We studied the effect of genotype, explant, age of explant, medium (plant growth regulators and gelling agents), and standardized an efficient regeneration protocol for inbred lines of Chinese cabbage (Brassica rapa ssp. pekinensis). Of the different concentrations of NAA and BA tested, the combination of $5\;\cal{mg/L}\;BA\;and\;0.5\;\cal{mg/L}$ NAA gave the highest frequency of shoot regeneration. The cotyledonary explants had more shoot regeneration frequency ($\ge40\%$) than the hypocotyl ex-plants. Besides, the cotyledonary explants, excised from the 4-day old seedlings, showed higher shoot regene-ration ($56.7\%$). Of the three gelling agents and their concentrations used, 16 g/L agar was found to be the best for shoot regeneration. Shoot regeneration frequency in-creased significantly by supplementing the medium with $4\;\cal{mg/L}\;of\;AgNO_3$ MS medium devoid of NAA showed higher frequency of rooting in the regenerated shoots than the ones supplemented with NAA. Our improved regeneration protocol will be especially useful for the genetic transformation of Chinese cabbage inbred lines to develop transgenic hybrids.
This experiment was carried out to establish micropropagation system in Fritillaria thunbergii Miq. Through the culture of bulblet scales, stems, node-buds and shoot tips with special reference to the effect of physiological age of explant and plant growth regulators on bulblet formation. Number of formed bulblets was significantly increased in node-bud or stem tissue compared to scals segments and on the medium supplemented with kinetin than BA containing medium. Optimum levels of kinetin for bulblet formation from node-bud taken from above 3 cm shoot length and stem segments excised from below 3 cm shoot length were 5.0 mg /L and $1.0{\sim}3.0\;mg$ /L kinetin, respectively. Interesting phenomenon was observed, the direct formation of bulblets from the axilliary bud of cultured explants. Bulblet forming capacity in stem tissue was depended on stem age, young stem had high regeneration ability compared to old stem taken from above 10 cm shoot length. 1.0 mg /L kinetin was optimum concentration for the formation of bulblets from old stem segments. Stem tissue taken from underground growing plant was promoted coampare to shoot tips or bulb scale segments. Optimum concentration of sucrose was $5{\sim}7%$. Summariged above results revealed that effective explant for micropropagation was stem and /or node-bud tissue excised from less than 3 cm plant height compared to those of bulb scale segments which showed high contamination after culture. Maximum multiplication rate of young stem and /or node-bud segment was about 20 times. Kinetin requirement for stimulation of bulblet formation from cultured explant depended on source of explants but favorable levels of kinetin for organogenesis ranged from 1.0 mg /L to 5.0 mg /L.

This study was carried out to investigate rooting rate, plagiotrophic growth and root-promoting effect of IBA in Taxus cuspidata S, et Z. cuttings. The results obtained were summarized as follows ; 1. As the ortet ages of cuttings were increased in 5, 10, to 20 years, the rooting rates were decreased in order of 85.7%, 81.7% and 62.4%, and the number of primary roots were also decreased in 5.3, 3.7 and 2.9, respectively. 2. Rooting rate by cutting types of lateral shoot were higher than those of main shoot type, significant at the 1% level. 3. IBA(l00ppm) treatment promoted the rooting rates and the number of primary roots. 4. As the ortet ages were increased in 5, 10 to 20 years, angles of central shoot of rooted cuttings which mesured cyclophysis were decreased in order of $75.9^{\circ}$, $68.5^{\circ}$ and $59.6^{\circ}$, respectively, significant at the 1% level. 5. Average angles showed $77.0^{\circ}$ for main shoot cutting, $65.9^{\circ}$ for upper lateral shoot and $61.7^{\circ}$ for lower lateral, which was significant at 1% level, and lateral shoot cutting showed more topophysis than main shoot cutting. 6. Shoot length of rooted cuttings from main shoot cutting was longer than that from lateral shoot significant at the 1% level. 7. Relationship between some characteristics showed highly positive correlation.

The response of dissolved oxygen (DO) concentrations caused significant change in root number, root length, coleoptile length, shoot length and leaf age of seedlings. The genotypic difference in the effect of DO also highly significant (P<0.01) for all of the seedling traits. The number and length of root were extremely inhibited at the condition of $0.39\pm0.09$ DO concentration. While the coleoptile elongated markedly in the lowest DO concentrations, the shoot did not develop. The root growth was improved slightly at the $1.39\pm0.27mg L^{-1}$, however, there were no difference among genotypes at these two low DO concentrations. As the DO concentration become higher, the growth of root and shoot was improved remarkably. Root number, root length and shoot length was significantly different between $20\;and\;30^{\circ}C$ in DO rich and normal conditions, the development of those traits were apparently accelerated in high water temperature, however those traits of seedlings in DO deficiency were not different between the two temperatures except for shoot length. On the other hand the coleoptile length was not affected by the stagnant water temperature; it was stimulated by the low DO concentration. The competition of DO was greater as the seedling density was increased in the stagnant water, therefore the seedlings grown under high density have long and white coleoptiles, and the growth of roots and shoots was retarded severely.
To evaluate the effects of cultivar, environment, age and cultivation times on ginsenoside content among 8 wild populations of American ginseng (Panax quinquefolium), the concentrations of 6 ginsenosides in root were determined at the time of collection (T0) of plants from the wild and 1 year after (T1) transplanting the roots to each of two different forest garden locations. Both location and population had significant effects on root and shoot growth. Overall, ginsenoside Rb1 was most abundant. The second most abundant ginsenoside were Re and Rg1, however the contents of them were not significantly different from each other. Concentrations of Rg1 and Re were inversely related. Ginsenoside Re was influenced by population and location. Ginsenoside Rg1, Rb1, Rc, Rb2 and Rd were influenced by population, location and age. Ginsenoside levels were consistently lower but growth was consistently higher at the more intensively managed garden location.
Present study describes a method on the application of efficient tissue culture systems for the micro-propagation of juvenile and mature sawtooth oak(Quercus acutissima). Nodal segments with axillary buds were used as initial explant sources. WPM(Woody Plant Medium) was the best in growth and proliferation of shoot among the media tested. Although the single effect of zeatin revealed on two dorminant shoot elongation with normal growth until the elevation of levels up to 3.0mg/l, BAP($N^6$-benzyl amino purine) usually showed better response than zeatin on shoot multiplication and/or elongation. In addition, the incorporation of BAP and zeatin onto the culture media represents more effectiveness in shoot proliferation and its growth. Optimum concentrations of BAP and zeatin were 0.5 and 0.05~1.0mg/l, respectively. Ninety percent of the proliferated shoots was rooted on half-strength GD (Gresshoff and Doy, 1972) medium containing 0.5mg/l IBA(indole butyric acid) in 4 weeks after culture. More than 70% of the rooted plantlets survived after 5 months of transplanting into artificial soil mix containing equal amount of peatmoss and perlite. Among 27 plus tree clones which were grafted twice onto the juvenile rootstocks, only 4 clones revealed the possibility for shoot multiplication through tissue culture system. The capacity for the micropropagation using mature explant sources was highly depended on clonal differences compared with those of octet age. More than 90% of rooting ratio was obtained from the best responding clone. Among the 7 rooting media tested, GD medium was the best far rooting. The most effective rooting was obtained on half-strength GD medium containing 0.2 to 2.0mg/l IBA. More than 60% of rooted plantlets survived after 5 months of transplanting into the artificial soil mix.

This study was conducted to determine the effects of embryo age, growth regulators, low temperature treatment on regeneration, plant growth, and heading in immature embryo culture of Barley. Shoot and root development was more in older than in younger excised embryos, and more in a medium without kinetin than in one with kinetin. The rate of survival was higher on medium without growth regulators and lower on medium with kinetin 5 mg/L because of retarded plant growth. Embryo age and media did not completely replace low temperature treatment. Twenty- and 14 day-old embryos responded by flowering after 4 weeks of vernalization. Embryo culture at 14- and 20 days after anthesis coupled with 4-week cold treatment shortens generation time of barley. When 20 day immature embryos of barley (cv. Olbori) was treated low temperature treatment for 4 weeks, the rate of heading was better than the other treatment.
Leaflets of Allium sativum L. (garlic) and leaf scales Allium cepa L. (onion) were cultured on the Murashige and Skoog(MS) medium to study the effects of plant growth regulators, temperature and light on the formation of shoot and root. And also, the potentiality of shoot or root formation of tissues in accordance with the leaf age and the region was discussed. In the experiment with garlic, the formation of shoot and root was more effective on the medium containing $BA\;2mg/{\ell}$ and $NAA\;1mg/{\ell}$ and it occured on the basal region of outer leaflets. The shoot formation was more effective at $20^{\circ}C$ than $28^{\circ}C$ under 16-hour daylength, and it was more effective in darkness than light condition at $28^{\circ}C$. The results of shoot and root formation on the new cloves of garlic harvested in 1982 were similar to those the old cloves of garlic harvested in 1981. In the experiment with leaf seales of onion, when the tissues of dormant leaf scales of onions harvested in 1981 were explanted on the MS medium, a few shoots were formed on the medium containing $BA\;2mg/{\ell}$ and $NAA\;1mg/{\ell}$. In this case, there were no differences on the shoot formation among the age and region of leaflets. On the culture of leaf scales, the shoot was not formed on the leaf scales located at outer or middle region, but it was formed on the medium containing BA accompanied with NAA at inner scales. On the medium complemented with BA alone, the tissues were not differentiated, but on the medium complemented with NAA, the callus was formed from tissues and then the root was formed through the callus.

In this study, basic data were obtained to determine the optimal cultivation method to achieve stable fruiting and yield increase in "Cheonhwang" jujube trees. Accordingly, thefructification, fruit characteristics, and yield according to tree age were analyzed. The mean (and range of) tree height, crown area, stem diameter near the root, clear stem length, number of main branches, and distance between main branches were 235.6 (217.4-253.8) cm, 3.5 (3.1-4.1) m2, 5.5 (4.0-7.1) cm, 70.6 (66.2-72.7) cm, 9.7 (8.6-10.5), and 10.4 (7.9-14.2) cm, respectively. Correlationanalysis results indicated that tree age was positively correlated with crown area, stem diameter near the root, and clear stem length but not with the number of main branches. The mean number (and range) of fruit per fruit-bearing mother shoot and tree were 18.3 (16.7-18.3) and 170.7 (157.9-178.3), respectively. Tree age was not significantlycorrelated with fruiting characteristics (i.e., the numbers of fruit-bearing mother shoots per main branch, fruit-bearing shoots per fruit-bearing mother shoot, fruit per fruit-bearing shoot, and fruit per tree). Given that the shape of jujube trees is constantly managed according to the growing area and greenhouse type, the tree growth characteristics were more affected by tree management techniques than by tree age. The mean (and range of) fruit weight, fruit hardness, and soluble solid content were 28.6 (27.7-30.3) g, 29.4 (28.5-30.4) N, and 20.4 (19.3-21.0) °brix, respectively, and these fruit characteristics were not significantly correlated with tree age.The average yield per tree of the 'Cheonhwang' jujube cultivar was 4.9 (4.8-5.0) kg, which was not significantly correlated with tree age.

A breeding program in progress at Suweon Livestock Experiment Station, RDA, involves the hybridization of italian ryegrass (Lm) and several leading forage grasses in an effort to combine the nutritive, productivity and palatability qualities of Lm with the adaptive and cold resistance qual; .les of several leading grasses. In order to study the fate of the hybridization between remotely related species, immatured hybrid embryos were cultured on media. The emasculated plants of Lm were Sikem and Tetrone. Reweille, 2n=14, of perennial ryegrass (Lp.), Forager, 2n=42, of tall fescue(Fa), First, 2n=14,of meadow fescue(Fp), Potomac, 2n=28, of orchardgrass(Dg), and Richmond, 2n=42, of timothy(Ph.p) were used as pollinators. Embryos were isolated on 4, 8, 12, 16, and 20 days after pollination and cultured them on modified Murashige and Skoog media. Calluses and plantlets have been obtained after 8 days old embryos crossing between $Lm{\times}Fa$, after 12 days embryos crossing between $Lm{\times}Lp$, Dg, and Ph.p, and after 16 days embryos crossing between $Lm{\times}Fp$. Both callus and shoot formation occurred on 6 % or less of the plated embryos of $Lm{\times}Fp$, Dg, and Ph.p. Embryoderived callus forming shoots have been obtained from 4 days old embryo crossing between $Lm{\times}Dg$ which has not successfully been done anywhere as far as we know. It means that hybrid plants of species without crossability can be obtained through the use of immatured embryo culture. Some of plated embryos developed directly shoots from embryos and the others shoot-forming callus. Cross between related species showed a high frequency of directly shoot formation from plated embryos and cross between remotely related species a high frequency of callus formation.
