Several [2]- and [3]rotaxanes bearing some functional groups on their wheel components and spacers with different lengths between two ammonium centers on their dumbbell components were prepared in good yields from dibenzo-24-crown-8-ether derivatives and dumbbell-shaped bis(sec-ammonium salt)s having a centrally located disulfide linkage, by utilizing the reversible thiol-disulfide interchange reaction. The shuttling behaviors of the [2]rotaxanes were investigated by $^1H$ NMR by use of the spin polarization transfer-selective inversion recovery technique. It was found that the change in spacer length in the axle resulted in a drastic change in shuttling rate of the [2]rotaxanes, although the introduction of the functional groups to the wheels did not affect the shuttling behavior at all.
As anode fabrication with different materials has been proven to be a successful alternative for enhancing power generation in the microbial fuel cells, a new approach to improved performance of MFCs with the use of menadione/carbon powder composite-modified carbon cloth anode has been explored in this study. Menadione has formal potential to easily accept electrons from the outer membrane cytochromes of electroactive bacteria that can directly interact with the solid surface. Surface bound menadione was able to maintain an electrical wiring with the trans-membrane electron transfer pathways to facilitate extracellular electron transfer to the electrode. In a single chamber air cathode MFC inoculated with aerobic sludge, maximum power density of $1250{\pm}35mWm^{-2}$ was achieved, which was 25% higher than that of an unmodified anode. The observed high power density and improved coulomb efficiency of 61% were ascribed to the efficient electron shuttling via the immobilized menadione.
Serine and arginine-rich (SR) proteins are RNA-binding proteins (RBPs) known as constitutive and alternative splicing regulators. As splicing is linked to transcriptional and post-transcriptional steps, SR proteins are implicated in the regulation of multiple aspects of the gene expression program. Recent global analyses of SR-RNA interaction maps have advanced our understanding of SR-regulated gene expression. Diverse SR proteins play partially overlapping but distinct roles in transcription-coupled splicing and mRNA processing in the nucleus. In addition, shuttling SR proteins act as adaptors for mRNA export and as regulators for translation in the cytoplasm. This mini-review will summarize the roles of SR proteins as RNA binders, regulators, and connectors from transcription in the nucleus to translation in the cytoplasm.
본 연구의 목적은 넙치 인지질가수분해효소(PLC-${\delta}1$) 3가지 타입의 세포내 특성을 규명하고자 하였다. 일반적으로 인지질가수분해효소(PLC)의 신호전달경로는 핵, 세포막, 세포질에 분포한다고 알려져 있으나, 핵내 위치 메커니즘은 여전히 불분명하다. PoPLC-${\delta}1A$, PoPLC-${\delta}1B$ (Sf)과 PoPLC-${\delta}1B$ (Lf)의 3타입의 유전자들은 각각 핵위치 신호(NLS)와 핵방출서열(NES)을 포함하고 있다. 본 연구에서는, 넙치 3가지 타입 인지질가수분해효소(PLC-${\delta}1$)의 세포내 위치이동 메커니즘 분석을 위해 GFP 벡터에 유전자를 삽입하여 ionomycin과 thasogargin처리 후 세포위치와 이동양상을 공초점 레이저 주사현미경으로 관찰하였다. PoPLC-${\delta}1A$는 PoPLC-${\delta}1B$ (Lf)와 PoPLC-${\delta}1B$ (Sf)가 원형질막에 국한되어 분포할때 세포질과 세포막보다 세포 소기관에 분포되어 있었다. PoPLC-${\delta}1B$ (Lf) 및 PoPLC-${\delta}1$ (Sf)이 핵 세포질내 이동양상을 보이지 않을 때, PoPLC-${\delta}1A$은 ionomycin과 thapsigargin 처리에 의해 핵 내에 축적되는 양상을 나타냈다. 이런 결과는 손상되지 않은 기능적 NES 서열을 포함하는 PoPLC-${\delta}1A$가 어류에서 핵 세포질 내 왕복 및 이동의 주된 역할을 한다는 것을 보여주고 있다.
Insulating effects on Au electrode according to the thickness and density of coated materials are well-known. To do electrochemical immunoassay reproducibly the glod electrode would be coated with self-assembled monolayers and antobodies. To get reproducibility, the antobody monolayer should be packed at highest density so that the amount of immobilized antibody at defined area should be the same. The calix[4]crown-5 SAMs could provide the basis for the antibodies to be immobilized reproducibly and at highest density. But the insulating effect would be highest too. We proved that the compactly packed protein monolayers on SAMs inhibited the electron transfer by block the free shuttling of redox molecules. The inhibition was minimized by inserting redox molecules in between the proteins during immobilization process. In this paper, we demonstrated that the calix[4]crown-5 SAMs would provide the protein monolayers with highest density and new method to minimize the insulating effect by inserted redox molecules in between the compactly packed protein monolayers.
We established an efficient transformation method for thermophile Geobacillus kaustophilus HTA426 using conjugative transfer from Escherichia coli of host-mimicking plasmids that imitate DNA methylation of strain HTA426 to circumvent its DNA restriction barriers. Two conjugative plasmids, pSTE33T and pUCG18T, capable of shuttling between E. coli and Geobacillus spp., were constructed. The plasmids were first introduced into E. coli BR408, which expressed one inherent DNA methylase gene (dam) and two heterologous methylase genes from strain HTA426 (GK1380-GK1381 and GK0343-GK0344). The plasmids were then directly transferred from E. coli cells to strain HTA426 by conjugative transfer using pUB307 or pRK2013 as a helper plasmid. pUCG18T was introduced very efficiently (transfer efficiency, $10^{-5}-10^{-3}\;recipient^{-1}$). pSTE33T showed lower efficiency ($10^{-7}-10^{-6}\;recipient^{-1}$) but had a high copy number and high segregational stability. Methylase genes in the donor substantially affected the transfer efficiency, demonstrating that the host-mimicking strategy contributes to efficient transformation. The transformation method, along with the two distinguishing plasmids, increases the potential of G. kaustophilus HTA426 as a thermophilic host to be used in various applications and as a model for biological studies of this genus. Our results also demonstrate that conjugative transfer is a promising approach for introducing exogenous DNA into thermophiles.

We describe a redox-enhanced electric double-layer capacitor (EDLC) that turns the electrolyte in a conventional EDLC into an integral, active component for charge storage-charge is stored both through faradaic reactions with soluble redox-active molecules in the electrolyte, and through the double-layer capacitance in a porous carbon electrode. The mixed-redox electrolyte, composed of vanadium and iodides, was employed to achieve high power density. The electrochemical reaction in a supercapacitor with vanadium and iodide was studied to estimate the charge capacity and energy density of the redox supercapacitor. A redox supercapacitor with a mixed electrolyte composed of 0.75 M NaI and 0.5 M $VOSO_4$ was fabricated and studied. When charged to a potential of 1 V, faradaic charging processes were observed, in addition to the capacitive processes that increased the energy storage capabilities of the supercapacitor. The redox supercapacitor achieved a specific capacity of 13.44 mAh/g and an energy density of 3.81 Wh/kg in a simple Swagelok cell. A control EDLC with 1 M $H_2SO_4$ yielded 7.43 mAh/g and 2.85 Wh/kg. However, the relatively fast self-discharge in the redox-EDLC may be due to the shuttling of the redox couple between the polarized carbon electrodes.
The potential for humic substances to serve as terminal electron acceptors in microbial respiration and the effects of humic substances on microbial azoreduction were investigated. The dissimilatory azoreducing microorganism Shewanella decolorationis S12 was able to conserve energy to support growth from electron transport to humics coupled to the oxidation of various organic substances or $H_2$. Batch experiments suggested that when the concentration of anthraquinone-2-sulfonate (AQS), a humics analog, was lower than 3 mmol/l, azoreduction of strain S12 was accelerated under anaerobic condition. However, there was obvious inhibition to azoreduction when the concentration of the AQS was higher than 5 mmol/l. Another humics analog, anthraquinone-2-sulfonate (AQDS), could still prominently accelerate azoreduction, even when the concentration was up to 12 mmol/l, but the rate of acceleration gradually decreased with the increasing concentration of the AQDS. Toxic experiments revealed that AQS can inhibit growth of strain S12 if the concentration past a critical one, but AQDS had no effect on the metabolism and growth of strain S12 although the concentration was up to 20 mmol/l. These results demonstrated that a low concentration of humic substances not only could serve as the terminal electron acceptors for conserving energy for growth, but also act as redox mediator shuttling electrons for the anaerobic azoreduction by S. decolorationis S12. However, a high concentration of humic substances could inhibit the bacterial azoreduction, resulting on the one hand from the toxic effect on cell metabolism and growth, and on the other hand from competion with azo dyes for electrons as electron acceptor.
Purpose: The purpose of this study was to understand and describe the illness experience of married Korean women with epilepsy. Methods: Data were collected during 2015~2016 through individual in-depth interviews with 12 married women with epilepsy. Verbatim transcripts were analyzed using Giorgi's phenomenological analysis to uncover the meaning of the illness experience of the participants. Results: The study results showed that the illness experience of married Korean women with epilepsy was clustered into a specific description of situated structure and a general description of situated structure. Six themes from 20 meaning units were identified: 1) Undermined self-esteem with stigma of being epileptic; 2) Limited social interaction; 3) Suffering sorrow as a 'disqualified being'; 4) Shuttling back and forth across the boundary between healthy and epileptic; 5) Desperate struggle to meet the expectation of given role; 6) Self-empowering through self-restriction and realization. Conclusion: The findings from this study show that both the enacted and felt stigma of epilepsy impact on the life of married Korean women with epilepsy. Although the participants face social and interpersonal restriction and prejudices, they try their best to fulfill their role rather than to be cared for as patients. As the stigma and hardships of the participants are related to lack of knowledge, health professionals should focus not just on clinical intervention but also on providing targeted educational programs and counseling for these women to dispel the stigma of the disease and to increase their quality of life.

Transcription factor Nrf2 is essential for the antioxidant responsive element (ARE)-mediated induction of phase II detoxifying and oxidative stress enzyme genes. Detailed analysis of differential Nrf2 activity displayed in transfected cell lines ultimately led to the identification of a new protein, which we named Keap1, that suppresses Nrf2 transcriptional activity by specific binding to its evolutionarily conserved amino-terminal regulatory domain. The closest homolog of Keap1 is a Drosophila actin-binding protein called Kelch, implying that Keap1 might be a Nrf2 cytoplasmic effector. We then showed that electrophilic agents antagonize Keap1 inhibition of Nrf2 activity in vivo, allowing Nrf2 to traverse from the cytoplasm to the nucleus and potentiate the ARE response. We postulate that Keap1 and Nrf2 constitute a crucial cellular sensor for oxidative stress, and together mediate a key step in the signaling pathway that leads to transcriptional activation by this novel Nrf2 nuclear shuttling mechanism. The activation of Nrf2 leads in turn to the induction of phase II enzyme and antioxidative stress genes in response to electrophiles and reactive oxygen species.
