Inheritance of Resistance to Bacterial Blight in Tongil Type Rice Cultivar 'Changseongbyeo'

  • The inheritance of resistance to bacterial blight in a Tongil-type rice cultivar 'Changseongbyeo' was studied using a Korean isolate HB9101. which belongs to race $K_1$. The resistance of Changseongbyeo to the HB9101 was controlled by a single dominant gene. This gene was allelic with Xa-1 gene and linked to Ig gene on chromosome 4 with the recombination value of 7.5%.

  • Inheritance pattern of resistance to Bombyx mori nuclear polyhedrosis virus (BmNPV) was studied in an Indian silkworm stock TX by single back-cross test method. The resistant parent [TX], susceptible parent [HM], their Fl, F2, and Fl progeny back-crossed to TX [BC(R)] and HM [BC(S)] were inoculated per os with a fixed concentration of BmNPV($0.5{\times}10^{th} PIB/ml$) on the first day of second stadium. The cumulative mortality was recorded until day $10^{\times}$ post-inoculation. The results show that the resistance to BmNPV in TX fellow mono Mendelian inheritance pattern. The resistance dominated over the susceptibility at Fl. At F2, the resistant and susceptible offspring segregated in 3:1 ratio whereas at BC(S), the resistant and susceptible offspring segregated in 1:1 ratio. The response of BC(R) was more or less like the resistant parent TX which confirms the involvement of a major dominant gene conferring resistance to BmNPV in TX. The possible mechanism of inheritance of resistance in TX is discussed.

Inheritance of Golden Coloration in the Zebrafish, Danio rerio (Zebrafish (Danio rerio)의 체색 변이에 관한 유전 분석)

  • The study has been conducted in order to understand the inheritance of body color in the wild type zebrafish (zebra danio), Danio rerio, and its golden mutant (golden danio). The body color was also studied to determine the effect of golden coloration on the survival rate of zebrafish eggs and larvae up to 15 days after fertilization. Reciprocal monohybrid crosses between the wild and the golden type of zebrafish indicated that golden coloration was controlled by a single gene which had two alleles. Transmission of these alleles from parents to their progenies followed the principles of dominance and segregation based on Mendelian inheritance. Similar results from the reciprocal crosses implied that a locus for golden coloration was located on an autosomal chromosome. On the other hand, average survival rates from four different types of mating between, and within, zebra and golden danio suggested that golden coloration seemed to be associated with the survival rate of zebrafish, especially in its early embryonic stage. This indicated that homozygous recessive golden mutation was likely to weaken the golden danio's chance of survival.

Genetic Variation in Exon 3 of Human Apo B mRNA Editing Protein (apobec-1) Gene

  • We have investigated the genetic variation in the human apo B mRNA editing protein (apobec-1) gene. Exon 3 of the apobec-1 gene was amplified by polymerase chain reaction. After detection of an additional band by single strand conformational polymorphism (SSCP) analysis, sequencing of the SSCP-shift sample revealed a single-base mutation. The mutation was a CGG transversion at codon 80 resulting in a lleRMet substitution. This substitution was confirmed by restriction fragment length polymorphism analysis since a Pvull site is abolished by the substitution. Population and family studies confirmed that the inheritance of the genotypes for apobec-1 gene polymorphism is controlled by two codominant alleles (P1 and P2). A significant difference in plasma triglyceride was detected among the different apobec-1 genotypes in the CAD patients (P<0.05). Our study could provide the basis for elucidating the interaction between genetic variation of the apobec-1 gene and disorders related to lipid metabolism.

  • The inheritance and allelism tests of the genes resistant to bacterial blight in some rice breeding lines and varieties were studied. Resistance to isolate JB 8206 was found to be controlled by a single dominant gene in the rice varieties such as, Cheongcheongbyo, Yeongpungbyo, Nampungbyo, Samgangbyo, Hangangchalbyo, and Milyang 42. The resistance in varieties like Pungsanbyo and Baegyangbyo, to isolates JB 8206 and KN 8298 appeared to be governed by a single dominant gene. Evidence from the allelism test indicates that Pungsanbyo, Cheongcheongbyo, Milyang 30, and Baegunchalbyo may have the same dominant gene for resistance to isolate JB 8206, and that Suweon 312, Baegyangbyo as well as Baegunchalbyo may have the other same dominant gene.

  • Benign recurrent intrahepatic cholestasis (BRIC) is a rare autosomal recessive inherited disorder characterized by multiple recurrent episodes of severe cholestatic jaundice without obstruction of extrahepatic bile duct. We present the case of a 7-year-old boy with BRIC confirmed by mutation analysis in the ATP8B1 gene and typical clinical manifestation. Despite inheritance of BRIC, we detected a mutation on only one allele. To our knowledge, this is the first report of BRIC with a confirmed single heterozygote novel mutation in the ATP8B1 gene in Korea.

  • This experiment was conducted to characterize the inheritance of petal color, flower-eye color, flower shape, leaf variegation pattern, and leaf-end shape in Cyclamen persicum. The segregation of $F_2$ and backcross populations from crosses between inbred lines was tested for Mendelian inheritance mode. From four different combinations of crosses, it was found that the petal color was controlled by a single incomplete dominant gene. The other characters, flower-eye color, flower shape, leaf variegation pattern and leaf-end shape were confirmed to be controlled by single complete dominant genes.

  • Monogenic disorders are traditionally attributed to the presence of mutations in a single gene. However, recent advancements in genomics have revealed instances where the phenotypic expression of apparently monogenic disorders cannot be fully explained by mutations in a single gene alone. This review article aims to explore the emerging concept of digenic or oligogenic inheritance in seemingly monogenic disorders. We discuss the underlying mechanisms, clinical implications, and the challenges associated with deciphering the contribution of multiple genes in the development and manifestation of such disorders. We present relevant studies and highlight the importance of adopting a broader genetic approach in understanding the complex genetic architecture of these conditions.

  • The inheritance of stem trichome in red pepper(Capsicum annuum L.) was studied. The parents, $F_1$, $F_2$, $BC_1F_1$ of two combinations, 342 ${\times}$ Kimjanggochu, Namji ${\times}$ Hungarian Wax, were used. Stem pubescent character is controlled by a single gene and pubescent character is dominant to glabrous character.

  • To classify the inheritance of resistance to potato virus Y, crosses between susceptible flue-cured tobacco variety NC 95 and resistant variety McNair 30 were conducted. The parents, $F_1$ plants, $F_2$ populations, and haploid plants derived from anthers of $F_1$ plants were screened for a resistance of two potato virus Y strains (PVY-VB and PVY-VN) isolated in Korea. The Chi-square values for the $F_3$ populations and haploids of $F_1$ fitted 1 :3 and 1 :1 ratios of resistant to susceptible for two strains, respectively. Therefore, it was found that the resistance of McNair 30 for the potato virus Y was controlled by a single recessive gene. Moreover the resistance to two strains screened was inherited dependently.
