Breeding of New Silkworm Variety Golden silk, a Yellow Cocoon Color for Spring Rearing Season (황색실크 생산용 특수누에 품종 '골든실크' 육성)

  • A new silkworm variety Golden silk for spring rearing season is $F_1$ hybrid between Jam311, a Japanese race bred from introduction breeding and Jam312, a Chinese race from introduction breeding. In the local adaptability test performed at 8 local areas in spring of 2006, "Golden silk" was showed 7 percent higher in hatchability than authorized silk-worm races laval. "Golden silk" was receded higher 1.7 percent, 2.2 percent in pupation rate, best cocoon rate respectively than Chunsujam. The larval period shortened for 1.12 day even though low record to the several items including productivity than Chunsujam. "Golden silk" was showed 1,031 m, 2.97 denier in length of cocoon filament, weight of cocoon filament respectively. The concentration of Deoxynojirimycin(DNJ) was measured 4.34 mg in "Golden silk", high then to "Chunsujam"(3.81 mg). In the Paecilomyces tenuipes production ability test, the pupal weight of "Golden silk" was 1.20g.

Scanning Electron Microscopic Studies on Leaf Surface Trichomes in Mulberry and Its Influence on Rearing Performance of Silkworm Bombyx mori L.

  • The type of trichomes, their density and pattern of distribution on leaves of 16 genotypes of mulberry, belonging to both diploid and polyploid categories, were studied by scanning electron microscope. The present investigation was undertaken to find out the relationship of physical attributes, especially the density and trichome types with higher acceptability and better rearing performance by the silkworm Bombyx-mori L. Two types of trichomes glandular and non-glandular types were observed on both the leaf surfaces of all the mulberry genotypes studied. In general, greater densities of trichomes were observed on the abaxial surface than the adaxial surface of leaves in most of the genotypes. Distribution of glandular trichomes were more in abaxial surface and non-glandular trichomes were more in adaxial surface. Overall, distribution of glandular and non-glandular trichomes per unit area of leaf did not follow any regular pattern. When leaves of those genotypes were fed to silkworms, trichome density was found to be significantly negatively correlated with the survival of larvae i.e., effective rate of rearing, but trichome density did not influence the economic characters of rearing. As the distribution of glandular trichomes (GT) and non-glandular trichomes (NGT) did not follow any definite pattern, no relation could be established between the GT and NGT densities with silkworm rearing performance. However, the ratio of GT and NGT in a particular genotype influenced the rearing parameters, higher the ratios better the rearing performance. High GT and NGT ratio (>1.00) was found positively significant when correlated with economic parameters viz., larval weight, single cocoon weight and single shell weight. The study is useful in screening different mulberry genotypes for their better acceptability to silk-worm and higher rearing performance at the early stage of selection without actually conducting the rearing.

Evaluation of Relative Fluoride Toxicity and Its Impact on Growth, Economic Characters and Fecundity of the Silkworm, Bombyx mori L.

  • A detailed evaluation of relative toxicity of fluoride to die 5th instar larvae of PM and NB$_4$D$_2$ races of silkworm and the effects of lethal, sublethal and prevailing levels of fluoride in groundwater on the growth, economic characters and fecundity of the silkworm, Bombyx mori (Lepidoptera: Bombycidae) were studied. The feeding of mulberry, Morus alba leaves treated with lethal and sublethal doses of fluoride to PM and NB$_4$D$_2$ races of silkworms from the beginning of the 5th instar to the end of the feeding period resulted in significant reduction in growth, single cocoon weight, single shell weight, silk index, average filament length, and fecundity when compared to controls. These manges were more pronounced on exposure to lethal dose than sublethal dose and in general, the changes induced by fluoride were more striking in NB$_4$D$_2$ than PM, indicating the greater resistance of PM to higher fluoride levels. Groundwater quality in sericulturally important villages of Karnataka and Andhra Pradesh States has been studied with special reference to the presence of fluoride. On exposure to dose of prevailing levels in the waters of sericulturally important areas, die changes observed on growth, economic characters and fecundity were slightly lower when compared to controls and the decrease was found to be insignificant (P$\geq$0.05). It was concluded that, though minute dose (4.0 ppm) of fluoride did not have any toxic impact, it is toxic at higher concentrations to silkworms.

  • Flacherie of all other silkworm diseases greatly affects cocoon crop as it is far-reaching and wide spreading. Fleacherie which kills silkworms caused by bacteria can be classified as bacterial digestive organ disease, and "Sotto" disease. Bacterial digestive organ disease is caused by the bacteria living in the silkworms alementary canal and a majority of flacherie belongs to this disease. Septicemia is caused by bacterias breeding in silkworms body fluid but its attach is comparatively limited during the larva period. "Sotto" disease is caused by eating mulberry leaves infected with bacteria which produce toxin and silkworms are intoxicated and killed by the toxin. The cause of flacherie is mainly due to a poor environment. The unclean and unsanitary silkworm rearing beds help bacterias breeding and bacteria enter silkworms body through mouth organ or skin. The present study is to investigate various causes of flacherie by means of pulverization of silkworm. Filtrated fluid is extracted by centrifuge and hypodermic of peroral inoculation-is given to young and medium silkworms of spring and autumn. The gained results of the experiment are summarized as follows: 1. Silkworms infected with flacherie were pulverized and their filtrated fluid was extracted by centrifuge and inspected under microscope to find polyhedron from the fluid. 2. The experimenting group of peroral inoculation. a) From the third day of peroral inoculation silkworms appetite generally decreased and ate less compared with the control group. b) After 7 days of the inoculation silkworms suffered from empty head, loose bowels and fainting. c) Some of the silkworms still ate but as were shown in Fig. 3 and 4 some dwarfish silkworms were found. d) There was no remarkable difference between 1st and 2nd instar inoculation groups. e) There was a tendency that the number of diseased silkworms was decerased as the increase of instars. 2. The experimental group of hypodermic inoculation a) Both of 3rd and 4th instar inoculation groups showed no remarkable singularity and the number of diseased silkworms decreased. b) The rate of diseased silkworms was comparatively low because the body fluid was acidy or toxin was hard soluble. Hypodermic inoculation could not give much harm to the silkworms compared to peroral inoculation.

  • 1. The varietal difference in amylase activity of the digestive fluid of the 5th instar larvae reared on the artificial diet was investigated, using the parent commercial silkworm varieties of Japanese strain. The amylase activity was large different among silkworm varieties. The activity was strong in Hansaeng-1, Jam 115 and Jam 117, medium in Hansaeng-3, Jam 113, Jam 119, and Jam 201, weak in Jam 107, Jam 121 and Gyeongchu. The amylase of the digestive fluid of ten parent commercial silkworm varieties is possible-(ae) type, compared with +(+$\^$ae/) type of Daizo. 2. To investigate the effect of a-amylase pre-treatment of the artificial diet, larvae were fed with the diet treated by a-amylase during 4th-5th instar periods. The blood sugar content and cocoon qualities were slightly higher in the experimental larvae than those in the control, while showing the slight less body weight, amylase activity and dietary efficient.

Studies on the Flacherie Virus in the Silkworm, Bombyx mori L. I. Resistance to Flacherie and Ina-flacnerie Virus in the Leading Silkworm Varieties in Korea (가잠의 바이러스성 연화병에 관한 연구 I. 장려품종에 대한 저항성 검정)

  • The infectious flacherie viruses are serious diseases in the silkworm, which affects the cocoon crops. However, there is only a few study on this diseases up-to-date in Korea and, in this experiment, the authors investigated the resistance of the silkworm varieties to flacherie and Ina-flacherie viruses by peroral infection. A cross, Hansaeng #1$\times$ Harisaeng #2, showed the highest resistance to the flacherie virus a Mudeung$\times$Geumho showed the lowest resistance among the examined varieties. It seemed that the varietal difference of resistance against Ina-flacherie virus was appeared and Jam117$\times$ Jam118, Gyeongchu$\times$Yeonil, Mudeung$\times$Geumho, Hansaeng#1$\times$Hansaeng#2 and Hansaeng#3 $\times$ Hansaeng #4 showed non susceptibility by peroral infection. However it was shown that Jam115 $\times$Jam116 had the lowest resistance to Ina-flacherie virus.

Studies on the Behavioral response in Silkworm larvae, Bombyx mori. Genetic Analysis and Correlated Response to Economic Characters of Walking Distance (가잠유충의 행동반응성에 관한 연구)

  • The objective of the present study was to investigate the behavioral responses of sillkworm (Bombyx mori) larvae to: the dominant effect of walking distace: and the relationships between walking distance and the various economic characters on yellow sheets. The means of walking distances of progeny from F2 and F1 F1 and F2 generations, which was a hybrid between the long and the short walking distances, were shorter than those of parents. This may indicate that the genes for the short walking distances were partially dominant over those for the longwalking. Estimated broad sense heritability of larval walking distance was different for each larval instar, which fell into the range of 52∼84%. Genetic correlation was apparent between walking distances of all larval stages and some economic characters. The number of cocoons per liter and the period from incubation to emergence have shown the positive genetic correlation with walking distances. The cocoon yield, the sing cocoon weight, and the cocoon shell weight have shown to be in the negative correlation with walking distances. No correlation was observed between walking distances of the mature larvae and any economic characters. The analysis by the path coefficient showed negative direct effect of walking distances on the cocoon yield, and positive indirect effect on the single cocoon weight and the cocoon shell weight.

  • This experiment was to know properties of Sericin Jam that development, growth of silkgland, content of sericin and composition of amino acid in hemolymph. The characters of Sericin Jam can he seen form this experiment. Hatching ratio was 85% in Sericin Jam and 95% in Jam 120. Especially hatching period of Sericin Jam was longer than Jam 120 and also hatching of Sericin Jam was ununiform. The larval duration of Sericin Jam was 20 days and 23 hours, and Jaw 120 was 21 days and 22 hours. In Sericin Jam, middle silkgland contain a great p arts in silkgalod and posterior silkgand is short and no curves. The period of mounting to emergence was 12 days in Sericin Jam. The period of pupa was 7day. It is property of Sericin Jam that the period is short. Cocoon was very thin and light in Sericin Jam. Weight of cocoon shell of Sericin Jam is 2.7cg. The sericin protein quantity was 100% in cocoon shell of Sericin Jam, about 28% in cocoon shell of Jam 120, however the sericin ratio per Sericin Jam cocoon was 34.6% compare to cocoon shell of Jaw 120 in sericin protein quantity. The amino acids in hemolymph of Sericin Jaw was much hidtidine, lysine, glut amic acid. And the amount of almost amino acids were increased depends upon development at t he 5th instar.

  • It has been reported that the effect of royal jelly on silkworms can be markedly resulted from feeding on royal jelly throughout all the instars. In 1965, I began the experiment in order to know whether it has a practical for rearing, only through feeding silkworms at 1st to 2nd instar, when a comparative little amount of mulberry leaves and labor is needed for rearing. In the method of my experiment, mulberry leaves with different concentration of royal jelly added, 2.5%, 5%, and 10%. respectively, are feeded on silkworms of Sulack ${\times}$ Soyang at 1st and 2nd instar in spring in 1965. However, in the result of the experiment there is not any effect on the survival. the growth speed, the body weight, of the larvae, and the weight of cocoon layer, the cocoon layer ratio, and number of silkworm eggs laid.

  • Normally the first generation of hybrid is supplied for farm rearing, precise and labor saving sex discrimination is needed. The new strain "Hansaengjam" is sex-limited (female:larval markings, male:no-markings) strain which was bred from both sex-limited strain Japanese originated Jam 153 and Chinese originated strain Jam 154. Productivity test of Hansaengjam during 2007 and 2009 showed high healthiness and cocoon yield. The Hansaengjam is evaluated as an excellent strain from healthiness, cocoon yield and other test results.
