Structural Optimization for LMTT-Mover Using the Kriging Based Approximation Model (크리깅 근사모델 모델을 이용한 LMTT 이동체의 구조최적설계)

  • LMTT (Linear Motor-based Transfer Techn-ology) is a horizontal transfer system for the yard automation, which has been proposed to take the place of AGV (Automated Guided Vehicle) in the maritime container terminal. The system is based on PLMSL (Permanent Magnetic Linear Synchronous Motor) that consists of stator modules on the rail and shuttle car. It is desirable to reduce the weight of LMTT in order to control the electronic devices with minimum energy. In this research, the DACE modeling, known as the one of Kriging interpolation, is introduced to obtain the surrogate approximation model of the structural responses. Then, the GRG(Generalized Reduced Gradient) method built in Excel is adopted to determine the optimum. The objective function is set up as weight. On the contrary, the design variables are considered as transverse, longitudinal and wheel beam's thicknesses, and the constraints are the maximum stresses generated by four loading conditions.

  • The actual observance of hours of rest became one of the main checking items of PSC after enter into force of Maritime Labour Convention in 20 August 2013. And the non-compliances of the rest hours stipulated in the provisions of Seafare's Act of Korea and MLC is occuring and this would be the great obstruction for the shuttle vessels operating between Korea and Japan. In this study, to find the method which could give solution for the observance of provisions of rest hours, the modified duty model, reduction of calling ports and substitute of port duty by shore assistance personnel were reviewed. To comply with the requirement of hours of rest by shuttle vessels operating between Korea and Japan, adoption of the method to reduce calling ports or substitution of port duty by shore personnel are recommended.

  • For the expression of the cry11Aa gene highly toxic to dipteran insects, we constructed two cyanobacteria-Escherichia coli(E. coli) shuttle vectors, pCYASK5-l and pCYASK5-2. These vectors were transformed into E. coli and selected with kanamycin. The expression of the cry11Aa gene in E. coli was characterized by SDS-polyacrylamide gel electrophoresis and Western blot analysis. Two E. coli transformants harboring pCYASK5-1 and pCYASK5-2 expressed the cry11Aa gene in size of 72 kDa and 64 kDa, respectively and showed over 89% mortality against Culex pipiens larvae.

Evaluating Applicability of SRTM DEM (Shuttle Radar Topography Mission Digital Elevation Model) in Hydrologic Analysis: A Case Study of Geum River and Daedong River Areas (수문인자추출에서의 SRTM DEM (Shuttle Radar Topography Mission Digital Elevation Model) 적용성 평가: 대동강 및 금강 지역 사례연구)

  • Shuttle Radar Topography Mission Digital Elevation Model (SRTM DEM) offers opportunities to make advances in many research areas including hydrology by providing near-global scale elevation measurements at a uniform resolution. Its wide coverage and complimentary online access especially benefits researchers requiring topographic information of hard-to-access areas. However, SRTM DEM also contains inherent errors, which are subject to propagation with its manipulation into analysis outputs. Sensitivity of hydrologic analysis to the errors has not been fully understood yet. This study investigated their impact on estimation of hydrologic derivatives such as slope, stream network, and watershed boundary using Monte Carlo simulation and spatial moving average techniques. Different amount of the errors and their spatial auto-correlation structure were considered in the study. Two sub-watersheds of Geum and Deadong River areas located in South and North Korea, respectively, were selected as the study areas. The results demonstrated that the spatial presentations of stream networks and watershed boundaries and their length and area estimations could be greatly affected by the SRTM DEM errors, in particular relatively flat areas. In the Deadong River area, artifacts of the SRTM DEM created sinks even after the filling process and then closed drainage basin and short stream lines, which are not the case in the reality. These findings provided an evidence that SRTM DEM alone may not enough to accurately figure out the hydrologic feature of a watershed, suggesting need of local knowledge and complementary data.

Deep X-ray Mask with Integrated Micro-Actuator for 3D Microfabrication via LIGA Process (3차원 LIGA 미세구조물 제작을 위한 마이크로 액추에이터 내장형 X-선 마스크)

  • We present a novel method for 3D microfabrication with LIGA process that utilizes a deep X-ray mask in which a micro-actuator is integrated. The integrated micro-actuator oscillates the X-ray absorber, which is formed on the shuttle mass of the micro-actuator, during X-ray exposures to modify the absorbed dose profile in X-ray resist, typically PMMA. 3D PMMA microstructures according to the modulated dose contour are revealed after GG development. An X-ray mask with integrated comb drive actuator is fabricated using deep reactive ion etching, absorber electroplating, and bulk micromachining with silicon-on-insulator (SOI) wafer. 1mm $\times$ 1 mm, 20 $\mu$m thick silicon shuttle mass as a mask blank is supported by four 1 mm long suspension beams and is driven by the comb electrodes. A 10 $\mu$m thick, 50 $\mu$m line and spaced gold absorber pattern is electroplated on the shuttle mass before the release step. The fundamental frequency and amplitude are around 3.6 kHz and 20 $\mu$m, respectively, for a do bias of 100 V and an ac bias of 20 $V_{p-p}$ (peak-peak). Fabricated PMMA microstructure shows 15.4 $\mu$m deep, S-shaped cross section in the case of 1.6 kJ $cm^{-3}$ surface dose and GG development at 35$^{\circ}C$ for 40 minutes.

Construction of a Novel Shuttle Vector for Tetragenococcus species based on a Cryptic Plasmid from Tetragenococcus halophilus

  • A cryptic plasmid (pTH32) was characterized from Tetragenococcus halophilus 32, an isolate from jeotgal, Korean traditional fermented seafood. pTH32 is 3,198 bp in size with G+C content of 35.84%, and contains 4 open reading frames (ORFs). orf1 and orf2 are 456 bp and 273 bp in size, respectively, and their translation products showed 65.16% and 69.35% similarities with RepB family plasmid replication initiators, respectively, suggesting the rolling-circle replication (RCR) mode of pTH32. orf3 and orf4 encodes putative hypothetical protein of 186 and 76 amino acids, respectively. A novel Tetragenococcus-Escherichia coli shuttle vector, pMJ32E (7.3 kb, Emr), was constructed by ligation of pTH32 with pBluescript II KS(+) and an erythromycin resistance gene (ErmC). pMJ32E successfully replicated in Enterococcus faecalis 29212 and T. halophilus 31 but not in other LAB species. A pepA gene, encoding aminopeptidase A (PepA) from T. halophilus CY54, was successfully expressed in T. halophilus 31 using pMJ32E. The transformant (TF) showed higher PepA activity (49.8 U/mg protein) than T. halophilus 31 cell (control). When T. halophilus 31 TF was subculturd in MRS broth without antibiotic at 48 h intervals, 53.8% of cells retained pMJ32E after 96 h, and only 2.4% of cells retained pMJ32E after 14 days, supporting the RCR mode of pTH32. pMJ32E could be useful for the genetic engineering of Tetragenococcus and Enterococcus species.

Application of Graphene Nanoribbon Trench for C60 Fullerene Shuttle Device: Molecular Dynamics Simulations (풀러렌 셔틀 소자로 그래핀 나노리본 트렌치 응용에 관한 분자동력학 시뮬레이션 연구)

  • We investigated the position controlling C60 fullerene encapsulated into a graphene-nanoribbon trench via classical molecular dynamics simulations. The graphene-nanoribbon trench can provide nanoscale empty spaces, and a C60 encapsulated therein can be considered as media for a nanoelectromechanical shuttle device. The classical molecular dynamics simulations presented here provide information on the potential application of a graphene-nanoribbon trench in a C60 shuttle device. Driving forces applied to C60 resulted in its motion toward the edges of the graphene-nanoribbon trench, the suction forces induced at both edges were balanced with the driving forces, and finally, the C60 fullerene gradually settled on the edges of the graphene-nanoribbon trench after several oscillations. The results of the present simulation suggest the importance of graphene-nanoribbon trenches encapsulating fullerenes in a wide range of applications in the field of nanotechnology.

Vegetation Height and Age Estimation using Shuttle Radar Topography Mission and National Elevation Datasets (SRTM과 NED를 이용한 식생수고 및 수령 추정)

  • SAR (Synthetic Aperture Radar) technology, which is not influenced by cloud cover because of using electromagnetic wave of long wavelength, has an advantage in mapping the earth. NASA, recognizing these strong points of SAR, launched SRTM (Shuttle Radar Topography Mission), and acquired the topographic information of the earth. SRTM and NED (National Elevation Data) of USGS were used for the research and vegetation height map was produced through differentiating the two data. Correlation between SRTM-NED and planting year was analyzed to see the relationship. Strong correlation was detected and it shows the feasibility of estimating timber age and eventually creating timber age map from SRTM-NED. Additional analyses were conducted to check if the linearity is influenced by regional characteristics and forest uniformity. As results, the correlation between SRTM-NED and timber age is influenced by roughness of the terrain. Overall, this paper shows that timber age estimation using SRTM and NED can be sufficiently practical.

Characterization of Plasmid pKJ36 from Bifidobacterium longum and Construction of an E. coli-Bifidobacterium Shuttle Vector

  • Abstract The full sequence of the plasmid pKJ36, which was derived from Bifidobacterium longum KJ, was determined and analyzed to construct shuttle vectors between E. coli and Bifidobacterium. The plasmid pKJ36 was composed of 3,625 base pairs with a 65.1% G+C content. The structural organization of pKJ36 was highly similar to that of pKJ50, and the three major ORFs on pKJ36 showed high amino acid sequence homologies with those of pKJ50. The putative proteins coded by these three ORFs were designated as RepB (32.0 kDa, pI=9.25), MembB (29.0 kDa, pI=12.25), and MobB (39.0 kDa, pI=IO.66), respectively. The amino acid sequence of RepB showed a 57% identity and 70% similarity with that of the RepA protein of pKJ50. Upstream of the repB gene, the so-called iteron sequence was directly repeated four-and-ahalf times and a conserved dnaA box was identified. An amino acid sequence comparison between the MobB and MobA of pKJ50 revealed a 48% identity and 61 % similarity. A conserved oriT sequence with an inverted repeat identical to that of pKJ50 was also found upstream of the mobB gene. A hydropathy analysis of MembB revealed four possible transmembrane regions. The expressions of the repB and membB genes were confirmed by RT-PCR. The in vitro translation reaction of pKJ36 showed protein bands with anticipated sizes with respect to each putative gene product. S 1 endonuclease treatment and Southern hybridization suggested that pKJ36 replicates by a rolling circle mechanism via a single-stranded DNA (ssDNA) intermediate. A shuttle vector between E. coli and Bifidobacterium sp. was constructed using the pKJ36, pBR322, and staphylococcal chloramphenicol acetyl transferase (CAT) gene. The successful transformation of the Bifidobacterium strains was shown by Southern hybridization and PCR. The transformation efficiency differed from strain to strain and, depending on the electroporation conditions, with a range between $1.2{\times}10^1-2.6{\times}10^2{\;}cfu/\mu\textrm{g}$ DNA.X> DNA.
