Effect of Growth Retardant BX-112 on Growth, Floral Initiation, and Endogenous GA Levels in Sorghum

  • To define the relations between endogenous GA levels and growth and flowering in short-day plant sorghum, growth retardant BX-112 was applied to two sorghum genotypes, wild-type and phytochrome B mutant (phyB-1), which grows faster and flowers earlier than the wild-type. BX-112 and $GA_3$ were applied as a soil drench, and plant height, culm length, and date to floral initiation were investigated. Endogenous GAs contents were measured with GC-MS-SIM. BX-112 treatments inhibited shoot growth in both genotypes and drastically reduced $GA_1$ and $GA_8$ levels. With increasing BX-112 concentrations, $GA_1$ concentrations declined linearly, but caused the accumulation of intermediates from $GA_12$ to $GA_20$. This result implies that $GA_1$ is the major active endogenous GA in shoot elongation in a short day plant sorghum. The inhibition of plant growth in both of wild type and phyB-1 by BX-112 was very similar, while BX-112 effects on floral initiation in two types of plants differed significantly. Floral initiation of phyB-1 was not affected by BX-1l2, but that of wild-type was delayed as BX-1l2 concentration increased. Because BX-112 treatment causes accumulation of biosynthetic intermediates between synthetic pathway from $GA_12$ to $GA_20$ and because phyB-1 is altered in GA metabolism in this same region of the early C13-hydroxylation pathway, BX-112 may fail to block flowering of phyB-1.

Effects of Photoperiod, Light Intensity and Electrical Conductivity on the Growth and Yield of Quinoa (Chenopodium quinoa Willd.) in a Closed-type Plant Factory System

  • Quinoa (Chenopodium quinoa Willd.) is a plant native to the Andean region that has become increasing popular as a food source due to its high nutritional content. This study determined the optimal photoperiod, light intensity, and electrical conductivity (EC) of the nutrient solution for growth and yield of quinoa in a closed-type plant factory system. The photoperiod effects were first analyzed in a growth chamber using three different light cycles, 8/16, 14/10, and 16/8 hours (day/night). Further studies, performed in a closed-type plant factory system, evaluated nutrient solutions with EC (salinity) levels of 1.0, 2.0 or $3.0dS{\cdot}m^{-1}$. These experiments were assayed with two light intensities (120 and $143{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$) under a 12/12 and 14/10 hours (day/night) photoperiod. The plants grown under the 16/8 hours photoperiod did not flower, suggesting that a long-day photoperiod delays flowering and that quinoa is a short-day plant. Under a 12/12 h photoperiod, the best shoot yield (both fresh and dry weights) was observed at an EC of $2.0dS{\cdot}m^{-1}$ and a photosynthetic photon flux density (PPFD) of $120{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$. With a 14/10 h photoperiod, the shoot yield (both fresh and dry weights), plant height, leaf area, and light use efficiency were higher when grown with an EC of $2.0dS{\cdot}m^{-1}$ and a PPFD of $143{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$. Overall, the optimal conditions for producing quinoa as a leafy vegetable, in a closed-type plant factory system, were a 16/8 h (day/night) photoperiod with an EC of $2.0dS{\cdot}m^{-1}$ and a PPFD of $143{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$.

Effect of Long Day Period Before Short Day treatment on the Dry Matter Production and Flower Quality of Greenhouse-Grown Cut Chrysanthemum cv, Reagan Improved (단일처리전 장일처리 기간이 온실재배 스프레이 절화국의 건물생산과 절화품질에 미치는 영향)

  • This study was conducted under the condition of greenhouse to investigate the relationships between the period of long day (LD) treatment before short day treatment, the dry matter production and flower quality of cut chrysanthemum (Indicum group) cv. Reagan Improved at Wagenigen University in the Netherlands. Rooted cuttings of chrysanthemum (Indicum group) cv. Reagan Improved were transplanted on 6th, 13th, loth of September and all of them were treated with short-days (SD) on 21th September. The periods from planting until final harvest were 70, 77, 84 days after being planted for 1, 2, and 3 week-LD period, respectively. The time of flower initiation was similar in all treatments. The number of flowers per plant was greater in the plot of 3 week-LD period than that in the plot of 1 or 2 week-LD period. The fresh weight and dry weight of flowers also increased more in the plot of 3 week-LD period than that in the 1 week-LD period by 4g and 0.4g, respectively. The final dry matter production ($g{\cdot}m$^{-2}$ ) was greater in the treatment of 3 week-LD period than 1 or 2week-LD period. In this study, LD period before short day treatment did not affect the time of flower initiation and flower quality. As a result, 3 week-LD period before short day treatment was strongly recommanded for producing high quality cut flowers of greenhouse-grown chrysanthemum.

  • A new poinsettia cultivar "Early bird" was bred by the National Institute of Horticultural and Herbal Science (NIHHS) in 2007. A cross was made between 'Ichiban', a variety with deep lobed transitional leaves and bright red bracts, and 'Freedom Red', a vigorously growing dark red colored variety in 2004. "Early bird" was finally selected in 2007 after the investigation of the growth and flower characteristics from 2005 to 2007. "Early bird" has bright red colored, wide obovate bracts. Leafblade is dark green and ovate. Plant height is short and stem color is greenish. Response to short day is early and its bracts and transitional leaves are fully colored 6.5 weeks after short day commencement.

Studies on the Consumptive Use of Irrigated Water in Paddy Rice (수도 품종간의 필요수량 차이에 관한 연구)

  • This experiment was conducted to find out the consumptive use of irrigated water for calculation of duty water in paddy rice. Tall statured Japonica rice varieties, Nongbaek (early), Jinheung (medium) and Akibare (late), and short statured Tongil typed varieties, Josaeng Tongil(early), Suweon 264 (medium) and Suweon 258(late) were planted on the experimental farm of Kon-Kuk University in 1979. The results obtained in this study were as follows: 1. During the experimental period, the daily mean temperature was almost similar, the relative humidity was higher as much as 2.8%, the amount of rain fall was 100mm less and the pan evaporation was 70mm less compared with those of 30 years average, respectively. 2. The paddy soil was silty loam, which was suitable for the rice cultivation. 3. Varietal differences were find out for plant height, culm length, number of tillers, number of panicles, heading date, matured grain ratio, 1000-grain weight and rough rice yield. This difference might he the cause of varietal difference of the consumptive use of irrigated water during the rice growing period. 4. The evapotranspiration was gradually increased after transplanting and showed the peak from booting to heading stage of rice varieties. The average evapotranspiration through the whole growing period was 5.67-5. 80mm/day for tall statured Japonica varieties, and 5.99-6. 39mm/day for short statured Tongil typed varieties. 5. The ratio of evapotranspiration to pan-evaporation through the whole growing period was 1.49-1.50 for Japonica varieties, and 1.60-1.66 for Tongil typed varies. 6. Average amount of percolation in paddy field was 3. 62mm/day through the whole growing period of rice plant. 7. K-value in Blaney & Criddle formula was 0.94-0.98 for Japonica varieties and 1.02-1.08 for Tongil typed varieties, and coefficient consumptive water use (Kc-value) was 0.95-1.02 for Japonica varieties and 1.04-1.12 for Tongil typed varieties in this study.   • Flowering time (or heading date) is controlled by intrinsic genetic programs in response to environmental cues, such as photoperiod and temperature. Rice, a facultative short-day (SD) plant, flowers early in SD and late in long-day (LD) conditions. Casein kinases (CKs) generally act as positive regulators in many signaling pathways in plants. In rice, Heading date 6 (Hd6) and Hd16 encode $CK2{\alpha}$ and CKI, respectively, and mainly function to delay flowering time. Additionally, the major LD-dependent floral repressors Hd2/Oryza sativa Pseudo-Response Regulator 37 (OsPRR37;hereafter PRR37) and Ghd7 also confer strong photoperiod sensitivity. In floral induction, Hd16 acts upstream of Ghd7 and CKI interacts with and phosphorylates Ghd7. In addition, Hd6 and Hd16 also act upstream of Hd2. However, whether CKI and $CK2{\alpha}$ directly regulate the function of PRR37 remains unclear. Here, we use in vitro pull-down and in vivo bimolecular fluorescence complementation assays to show that CKI and $CK2{\alpha}$ interact with PRR37. We further use in vitro kinase assays to show that CKI and $CK2{\alpha}$ phosphorylate different regions of PRR37. Our results indicate that direct posttranslational modification of PRR37 mediates the genetic interactions between these two protein kinases and PRR37. The significance of CK-mediated phosphorylation for PRR37 and Ghd7 function is discussed.

Changes in the Growth of Young Rice Seedlings in the Root Extension Stage under Different Growth Conditions (벼 착근기 생육 환경에 따른 어린모의 생육변화 분석)

  • Root extension is the most important growth change that occurs during cultivation. We analyzed growth changes according to young seedling age, temperature, and the degree of root cutting in order to identify factors affecting rooting after transplanting. Root cutting did not affect plant height growth rate, root growth rate was increased in plants that experienced root cutting, and 14-day-old seedlings exhibited a higher growth rate than 7-day-old seedlings. Growth temperature experiments revealed that elongation was high at 25℃ and 28℃, but tended to be low at 18℃, and root elongation was high at 22-28℃ for 7-day-old seedlings and 22-25℃ for 14-day-old seedlings. Nitrogen absorption decreased in the following growth temperature order: 25, 28, 22, 18℃, and differences in nitrogen absorption under different growth temperatures tended to be lower in 7-day-old seedlings. The amount of nitrogen taken up by roots did not differ significantly between the short root treatment and the control, and 7-day-old seedlings tended to start nitrogen absorption faster than 14-day-old seedlings. Root vitality was highest in short-rooted 7-day-old seedlings with 3 cm of root remaining, and vitality also tended to be high in short-rooted 14-day-old seedlings.

Effects of Electric Current on Flowering in Pharbitis and Floral Stimulus activity in the Phloem Exudate of Cotyledons

  • Direct current (DC) applied to cotyledons during a 16 h inductive dark period inhibited the flowering in the short-day plant, Pharbitis nil Choisy cv. Violet. The inhibitory effect of DC was more profound when the current flowed from roots to cotyledons, showing its polarity-dependent action. The second half on the inductive dark period was more sensitive to DC stimulus. The flowering was significantly depressed only when DC stimuli were applied to the translocation path of the floral stimulus from the induced cotyledon to the apex, suggesting that the transport of floral stimulus was damaged by the DC treatment. The vegetative apex culture bioassay system showed that a significant level of the floral stimulus activity existed in the phloem exudate from the cotyledons which would fail to form their own floral buds. These results strongly support the hypothesis that DC partially impede, at least temporarily, the transmission path of the floral stimulus from florally-induced cotyledon to the apex, rather than depressing in situ synthesis of the floral stimulus.

Varietal Differences of the Yield and Growth Characteristics under the Different Temperature and Photoperiod Conditions in Winter Wheat (소맥에 있어서 온도 및 일장변화에 따른 생육 및 수량의 품종간 차이)

  • Eight winter wheat varieties were tested to study the varietal differences on yield and the other characteristics at four different treatments with two different photoperiod and two different temperature levels. In heading date, Changkwang and Parker exhibited the moot sensitive responses to short day length, but Yecora 70. Suweon 169 and Bezostaya were insensitive in general. Culm length was greatly increased at short day length in Bezootaya while Yukseong 3 expressed same responses under low temperature condition. However, little variation of culm length was observed in Yecora 70 regardless the day length. Blueboy, Changkwang and Yecora 70 exhibited the minimal variations in culm length due to the temperature treatment. High yieldings were produced in Blueboy, Suweon 169 and Parker under short day condition, but Parker, Yecora 70. Yukseong 3 and Study produced high yields due to the influences of low temperature. Yield increase was primarily due to the increased number of spikes per plant and number of spikes per plant and 1, 000 grain weight under the low temperature conditions.
