Multiple shoot induction and plant regeneration from axillary buds of Magnolia 'Vulcan'

  • An efficient protocol for multiple shoot induction and plant regeneration from axillary bud culture of Magnolia 'Vulcan' was developed in the present study. Primary shoots were obtained from axillary bud explants cultured on Murashige and Skoog (MS) medium containing 1.0 mg/L 6-benzylaminopurine (BA). To induce multiple shoots effectively, primary shoot tips were cultured on MS medium supplemented with different concentrations of BA and zeatin at 0, 0.2, 0.5, and 1.0 mg/L. Of these treatments, the MS medium with 0.5 mg/L BA resulted in the highest number of shoots per explant with an average value of 5.9, and it produced the greatest shoot height at 4.8 cm after 12 weeks of culturing. In the rooting of in vitro produced shoots, the greatest percentage of explants forming roots (91.3%), number of roots per explant (9.7), and root length (2.8 cm) were obtained in half-strength MS medium supplemented with 6.0 mg/L indole-3-butyric acid (IBA). Regenerated plantlets were successfully acclimatized and hardened off inside the culture room with 87.5% survival rate. Plants were transferred to a greenhouse with a 97.2% survival rate. The highly efficient shoot multiplication and plant regeneration system reported herein can be used for large-scale clonal propagation of valuable Magnolia species or cultivars.

Selection of the Most Sensitive Waveband Reflectance for Normalized Difference Vegetation Index Calculation to Predict Rice Crop Growth and Grain Yield

  • A split-plot designed experiment including four rice varieties and 10 nitrogen levels was conducted in 2003 at the Experimental Farm of Seoul National University, Suwon, Korea. Before heading, hyperspectral canopy reflectance (300-1100nm with 1.55nm step) and nine crop variables such as shoot fresh weight (SFW), leaf area index, leaf dry weight, shoot dry weight, leaf N concentration, shoot N concentration, leaf N density, shoot N density and N nutrition index were measured at 54 and 72 days after transplanting. Grain yield, total number of spikelets, number of filled spikelets and 1000-grain weight were measured at harvest. 14,635 narrow-band NDVIs as combinations of reflectances at wavelength ${\lambda}l\;and\;{\lambda}2$ were correlated to the nine crop variables. One NDVI with the highest correlation coefficient with a given crop variable was selected as the NDVI of the best fit for this crop variable. As expected, models to predict crop variables before heading using the NDVI of the best fit had higher $r^2$ (>10\%)$ than those using common broad- band NDVI red or NDVI green. The models with the narrow-band NDVI of the best fit overcame broad- band NDVI saturation at high LAI values as frequently reported. Models using NDVIs of the best fit at booting showed higher predictive capacity for yield and yield component than models using crop variables.

Introduction, Development, and Characterization of Supernodulating Soybean Mutant -Shoot Factor Regulation of Nodule Development in Supernodulating Soybean Mutant-

  • Nodule development was regulated partially by host plant factors originating in the shoots and roots. This study was performed to identify the origin of the factors regulating nodulation in supernodulating soybean (Glycine max [L.] Merr.) mutant 'SS2-2' which was isolated recently from ethyl methanesulfonate (EMS) mutagenesis of 'Sinpaldalkong 2'. Self- and reciprocal-grafts were made among three soybean genotypes which consisted of two supernodulating mutants, SS2-2 and 'nts 382', and a normal nodulating Sinpaldalkong 2. Self-grafted supernodulating mutants were characterized by greater nodule number, nodule dry weight, and $C_2$H$_2$ reduction activity than self-grafted wild types. They were also characterized by relatively higher nodule to root dry weight. Significant shoot genotypic effects were observed on nodule number, nodule dry weight, and $C_2\;H_2$ reduction activity per plant, whereas varying root genotypes had no effects. From this result, it is surmised that supernodulating characters are controlled by a graft-transmissible shoot factor, and mutant SS2-2 may have similar nodulation mechanism to the former supernodulating nts 382. In all grafts, both supernodulating mutants and Sinpaldalkong 2 maintained the similar balance between above ground and below ground parts regardless of significant differences in partitioning of dry matter into root and nodule between supernodulating mutants and Sinpaldalkong 2.

Allelopathic effects of red pepper (Capsicum annuum L.) and coriander (Coriandrum sativum L.) on early seedling growth of wheat (Triticum aestivum L.)

  • A pot experiment was conducted to assess the effects of red pepper (Capsicum annuum) and coriander (Coriandrum sativum) on seedling growth of wheat (Triticum aestivum). The aqueous extracts treatment of red pepper and coriander showed a significant (p < 0.05) reduction in root, shoot and seedling length, number of leaves and seedling dry weight of wheat (T. aestivum) as compared to control. The inhibitory different effect on growth of wheat (T. aestivum) was directly proportional to the increasing concentration (1, 2, 3, 4 and 5%) of aqueous extracts of red pepper and coriander as compared to control treatment (0%). The root, shoot, seedling length and number of leaves of T. aestivum significantly p < 0.05 decreased at 5% concentration of red pepper as compared to control. The root, shoot and seedling growth of T. aestivum was also significantly reduced at 1, 2, 3, 4 and 5% concentration of coriander as compared to control. The root, shoot and leaves dry weight of T. aestivum at 5% coriander extract treatment concentration decreased as compared to control. The tolerance in seedlings of T. aestivum to red pepper and coriander extract treatment was dose dependent as compared to control. The seedlings of T. aestivum showed low percentage of tolerance to pepper extract treatment than coriander extract treatment.

In Vitro Plant Regeneration from Stolen Node Explant in Eremochloa Ophiuroides (Munro) Hack

  • In vitro shoot regeneration and multiple shoot induction has been obtained from the stolen node explants in Eremochloa ophiuroides (Munro) Hack. The highest number of shoots ($10.66{\pm}0.21$) was observed from initial explants after one month culture duration on Murashige and Skoog (MS) medium containing 6-benzyladenine (BA: 0.5 mg/l). First generation shoot was excised and sub-cultured on the same fresh media for further multiplication of shoots. An enhanced number of second round shoots ($15.33{\pm}0.21$) was obtained compared to the initial culture media containing BA (0.5 mg/l). The number of shoots/stolon node was higher among all the concentrations of BA than kinetin (KN). In vitro regenerated shoots were successfully rooted in the phytohormone free MS medium. Plantlets generated with roots were transferred to pots containing compound mixture of soil and kept in green house conditions. Acclimatized plants showed 100% survival rate with normal morphology in green house conditions. The present study demonstrates the effect of explant and different plant growth regulators towards in vitro response in E. ophiuroides. Moreover, the study reveals the effect of cytokinin on induction of shoot number per stolen node explant in E. ophiuroides.

Four Novel PWM Shoot-Through Control Methods for Impedance Source DC-DC Converters

  • This study proposes four novel pulse width modulation (PWM) shoot-through control methods for impedance source (IS) galvanically isolated DC-DC converters. These methods are derived from a PWM control method with shifted shoot-through introduced by the authors in 2012. In contrast to the baseline solution, where the shoot-through states are generated by the simultaneous conduction of all transistors in the inverter bridge, our new approach is based on the shoot-through generation by one inverter leg. The idea is to increase the number of soft-switched transients and, therefore, decrease the dynamic losses of the front-end inverter. All the proposed approaches are experimentally verified through an insulated-gate bipolar transistor-based IS DC-DC converter. Conclusions are drawn in accordance with the results of the switching loss analysis.

콩 유묘의 조직배양에 의한 식물체 분화 (Plant Regeneration from in vitro Tissue Culture of Soybean Seedling)

  • 콩에서의 조직배양 가능성을 알아보고자 콩 유묘를 기내에서 배양시킨 뒤, 여러 부위를 채취하여 각기 다른 배지 치상한 결과는 다음과 같다. 1. 줄기정단조직 배양에서는 대부분 shoot가 분화되어 완전한 식물체를 얻을 수 있었다. 2. 자엽절 조직배양에서는 multiple shoots가 나타나 대량증식에 유용했으나 배지에 따른 영향이 컸다. 3. 배축 및 상배축 조직배양에서는 callus만 분화되엇다. 4. 엽조직배양에서는 callus 분화 후 계대배양했을 때 shoot가 분화되었으나 완전한 식물체를 얻기는 힘들었다.

무 원종 대량생산을 위한 경정배양 (Shoot-tip culture for massive production of radish foundation seeds)

  • 순도가 균일한 원종을 대량생산하기 위해 원원종 종자를 이용하여 조직배양 기술 중 경정배양을 이용하였다. 실험 재료로는 원원종 계통 RA2와 RA4 계통을 사용하였다. BA 호르몬 사용의 경우 RA2 계통의 유묘에서는 BA 1.33 uM 첨가 배지에서 평균 14.67개로, 성묘의 경우 BA 1.78 uM 첨가 배지에서 평균 11.33개로 가장 많은 개수의 multi-shoots이 형성되었으며, RA4 계통 경우 유묘에서는 BA 2.22 uM 첨가 배지에서 평균 11.67개로, 성묘에서는 BA 1.33 uM 첨가 배지에서 평균 13.67개로 가장 많은 multi-shoots이 형성되었지만 BA 농도별로 유의수준에서 거의 차이가 없는 것으로 나타났다. TDZ 호르몬 사용의 경우 RA2 계통은 TDZ 농도가 증가할수록 multi-shoots의 수가 증가하여 TDZ 0.45 uM 첨가 배지에서 가장 많은 multi-shoots가 형성되었으나(유묘 7.0개, 성묘 3.0개), TDZ 2.25 uM 부터는 거의 multi-shoots가 형성되지 않았다. RA4 계통은 유묘의 경우 거의 multi-shoots이 생성되지 않았고 성묘의 경우 TDZ 0.23, 0.45 uM 첨가 배지에서 3.7개의 multi-shoots이 생성되었으나 그 이상의 농도에서는 유묘에서와 같이 생성되지 않았다. 생성된 multi-shoots를 계대배양하여 생장한 조직배양묘를 SSR marker를 사용하여 원종계통과 비교분석한 결과 생성된 조직배양묘에서 변이의 양상이 약하게 보인 것이 있었으나 돌연변이체로 나타난 것은 없었다. 또한 뿌리 발근에 있어서는 RA2, RA4 계통 모두 대체적으로 IBA 4.9 uM 첨가된 배지에서 발근율, 뿌리 수, 뿌리발달 정도 등이 가장 좋았다.

국화 '백마'의 플러그 육묘시 삽목조건이 삽목묘와 절화 생육에 미치는 영향 (Effects of Cutting Condition on Growth of Rooted Cuttings and Cut Flower in Plug Cutting of Dendranthema grandiflorum 'Baekma')

  • 본 연구는 스탠다드 국화 '백마' 품종의 플러그 묘 생산에 있어서 삽수의 조건이 지상부 생육과 발근 및 정식 후의 절화 생육에 미치는 영향을 알아보고자 실시하였다. 삽수에 잎을 많이 부착할수록 삽목묘의 지상부 생육이 양호하였고, 잎을 4-6장 부착하여 삽목하는 것이 다른 처리에 비해 발근에 효과적이었다. 삽수길이에 따른 삽목묘의 지상부 생육은 큰 차이가 없었으며, 삽수길이를 5-7cm로 처리하는 것이 발근상태가 양호하였다. 삽수 굵기는 3.1mm 처리에 비해 3.6mm와 4.2mm 처리에서 삽목묘의 지상부 생육과 발근이 양호하였다. NAA 처리농도가 높을수록 삽목묘의 지상부 생육이 억제되었으며, 뿌리의 길이도 짧아졌다. NAA $100mg{\cdot}L^{-1}$를 1시간 처리하여 삽목하는 것이 삽목묘의 지상부 생육과 발근에 효과적이었다. 피트모스와 펄라이트를 2:2로 혼합한 배지에서 다른 처리에 비해 뿌리의 길이, 수 및 무게 등이 뿌리의 생육이 양호하였다. 잎이 적은 것보다 4-6개 부착된 삽수의 삽목묘를 정식했을 때, 절화장이 길고, 무게가 무거웠으며, 엽수도 많았다. 삽수길이에 있어서 7-9cm 처리가 절화장, 절화무게, 설상화 수 등 전반적인 절화의 생육에 효과적이었다. 따라서 '백마' 플러그 묘 생산시 7cm 길이에 3.6mm 굵기의 4-6장의 잎을 부착한 삽수를 삽목하는 것이 삽목묘의 지상부 생육과 발근 및 정식 후의 절화 생육을 위해 바람직하였다.

The growth and productivity of native Indonesian rice progenies and its relationship with root development during dry-season

  • One of the problems in increasing rice production is getting lines or varieties that have high adaptability so that able to produce maximum production in a variety of environmental conditions. One strategy that can be done to get adaptive varieties is through the improvement of native varieties. This research was conducted in rain fed paddy field, Aceh province, Indonesia from June to September 2014. The texture of the soil was clay-loam with the soil pH ranged from 6.5-6.7. Five potential progenies of rice from crossing between native Indonesian rice with introduced rice varieties consist of C3, C4, S3, S5, S6 were used in this study. Besides that, one national rice variety Ciherang also used as a comparison. The plants were growth in the plot with the size of $2m{\times}1.4m$ with plant distance was $20cm{\times}20cm$. The fertilizers used in this study were Urea, NPK, and KCl. Randomized block design with 6 rice progenies/variety and 3 replications were used in this study. There were 18 experimental units and each experimental unit had 10 samples for the sources of data. The variables that were observed in this study including plant height at harvesting time, number of productive tiller, the percentage of empty grains and filled grain per panicle, weight of filled grains per hill, weight of filled grain per plot and yield potential per hectare. Analyzed were also conducted for the depth of root penetration, dry-root weight, dry-shoot weight, shoot-root weight ratio and its correlation with the weight of filled grain per hill. The research results show that there was significant difference on plant height at harvesting time, number of productive tillers, the percentage of empty grains and filled grain per panicle, weight of filled grains per hill, weight of filled grain per plot and yield potential per ha (p>0.01) among the treatments. In addition, depth root penetration, dry-root weight, dry-shoot weight, shoot-root weight ratio also had significant difference (p>0.01) among the treatment. The highest plant at harvesting time was found in S6, reaching 129.8 cm and the shortest plants was found in C3 reaching 107.5 cm. The largest number of productive tillers and the highest percentage of filled grains per panicle were found in Ciherang reaching 10.5 tillers and 80.7% respectively. Ciherang also had the heaviest weight of filled grains per hill and per plot reaching 21.1 g and 2.18 kg respectively. Whereas, S6 had the lowest number of tillers and the lowest percentage of filled grain per panicle.. The highest yield potential per ha was found in Ciherang reaching 7.79 tons. Among the progenies, S5 had the highest yield potential reaching 5 tons/Ha. The result also showed significant relationship between shoot-root weight ratio with weight of filled grains per hill. The highest value of shoot-root weight ratio (1.57) in Ciherang is thought had closed relationship with its yield potential.
