Electrochemical Impedance Characteristics of a Low-Temperature Single Cell for CO2/H2O Co-Reduction to Produce Syngas (CO+H2)

  • In this study, the electrochemical impedance characteristics of CO2/H2O co-reduction to produce CO/H2 syngas were investigated in a low-temperature single cell. The effect of the operating conditions on the single-cell performance was evaluated at different feed concentrations and cell voltages, and the corresponding electrochemical impedance spectroscopy (EIS) data were collected and analyzed. The Nyquist plots exhibited two semicircles with separated characteristic frequencies of approximately 1 kHz and tens of Hz. The high-frequency semicircles, which depend only on the catholyte concentration, could be correlated to the charge transfer processes in competitive CO2 reduction and hydrogen evolution reactions at the cathodes. The EIS characteristics of the CO2/H2O co-reduction single cell could be explained by the equivalent circuit suggested in this study. In this circuit, the cathodic mass transfer and anodic charge transfer processes are collectively represented by a parallel combination of resistance and a constant phase element to show low-frequency semicircles. Through nonlinear fitting using the equivalent circuit, the parameters for each electrochemical element, such as polarization resistances for high- and low-frequency processes, could be quantified as functions of feed concentration and cell voltage.

Single-cell PCR on protargol-impregnated euplotid ciliates: a combined approach of morphological and molecular taxonomy

  • Ciliates are considered one of the most diverse protozoa and play significant roles in ecology. For successful taxonomic study of these microscopic eukaryotes, a staining procedure is necessary, due mainly to intrinsic difficulties in recognizing characteristics from living cells. Although molecular taxonomy has been used to resolve the ambiguities associated with traditional morphology-based taxonomy, extraction of genomic DNA from stained ciliate cells is not available yet. In the present study, we describe a method to extract genomic DNA from a single protargol-impregnated euplotid cell. By using $HgCl_2$ as a fixative and modulating the exposure time of bleach solution in the protargol impregnation, high-quality genomic DNA can successfully be extracted from a stained single cell with minimal loss of morphological integrity. This technique will contribute to the effectiveness of combined approaches of molecular and morphological taxonomy from single ciliate cells.

Development of PCR based approach to detect potential mosaicism in porcine embryos

  • Direct injection of genome editing tools such as CRISPR/Cas9 system into developing embryos has been widely used to generate genetically engineered pigs. The approach allows us to produce pigs carrying targeted modifications at high efficiency without having to apply somatic cell nuclear transfer. However, the targeted modifications during embryogenesis often result in mosaicism, which causes issues in phenotyping founder animals and establishing a group of pigs carrying intended modifications. This study was aimed to establish a genomic PCR and sequencing system of a single blastomere in the four-cell embryos to detect potential mosaicism. We performed genomic PCR in four individual blastomeres from four-cell embryos. We successfully amplified target genomic region from single blastomeres of 4-cell stage embryo by PCR. Sanger sequencing of the PCR amplicons obtained from the blastomeres suggested that PCR-based genotyping of single blastomere was a feasible method to determine mutation type generated by genome editing technology such as CRISPR/Cas9 in early stage embryos. In conclusion, we successfully genotyped single blastomeres in a single 4-cell stage embryo to detect potential mosaicism in porcine embryos. Our approach offers a simple platform that can be used to screen the prevalence of mosaicism from designed CRISPR/Cas9 systems.

Basal cell carcinoma and squamous cell carcinoma in a single tumor in the anterior auricular area

  • The concurrence of basal cell carcinoma (BCC) and squamous cell carcinoma (SCC) in a single tumor is rarely encountered. We report a case of BCC and SCC in a single tumor in the anterior auricular area. A 70-year-old woman had been diagnosed with BCC by a punch biopsy performed at a dermatology clinic. We performed wide excision of the tumor with an ulcer in the anterior auricular area. Analysis of the biopsy specimen revealed the presence of both BCC and SCC in the tumor. This case illustrates that it is necessary to establish a precise diagnosis and formulate appropriate surgical and treatment plans considering the possibility that two carcinomas may coexist, although the possibility is low in patients with skin cancer.

  • Cell migration is one of the essential mechanisms responsible for complex biological processes. Intensive researches have begun to elucidate the mechanisms and search intriguing conditions for efficient control of cell migration. One general mechanism that is widely applicable for cells including Escherichia coli, amoebae and endothelial cell is chemotaxis. The single cell study for bacterial chemotaxis has an advantage over studies with the population of cells in providing a clearer observation of cell migration, which leads to more accurate assessments of chemotaxis. In this paper, we propose a three-dimensional model considering a single bacterium to study its chemotaxis. The semi-implicit Fourier spectral method is applied for high efficiency and numerical stability. The simulation results reveal rich dynamics of cell migration and provide quantitative assessments of bacterial chemotaxis with various chemoattractant gradient fields.

  • We present a method of surface functionalization of a single layer graphene for linking and detecting MDA-MB-231 human breast cancer cell. The methodology is done by utilizing 1-pyrenebutanoic acid and succinimidyl ester for immobiling CD44 antibodies. This work shows that the single layer graphene is an efficient fixing substance to capture the MDA-MB-231 human breast cancer cell, selectively. The immobilization method of the cancer cell on the graphene layer will be an effective cell counting system. Moreover usage of the linking with non-covalent bonding is expected to develope a sensor scheme of electrical cell-detecting diagnosis system.

Integration of Single-Cell RNA-Seq Datasets: A Review of Computational Methods

  • With the increased number of single-cell RNA sequencing (scRNA-seq) datasets in public repositories, integrative analysis of multiple scRNA-seq datasets has become commonplace. Batch effects among different datasets are inevitable because of differences in cell isolation and handling protocols, library preparation technology, and sequencing platforms. To remove these batch effects for effective integration of multiple scRNA-seq datasets, a number of methodologies have been developed based on diverse concepts and approaches. These methods have proven useful for examining whether cellular features, such as cell subpopulations and marker genes, identified from a certain dataset, are consistently present, or whether their condition-dependent variations, such as increases in cell subpopulations in particular disease-related conditions, are consistently observed in different datasets generated under similar or distinct conditions. In this review, we summarize the concepts and approaches of the integration methods and their pros and cons as has been reported in previous literature.

싱글입자-마이크로캡슐형 전자종이의 셀갭 차이에 따른 구동 특성평가 (Evaluation of Driving Properties by Cell-gap Difference of Single Particle-Microcapsule Type Electronic Paper)

  • We fabricate a single particle-microcapsule type electronic paper using electrophoresis, which is different with a reported dual particle-microcapsule type and of which electro-optical researches are not reported. So we analyzed a basic properties, such as reflectivity, response time, and driving voltage. Our display panels having various cell-gaps of $30{\mu}m$, $34{\mu}m$, $38{\mu}m$, $42{\mu}m$, and $46{\mu}m$ are inspected. As a results, a driving voltage is defined to 10 V and desirable cell-gap is $30{\mu}m$ or $34{\mu}m$. Considering a mechanical strength, the optimum cell-gap is $34{\mu}m$ for the single particle type electronic paper.
