This experiment was attempted to investigate the effect of the specific gravity of silkworm eggs on the hatching and the practical hatching ratio depend upon six egg production companies for two silkworm races and their reciprocal crosses. 1. The hatching and the practical hatching ratio in the specific gravity of medium and heavy eggs were higher than in the specific gravity of light eggs. 2. Compare with Japanese or Chinese mother races each other, it was inclined that the former seems to be higher than the latter on the hatching ratio in the specific gravity of light and medium eggs, but the practical hatching ratio was high only in the specific gravity of light eggs. 3. Chinese mother races were different in the practical and the hatching ratio between the specific gravity of eggs. On the contrary in case of Japanese mother races were no difference for the hatching ratio but difference in the practical hatching ratio between the specific gravity of eggs. 4. On the egg production company, in case of the specific gravity of medium and light eggs, the hatching and the practical hatching ratio were high significance, but no difference in the specific gravity of heavy eggs.
The following results were obtained through the experiments and study of fertility for silkworm eggs in vagina, oviduct, and ovarial base. The sample eggs were collected after leaving silkworm moth for one week from the date of treatment was given to oviporm orifice of the silkworm moth by heated needle in order to discontinue laying eggs. Those sample silkworm moths were divided into two groups. The first group was under the condition of immediately after the couplation and the second group was under the condition of after the laying eggs in some extend, and both of them were completed couplation. The sample eggs were screened in order to confirm the degree of color which normally be observed as very characteristic during hibernating. 1. About 5% of silkworm out of the total sample were identified as having 1-2 eggs. (See table 1). But, the charasteristic colour of hibernating egg was not observed. The colour was not much different as that of eggs in ovarian duct. 2. Immediately after the couplation, the eggs were not found in vagina under the condition of immediately after the couplation. 3. About 10% of the sample under the condition of after laying eggs in some extend were found having eggs in vagina or oviduct. 4. About 2% of the sample silkworm moth under the condition of immediately after the couplation were found having I fertilized egg in oviduct. 5. No fertilized egg was found in ovarian tube. It seemed that sperm did not enter ovarian tube. 6. Almost the entire eggs were found in ovarian tube under the condition of immediately after the couplation. But, eggs under the condition of after laying eggs in some extend were found around oviduct and vagina due to the continuous movement of silkworm eggs towards oviporus orifice. 7. Sperm is mainly found around seminal receptacle and vagina, and sperm goes into micropyle together with silkworm eggs which were moving towards oviduct.
The purpose of this treatise is to prove the presence of cystine in silk fiber through wide sampling throughout all the sericultural processes of Bombyx mori.; also to show that disulfide cross linkages exist in the silk fiber. The conclusions reached were as follows: 1. Crystalline cystine was obtained from silk fibroin using Folin's Method. 2. Analytical data showing the cystine content of silk fiber and its related materials were obtained using Sullvan's Method as follows: Material Percent Cystine A. Mulberry leaf protein 0.175 B. Silkworm egg 0.33 C. Silkworm Body, matured, fat extracted, without silk gland 0.41 D. Silk gland, matured 1.23 E. Silkworm feces none F. Silkworm pupa, fat extracted 0.30 G. Silkworm moth, fat extracted 0.60 H. Raw Silk 0.22 I. Fibroin 0.175 J. Sericin 0.30 3. The presence of cystine in the silkworm was substantiated the existence of 0.175 % methionine in mulberry leaves and 0.12% methionine in the silk gland. 4. Part of the sulfhydryl compounds in the silk gland is believed to transfer to serine and methionine, with the former being secreted into the liquid silk finally as silk fiber and the latter used for nutritive purposes in the growing of silk gland tissue. 5. The cystine content is variable by mulberry species, silkworm species, sex, breeding process, and other culturing environments. 6. Hybrid silkworms require more nutritive amino acids for effective growth than the original parents, and secrete less of them as silk fiber. 7. From such an observation, the amino acid composition of silk fiber is believed to be fairly flexible. Cystine if included in the amorphous part of the fiber, especially in sericin. 8. The result from enriching the silkworm diet with pure cystine or wool cystine did not result in any advantage, therefore it is believed that the natural cystine and methionine contents in the mulberry leafaregoodenoughforsilkwormnutrition. 9. The disulfide cross linkage in silk fiber was verified by using the Harris Method. Contraction took place following the treatment of the fiber with various salts and acids. Comparisons were made with wool fiber. 10. During these experiments, the fibrious structure of silk fiber and the net-globular liquid form were photographed microscopically. It is believed that the globules of liquid silk are net-formed by the inter attraction of the OH ion of the globular peptide and the H ion of water as shown by the hair cracking behavior of the film. The net-globular protein precipitation from the mulberry protein solution showed that mulberry is a proper diet for the formation of fibrous protein in the silk fiber. 11. The significance of the presence of cystine in silk fiber as emphasized in this paper should result in modification of the general conception that cystine is absent from this fiber. The hybrid ($CSR2{\times}CSR4$) eggs treated with acid were taken up for the study with an objective to develop long-term preservation schedule. The hybrid eggs obtained with two mating duration (3 h and 6 h) and oviposition period (6 h and 24 h) with two age groups of eggs (24 h and 36 h) were treated with Hydrochloric acid. These eggs were subjected to preservation at $5^{\circ}C$ in single step refrigeration and at $5^{\circ}C$ and $2.5^{\circ}C$ under double step refrigeration from $10{\sim}120$ days. These eggs were released from the cold storage as per the specified durations and incubated at standard conditions and allowed 2 h for hatching at 450 lux light. Hatchability was found to be significantly higher or on par with the control in three treatments (T1, T2 and T4) where the eggs are preserved continuously at $5^{\circ}C$ up to 30 days. However under double step refrigeration, hatching was not significantly affected in 20+60 day's combination of T1 treatment up to 80 days. Bioassay studies of the promising treatment i.e.. T1 with (20+60) days indicated that early stage loss and cocoon yield was found to be on par with the control. Hence this treatment was recommended for preservation of acid treated new bivoltine hybrid layings. Details of the hatchability and rearing performance of long term preservation of acid treated eggs are discussed.
In the hemolymph proteins of the silkworm reared with mulberry leaves and artificial diet, electrophoretic patterns of Storege Protein 1 (SP1) and Vitellogenin(VG), changes of relative concentration of SP1, JHA effects during the developmental stages, and estimation of the molecular weight of vitellin subunits were investigated in the present study. 1. Storage Protein 1 (SP1) showed female specificity from the middle stage of the fifth instar to the end of spinning stage in the silkworm reared with mulberry leaves and artificial diet. 2. Vitellogenin (VG) showed female specififcity just afrter pupation in the silkworm reared with mulberry leaves and artificial diet. 3. Storage Protein 2 (SP2) without female specificity was detected from the early stage of the fifth instar to the first or second day after pupation. 4. Vitellin was composed of two non-identical subunits (vitellin-heavy chain ; VTL-H, vitellinlight chain ; VTL-L) with mplecular weight of 186,000 and 41,000. Also, there were no differenes between the molecular weights of vitellin subunits obtained from silkworms reared with mulberry leaves and artificial diet. 5. Relative concentration of Storage Protein 1 (SP1) after topical application - 0.5, and 10$\mu\textrm{g}$ per body weight - JHA on the 60th hour of the fifth instar showed the highest increase with the treatment of 5$\mu\textrm{g}$ and a higher increase with the treatment of 10$\mu\textrm{g}$ compared with the control (no topical application of JHA).
Bioactive and chemical properties of silkworm powder (SP) degradation by fruit extract containing the proteolytic enzymes of kiwifruit, papaya, pineapple and pear were investigated. Silkworm powder was incubated with extracts from each fruit at $60^{\circ}C$ for 24 hr. Protein content was slightly higher in the SP treated with fruit extract than that in the control SP. Major minerals were K, Ca, Mg, and Zn. Major fatty acids were linolenic acid, oleic acid, and palmitic acid. When total protein patterns were analyzed by SDS-polyacrylamide gel electrophoresis (PAGE), silkworm protein was strongly degraded by the treatment of fruit extract from pineapple, papaya, and pear, but little silkworm degradation was observed in kiwifruit extract treatment. Fibriolytic activity was only detected in the SP by the fruit extract treatments from papaya and pear. DPPH radical scavenging activity was slightly stronger in the SP treated with fruit extract than that in silkworm powder. However, all these samples exhibiteda relatively low activity compared with the butylated hydroxytoluene (BHT). These results may provide the basic data for understanding the biological activities and chemical characteristics of SP treated with fruit extract for development of functional foods.
The aim of this work was to investigate the rate of hybrid vigor among the $F_1$ hybrids. The results obtained are as follows; 1. The rate of hybrid vigor in outbreedings(E$\times$J, & J$\times$C) was higher than that in inbreedings in all the metric characters, especially in moulting laval weight. 2. In outbreedings, the $F_1$ between Europe and Chinese strain showed considerably higher rate of hybrid vigor than that of Japanese and Chinese. 3. The hybrid vigor rate of moulting larvel weight gave rise to be significant differences among the strains due to the maternal effects from newly hatched larvae to pupae.

Mulberry (Morus spp. family: Moraceae) has prime importance in the sericulture industry, and its foliage is the only natural feed of the silkworm Bombyx mori L. Traditional classification methods using morphological traits were largely unsuccessful in assessing the diversity and relationships among different mulberry species because of environmental influences on the traits of interest. For these reasons, it is difficult to differentiate between the varieties and cultivars of Morus spp. In the present study, inter-simple sequence repeat (ISSR) markers were used to investigate the genetic diversity of 48 mulberry samples genotyped using nine ISSR primers. The ISSR markers exhibited polymorphisms (53.2%) among mulberry genotypes. Furthermore, similarity coefficient estimated for these ISSR markers was found to vary between 0.67 and 0.99 for the combined pooled data. The phenogram drawn using the UPGMA cluster method based on combined pooled data of the ISSR markers divided the 48 mulberry genotypes into seven major groups. No genetic association was found in the collection area, and there was a mixed pattern between the mulberry lines. The hybridization between different mulberry species is highly likely to be homogenized due to natural hybridization.
Many of studies on the transovarial transmission of occult virus and their activation due to various stresses such as cold or heat treatment, chemical feeding, and nutritional deficiency, etc., in the silkworm, Bombyx mori L. have been made, but any attempts have been not made to control virus diseases by detection of the occult virus-carried moths in the production of silkworm egg of hybrids, because of difficulty to detect occult virus in any stage. Therefore, it may be worth while to disclose whether a sublethal infection of the moths from which active virus are detectable, has the same level of induction rate as that of occult virus activation, thus to apply its results for the reduction of the occurence of virus diseases in silkworm rearing. For these purposes, the following experiment was conducted as one of preliminary steps. In this study, investigations on the generation-to-generation transmission of occult virus and a sublethal infection, and the role of chromosomal gene of the host, Jam 103 and Jam 104 in the Previous generation, and Jam 103 x Jam 103 and Jam 104 f Jam 104 in the next generation were made for the induction of virus diseases due to the transmitted virus. The frequency of cytoplasmic polyhedrosis due to the induction in the F$_1$ generation was markedly higher in the cross-batches, male$\times$female and male$\times$female in which inoculated individuals were used as fem ale parents than in the cross-batches, male$\times$female and male$\times$female in which virus has been not inoculated or inoculated only to male in the previous generation. The tendency of increasing rate was observed in any treatments; such as the inoculations of cytoplasmic polyhedrosis virus (10$\^$5/, 10$\^$6/ 10$\^$7, and 10$\^$8//ml ill different concentration of inocula) , cold-treatment (5$^{\circ}C$, 12hrs or 24hrs), and formalin-feeding treatment (2% or 3%). The shape of polyhedra (tetragonal in outline) examined in the F, larvae was identified as that of the inoculated polyhedra with partial application of immunofluorescent techniques. These results suggests that the cytoplasmic polyhedrosis virus in B. meri L. are transmitted to the next generation through the egg, apparently in the occult state. And the experimental results of various cross-batches revealed the egg cytoplasm plays an important part i the transmission of the occult virus of the cytoplasmic polyhedrosis virus,
The cost of artificial diets for silkworm is very expensive, so that it is essential to lower it in order to extend the period of silkworm rearing on artificial diets. In the present paper, the utilization of several kinds of grass powders was tried to use as an ingredient of artificial diet to reduce its production cost. The value of grass powder as an ingredient of artificial diet was assessed on the base of ingestability and growth of silkworms. As a result it was found it was found that they varied with the kinds and harvesting season of grasses. But it is appeared to be possible to use a grass powder as an ingredient for silkworm diet, when it contains 10~20% of grasses. Moreover, to cut the cost of diet, it is recommanded that the diet containing grass powder could be a applied to the polyphagous silkworm variety.
