A Study on the Protective Effects of Siegesbeckiae Herba on Neurotoxicity Induced by N-methyl-D-aspartic acid(NMDA) (희렴(??)이 NMDA로 유발된 신경세포 손상에 미치는 효과)

  • Objectives : Siegesbeckiae Herba's effect on the protection of nerve cells was tested, and the effects were compared between Siegesbeckia glabrescens Makino, the state of which is spica imported from China, and original Korean leaves of it. Methods : After damaging nerve cells by exposing them on NMDA (N-methyl-D-aspartic acid) and KA(kainic acid), Siegesbeckiae Herba's effect on cell death, inhibition rate, glutamate separation, and ROS(reactive oxygen species) production were examined. Results : 1. Siegesbeckiae Herba inhibited the cell death exposed to NMDA. 2. Siegesbeckiae Herba inhibited the amount of glutamate separated from nerve cells exposed to NMDA. 3. Siegesbeckiae Herba inhibited the production of ROS induced by NMDA. 4. Siegesbeckiae Herba did not inhibit the cell death exposed to KA. 5. Chinese Siegesbeckiae Spica had no inhibition effect on cell death. Conclusions : Siegesbeckiae Herba was effective in inhibiting the death of nerve cells exposed to NMDA, and in protecting nerve cells from various damages in nerve cell diseases. Because Chinese Siegesbeckiae Spica did not show such effects, it is necessary to closely examine those effects according to the used parts.

The Effects of Siegesbeckiae Herba on EDRF in the Carotid Artery of the Rabbit (희렴이 가토(家兎)의 혈관내피세포성(血管內皮細胞性) 이완인자(弛緩因子)에 미치는 영향(影響))

  • This study was undertaken to define the mechanism of Siegesbeckiae Herba-induced relaxation in rabbit common carotid artery contracted by agonists. In order to investigate the effect of Siegesbeckiae Herba on contracted rabbit carotid arterial strips, transverse strips with intact or damaged endothelium were used for the experiment using organ bath. To analyze the mechanism of Siegesbeckiae Herba-induced relaxation, Siegesbeckiae Herba extract infused into contracted arterial strips induced by agonists after treatment of lanthanum chloride, indomethacin, atropine, $N\omega-nitro-{_L}-arginine$, cobalt chloride or methylene blue. The relaxation effect of Siegesbeckiae Herba was dependent on the presence of endothelium, showing that Siegesbeckiae Herba-induced relaxation was not observed in the strips without endothelium. The endothelium-dependent relaxation induced by Siegesbeckiae Herba was suppressed by the pretreatment of lanthanum chloride, $N\omega-nitro-{_L}-arginine$, cobalt chloride or methylene blue, but it was not observed in the strips pretreated with indomethacin or atropine. These results demonstrated that Siegesbeckiae Herba may inhibit agonist-induced contraction through an increase in the cyclic GMP by the production of nitric oxide in the vascular endothelial cells.

Quantitative Analysis of Kirenol in Siegesbeckia glabrescens and S. pubescens by HPLC-UV (HPLC-UV에 의한 진득찰과 털진득찰의 Kirenol 정량분석)

  • Many diterpenoids from Siegesbeckia species (Compositae) and their anti-inflammatory actions have been examined. In this research, high-performance liquid chromatography-ultraviolet spectrophotometer (HPLC-UV) method was used to compare the quantitative level of kirenol (ent-pimarane-type diterpenoid) in the aerial parts of Korean S. glabrescens and S. pubescens and the Chinese Siegesbeckiae Herba. Fingerprints of the two HPLC chromatograms of Korean S. glabrescens and S. pubescens were similar, but considerably different from Chinese Siegesbeckiae Herba. The content of kirenol in S. pubescens ($16.51{\pm}0.10$ mg/ml dry weight as mean${\pm}$RSD) was higher than S. glabrescens ($13.48{\pm}0.12$ mg/g dry weight). These values were considerably higher than the Chinese Siegesbeckiae Herba ($1.55{\pm}0.74$ mg/g dry weight). Thin layer chromatography (TLC) analysis demonstrated the containing of kirenol in the three plant materials, but the presence of siegeskaurolic acid (entkaurane-type diterpenoid) only in the Chinese Siegesbeckiae Herba.

An Experimental Study On The Effect of Herba Segesbeckiae on the Intravascular Coagulation Induced by Endotoxin Rats (희첨이 Endotoxin으로 유발(誘發)된 흰쥐의 혈전(血栓)에 미치는 영향(影響))

  • In order to investigate the effect of the Herba Segesbeckiae on the thrombosis induced by endotoxin in rats. The rats were treated with solid extract of Herba Siegesbeckiae 41.9mg /200g(Sample) which were administered orally. And the thrombosis was induced by injecting endotoxin in the caudal vein rats. And then the number of platelets, concentration of fibrinogen, prothrombin time and FDP(fibrinogen degradation product) were measured. The results were as follows; 1. Platelet was increased significantly in sample group compared with control group. 2. Prothrombin time was shortened significantly in sample group compared with control group. 3. Fibrinogen was increased significantly in sample group compared with control group. 4. FDP was decreased significantly in sample group compared with control group. According to the above results, Herba Siegesbeckiae had significant effects on number of Platelets, Prothrombin time, Fibrinogen and FDP. therefore, it is considered that Herba Siegesbeckiae can be applied to diseases related to thrombosis.

Effects of Siegesbeckiae Herba Extract on the Blood Pressure of Dogs and Chickens(II) (회첨 엑기스의 혈압에 미치는 영향(II))

  • This study was performed in dogs and chickens to observe differences of responses by species of animals to hypotensive action of Siegesbeckiae Herba, using its ethanol extract. Siegesbeckiae Herba e ethanol extract (SGEE), when injected into the vein of dogs or chickens, elicited depressor actions in both a animals. The hypotensive effects of SGEE in dogs and chickens were markedly inhibited by the pretreatment of atropine, but not affected by bethanidine, propranolol and diphenhydramine. Phentolamine significantly weakened the action of SGEE in dogs, but did not alter the action of SGEE in chickens. SGEE inhibited the pressor action by norepinephrine or carotid artery occlusion, whereas it did not change the pressor action b by angiotensin in dogs.

  • To investigate the effects of Siegesbeckiae Herba on the blood pressure, this study was carried out in the whole and the spinal rabbits, using its water and ethanol extracts. When Sigesbeckiae Herba water extract (SGWE) and ethanol extract (SGEE) were administered into the whole rabbits by route of e ear vein, both of them produced fall of blood pressure. The difference between these two extracts was that SGEE is more potent than SGWE. The depressor effects of SGWE and SGEE were not affected by vagoto minization but inhibited by pretreatment of atropine. The depressor responses of the whole rabbits to intravenous SGWE and SGEE were weakened by treatment of animals with bethanidine or phentolamine but not by propranolol. Pretreatment of the whole rabbits with diphenhydramine significantly weakened the d depressor effects of SGEE. Infusion of SGWE and SGEE in the whole rabbits did not influence the pressor effects caused by angiotensin, norepinephrine or carotid artery occlusion. SGEE, when given into the lateral ventricle of the whole rabbits or into the vein of the spinal rabbits, elicited fall of blood pressure, respectively.

Effects of Siegesbeckiae Herba on rheumatoid arthritis induced by collagen in MRL/MpJ-Ipr-Ipr Mice (희첨이 RA 병태 모델에서 관련인자 및 조직학적 변화에 미치는 영향)

  • For evaluating the effect of Siegesbeckiae Herba (here after abbreviated as SBH) on rheumatoid arthritis, the experiment was carried out; Incidence of rheumatoid arthritis, IRA(indices of rheumatoid arthritis), immunophenotypes by flow cytometer and histopathological changes in MRL/MpJ-Ipr-Ipr Mice in vivo were studied. The results were obtained as follows: 1. Incidence of RA in MRL/Ipr mice was suppressed to 60% of control by SBH. 2. IRA was significantly reduced for IgG3 and IgM at 20 weeks of age and for IgG2b at 12 and 20 weeks of age in mice by SBH compared with control. 3. Immunophenotypes such as CD4/sup +//CD25/sup +/, CD8/sup +//CD3e/sup +/, CD69/sup +//B220/sup +/, NK/sup +//CD3e/sup +/ were significantly increased by SBH compared with control. 4. In histopathological analysis, SBH suppressed the progression of PMN(polymorphonuciear leukocyte), leukocyte and fibroblast infiltration, and subsynovial soft tissue edema frequently showing in the early stage of the arthritis, and also effectively reduced the degeneration of cartilages and degenerative bone symptoms. These results suggest Siegesbeckiae Herba can be effectively applied to the treatment of rheumatoid arthritis and it is still necessary to isolate effective compound from Siegesbeckiae Herba in the near future.

Exmination of Anti-Obesity Effect of Regional Special Natural Products of Anthrisci radix, Psoraleae semen, Siegesbeckiae herba and Corni fructus (지역 특산 천연산물 전호, 파고지, 희첨 및 산수유의 항비만효과 규명)

  • 4 kinds of Regional Special Natural Products (RSNPs), such as Anthrisci radix, Psoraleae semen, Siegesbeckiae herba and Corni fructus were examined to verify for anti-obesity effect. $PPAR\gamma$ (peroxisome proliferator-activated receptor $\gamma$) from 3T3-L1 cell concerning adipocyte differentiation was suppressed by different concentraton of 4 RSNPs with western blot, when treated RSNPs' extract and MDI (IBMX, Dexamethasone, Insulin) at the same time. Also, SREBP-1 (Sterol regulatory element binding protein) controlling lipogenesis and $PPAR\gamma$ expression levels were reduced by these 4 RSNPs' extract, when these chemicals after differentiation of 3T3-L1 cell. And lipid droplets were reduced by 7.5%, 14.4%, 18.3% and 30% at different concentration of Anthrisci radix from Oil Red O staining. Also, it was reduced by 2%, 4.9%, 9.3% and 38% at different concentration of Psoraleae semen. For Siegesbeckiae herba, it was inhibited by 1.4%, 6.4%, 16.4% and 30.1%, respectively. And Corni fructus was also showed by 0.9%, 6.3%, 13.7% and 33% at same concentration of Siegesbeckiae herba. These 4 kinds of RSNPs were expected for a useful material for anti-obesity materials.

A Study on the Diuretic Action of Sigesbeckiae Herba Ethanol Extract in Dogs (희점(稀簽) 에탄올엑기스의 이뇨작용(利尿作用)에 관(關)한 연구(硏究))

  • A study on the diuretic action of Siegesbeckiae herba in dogs was made using a clearance technique with ethanol extract. Siegesbeckiae herba ethanol extract (SGEE) administered intravenously in doses 6.0 and 60.0mg/kg elicited diuresis and an increase in sodium and potassium excretion, and it simultaneously produced a decrease in the reabsorption rate of sodium and potassium in renal tubules. Neither renal plasma flow nor glomerular filtration rate changed significantly. SGEE, when infused directly into a renal artery in doses of 0.5 and 1.5mg/kg min, exhibited identical results to the intravenous responses confined only to the infused kidney. The above observations lead to the suggestion that SGEE elicits diuresis in dogs by decreasing the reabsorption rates of sodium and potassium in renal tubles.

  • The method extracts of Patriniae radix, Dianthii herba, Melandrii herba, Echinopii radix, Siegesbeckiae herba and Magnoliae cortex showed a significant elevation of serum transam inase activity accompanied by fatty degeneration and Kupffer accompanied by fatty degeneration and Kupffer cell activation in hepatic cells.
