand-to-sit, and step up/down) in ten healthy males and compares their performance. To compare the performance of the methods, two sets of kinematic variables were calculated using different two marker clusters, and the difference was obtained. Results showed that the compensation method with skin marker displacement had less differences by 30~60% compared to without compensation. And, it had significantly less difference in some kinematic variables (7 of 18) by 25~40% compared to the compensation method with joint angle. This study supports that compensation with skin marker displacement reduced the motion analysis STA errors more reliably than with joint angle in lower extremity motion analysis.

Elevated Aurora Kinase A Protein Expression in Diabetic Skin Tissue

  • Background Aurora kinase A (Aurora-A) plays an important role in the regulation of mitosis and cytokinesis. Dysregulated Aurora-A leads to mitotic faults and results in pathological conditions. No studies on Aurora-A expression in human diabetic skin tissue have been reported. In light of this, we explored the expression of Aurora-A in human diabetic skin tissue. Methods Aurora-A protein was evaluated by western blotting in 6 human diabetic skin tissue and 6 normal skin specimens. Results Increased expression of Aurora-A protein was detected in all diabetic skin tissue samples in both western blot analysis and immunohistochemical staining. However, in the case of the normal skin tissue, no bands of Aurora-A protein were detected in either the western blotting analysis or the immunohistochemical staining. Conclusions Thus far, there have been no studies on the expression of Aurora-A in diabetic skin tissue. However, we believe that oxidative DNA damage related to the expression of Aurora-A protein and Aurora-A could be involved inhuman diabetic skin tissue.

토마토추출물의 흰쥐 건성피부에 미치는 효과 (Effects of Tomato Extracts on Detergent-Induced Dry Skin in Rats)

  • 본 연구는 토마토 추출물의 주방세제 유도성 건성피부에 대한 효능을 실험쥐에 식이방법과 피부도포방법으로 처리하여 확인하였다. 약 3주간의 주방세제 처리에 의한 건성 증후는 홍반의 심각한 상태를 나타내는 4정도의 visual scoring이 모든 군에서 확인되었다. 그러나 약 1주간의 토마토추출물의 식이와 피부도포를 통해 상당히 완화되거나 정상 피부에 가까운 0.7-1.0정도의 visual scoring이 확인되었다. 또한 광학현미경을 통해 건성의 또 다른 증후인 과각화 현상이 토마토추출물의 피부도포와 식이로 현저히 감소하는 것으로 확인되었다. 이러한 토마토추출물에 의한 건선성 증후의 완화는 피부 단백질 함량의 양적 변화를 유도하는 것으로 확인되어 토마토추출물의 건선성 피부의 예방과 개선에 주요한 기전으로 이해된다.

피부색 칼라 분포 특성을 이용한 조명 색도 검출 (Estimation of Illuminant Chromaticity by Analysis of Human Skin Color Distribution)

  • 본 논문에서는 영상을 촬영한 장면의 조명 색도를 추정하는 방법을 제안한다. 기존의 피부색을 이용한 조명 색도 추정 방법은 Storring, Bianco 등이 제안하였다. Storring은 피부색 분포 특성, 완전 복사체 궤적을 이용하였으나 완전 복사체 궤적과 CIE-xy 자료의 연계성이 저하되는 문제가 있다. Bianco 등은 표준조명에서의 피부색 분포와 입력 영상의 피부색 분포를 대조하여 조명 색도를 추정하였다. 이 방법은 다양한 조명에서의 피부색을 최대한 많이 측정하고 확보하기가 어렵다. 제안한 방법은 피부색도 정보와 조명색도 간의 관계를 분석하여 임의의 입력 영상에 대한 조명 색도를 추정할 수 있다. 추정 방법은 분석단계와 테스트 단계로 구분되며, 데이터 셋을 분석 군과 테스트 군으로 분류하여 사용하였다. 분석 군의 모든 입력영상으로부터 각각 피부색 영역을 구하여 피부 색도를 계산한다. 피부 색도의 평균값 집합과 기준 조명 색도 집합의 상관 관계를 분석하여 사상을 구한다. 계산된 사상을 분석 군의 모든 입력 영상에 적용하여 조명 색도를 추정하고 기준 조명색도와의 오차를 계산하고, 오차의 변화가 없을 때까지 상기 과정을 반복하여 안정적인 사상을 구한다. 구해진 사상을 분석 단계와 유사하게 테스트 군 영상에 적용하여 조명 색도를 추정한다. 피부영역과 조명 기준정보가 포함된 데이터 셋이 독립적으로 존재하지 않기 때문에 인텔 TAU 데이터셋의 영상 일부를 이용하여 실험 데이터 셋을 구성하였다. 유사한 이론 기반의 기존 방법인 Finlayson에 비하여 40% 이상, Zhang에 대해서는 11%, Kim에 대해서는 16% 정도의 성능개선을 보였다.
