Morphological studies on the dendritic cells in the mammary gland II. Electron microscopic observations (유선조직내에 출현하는 dendritic cell의 형태학적 연구 II. 전자현미경적 관찰)

  • In order to investigate the morphological characteristics of dendritic cells in the mammary gland of the mouse (C57 BL/6), rat(W), rabbit and cat, the fine structures of the dendritic cells have been observed by the electron microscope. The results obtained were summarized as follows: The dendritic cells with the well-developed processes had an irregular shape, and lacked the desmosome. The pinocytotic vesicles and tubular invaginations of the cell membrane were frequently observed, and the mitochondria with the well-developed cristae were located in the restricted region in the cytoplasm of the dendritic cells. The nuclei of dendritic cells were indented, In the mice and rats, the dendritic cells had a few Langerhans cell granules(Birbeck granules). From the above results, it is confirmed that the ATPase-positive dendritic cells in the mammary gland are the Langerhans cells.

Proton Conducting Behavior of a Novel Composite Based on Phosphosilicate/Poly(Vinyl Alcohol)

  • A series of proton conductive composite membranes based on poly(vinyl alcohol) and phosphosilicate gels powders were successfully prepared. The proton conductivity of these composite was attributed to the phosphosilicate gel, which derived from tetraethoxysilane and phosphoric acid by sol-gel process at a molar ratio of P/Si = 1.5. The proton conductivity increased with increasing both the content of phosphosilicate gel and relative humidity. Temperature dependence of conductivity showed a Vogel-Tamman-Fulcher type behavior, indicating that proton was transferred through a liquidlike phase formed in micropores of phosphosilicate gel. The high conductivity of 0.065 S/cm with a membrane containing 60 wt$\%$ of the gel was obtained at $60^{\circ}C$ at $90\%$ relative humidity.

  • Hepatocyte growth factor(HGF) and c-Met play an important role in the control of tumor growth and invasion, and they are known to be good prognostic indicators of patient outcome. Epigallocatechin-3-gallate (EGCG) has been shown to have chemopreventive and therapeutic properties by modulating multiple signal pathways regarding the control of proliferation and invasion of cells. In this study, we evaluated the role of c-Met in EGCG-induced inhibition of invasion and apoptosis in an oral cancer cell line. In KB cells where c-Met was knocked down with siRNA, we performed invasion assay and FACS with Annexin V-FITC/PT staining. In addition, we checked the change of mitochondrial membrane potential(MMP) and the generation of reactive oxygen species(ROS). EGCG-induced inhibition of invasiveness was significantly decreased after the knock-down of c-Met. EGCG-induced apoptosis, MMP change and ROS generation was also reduced in c-Met knock-ed-down KB cells. These results suggest that c-Met is involved in EGCG-induced apoptosis and inhibition of invasiveness of oral cancer cell line.

Fabrication and yield improvement of oxide semiconductor thin film gas sensor array (산화물 반도체 박막 가스센서 어레이의 제조 및 수율 개선)

  • A thin film oxide semiconductor micro gas sensor array which shows only 60㎽ of power consumption at an operating temperature of 30$0^{\circ}C$ has been fabricated using microfabrication and rnicrornachining techniques. Excellent thermal insulation of the membrane is achieved by the use of a double la! or structure of 0.1${\mu}{\textrm}{m}$ thick Si$_3$N$_4$ and 1${\mu}{\textrm}{m}$ thick phosphosilicate glass(PSG) prepared by low pressure chemical vapor deposition(LPCVD) and atmospheric-pressure chemical-vapor deposition(APCVD), respectively. The sensor way consists of such thin film oxide semiconductor sensing materials as 1wt.% Pd-doped SnO$_2$, 6wt.% AI$_2$O$_3$-doped ZnO, WO$_3$ and ZnO. The thin film oxide semiconductor micro gas sensor array exhibited resistance changes usable for subsequent data processing upon exposure to various gases and the sensitivity strongly depended on the sensing layer materials. Heater Part of the sensor structure has been modified in order to improve the process yield of the sensor, and as a result of modified heater structure improved process yield has been achieved.

MOF-based membrane encapsulated ZnO nanowires for H2 selectivity (MOF 기반 멤브레인 기능화된 ZnO 나노선의 수소 가스 선택성)

  • 가스센서는 사내 및 산업 환경에서의 유독성 또는 폭발성 가스 검출, 환경 모니터링, 질병 진단 등 매우 다양한 응용분야에서 큰 관심을 가지고 있다. 반도체 금속산화물(SMOs) 기반의 센서 분야에서는 이들의 감도 및 선택성을 향상시키기 위해 많은 노력을 기울이고 있다. 이는 센서의 선택성을 부여하게 되면 다양한 가스들이 존재하는 환경에서도 검출자가 원하는 가스만의 응답을 얻을 수 있기 때문이다. 본 연구에서는 MOF(Metal-Organic Framwork) 기반 멤브레인으로 ZIF-8(Zeolitic Imidazolate Frameworks 구조들 중 하나) 멤브레인 쉘 층을 이용하여 ZnO 나노선에 형성하였다. ZnO 나노선은 VLS공정 (Vapor-Liquid-Solid)을 이용하여 패턴된 전극을 갖는 $SiO_2$-grown Si 웨이퍼 상에 성장되었고, 성장된 ZnO 나노선은 2-methyl imidazole과 methanol이 포함된 고용체에 넣고 폐쇄된 압력용기 속에서 가열시켜 얻게 된다. 이렇게 얻어진 ZIF-8@ZnO 나노선의 ZIF-8 멤브레인은 분자 체 구조(molecular sieving structure)를 갖게 되며, 이들의 pore 크기는 약 $3.4{\AA}$을 갖는다. 따라서 이보다 더 큰 동적 직경을(kinetic diameter) 갖는 가스 종은 이 멤브레인을 통과할 수 없음을 나타내므로 제작된 시편은 $H_2$(kinetic diameter : $2.89{\AA}$), $C_7H_8$(kinetic diameter : $5.92{\AA}$), 그리고 $C_6H_6$(kinetic diameter : $5.27{\AA}$) 가스들을 각각 사용함으로써 ZIF-8@ZnO 나노선의 센서 특성을 조사했으며, 보다 정확한 비교를 위해 순수한 ZnO 나노선 역시 동일한 조건에서 측정되었다. 결과를 통해, 수소 가스를 제외한 다른 가스들에 대해서는 반응을 하지 않고, 오직 수소 가스에 대해서만 반응을 나타냈으며, 순수 ZnO 나노선의 수소 감응도보다 낮은 감응도를 나타내었다. 이는 멤브레인 쉘 층을 형성함으로써 ZnO 나노선의 표면적이 감소해 가스 분자와의 접촉점을 감소시키기 때문이라고 판단된다. 이와 같은 MOF 멤브레인의 캡슐화 전략은 가스센서뿐 아니라 바이오 센서 및 광촉매 등과 같은 이온 선택성을 필요로 하는 다양한 응용분야에 적용될 수 있을 것으로 기대된다.

Cripto Enhances Proliferation and Survival of Mesenchymal Stem Cells by Up-Regulating JAK2/STAT3 Pathway in a GRP78-Dependent Manner

  • Cripto is a small glycosylphosphatidylinositol-anchored signaling protein that can detach from the anchored membrane and stimulate proliferation, migration, differentiation, vascularization, and angiogenesis. In the present study, we demonstrated that Cripto positively affected proliferation and survival of mesenchymal stem cells (MSCs) without affecting multipotency. Cripto also increased expression of phosphorylated janus kinase 2 (p-JAK2), phosphorylated signal transducer and activator of transcription 3 (p-STAT3), 78 kDa glucose-regulated protein (GRP78), c-Myc, and cyclin D1. Notably, treatment with an anti-GRP78 antibody blocked these effects. In addition, pretreatment with STAT3 short interfering RNA (siRNA) inhibited the increase in p-JAK2, c-Myc, cyclin D1, and BCL3 levels caused by Cripto and attenuated the pro-survival action of Cripto on MSCs. We also found that incubation with Cripto protected MSCs from apoptosis caused by hypoxia or $H_2O_2$ exposure, and the level of caspase-3 decreased by the Cripto-induced expression of B-cell lymphoma 3-encoded protein (BCL3). These effects were sensitive to down-regulation of BCL3 expression by BCL3 siRNA. Finally, we showed that Cripto enhanced expression levels of vascular endothelial growth factor (VEGF), fibroblast growth factor (FGF), and hepatocyte growth factor (HGF). In summary, our results demonstrated that Cripto activated a novel biochemical cascade that potentiated MSC proliferation and survival. This cascade relied on phosphorylation of JAK2 and STAT3 and was regulated by GRP78. Our findings may facilitate clinical applications of MSCs, as these cells may benefit from positive effects of Cripto on their survival and biological properties.

Freehang 방법을 이용한 DLC 필름의 탄성 특성 평가

  • 박막의 탄성 특성을 평가하는 방법으로 nano-indentation, Brillouin light scattering measurement, ultrasonic surface wave measurement, bulge test, vibration membrane method 등 여러 가지가 제시되어 왔다. 이러한 방법들은 필름의 두께가 일정 두께 이상이 되어야 정확한 측정이 가능한 방법으로 매우 얇은 박막에서도 탄성특성을 평가할 수 있는 freehang, bridge 방법이 제시되었으며, 이 방법은 간단한 식각 공정을 통해 매우 얇은 박막에도 적용시킬 수 있다는 장점을 가지고 있다. 본 연구에서는 아주 얇은 박막에서도 탄성특성을 평가할 수 있는 freehang 방법을 이용하여 순수한 Diamond-like carbon (DLC) 필름과 Sidl 첨가된 DLC 필름의 탄성 특성을 평가하고자 한다. 실험에서 사용한 필름은 rf-PACVD 장비를 이용하여 증착하였다. 이때 전극과 플라즈마 사이의 바이어스 음전압은 -400 Vb로 합성압력은 10mTorr로 고정하였다. 사용한 반응 가스는 벤젠(C6H6), 그리고 벤젠과 희석된 실렌(SiH4 : H2 = 10 : 90)이며, 희석된 실렌의 첨가량을 조절하여 필름 내에 일정량의 Si을 함유시켰다. 각각의 조건에서 증착시간을 조절하여 필름의 두께를 변화시켰으며, KOH(5.6mol) 용액을 이용하여 습식 식각을 함으로써 freehang을 제작하였다. 이때 식각액에 의한 DLC 필름의 손상은 관찰되지 않았다. 필름의 잔류 응력을 측정하기 위해 200$\pm$10 혹은 100$\pm$5$\mu\textrm{m}$ 두께의 얇은 (100) Si wafer를 5$\times$50 mm2의 strip 형태로 절단하여 사용하였다. 필름의 압축 잔류 응력에 의해 발생한 필름/기판 복합체의 곡률은 laser 반사법과 $\alpha$-step profiler를 이용하여 측정하였으며, 이 결과를 Brenner 등에 유도된 식을 이용하여 잔류 응력을 계산하였다. 또한 제작된 frddhang은 광학 현미경과 전자주사현미경에 의해 관찰되었다. 이렇게 제작된 freehang을 이용하여 필름이 기판에 부착되기 위해 필요한 변형률을 측정하고, 독립적으로 측정된 필름의 잔류 응력을 박막의 응력-변형률 관계식에 적용하여 biaxial elastic modulus, E/(1-v)를 구할 수 있었다. 측정 결과 필름의 잔류 응력과 biaxial elastic modulus는 필름의 두께가 감소함에 따라 감소하는 경향을 나타냈으며, 같은 두께의 필름인 경우, 식각 깊이에 따른 biaxial elastic modulus 의 변화를 통해 최적의 식각 깊이를 알 수 있었다.

Identification of the genes which related cold (low temperature) stress in Bombyx mori

  • Stress may be defined as any modification of environmental parameters that leads to a response by biological organisms. Stresses that affect biolpgical structures may be nonthermal, such as ultraviolet radiation, pH, or salinity, or thermal. Temperture is one of the major stresses that all living organism face. The major effects of cold(low emperature) are decrease of membrane fluidity and the stabilization of secondary structures of RNA and DNA in the cells, which may effect the efficiency of translation, transcription, and DNA replication. In this study, we focus on discovering the genes that are expressed by the cold(low temperature) stress in the silkworm. In cold(low temperature) stress test, we found 100% survive from cold stress at $0^{\circ}C$ up to 12h and $-5^{\circ}C$ up to 2h, and then, survive rate was rapidly decrease in silkworm. Thereafter two whole genes have selected by SSH(Suppression subtractive hybridization). (GenBank accession : GQ149511, GQ338156)

Preparation and Characterization of Proton Conductive Phosphosilicate Membranes Based on Inorganic-Organic Hybrid Materials

  • A series of proton conductive inorganic-organic hybrid membranes doped with phosphoric acid ($H_3PO_4$) and/or triethylphosphate (PO(OEt)$_3$) have been prepared by sol-gel process with 3-glycidoxypropyltrimethoxysilane (GPTMS), 3-aminopropyltriethoxysilane (APTES) and tetraethoxysilane (TEOS) as precursors. High proton conductivity of 3.0 ${\times}$ $10^{-3}$ S/cm with composition of 50TEOS-30GPTMS-20APTES-50$H_3PO_4$ was obtained at 120 ${^{\circ}C}$ under 50% relative humidity. Thermal stability of membrane was significantly enhanced by the presence of SiO$_2$ framework up to 250 ${^{\circ}C}$. XRD revealed that the gels are amorphous. IR spectra showed a good complexation of $H_3PO_4$ in the matrix. The conductivity under 75% relative humidity was significantly improved by addition of APTES due to the increase in concentration of defected site in hybrid matrix. The effect of PO(OEt)$_3$, humidifying time, and heat-treatment were also investigated. PO(OEt)$_3$ had no improvement on conductivity and conductivity increased with humidifying time, however, decreased with heating temperature.
