Anticancer effects of D-pinitol in human oral squamous carcinoma cells

  • D-pinitol is an analog of 3-methoxy-D-chiro-inositol found in beans and plants. D-pinitol has anti-inflammatory, antidiabetic, and anticancer effects. Additionally, D-pinitol induces apoptosis and inhibits metastasis in breast and prostate cancers. However, to date, no study has investigated the anticancer effects of D-pinitol in oral cancer. Therefore, in this study, whether the anticancer effects of D-pinitol induce apoptosis, inhibit the epithelial-to-mesenchymal transition (EMT), and arrest cell cycle was investigated in squamous epithelial cells. D-pinitol decreased the survival and cell proliferation rates of CAL-27 and Ca9-22 oral squamous carcinoma cells in a concentration- and time-dependent manner. Evidence of apoptosis, including nuclear condensation, poly (ADP-ribose) polymerase, and caspase-3 fragmentation, was also observed. D-pinitol inhibited the migration and invasion of both cell lines. In terms of EMT-related proteins, E-cadherin was increased, whereas N-cadherin, Snail, and Slug were decreased. D-pinitol also decreased the expression of cyclin D1, a protein involved in the cell cycle, but increased the expression of p21, a cyclin-dependent kinase inhibitor. Hence, D-pinitol induces apoptosis and cell cycle arrest in CAL-27 and Ca9-22 cells, demonstrating an anticancer effect by decreasing the EMT.

  • This paper suggests study of basic structure design and structural analysis for the twin car-ferries. The rules and methodology for the analysis of strength of medium and small high speed vessels with a length of more than 50m and a width / ratio of more than 12, such as car-ferries, have not been clarified yet. Therefore, in this paper, the scantling of the members is based on the Korea Classification standards, and the car-ferries standards were additionally applied to verify the structural strength of the design. The results of this study are expected to be useful as basic data related to structural design and structural analysis of high speed twin car-ferries.

  • Until now, 53 species belonging to 22 genera of 6 subfamily of family Histeridae have been recorded in Korea. As a result of this study, we newly add two subfamily, three genera, and three species, Niponius osorioceps Lewis, 1885, Plegaderus (Plegaderus) marseuli Reitter, 1877, and Trypeticus fagi (Lewis, 1884), to Korea along with a brief description and illustration of the diagnostic characters.

  • Researchers indicates several issues as to performance evaluation methods for public health centers. Firstly, health centers are passively involved in the evaluation process, the performance indicators are activity-oriented, and mostly unrealistic. Balanced Scorecard is one of the methods for evaluating organizational performance, which is utilized at current in many industries including public sector. The purposes of this study is to apply balanced scorecard approach to a public health center and to develop performance indicators as well as their vision and strategies. For developing performance indicators, researchers selected K City Public Health Center and implemented brainstorming with members of health promotion team. Through team process they suggested goals, strategies and several indicators they considered proper to accomplish their shared vision. And then appropriateness of the indicators were evaluated by professional researchers in health promotion field for consensus building by email questionnaire. Based on survey and professional consensus meeting, 11 performance indicators were tailored in four perspectives as well as 6 strategies and 10 strategic goals, which are steps for accomplishing shared vision of health promotion team. For details, refer to the paper. Most members of health promotion team were satisfied with the indicators. However issues such as low level of recognition and familiarity with a new concept of BSC, and cultural resistance to strategic approach in public organizations should be addressed for future application and dissemination of BSC technique to public health organizations.

A Study on the Pulmonary Hemodynamic Changes before and after PEEP in ARDS (호흡부전증에세 호기말 양압호흡법 치료 전후의 폐혈류역학적 변화에 관한 연구)

  • The purpose of this study is to observe the changes of pulmonary capillary permeability and various hemodynamic parameters before and after Positive End-Expiratory Pressure(PEEP) in Adult Respiratory Distress Syndrome (ARDS). Using a canine oleic acid induced ARDS model, we measured the pulmonary capillary permeability with a slope of lung: heart radioactivity ratio, hemodynamic parameters with Swan-Ganz catheter and blood gas tensions. 1) In normal and ARDS dogs, the PEEP didn't significantly influence the slope of lung: heart radioactivity ratio. But in ARDS group the slope index was increased compaired with that of control group (p<0.05). 2) Also in ARDS group, $PaO_2$ was significantly decreased, and $PaCO_2,\;PvCO_2$, MPAP, $AaDO_2$, Qs/Qt were significantly increased compared with those of control group (p<0.05). 3) In normal dogs, the PEEP didn't influence blood pH or gas tension, $AaDO_2$, Qs/Qt, or hemodynamics. 4) In ARDS dogs, however, the PEEP significantly increased $PaO_2$ and decreased $AaDO_2$, Qs/Qt (p<0.05).

Saccharomyces cerevisiae KNU5377 with Multiple Stress Tolerance and its Potential as a Worldwide On-site Industrial Strain for Alcohol Fermentation

  • Saccharomyces cerevisiae KNU5377 was examined to assay the recovering capacity against heat and other stressors. Along with a particular fermentation ability that is able to produce ethanol even at high temperature such as $40^{\circ}C$ with a comparable rate to the fermentation at $33^{\circ}C$, this strain also exhibited higher viability than a reference strain owing to its own thermotolerance that conferred the survival after the severe heat shock at $60^{\circ}C$ for 30 minutes. Furthermore, this strain showed outstanding tolerances against $H_2O_2$, ethanol and some chemical compounds. But, especially due to the thermotolerance, this strain has been suspected of other species of yeast. However, ITS (internally transcribed spacer) 1 and 2 sequencing data confirmed this strain was a typical strain of S. cerevisiae. The outstanding tolerances to various environmental stressors Indicate this S. cerevisiae KNU5377 is enough to use both as an on-site potential strain for world-wide alcohol fermentation industry and as a model strain for researches into the routes to acquire the tolerance to various stressors.

Development of Semi-Solid Apple Baby Food using High Pressure Processing and Quality Evaluation (초고압을 이용한 반고형 사과 이유식 개발 및 품질평가)

  • For the purpose of develop semi-solid baby food, we investigated ingredients, processing aids as a thickener, processing suitability test, natural antibrowning agent, packing method, novel preservation technology, storage test and quality evaluation. In results, semi-solid baby food was developed. It's formulation is eco-friendly apple as a main ingredients, natural rhubarb extract 5% and thickener 4%. Thickener was used brown rice puffing powder and sweet pumpkin puffing powder. High pressure processing(HPP) and pouch packaging was used as processing and novel preservation technology. In results of storage test and quality evaluation, shelf-life of semi-solid baby food was 15 days in $5^{\circ}C$ cold storage and hedonic score of sensory evaluation was 4.3/5.0. Taste of them was fresh and sweet.

  • Homocysteine is a sulfur amino-acid produced during the metabolism of the essential amino acid methionine. Moderately increased plasma total homocysteine concentration have been implicated as a risk factor for occlusive vascular disease. Smoking is known to be one of the most significant factors leading to elevated plasma homocysteine concentration. However, the main component of a cigarette, nicotine has been not studied whether it is linked directly to the increase of homocysteine concentration in blood. The metabolism of homocysteine is closely linked to that of its cofactors, folate. Here, the effects of nicotine and folic acid on amount of plasma homocysteine were studied. The concentration of homocysteine was increased by about 70% in rat plasma after nicotine treatment for one month. This increased concentration of homocysteine was reduced by about 60% at 6 hours later after folate treatment. Thus, nicotine should be directly involved in increasing the concentration of plasma homocysteine. Also it is suggested that these results can be and applied and used for controlling withdrawal symptoms after stopping smoking as one of oriental medicine formulas.

The Current Incidence of Viral Disease in Korean Sweet Potatoes and Development of Multiplex RT-PCR Assays for Simultaneous Detection of Eight Sweet Potato Viruses

  • Sweet potato is grown extensively from tropical to temperate regions and is an important food crop worldwide. In this study, we established detection methods for 17 major sweet potato viruses using single and multiplex RT-PCR assays. To investigate the current incidence of viral diseases, we collected 154 samples of various sweet potato cultivars showing virus-like symptoms from 40 fields in 10 Korean regions, and analyzed them by RT-PCR using specific primers for each of the 17 viruses. Of the 17 possible viruses, we detected eight in our samples. Sweet potato feathery mottle virus (SPFMV) and sweet potato virus C (SPVC) were most commonly detected, infecting approximately 87% and 85% of samples, respectively. Furthermore, Sweet potato symptomless virus 1 (SPSMV-1), Sweet potato virus G (SPVG), Sweet potato leaf curl virus (SPLCV), Sweet potato virus 2 ( SPV2), Sweet potato chlorotic fleck virus (SPCFV), and Sweet potato latent virus (SPLV) were detected in 67%, 58%, 47%, 41%, 31%, and 20% of samples, respectively. This study presents the first documented occurrence of four viruses (SPVC, SPV2, SPCFV, and SPSMV-1) in Korea. Based on the results of our survey, we developed multiplex RT-PCR assays for simple and simultaneous detection of the eight sweet potato viruses we recorded.

  • The main theme of this research is a case of e-Business implementation in part manufacturing industry, especially in a PCB manufacturing company. The characteristics of part manufacturing industry are as follows. First, an ERP system runs as a legacy system that is ready to be combined with e-Business system. Secondly, the number of customers is very small. The customers are not many individuals but only a few big electronic enterprises that are strategically affiliated with the part manufacturing company. This means that the e-Business of the part manufacturing industry needs to focus on sharing pertinent information throughout the transactions with the customers, not on data-warehousing or data-mining customers' potential needs or requests. In this paper, we extracted e-Business opportunity domains from a PCB manufacturing company, a typical part manufacturing industry. We are intended to enhance information sharing between customers and the company, and provide functions of transactions necessary in the whole value chain from order to shipment. Implementing the e-Business system on the Web can increase the visibility of customers, and further, the company can be transformed into an extended enterprise where the relationship with the customers becomes very close and interleaved. Also, the Cyber Office functionality of the e-Business system can support the salespersons effectively, so that they can spend more time on customer satisfaction. Such efforts, in the future, can be a basis for active adaptation to the industry transformations such as forming e-community and participating in the marketplace.
