In the environment, chromium often exists in a highly mobile and toxic form of Cr(VI). Therefore, the reduction of Cr(VI) to less toxic Cr(III) is considered an effective remediation strategy for Cr(VI)-contamination. In this study, the biological reduction of hexavalent chromium was examined at the concentrations of 0.01 mM, 0.1 mM, and 1 mM Cr(VI) by the dissimilatory metal-reducing bacterium, Shewanella sp. HN-41 in the presence of ferric-citrate. With the relatively condensed cell densities, the aqueous phase Cr(VI) was reduced at the proportions of 42%, 23%, and 31%, respectively for the 0.01 mM, 0.1 mM, and 1 mM Cr(VI) incubations, while Fe(III)-citrate was reduced at 95%, 88%, and 73%, respectively. Although the strain HN-41 was not considered to reduce Cr(VI) as the sole electron acceptor for anaerobic metabolism in the preliminary experiment, it has been presumed that outer-membrane c-type cytochromes such as MtrC and OmcA reduced Cr(VI) in the presence of ferric-citrate as the electron acceptor. Since this study indicated the potential of relatively high cell density for Cr(VI) reduction, it might propose a bioremediation strategy for Cr(VI) removal from contaminated waters using engineered systems such as bioreactors employing high cell growths.
2022년 7월, 국내의 저염분 해수 흰다리새우 양식장에서 양식 중이던 흰다리새우가 이상유영과 식욕부진을 보이며 지속적으로 폐사하였다. 일일 평균 폐사율이 0.1%로 완만한 폐사를 보였으나, 2달 넘게 폐사가 지속되었고 성장 지체로 인한 경제적인 피해가 가중되었다. 질병 진단 과정 중, 폐사 새우의 간췌장에서 세균(KNUAF-SHP3)이 분리되었다. 본 세균의 16S rRNA 유전자의 시퀀스 분석 결과, 분리주 KNUAF-SHP3은 Shewanella에 속하는 균으로 동정이 되었고, S. algae MARS 14, S. chilikensis JC5T와 근연 관계를 보이며 같은 그룹으로 분류되었다. 실험 동물을 활용한 인위 감염 실험에서는 상대적으로 고농도(2.1×108CFU/ml와 2.1×109CFU/ml)를 적용한 군에서는 모든 새우들이 감염 증상을 보이며 감염 후 7일 내에 100% 폐사율을 나타내었다. 본 연구는 Shewanella 세균이 저염분 해수 양식 흰다리새우에서의 잠재적인 기회 감염균이 될 수 있음을 기술하고 있다.
The bacterial communities in the intestinal tracts of earthworm were investigated by culture-dependent and -independent approaches. In total, 72 and 55 pure cultures were isolated from the intestinal tracts of earthworms under aerobic and anaerobic conditions, respectively. Aerobic bacteria were classified as Aeromonas (40%), Bacillus (37%), Photobacterium (10%), Pseudomonas (7%), and Shewanella (6%). Anaerobic bacteria were classified as Aeromonas (52%), Bacillus (27%), Shewanella (12%), Paenibacillus (5%), Clostridium (2%), and Cellulosimicrobium (2%). The dominant microorganisms were Aeromonas and Bacillus species under both aerobic and anaerobic conditions. In all, 39 DNA fragments were identified by polymerase chain reaction-denaturing gradient gel electrophoresis (PCR-DGGE) analysis. Aeromonas sp. was the dominant microorganism in feeds, intestinal tracts, and casts of earthworms. The DGGE band intensity of Aeromonas from feeds, intestinal tracts, and casts of earthworms was 12.8%, 14.7%, and 15.1%, respectively. The other strains identified were Bacillus, Clostridium, Enterobacter, Photobacterium, Pseudomonas, Shewanella, Streptomyces, uncultured Chloroflexi bacterium, and uncultured bacterium. These results suggest that PCR-DGGE analysis was more efficient than the culturedependent approach for the investigation of bacterial diversity and the identification of unculturable microorganisms.
Histamine은 적색육 어류의 histidine이 어육 중의 Morganella morganii, Hafnia alvei 및 Klebsiella pneumoniae와 같은 부패세균에 의해 탈탄산 되어 초기에 형성되는 것으로 allergy성 식중독을 일으킬 수 있다. 이는 적색육 어류인 고등어의 선도저하 시에 많이 생성된다. 그리고 부패 후기에는 histamine을 분해하는 세균도 존재하는 것으로 알려져 있다. 그러므로, histamine 식중독의 잠재력을 지닌 자반고등어로 인한 식중독 사고 예방과 그 위생 대책을 수립하는데 필요한 자료를 얻고자 자반고등어에서 histamine 분해능을 가진 균을 분리, 동정하였다. 시료는 대형마트에서 시판되는 상태로 구입하였다. 질소원과 탄소원으로써 histamine만을 첨가한 제한배지를 사용하여 histamine 분해능을 가진 균을 분리하였다. 그리고 Cram staining, oxidase, catalase, citrate, TSI test, $H_{2}S$ reaction 및 indole 생성 등의 기본적인 생화학적 동정시험을 거쳐 10종의 시험균주를 선택하였다. 이 균주들을 16SrRNA gene 염기서열 비교에 의한 계통발생학적 분석을 이용하여 동정 하였다. 그 결과, Pseudomonas putida strain RA2, Halomonas marina, Uncultured Arctic sea ice bacterium clone ARKXV1/2-136, Halomonas venusta, Psychrobacter sp. HS5323, Pseudemonas putida KT2440, Rhodococcus erythropolis, Klebsiella terrigena (Raoultella terrigena), Alteromonadaceae bacterium T1, Shewanella massilia의 10종이 모두 동정 되 었으며, 각각 $100\%,{\;}100\%,{\;}99\%,{\;}99\%,{\;}99\%,{\;}99\%,{\;}100\%,{\;}95\%,{\;}99\%,{\;}100\%$의 상동성을 보였다. Histamine분해능의 존재를 탁도측정법과 효소법에 의해 확인한 결과, 분리된 10종 모두의 histamine 분해능이 재확인 되었고, 그 중 Shewanella massilia가 최대의 histamine 분해능을 보이는 것으로 확인되었다. 이 결과로 자반고등어 시판 제품에는 다수의 histamine 분해 세균이 존재하는 것을 확인할 수 있었으며, 이 세균을 활용한다면 식품 내 존재하는 histamine을 효과적으로 분해할 수 있을 것이라 예상된다.
Frog culture industry is not yet familiar but has much potential. Generally, in farm, the population density is higher than that of in nature and frog farm is not the exception. But when population density is high, it can easily leads to stressful condition, poor sanitation. When a disease occur, it is a primary factor that makes the population more susceptible and the results more grave. Because of severe Rhabditoidea- helminth infection and subsequent bacterial septicemia, 50~70% of the total population had been died in a farm in Jeong-sun in Gangwon-do and Chungju in Chungcheongbuk-do from late June, 2012 to September, 2012. Diseased frogs showed ruptured lung, bloody ascites, liver discoloration, myocardium weakness, congested kidney, microcytic anemia and so on. Enterobacteriacea, Citrobacter.sp, Cupriavidus metallidurans, Acinetobacter.sp were isolated as major bacterium that had caused septicemia in frogs. Among isolated bacterium, Cupriavidus metallidurans, Ewingella americana, Shewanella aquimarina and Pseudoalteromonas sp. have not reported as potential pathogens in frogs before. It is a good example that severe helminth infection in frogs can lead to secondary infection of bacteria.
Shewanella oneidensis MR-1 has the ability to inhale certain metals and chemical compounds and exhale these materials in an altered state; as a result, this microorganism has been widely applied in bioremediation protocols. However, the relevant characteristics of cell growth and biosynthesis of PuFAs have yet to be thoroughly investigated. Therefore, in this study, we have attempted to characterize the growth and fatty acid profiles of S. oneidensis MR-1 under a variety of temperature conditions. The fastest growth of S. oneidensis MR-1 was observed at $30^{\circ}C$, with a specific growth rate and doubling time of $0.6885h^{-1}\;and\;1.007 h$. The maximum cell mass of this microorganism was elicited at a temperature of $4^{\circ}C$. The eicosapentaenoic acid (EPA) synthesis of S. oneidensis MR-1 was evaluated under these different culture temperatures. S. oneidensis MR-1 was found not to synthesize EPA at temperatures in excess of $30^{\circ}C$, but was shown to synthesize EPA at temperatures below $30^{\circ}C$. The EPA content was found to increase with decreases in temperature. We then evaluated the EPA biosynthetic pathway, using a phylogenetic tree predicted on 16s rRNA sequences, and the homology of ORFs between S. oneidensis MR-1 and Shewanella putrefaciens SCRC-2738, which is known to harbor a polyketide synthase (PKS)-like module. The phylogenetic tree revealed that MR-1 was very closely related to both Moritella sp., which is known to synthesize DHA via a PKS-like pathway, and S. putrefaciens, which has been reported to synthesize EPA via an identical pathway. The homology between the PKS-like module of S. putrefaciens SCRC-2738 and the entire genome of S. oneidensis MR-1 was also analyzed, in order to mine the genes associated with the PKS-like pathway in S. oneidensis MR-1. A putative PKS-like module for EPA biosynthesis was verified by this analysis, and was also corroborated by the experimental finding that S. oneidensis MR-1 was able to synthesize EPA without the expression of $dihomo-{\gamma}-linoleic$ acid (DGLA) and arachidonic acid (AA) formed during EPA synthesis via the FAS pathway.
Although, microbial arsenic mobilization by dissimilatory arsenate-reducing bacteria (DARB) and the practical use to the removal technology of arsenic from contaminated soil are expected, most previous research mainly has been focused on the geochemical circulation of arsenic. Therefore, in this review we summarized the previously reported DARB to grasp the characteristic for bioremediation of arsenic. Evidence of microbial growth on arsenate is presented based on isolate analyses, after which a summary of the physiology of the following arsenate-respiring bacteria is provided: Chrysiogenes arsenatis strain BAL-$1^T$, Sulfurospirillum barnesii, Desulfotomaculum strain Ben-RB, Desulfotomaculum auripigmentum strains OREX-4, GFAJ-1, Bacillus sp., Desulfitobacterium hafniense DCB-$2^T$, strain SES-3, Citrobacter sp. (TSA-1 and NC-1), Sulfurospirillum arsenophilum sp. nov., Shewanella sp., Chrysiogenes arsenatis BAL-$1^T$, Deferribacter desulfuricans. Among the DARB, Citrobacter sp. NC-1 is superior to other dissimilatory arsenate-reducing bacteria with respect to arsenate reduction, particularly at high concentrations as high as 60 mM. A gram-negative anaerobic bacterium, Citrobacter sp. NC-1, which was isolated from arsenic contaminated soil, can grow on glucose as an electron donor and arsenate as an electron acceptor. Strain NC-1 rapidly reduced arsenate at 5 mM to arsenite with concomitant cell growth, indicating that arsenate can act as the terminal electron acceptor for anaerobic respiration (dissimilatory arsenate reduction). To characterize the reductase systems in strain NC-1, arsenate and nitrate reduction activities were investigated with washed-cell suspensions and crude cell extracts from cells grown on arsenate or nitrate. These reductase activities were induced individually by the two electron acceptors. Tungstate, which is a typical inhibitory antagonist of molybdenum containing dissimilatory reductases, strongly inhibited the reduction of arsenate and nitrate in anaerobic growth cultures. These results suggest that strain NC-1 catalyzes the reduction of arsenate and nitrate by distinct terminal reductases containing a molybdenum cofactor. This may be advantageous during bioremediation processes where both contaminants are present. Moreover, a brief explanation of arsenic extraction from a model soil artificially contaminated with As (V) using a novel DARB (Citrobacter sp. NC-1) is given in this article. We conclude with a discussion of the importance of microbial arsenate reduction in the environment. The successful application and use of DARB should facilitate the effective bioremediation of arsenic contaminated sites.
새만금 간척사업이 진행중인 만경, 동진강 하구의 조간대 갯벌에 대한 중금속 분포 및 거동 특성을 조사한 결과를 요약하면 다음과 같다. 표층퇴적물의 중금속 함량은 1994년 관측자료보다 전체적으로 높은 농도를 나타내었고, 시화호 퇴적물보다는 대체적으로 낮은 농도수준을 나타내었다. 퇴적물의 중금속 농도는 시화호 보다 Cu 1/4배, Zn 1/3배, Cr 1/2배 이하의 낮은 값이었다. 농축계수는 Pb와 Zn을 제외하고는 1 이하의 낮은 값을 나타내었고, 시화호에서 나타난 결과와 유사하였다. 중금속 과잉량에서는 Cu, Cr, Fe, Mn은 부족하였으며, Pb와 Zn은 농축되어 있는 것으로 나타났다. 중금속과 유기물량과의 관계에서 유기물 농도와 중금속간에는 높은 정의 상관성을 나타내고 있는데, 강열감량과의 상관분석에서 모든 중금속이 좋은 상관성 (r=0.424$\~$0.839)을 보였고, 특히 산화 Fe, Mn, Ai은 0.7 이상의 높은 정의 상관계수를 나타내었다. 중금속과 입도와의 상관조사에서 입도가 세립할수록 중금속의 함량이 증가하였으며, 실트와의 상관분석에서 Cu와 Pb를 제외한 다른 중금속은 0.552$\~$0.732의 정의 높은 상관계수를 나타내고 있었다. 새만금 하구퇴적물의 중금속 오염정도는 시화호 퇴적물과 비교하여 많이 낮았는데, 이는 시화호의 경우 연안에 위치한 공단으로부터 배출되는 폐수의 영향을 직접적으로 받고 있는 반면, 만경강과 동진강하구 주변에는 산업단지가 조성되어 있지 않으며, 또한 생활하수를 배출하는 전주와 익산도 하구와는 상대적으로 떨어진 곳에 위치하고 있기 때문으로 판단된다.비하여 포화지방산의 비율은 감소하였으나 불포화지방산의 비율은 증가하였으며, 그 변동율은 C. calcitrans의 경우가 S. costatum보다 높았다. 그러나 V. alginolyticus와 혼합배양한 경우 C. calcitrans는 포화지방산이 증가한 반면 불포화지방산은 감소하는 경향을 나타내었고, S. costatum은 순수 배양한 조체와 큰 차이를 보이지 않았다. 그래서 C. calcitrans는 Shewanella sp.
