Effects of Daegeum-eumja, Igwi-tang and Sihosogan-san on Gastric Mucosal Lesions Induced by Alcohol, Indomethacin and Burn-stress in Mice. (Alcohol, Indomethacin 및 Burn-stress로 유발된 생쥐의 위점막 손상에 대한 대금음자(對金飮子), 익위탕(益胃湯), 시호소간산(柴胡疎肝散)의 효과)

  • Objectives : This study was conducted to investigate the effects of Daegeum-eumja, Igwi-tang, and Sihosogan-san on gastric mucosal lesions induced by alcohol, indomethacin, and burn-stress in mice. Methods : Experimental mice were divided into six groups. The normal group (NOR) did not receive any treatment to elicit gastropathy. In the control group (GE), gastropathy was elicited by alcohol, indomethacin, and stress. In the misoprostol group (MS), misoprostol was administered after gastropathy was elicited by alcohol, indomethacin, and stress. In the Daegeum-eumja group (DG), Daegeum-eumja was administered after gastropathy was elicited by alcohol, indomethacin, and stress. In the Igwi-tang group (IW), Igwi-tang was administered after gastropathy was elicited by alcohol, indomethacin, and stress. In the Sihosogan-san group (SH), Sihosogan-san was administered after gastropathy waselicited by alcohol, indomethacin, and stress. The effects on gastric mucosal lesions were evaluated by the morphological change of gastric mucosa, the anti-oxidant effect, HSP 70, $NF-{\kappa}B$ p65, $I{\kappa}B$, COX-1, PNA, BrdU, and iNOS. Results : Hemorrhage erosion, HSP70, and $NF-{\kappa}B$ in the DG, IW and SH groups decreased more than that of the control. The $I{\kappa}B$, COX-1, PNA, BrdU, and iNOS in the DG, IW, and SH groups increased more than that of the control. DG showed the most effect against gastric mucosal lesions induced by alcohol; IW against gastric mucosal lesions induced by indomethacin; and SH against gastric mucosal lesions induced by burn-stress. Conclusions: Daegeum-eumja, Igwi-tang, and Sihosogan-san extracts have excellent effects on gastric mucosal lesions induced by alcohol, indomethacin, and burn-stress, respectively.

Anti-inflammatory effect of Sihosogan-tang via inhibition of NF-κB and MAPK cascade (NF-κB와 MAPK억제를 통한 시호소간탕(柴胡疏肝湯)의 항염증효과)

  • Objectives : Sihosogan-tang (SST) is one of the traditional herbal formula and also one of the Korean medical insurance medicines. It commonly used in the treatment of hepatitis, chronic gastritis, intercostal neuralgia, pleurisy, and depression in East Asia. In the present study, we have demonstrated the anti-inflammatory effects of SST in macrophage cell line. Methods : To investigate mechanism of the anti-inflammatory effect of SST, we examined the productions of nitric oxide (NO) and pro-inflammatory cytokines, and the expressions of inducible NO synthase (iNOS), nuclear factor-κ B (NF-κB) and mitogen-activated protein kinase (MAPK) on RAW 264.7 cells activated by LPS. Results : SST significantly inhibited the expression of iNOS increased by LPS, and also significantly inhibited the production of NO. In addition, SST significantly inhibited pro-inflammatory cytokines such as TNF- α and interleukines. SST inhibited the expression of NF-κB and MAPK activation. Conclusions : These results suggest that SST ameliorates inflammatory response in LPS-activated RAW 264.7 cells through the inhibition of the NF-κB and MAPK pathway. Therefore, this study supplies objective evidence for the anti-inflammatory effect of SST.

Comparative Study of the Neuroprotective Effect of Sihogyeji-tang, Sihosogan-tang, and Sihocheonggan-tang on an MPTP-Induced Parkinson's Disease Mouse Model (MPTP로 유도된 파킨슨병 생쥐 모델에 대한 시호계지탕, 시호소간탕, 시호청간탕의 신경세포 보호 효과 비교 연구)

  • Objectives : Parkinson's disease (PD) is the second most common neurodegenerative disorder worldwide and is characterized by the loss of the dopaminergic neurons in the substantia nigra (SN). In a previous in vitro study, we demonstrated that Sihogyeji-tang (SG), Sihosogan-tang (SS), and Sihocheonggan-tang (SC) have the potential to be candidate medicines for PD. This study aimed to compare the neuroprotective effect of SG, SS, and SC using 1-methyl-4-phenyl-1,2,3,6-tetrahydrophridine (MPTP)-induced PD mouse model. Methods : Eight-week-old male C57BL/6 mice were intraperitoneally administered with 30 mg/kg of MPTP for 5 days and orally administered SG, SS and SC for 12 days from the first MPTP injection. Motor function was assessed using the pole test and the rotarod test. Dopaminergic neuronal survival in the SN and striatum was evaluated through tyrosine-hydroxylase immunohistochemistry. Results : MPTP administration resulted in behavioral impairment and dopaminergic neuronal death in the SN and striatum. In the pole test, treatment with SG, SS, and SC alleviated the MPTP-induced motor dysfunction on day 5 and 12. In the rotarod test, SS and SG alleviated the MPTP-induced motor dysfunction on day 5, while only SS showed improvement on day 12. SS and SG significantly protected dopaminergic neurons in the SN from MPTP toxicity, and all three compounds (SG, SS, and SC) showed significant protection in the striatum. Notably, SS demonstrated superior efficacy in suppressing MPTP-induced motor dysfunction and dopaminergic neuronal death compared to SG and SC. Conclusions : These findings suggest that SS is the most effective formula among SG, SS, and SC for PD, indicating its potential role in the treatment of PD.

Analysis of Herbal formulation on Wolgug-hwan Series (동의보감(東醫寶鑑) 월국환가미방(越鞠丸加味方)의 방제학적(方劑學的) 분석(分析))

  • Objectives : This study was performed to analysis the composition of herbal formulations about a series of Wolgug-Hwan(WGH, formed by Gukchur-won and Sagung-san) in Dongeuibogam written by Heo Jun. Methods : The series of WGH in Dongeuibogam were selected by 'Prescription Lineage Graph' (http://164.125.206.43/PrescriptionLineageGraph.aspx) established by prof. Lee at Pusan National University. In the analysis of a series of WGH, we used some formulation books including classics. Results : Gaeuldogi-tang(GDT) is formed by WGH, Yeonggangchulgam-tang(YGT), Yugil-san (YIS) and Gungchul-san. Cheongul-san(CUS) is formed by WGH, Ijin-tang(IJT), YGT and Haegeum -hwan(HGH). Sosikcheongul-san(SCS) is formed by CUS and IJT. Jeungmiijin-tang(JIT) is formed by WGH, IJT, Sambaek-tang and Gyullyeonjichul-hwan. Sihoeokgan-tang(SET) is formed by WGH, Sihosogan-san(SSS) and Jigolpi-eum. Wolgukbohwa-hwan(WBH) is formed by WGH, Bohwa-hwan, Mokyangjichul-hwan and Hwangnyeon-tang(HNT). Chimsa-hwan(CSH) is formed by WGH, Injinho-tang(IHT), Yungmisamneung-hwan, Jeungmisamul-tang and Sinseongugi-tang. Conclusions : SCS, CUS, or JIT has been used frequently in case of phlegm-fire. These are formulated by the combination of WGH and IJT with Samseon-san or HGH or Jichul-hwan according to the special symptoms, respectively. SET which is formulated by WGH and SSS has been used in case of stagnation of liver-qi. GDT, WBH or CSH has been used in damp-heat. These are formulated by the combination of WGH with YIS, HNT, or IHT, respectively.

Bibliographical Review on Oriental Medicine Herbal Treatment for Uterine Carcinoma (자궁암 치료 한약물에 대한 문헌적 고찰)

  • To make a bibliographical review on the most frequently used prescriptions and herbs in treating the uterine carcinoma. 43 texts in China and 6 texts in Korea published since 1980 were reviewed for this objective. 590 Oriental Herbal Medicine formulas composed of 457 herbs were investigated. The frequency and efficacy of each herb used were statistically analyzed. The most frequently used herbal medicine formulas were Danchisoyosan in hepatic Qi stasis, Sihosogan-tang mixed with Lungeumdaebo-tang in damage of conception and thoroughfare vessel, Danchisoyosangagam in blood heat, Hoangyunhaedok-tang in pattern of retained dampness-heat, Jibapjihang-tang in iver-kidney yin deficiency pattern, Naebohyunhapsamyoungbaekchulsan in spleen-kidney yang deficiency pattern. The herbal medicines most frequently used were; Angelicae gigantis radix(126 times), Phellodendri cortex(91 times), Borneolum(87 times), Glycyrrhizae radix and Paeoniae radix alba(82 times), Realgar and Alumen(80times), Poria(77 times), Astragali radix(75 times), Moschus and Myrrha(74 times), Coicis semen and Olibanum(73 times), Codonopsis pilosulae radix and Scutellaria bardata Don(70 times) etc. The effect of the most frequently used herbs were : heat-clearing medicine(12), blood-circulation and stasis-dispelling medicine(9), tonifying and replenishing medicine(5), ointments(5), heat-clearing and blood-cooling medicine(3) and etc. The most frequently used herbs may be applied in future animal studies and clinical trials. Also this data can be used as a reference in formulating new Oriental Herbal Medicine formulas for uterine carcinoma.
