Finite element model updating of an arch type steel laboratory bridge model using semi-rigid connection

  • This paper presents finite element analyses, experimental measurements and finite element model updating of an arch type steel laboratory bridge model using semi-rigid connections. The laboratory bridge model is a single span and fixed base structure with a length of 6.1 m and width of 1.1m. The height of the bridge column is 0.85 m and the maximum arch height is 0.95 m. Firstly, a finite element model of the bridge is created in SAP2000 program and analytical dynamic characteristics such as natural frequencies and mode shapes are determined. Then, experimental measurements using ambient vibration tests are performed and dynamic characteristics (natural frequencies, mode shapes and damping ratios) are obtained. Ambient vibration tests are performed under natural excitations such as wind and small impact effects. The Enhanced Frequency Domain Decomposition method in the frequency domain and the Stochastic Subspace Identification method in the time domain are used to extract the dynamic characteristics. Then the finite element model of the bridge is updated using linear elastic rotational springs in the supports and structural element connections to minimize the differences between analytically and experimentally estimated dynamic characteristics. At the end of the study, maximum differences in the natural frequencies are reduced on average from 47% to 2.6%. It is seen that there is a good agreement between analytical and experimental results after finite element model updating. Also, connection percentages of the all structural elements to joints are determined depending on the rotational spring stiffness.

Identification of Major Metabolites of New Platinum(II) Complexes in Rats (새로운 백금 착체(II) 화합물의 흰쥐 혈장에서 대사체 확인)

  • KHPC-002 $[(trans-l-diaminocyclohexane-bis-l,2(diphenylphosphinoethane)platinum)\;{\cdot}2NO_3]$ and $KHPC-006[(cis-diaminocyclohexane-bis-1,2(diphenylphosphinoethane)platinum)\;{\cdot}2NO_3]$ were synthesized as candidates for third platinum antitumor agent. Before their pharmacokinetic study, we optimized the analytical condition with HPLC and identified the major metabolites in the rat plasma. HPLC analysis by $C_{18}$ reverse-phase column showed that standard peak of both compounds appeared rapidly at around 1 minutes, whereas metabolites of KHPC-002 and KHPC-006 which were extracted from plasma after single I.V. administration in rats or incubation for 24 hr at $37^{\circ}C$ showed retention time of $10{\sim}11$ minutes. These metabolites were identified as the major compound by Matrix Associated Laser Deposition/Ionization (MALDI), which only lose the 2 molecules of $NO_3$. Based on these results, we suggest that the major metabolites of KHPC-002 and KHPC-006 were [trans-l-diamino-cyclohexane-bis-l,2(diphenylphosphinoethane)platinum] and [cis-diaminocyclohexane-bis-l.2(diphenylphosphinoethane)platinum], respectively.

Purification and Characterization of 2,4-Dichlorophenol Oxidizing Peroxidase from Streptomyces sp. AD001

  • Streptomyces sp. AD001 is a Gram-positive soil actinomycetes secreting an uncharacterized 2,4-dichlorophenol (DCP) oxidizing enzyme, whose activity is similar to the previously known Actinomycetes lignin-peroxidase (ALiP). This extracellular peroxidase was purified from Streptomyces sp. AD001 as a single protein band on an SDS-PACE by ammonium sulfate fractionation, Q-sepharose, concanavalin A, and Bio-Gel HTP column chromatographies. The molecular mass of the purified peroxidase was determined by SDS-PAGE to be 45.2 kDa, and 49.7 kDa with MALDI-TOF-MS, respectively. The highest level of peroxidase activity was observed at pH 7.5 and $30^{\circ}C$. The amino terminal sequence of the purified peroxidase (G-E-P-E-E-G-N-V-D-G-T-L) showed no significant homologies to my known proteins, suggesting that Streptomyces sp. AD001 may secrete a novel kind of bacterial peroxidase Initial rate kinetic data of the 2,4-DCP oxidation were best modeled with a random-binding bireactant system.

  • Aspergillus niger was selected as a strain producing the potent raw starch hydorlyzing enzyme. These experiments were conducted to investigate the conditions of the glucoa- mylase production, the purification of the enzyme, some characteristics of the purified enzyme and hydrolysis rate on various raw starches such as com, rice, potato, glutinous rice, sweet potato, wheat and barley. The optimum cultural temperature and time for the enzyme production on wheat bran medium were $30^{\circ}C$ and 96hrs, respectively. The respective addition of yeast extract and nutrient broth on wheat bran medium increased slightly the enzyme production. The enzyme was purified by ammonium sulfate fractionation and DEAE-cellulose column chromatography. The specific activity of the purified enzyme was 30.7u/mg-protein and the yield of enzyme activity was 25.8%. The purified enzyme showed a single band on polyacrylamide disc gel electrophoresis and its molecular weight was estimated to be 56,000 by SDS-polyacrylamide disc gel electrophoresis. The isoelectric point for the purified enzyme was pH3.7. The optimum temperature and pH were $65^{\circ}C$ and pH 4.0, respectively. The purified enzyme was stable in the pH range of pH 3.0-9.5 and below $45^{\circ}C$, and its thermal stability was slightly increased by the addition of $Ca^{2+}$. The purified enzyme was activated by $Co^{2+},\;Sr^{2+},\;Mn^{2+},\;Fe^{2+},\;Cu^{2+}$. Raw rice starch, raw corn starch, raw glutinous rice starch, raw sweet potato starch, raw wheat starch and raw barley starch showed more than 90% hydrolysis rate in 48hrs incubation. Even raw potato starch, most difficult to be hydrolyzed, showed 80% hydrolysis rate. The purified enzyme was identified as glucoamylase.

  • Quantitative analysis was achieved by gas-liquid chromatographic method (GLC) with a single column system of OV-17 for 16 of free amino acids in Korean green tea and the contents of mineral in it was determined by atomic absorption flame emission. The results are summarized as follows: 1) Korean green tea contained Mn 1% or over out of the total ash content and $0.05{\sim}0.20%$ in the water extraction, as the major mineral. 2) Ba, Cr, Ni, Pb and V were analyzed also by small quantities relatively and Co, Tin and Ywere not detected in the water extraction. 3) GLC indicated the presence of 16 components in free ammo acids. 4) The quantities of free amino acids in Korean green tea were determined $2.96{\sim}6.61%$ Alanine, $1.01{\sim}l.89mg%$ Glycine, $2.07{\sim}7.81mg%$ Valine, $1.27{\sim}8.76mg%$ Leucine and Isoleucine, $94.31{\sim}316.27mg%$ Threonine, $9.10{\sim}39.91mg%$ Serine, $2.18{\sim}36.76mg%$ Hydroxyproline, $2.72{\sim}5.90mg%$ Proline, $39.64{\sim}70.02mg%$ Aspartic Acid, $25.93{\sim}101.28mg%$ Glutamic Acid and Lysine, $8.32{\sim}18.30mg%$ Phenylalanine and Tyrosine in trace amount. 5) The total free amino acid contents in Korean green tea ranged from 207.24mg% to 516.06mg% and Moo-Deoung tea contained outstandingly high, 516 mg% or over.

  • The feasibility of foam separation to remove protein produced from fish culture water was investigated, By assuming foam separation column as a single well-mixed pool, a simplified model for foam separator conditions was alse studied under the batch operation. The model indicated that the protein removal could be described as a first-order reaction whose rate increases with both superficial air velocity and protein concentration in the bulk solution. from ,the results of an experimental study on the effects of superficial air velocity, the protein concentration, temperature, and pH on protein removal, the superficial air velocity and initial protein concentration in bulk solution were found to be important operational factors. Other experimental results important that foam separator operated under batch conditions would be considered as a completely mixed reactor. The protein removal rate by foam separation process was increased proportionally with the superficial air velocity, and the adsorptive removal rate of protein was found to obey Langmuir adsorption formula. The preposed simplified model was verified with the experimental data obtained from this study. Under the experimental range used, both temperature and pH did not affect the protein removal.

  • Effects of cavitation and hydraulic flip in circular and elliptical nozzles on the flow characteristics have been studied. Spray tests were conducted using injectors with different ratios of an orifice length(L) to a diameter(d) and of a major axis diameter(a) to a minor axis diameter(b). With the increment of an injection pressure drop, discharge coefficients slightly decreased in cavitation flows, and those suddenly dropped and were almost constant in hydraulic flip flows. For elliptical nozzles with L/b > 8 and L/a < 8, discharge coefficients and flow patterns showed different results from those in previous circular nozzles. When a flow in the elliptical nozzle was under steady condition, as the liquid column went downstream from the nozzle, its spray angle a little decreased in the plane of a major axis and increased in the plane of a minor axis.

Effect of Functional Monomer on Retention Factor of Chiral Racemate (기능성 단량체가 키랄 물질의 체류인자에 미치는 영향)

  • In this work, molecular imprinted polymers (MIPs) using the template of the N-CBZ (carbobenzyloxy)-L-phenylalanine, MAA and 4-VPY as a monomer, EGDMA as a crosslinker and AIBN as an initiator were considered. The prepared polymer particles $(Ca.\;25-35\;{\mu}m)$ were packed into a chromatographic column $(3.9\;\times\;150\;mm)$. The chromatographic characteristics of the retention on the MIP were experimented with acetonitrile as a mobile phase at the flow rate of mobile phase, 0.5 ml/min. The retention factors and resolutions of chiral racemate of the N-CBZ-D, L-phenylalanine were measured. The results showed that the retention factor and resolution by the two co-monomer imprinting polymer were higher than the single monomer imprinting polymers, which indicated an increase in the affinity of the MIP with the sample as a result of the cooperation effect of the binding sites.

Symbolic computation and differential quadrature method - A boon to engineering analysis

  • Nowadays computers can perform symbolic computations in addition to mere number crunching operations for which they were originally designed. Symbolic computation opens up exciting possibilities in Structural Mechanics and engineering. Classical areas have been increasingly neglected due to the advent of computers as well as general purpose finite element software. But now, classical analysis has reemerged as an attractive computer option due to the capabilities of symbolic computation. The repetitive cycles of simultaneous - equation sets required by the finite element technique can be eliminated by solving a single set in symbolic form, thus generating a truly closed-form solution. This consequently saves in data preparation, storage and execution time. The power of Symbolic computation is demonstrated by six examples by applying symbolic computation 1) to solve coupled shear wall 2) to generate beam element matrices 3) to find the natural frequency of a shear frame using transfer matrix method 4) to find the stresses of a plate subjected to in-plane loading using Levy's approach 5) to draw the influence surface for deflection of an isotropic plate simply supported on all sides 6) to get dynamic equilibrium equations from Lagrange equation. This paper also presents yet another computationally efficient and accurate numerical method which is based on the concept of derivative of a function expressed as a weighted linear sum of the function values at all the mesh points. Again this method is applied to solve the problems of 1) coupled shear wall 2) lateral buckling of thin-walled beams due to moment gradient 3) buckling of a column and 4) static and buckling analysis of circular plates of uniform or non-uniform thickness. The numerical results obtained are compared with those available in existing literature in order to verify their accuracy.

A Measurement Study of Body Types for Clothing Construction (의복구성(衣服構成)을 위한 계량적(計量的) 체형연구(體型硏究))

  • To study the body trunk form which is essential to clothing constructions, necessary body sizes were measured indirect1y from 216 unmarried women from 19 to 24 years old by a photographic network method. The results are obtained as follow; 1) In this study of body type measurements, the four factors of bust, abdomen, column and hip size have been divided into four-stage intervals and given the body type codes 1, 2, 3 and 4, with 1 designating the size difference of the factor with the minimum size difference of the factor with the maximum size difference from waist size. These body type codes can be easily, linguistically interpreted. In this study, among $4^4=256$ possible body types, 54 types appeared frequently. These 54 types are integrated into 6 groups of types by clustering analysis. Representative types of each group are types 3333 and 2333 in group 1, 2233, 1233 and 1332 in group 2, 2222, 1222 and 1223 in group 3, 2223 in group 4, 2323 and 2322 in group 5, and 3323, 3223 and 3322 in group 6. In the types of groups 1, 2, and 3, the change is mainly in bust size, and in the types of group 4. is single type. In the types of group 5 the change is mainly in hip size, and in the types by groups 6 the change is mainly in abdomen size and hip size. 2) The analysis by measuring the four elements of body trunk in reference to waist size is scientific and grading of similar styles is possible in clothing construction. 3) Assuming that the human body is oval, using the calculating method, the size of girth, which cannot be measured by the indirect measuring method, is obtained approximate to actual size, with an error of ${\pm}2.8cm$.
