Bone Homeostasis and Gut Microbial-Dependent Signaling Pathways

  • Although research on the osteal signaling pathway has progressed, understanding of gut microbial-dependent signaling pathways for metabolic and immune bone homeostasis remains elusive. In recent years, the study of gut microbiota has shed light on our understanding of bone homeostasis. Here, we review microbiota-mediated gut-bone crosstalk via bone morphogenetic protein/SMADs, Wnt and OPG/receptor activator of nuclear factor-kappa B ligand signaling pathways in direct (translocation) and indirect (metabolite) manners. The mechanisms underlying gut microbiota involvement in these signaling pathways are relevant in immune responses, secretion of hormones, fate of osteoblasts and osteoclasts and absorption of calcium. Collectively, we propose a signaling network for maintaining a dynamic homeostasis between the skeletal system and the gut ecosystem. Additionally, the role of gut microbial improvement by dietary intervention in osteal signaling pathways has also been elucidated. This review provides unique resources from the gut microbial perspective for the discovery of new strategies for further improving treatment of bone diseases by increasing the abundance of targeted gut microbiota.

  • This paper proposes a kind of function-concentrated signaling service equipment structure, in which various kind of signaling service are efficiently made for call processing control. Based upon the typical switching system models, a philosophical study on the capacity, features of structure in world systems, and interfaces with peripheral equipments has been performed in order to provide the rationale on the merits or efficiency of the proposed digital signaling service equipment.

  • Railway is an important transportation tool for nation's industrial development and public daily life. These days the concern about railway has remarkably been rising due to the April 2004 opening of KTX, Korea High Speed Rail, together with Trans-Continental Railways linking North Korea, China, Russia, through European countries. In the meantime, the issues of train safety and efficiency, arising from the different on-board signaling systems installed on different kinds of trains, have been raised incessantly. In this paper CTCS(Continental Train Control System) is suggested, which can accommodate such various signaling functions as ATP, ATC, ATS, etc. into one single type of on-board signaling system and operate without changing ground signaling systems on the junctions of the Trans-Continental Railways. The development of this CTCS would cause speed-up of exiting lines and possession of railway core solutions for establishing Northeast Asia logistics center bases, which could be an important cornerstone for domestic railway improvements and Korea's role of Northeast Hub.

  • Common Channel Signaling(CCS) networks need special controls to avoid overload of signaling networks and degradation of call process-ing rate, since they play an important role of controlling communication transfer networks. Congestion control and flow control mechanisms are well described in ITU-T recommendation on Signaling System No.7(SS7). For the practical provisions, however, we need an analysis on the relation among service objects, system requirements and implementation of congestion control algorithms. SS7 provides several options for controlling link congestion in CCS networks. In this paper we give a general queueing model of congestion control algorithm which covers both the international and national options. From the queuing model, we obtain the performance parameters such as throughput, message loss rate and mean delay for the international option. To show the performance of the algorithm, some numerical results are also given.

  • In this paper, the inductive power collector using electromagnetic induction for vehicle such as the electric railway vehicle system is suggested and some ideas for power collector design to improve the power transfer performance are presented. The inductive power of secondary part is related to amount of linked flux to secondary part by the length of air-gap, which is expected by such a system parameter as mutual inductance. This paper will study for the transfer characteristic of power from input to output and equation including mutual inductance. And also, effect of leakage inductance variation of inductive power collector according to structure of winding was considered.

  • In this paper we introduce the method, procedure and result of RAM prediction for interlocking system which is applied for AREX signaling system. For RAM prediction, we breakdown the interlocking system to the LRU level and select the LRUs of which failure can cause the service delay. The prediction of reliability is based on the Reliability Block Diagram which is the functional diagram composed of selected LRUs and finally, the availability of interlocking system is estimated from the combination of reliability and maintainability.

Predicting the Role of Osteal Macrophages and Osteocytes in Bone Tissue Network Using a Mathematical Modeling (수학적 모델링을 이용한 골조직 세포 네트워크에서 Osteal Macrophage와 골세포의 역할 예측)

  • The aim of this study was to investigate the role of osteal macrophages (osteomac) and osteocytes in bone remodeling using a mathematical model. We constructed the bone system with pre-osteoblasts, osteoclasts, osteocytes, and osteomac. Each link of the parameters and ordinary differential equations followed the Graham's model in 2013 except for the parameters of osteomac signaling and osteocytes signaling to link preosteoblasts and osteoblasts. We simulated the changes in each cell and bone volume according to the changes in the parameters of osteomac signaling and osteocytes signaling. The results showed bone volume was unstable and decreased gradually when the effectiveness of osteocytes and osteomac dropped below a certain level. When the parameters of osteomac signaling and osteocytes signaling to link preosteoblasts and osteoblasts had a value less than 1, bone volume increased with the increase in the parameter of osteomac signaling to link preosteoblasts and osteoblasts. Moreover, although the parameter of osteocytes signaling to link preosteoblasts and osteoblasts, increased in case of a small parameter of osteomac signaling, bone volulme decreased. If the parameters of osteomac signaling to link preosteoblasts and osteoblasts were over a certain level, bone volume was positively maintained, despite the parameter of osteocyte signaling to link preosteoblasts and osteoblasts. We suggested the osteomac may affect bone remodeling and may play an important role in bone cell network.

Inhibitors of AKT Signaling Pathway and their Application

  • The AKT signaling pathway is a highly regulated cell signaling system that forms a network with other cell signaling pathways. Hence, the AKT signaling pathway mediates several important cellular functions that include cell survival, proliferation, cell migration, and et cetera. Irregularities that led overactive AKT signaling have been linked to many diseases such as cancer and metabolic-associated diseases. Hence, modulating the overactive AKT signaling pathway via inhibitor is a tantalizing prospect for treatment of cancer and metabolic-associated diseases. Two inhibitors of the AKT signaling pathway will be presented in this symposium: 1) Bisleuconothine A (BisA), a bisindole alkaloid that inhibit autophagy and 2) Ceramicine B (CerB), a limonoid that inhibit adipogenesis. The first topic is on a bisindole alkaloid, BisA and its mechanism in inducing autophagosome formation in lung cancer cell line, A549.(1) Since most autophagy inducing agents generally induce apoptosis, we found that BisA does not induce apoptosis even in high dose. BisA up-regulation of LC3 lipidation is achieved through mTOR inactivation. The phosphorylation of PRAS40, a mTOR repressor was suppressed by BisA. This observation suggested that BisA inactivates mTOR via suppression of PRAS40 phosphorylation. Interestingly, the phosphorylation of AKT, an upstream regulator of PRAS40 phosphorylation was also down-regulated by BisA. These findings suggested that Bis-A induces autophagosomes formation by interfering with the AKT-mTOR signaling pathway. The second topic is on CerB and its mechanism in inhibiting adipogenesis in preadipocytes cell line, MC3T3-G2/PA6.(2,3) CerB inhibits the phosphorylation of protein kinase B (AKT) at the Thr308 position but not the Ser473. Consequently, the phosphorylation of FOXO3 which is located downstream of AKT is also inhibited. Considering that FOXO3 is an important regulator of PPARγ which is a key factor in adipogenesis, CerB may inhibit adipogenesis via the AKT-FOXO3 signaling pathway. Taken together, both BisA and CerB highlighted the potential of AKT signaling pathway modulation as an approach to induce autophagy and inhibit the formation of fat cells, respectively.

The Vomeronasal Organ and Adjacent Glands Express Components of Signaling Cascades Found in Sensory Neurons in the Main Olfactory System

  • The vomeronasal organ (VNO) is a sensory organ that influences social and/or reproductive behavior and, in many cases, the survival of an organism. The VNO is believed to mediate responses to pheromones; however, many mechanisms of signal transduction in the VNO remain elusive. Here, we examined the expression of proteins involved in signal transduction that are found in the main olfactory system in the VNO. The localization of many signaling molecules in the VNO is quite different from those in the main olfactory system, suggesting differences in signal transduction mechanisms between these two chemosensory organs. Various signaling molecules are expressed in distinct areas of VNO sensory epithelium. Interestingly, we found the expressions of groups of these signaling molecules in glandular tissues adjacent to VNO, supporting the physiological significance of these glandular tissues. Our finding of high expression of signaling proteins in glandular tissues suggests that neurohumoral factors influence glandular tissues to modulate signaling cascades that in turn alter the responses of the VNO to hormonal status.
