Studies on the High Temperature Induced Stress on the Biochemical Profile and Fecundity of Daba and Laria Ecoraces of Tropical Tasar Silkworm Antheraea mylitta Drury (Lepidoptera: Saturniidae)

  • Tropical tasar silkworm Antheraea mylitta Drury experiences extreme temperature stress conditions during its life cycle particularly during diapauses and first crop. The present study witnessed the impact of high temperature on some biochemical profiles and egg production (fecundity) of semi-domesticated Daba and Shorea robusta (Sal) based wild ecorace Laria during seed cocoon (pupa) preservation. Cocoons of Daba and Laria were treated with high temperature at $40^{\circ}C$ for 10 days in a BOD incubator. The protein profile and carbohydrate content in the hemolymph and fat body and total haemocyte count (THC) in the hemolymph of pupa were investigated. Further, the fecundity and fertility of egg was assessed. Significant increase in the protein concentration was observed in the hemolymph with reduction in the fat body (p<0.05). The difference in protein concentration was highly significant between the semidomesticated Daba and wild ecorace Laria (p<0.05). High pupal mortality (20%) and reduced fecundity (10-15%) in Daba was noticed compared to wild Laria. Also an increased THC (>28000) was recorded in Laria. The study infers the potentials of wild ecoraces in sustaining the extreme temperature conditions and need of adopting suitable package of practices for the preservation of diapause seed cocoons during extreme summer conditions. There is possibility to introgression thermal stress resistant traits in the semi-domesticated races of tasar silkworm by resorting to conventional breeding plans with wild races and keeping the thermal stress induced response as markers.

  • By using PAGE, a study was made on the nonspecific esterase spectra of female genital organs and eggs in Bombyx mori L. The expression of 11 esterase bands was detected during ontogenesis of races and inter-races hybrids kept in Bulgaria. The gene activity of 9 esterase loci was assumed. Esterases specific for the spectrum of diapausing eggs were observed. In two esterase zones, intra- and inter-breed polymorphism was found. Based on the same breed specific expression, the existence of correspondence between esterase bands from spectra of different silkworm tissues and organs was suggested. Stage-specific expression of esterases in female genital glands, indicative of differentiated gene activity during ontogenesis, was established.

  • Of all factors that govern productivity in sericulture, the role of silkworm egg is of prime importance. The production of silkworm seed involves a long chain of interdependent and highly specialized conditions. Female moths of Bombyx mori L. commence egg deposition immediately when the moths were decoupled. More than 60% of eggs are deposited within 4 hrs after decoupling i.e. 62.33% in CSR-3, 61.67% in CSR-6, 62.50% in CSR-16 and 63.28% in CSR-17. Almost 90% of eggs were laid in the first 12 hrs (93.13% in CSR-3, 90.81% in CSR-6, 89.71% in CSR-16 and 93.17% eggs in CSR-17) after decoupling. Almost 8 to 10% of more eggs were laid when oviposition period was prolonged upto 48 hrs instead of 24 hrs. The present investigation showed that a significant increase of 45 (8.34%), 50 (9.98%), 41 (8.26%) and 45 (8.30%) (P < 0.01) numbers of more eggs were found between 24 to 48 hrs (T6 to T9) in CSR-3, CSR-6, CSR-16 and CSR-17 races respectively. The findings of present investigation is very important for sericulture industry while producing layings to keep for different hibernation schedule especially for autumn/late autumn crop, female may be allowed for prolonged oviposition to get 8 -10% more eggs and farmer may fetch 8 -10 kg. more cocoon yield/l00 dfls with more income.

  • The major dominant fluoride-endurance (Dfe) gene was introduced into the commercial varieties by crossing and pedigree selection to breed silkworm races that could normally develop in the area that polluted by fluoride. After backcrossed for two generations, the Dfe gene was made homozygous, and individuals with good economic characters were selected to generate next generation. After 8 generations of selection, their characters became stable, and the silkworm variety which is resistant to fluoride, Huayuan${\times}$Dongsheng, for spring rearing were bred.

  • At present, more than 300 races of the Silkworm are conserved and used as valuable genetic resources. But because of the uneffectiveness of manual data management, faster and systematic data base construction is needed. So, development of silkworm genetic resources management program has been begun and the result can be practically used. When developing the program, Visual basic was used for data input system construction, and MS Access for database. IIS(Internet Information System) and ASP(Active Server Page) was also used for searching data and information with Internet Web Server and Web Browser which is comfortable for constructing database and providing information. Data input item consists of 46 practical characteristics such as race name, moltinism, larval period and pupation percentage etc.. And these characteristics are classified with qualitative and quantitative character. Photographs of silkworm, cocoon and other related items were scanned and the image data was recorded on the database.

Evaluation of Relative Fluoride Toxicity and Its Impact on Growth, Economic Characters and Fecundity of the Silkworm, Bombyx mori L.

  • A detailed evaluation of relative toxicity of fluoride to die 5th instar larvae of PM and NB$_4$D$_2$ races of silkworm and the effects of lethal, sublethal and prevailing levels of fluoride in groundwater on the growth, economic characters and fecundity of the silkworm, Bombyx mori (Lepidoptera: Bombycidae) were studied. The feeding of mulberry, Morus alba leaves treated with lethal and sublethal doses of fluoride to PM and NB$_4$D$_2$ races of silkworms from the beginning of the 5th instar to the end of the feeding period resulted in significant reduction in growth, single cocoon weight, single shell weight, silk index, average filament length, and fecundity when compared to controls. These manges were more pronounced on exposure to lethal dose than sublethal dose and in general, the changes induced by fluoride were more striking in NB$_4$D$_2$ than PM, indicating the greater resistance of PM to higher fluoride levels. Groundwater quality in sericulturally important villages of Karnataka and Andhra Pradesh States has been studied with special reference to the presence of fluoride. On exposure to dose of prevailing levels in the waters of sericulturally important areas, die changes observed on growth, economic characters and fecundity were slightly lower when compared to controls and the decrease was found to be insignificant (P$\geq$0.05). It was concluded that, though minute dose (4.0 ppm) of fluoride did not have any toxic impact, it is toxic at higher concentrations to silkworms.

  • The general combining ability (GCV), the specific combining ability (SCA) and the reciprocal combining ability (RCA) effects wereobtained by 8$\times$8 diallel crosses of the silkworms with four Japanese races and four Chinese races, total eight lines. The result are as follows ; 1. The general combining ability (GVA) effects appeared high significant level in the total and the fifth instar periods (TP, FP), a female and a male cocoon weights (FW, MW), a female and a male cocoon layer weights (FL, ML) and a female and a male cocoon layer ratios (FR, MR). Only the reciprocal combining ability (RCA) effects appeared high significant level in the total and the fifth instar periods (TP, FP). 2. The Japanese original silkworm lines varied in the general combining ability effects from -0.864 to 0.578, and the Chinese original silkworm lines did in ones from -0.570 to 1.018. 3. The specific combining ability effects of the silkworm lines made in order of the crossing types of JL (Japanese lines) $\times$CL (Chinese lines)>CL$\times$JL>JL$\times$JL> CL$\times$CL in a total cocoon weights and a cocoon layer weights. 4. The reciprocal combining ability effects of the silkworm lines was in order of the crossing types of JL$\times$CL>JL$\times$JL> CL$\times$CL>CL$\times$JL in a total cocoon weights and cocoon layer weights.

Genomic Organization of Heat Shock Protein Genes of Silkworm Bombyx mori

  • The Hsp 20.8 and Hsp 90 cDNA sequence retrieved from NCBI database and consists of 764 bp and 2582 bp lengths respectively. The corresponding cDNA homologus sequences were BLAST searched in Bombyx mori genomic DNA database and two genomic contigs viz., BAAB01120347 and AADK01011786 showed maximum homology. In B. mori Hsp 20.8 and Hsp 90 is encoded by single gene without intron. Specific primers were used to amplify the Hsp 20.8 gene and Hsp 90 variable region from genomic DNA by using the PCR. Obtained products were 216 bp in Hsp 20.8 and 437 bp in Hsp 90. There was no variation found in the six silkworm races PCR products size of contrasting response to thermal tolerance. The comparison of the sequenced nucleotide variations through multiple sequence alignment analysis of Hsp 90 variable region products of three races not showed any differences respect to their thermotolerance and formed the clusters among the voltinism. The comparison of aminoacid sequences of B. mori Hsps with dipteran and other insect taxa revealed high percentage of identity growing with phylogenetic relatedness of species. The conserved domains of B. mori Hsps predicted, in which the Hsp 20.8 possesses ${\alpha}-crystallin$ domain and Hsp 90 holds HATPase and Hsp 90 domains.

Path Coefficient Analysis on Major Silk Quality Characteristics of the Silkworm, Bombyx mori L.

  • By means of correlation analysis and path coefficient analysis, 18 characteristics of 325 conserved silkworm races were studied for the selecting efficiency on main silk quality characteristics. The result showed that selecting lines with heavier cocoon weight are beneficial to raise the weight of have. Choosing strains with higher cocoon shell percentage, longer filament length and better reelability percentage is good for increasing non-broken filament length. Selecting lines with medium daily weight of have produced in the fifth instar, medium cocoon shell percentage and simultaneously selecting heavier cocoon shell weight are favorable to promote reelability percentage. Choosing strains with heavier cocoon weight, higher cocoon shell percentage, heavier daily weight of bave produced in the fifth instar and heavier weight of bave but shorter filament length may hopefully breed out a race with thick filament size. And selecting strains with heavier weight of have and fine filament size are beneficial to increase filament length. Selecting lines with higher cocoon shell percentage and heavier weight of bave are favorable to raise raw silk percentage of cocoon. There are no significant direct or indirect correlation coefficient between viability and major characteristics of silk quality. It is, therefore, feasible to breed a silkworm race with excellent vitality and good silk quality as well.
