Identification of Phellinus linteus by Morphological Characteristics and Molecular Analysis (형태적.분자생물학적 방법에 의한 Phellinus linteus의 동정에 관한 연구)

  • The context and upper surface of Phellinus basidiocarp become blackened, rimose and woody. The basidiocarp is sessile, dimidiate and elongate. The basidiospores are pigmented and ovoid to globose. Hymenial setae are $17{\sim}35{\times}6{\sim}8{\mu}m$. Nineteen isolates of Phellinus species, including Phellinus linteus, were used for sequencing of the internal transcribed spacer (ITS) region of the nuclear rDNA. Based on these sequence data, specific primers were designed for identification of Phellinus linteus isolates in Korea. The specific primers were within the ITS1 and ITS2 regions and were nested within the universal primers flanking the spacer regions. A total of four primers (the universal primers ITS-1F and ITS-4, and the specific primers PL-F and PL-R) were used for detection of Phellinus linteus collected in Korea. The length of the four amplification products of Phellinus linteus DNA were 800 bp (ITS-1F/ITS-4), two bands of about 720 bp (ITS-1F/PL-R and PL-F/ITS-4), and 610 bp (PL-F/PL-R). Among 23 isolates of Phellinus species collected in Korea, Thirteen isolates were identified as Phellinus linteus based on the presence of the four bands. The other species produced only the single ITS-1F/ITS-4 product.

An Etiological Study on the Anthracnose Fungus of Pepper Caused by Colletotrichum dematium in Korea (고추에서 분리(分離)한 탄저병균(炭疽病菌) Colletotrichum dematium에 대한 병원학적(病原學的) 연구(硏究))

  • In order to investigate an etiological characteristics of the pepper anthracnose fungus infected with Collectotrichum dematium, this experiment was undertaken. Isolation percent of the fungus was 55.3% from the seed sample of Jewon and 0.3% from that of Eumsung Kun, according to seed health blotter method. Although the acervulus was similar to the known two Colletotricum species, C. acutatum and C. gloeosporioides, setae was not only straightly extended above the acervulus, but also shaped as falcate conidia ranging $12{\sim}31.2{\times}2.4{\sim}4.8{\mu}m$. Acervului of the fungus were evenly scattered on the surface of pepper seed coat, and caused seedling blight after seed germination. Pre-and post-emergence seedling blight as well as foliar lesion and fruit rot was confirmed by inoculating conidial suspension. No typical anthracnose sysmptom was obtained from soybean seedlings, welsch onion and spinach seedlings upon inoculation. Therefore, this fungus is said to the undescribed form species of the fungus of pepper in Korea (Colletotricum dematium f. sp. capsicum).

  • The first zoeal stage of Palaemon ortmanni (Rathbun, 1902) is re-described and illustrated in detail from laboratory-hatched material. The first zoeas of Palaemon are more closely related to those of Palaemonetes than to Macrobrachium, Leptocarpus, Leander, Leanderites, and Brachycarpus by having the endopod of maxillule with terminal seta and the endopod of maxilla with 2+1 setae. A provisional key to the zoeas between three genera Palaemon, Macrobrachium, and Periclimenes known from Korean waters is included.

  • This study has been conducted to investigate the molting behavior and the effects of rearing temperature and day length on molting in the behavior and the effects of rearing temperature and day length on molting in the giant freshwater prawn, Macrobrachium rosenbergii reared in the laboratory. The results obtained were summarized as follow: 1. After pre-spawning molting, the protopodites of 1st, 2nd, 3rd, and 4th except 5th pleopod bore new breeding setae which conserve eggs in the brooding chamber and the basis of 3rd, 4th, and 5th pereiopods bore new breeding dresses which transport the ovulated eggs into the brooding chamber. 2. Adult females reared in 27.5-$29.4^{\circ}C$ molted 10-12 times per year at interval of 27-35 days, of which four or six moltings were common molting for growth and another four or five moltings were pre-spawning molting for spwaning and brooding. In winter season, pre-spawning molting did not happen to most of adult females in spite of the same temperature. 3. Duration of intermolt cycle was 31-38 days and 26-30 days at 25.3- $26.5^{\circ}C$ and 28.7- $30.4^{\circ}C$ of rearing temperature, respectively.

  • Tenuidraconema philippinensis, a new species of free-living marine nematode, collected from the shallow subtidal coarse sediments of the Philippines, is described. The new species differs from T. fiersi Decraemer, 1989 and T. koreensis Rho and Kim, 2004, by the following characteristics: the position of 12 cephalic adhesion tubes in both sexes (all 12 cephalic adhesion tubes inserted on the body annules), the number of posterior sublateral adhesion tubes (12 in male and 11 in female) and posterior subventral adhesion tubes (16 in male and 17 in female), and the absence of the intermingled somatic setae in male. This is the first discovery of the genus Tenuidraconema in the Philippines.

  • Korean nannastacids have recorded only eight species belonging to genera Campylaspis and Cumella. In this paper, Nannastacus nyctagineus Gamȏ, 1962, is redescribed and illustrated as a new species of Korean fauna in this study for the first time. Korean female specimens of Nannastacus nyctagineus correspond well with the original description by Gamȏ, 1962 based on only female specimens. Gamȏ has described that new species, Nannastacus pruinosus could be distinguished from N. nyctagineus based on only male specimens. The Korean male specimens of N. nyctagineus undoubtedly match well with the description of N. pruinosus with the following common features: the surface of the carapace is covered with numerous granules; pereonites and pleonites have similar patterns of the teeth on dorsal surface; the proportion and armature of the dactylus of pereopod 2 is similar; the number and pattern arrangement of the setae on the surface in antenna 1, pereopod 1, and uropod are the all most same. Nannastacus pruinosus Gamȏ, 1962 is proposed as a synonym of N. nyctagineus described in this study. This represents the first record on the genus Nannastacus from Korea.

A New Species of the Genus Incabates Hammer (Acari: Oribatida) from Jeju, Korea (한국산 Incabates (Acari: Oribatida)속의 1 신종)

  • An oribatid mite, Incabates barbatus sp. nov. in the family Haplozetidae is described herein from Korea. This new species is very similar to 1. major reported in Japan. However, prodorsal setae (ro & la) and head of sensillus are smooth in 1. major while barbs are found on the outer margin in the middle of rostral seta (ro) and lamellar seta (la) in addition to the presence of spinulae making the surface of head of sensillus rough in the new species. There are altogether 9 species described in the genus Incubates at present including the new species reported herein and keys to these 9 species are presented.

An Outbreak of Caparinia tripilis in a Colony of African Pygmy Hedgehogs (Atelerix albiventris) from Korea

  • In February 2010, dermatitis characterized by scale and self-trauma due to puritis was recognized in a group of 22 four-toed hedgehogs (Atelerix albiventris Wagner, 1841) from a local pet shop in Gwangju, Korea. Microscopic examinations of skin scraping samples showed numerous mites of all developmental stages. Morphologically, pedicels of adult mites were short and unjointed. Tarsal caruncles were bell-shaped on all legs of males while they were absent on legs III and IV of females. Three long setae on the third pair of legs in both sexes were present. Adult males had posterior end of the abdomen with trilobate projection on each side, each lobe with a long seta. Based on these features, the mites were identified as Caparinia tripilis. This is the first report of caparinic mite infestation in hedgehogs from Korea. Identification keys for the family Psoroptidae and the genus Caparinia are provided.

New Records of Two unknown Micro-filamentous Endophytic Green Algae in Korea: Phaeophila dendroides and Dilabifilum arthropyreniae (한국산 미기록 사상형 내생녹조 2종: 꼬인털속살이말 및 속살이마디말)

  • Morphological and molecular information about two microfilamentous endophytic algae of the Grateloupia lanceolata or Bryopsis sp. from Korea is given. Of two endophytes, Phaeophila dendroides is endophytic in Grateloupia lanceolata. It is green in color and composed of uniseriate branched filaments with long setae. Each cell had several pyrenoids. Undulate or twisted Phaeophila-type hair developed from vegetative cells. Dilabifilum arthropyreniae is endophytic in Bryopsis sp. The frond of this species consists of frequently irregular, branching uniseriate filaments. Each cell had a single pyrenoid without hairs. Sporangia were not observed. The ends of the filaments were curved. In the phylogenetic tree, based on tufA and ITS sequences, these two species are nested in the same clade as Phaeophila dendroides and Dilabifilum arthropyreniae, respectively. In this study, these two species are newly recorded in the Korean marine algal flora, based on the morphological and molecular data.

Colletotrichum Disease of Mungbean Sprout by Colletotrichum acutatum

  • Decayed samples of marketed mungbean sprout were collected from Sacheon, Suncheon, and Dangjin cities in Korea. Initial symptom on hypocotyls was dark-brown diamond speck, which developed into enlarged sunken brownish-black spot with irregular margin, followed by softening yellowish decay. Brown speck on cotyledon further developed into irregular lesions. This study isolated the fungus Colletotrichum sp. The fungal colony was pale orange, which turned greenish gray after 1 week at $25^{\circ}C$. Colony of reverse side in a petri dish was pink. Neither conidiomata nor setae were present in the culture. Typical fusiform conidia sized 7.5-15.0$\times$2.5-2.9 $\mu\textrm$were hyaline, aseptate, smooth, and had salmon color in mass. Conidiogenous cells were phialidic, hyaline, smooth, and cylindrical with terminal distinct collarette. Basedon these mycological characteristics, the casual organism was identified as Colletotrichum acutatum. This is first report of Colletotrichum acutatum in Korea.
