Metabolic Syndrome Risk by Dietary Fat Energy Ratio in Middle-aged Men - Using the 2012~2013 Korean National Health and Nutrition Examination Survey Data - (중년 남성의 지방 에너지비에 따른 대사증후군 위험도 비교 - 2012~2013년 국민건강영양조사 자료 이용 -)

  • This study aimed to compare energy nutrient intake, health related factors, physical characteristics, blood biochemical indices, prevalence of metabolic syndrome and odds ratio (OR) of metabolic syndrome based on dietary fat energy ratio. Subjects were 1,205 men aged 40~64 years. The average fat intake was 52.8 g. Subjects were divided into three groups (deficient, normal, excess) based on dietary fat energy ratio. The dietary fat energy rations of the three groups were 36.9%, 42.9% and 20.2%, respectively. Energy and protein intake were increased significantly with dietary fat energy ratio (p<0.001), whereas carbohydrate intake decreased (p<0.001). In health related factors, amount of smoking alone showed increase based on dietary fat energy ratio (p<0.001). In comparing physical characteristics, blood pressure and blood biochemical indices, excepting diastolic blood pressure, increased significantly based on dietary fat energy ratio (p<0.01~p<0.001). The rate that exceeded criteria in risk factors for metabolic syndrome was higher in the serum triglyceride (41.2%) and was lower in the waist circumference (22.2%). Prevalence of metabolic syndrome was 37.9%, and showed significant correlation to dietary fat energy ratio (p<0.05). The OR of metabolic syndrome was higher in deficient and excess group than in normal group, but it had no relationship between fat energy ratio and metabolic syndrome. The results of this study provide basic data to establish fat intake guidelines for prevention of metabolic syndrome in middle-aged men.

Separation of Immunoglobulin G from Colostrum by Reverse Micelles of Cationic Surfactant (양이온 계면활성제로 형성된 역미셀을 이용한 초유 Immunoglobulin G의 분리)

  • This study was carried out to separate immunoglobulin G(IgG) from colostrum using reverse micellar extraction of cationic surfactant and to suggest suitable extraction conditions. The reconstituted colostrum powder was solubilized into a reverse micellar phase containing CDAB(cetyldimethylethyl ammonium bromide) by mixing equal volume of the aqueous and organic phase with constant stirring. The solubilization of proteins from the aqueous to the organic phase was manipulated by pH and ionic strength of the aqueous phase and concentration of surfactant in the organic phase. Based on the SDS-PAGE and densitometry, about more than 90% of initial IgG was remained in the aqueous phase after reverse micellar extraction. Although the aqueous phase contained lactoferrin and bovine serum albumin as minor components, about 93% of the total protein was IgG. The efficient extraction was achieved by the reaction of sodium phosphate buffer(pH 8) containing 50 mM KCl and organic phase containing 100 mM CDAB. The separation of IgG using reverse micellar extraction was simple, highly efficient and easy to be scaled up.

Suppressed Cell Proliferation and Differentiation Following an Over-expression of Myostatin is Associated with Inhibited Expression of Insulin-like Growth Factor II and Myogenin in Rat L6 Myoblasts

  • Myostatin (MSTN) and insulin-like growth factors (IGFs) are a known inhibitor and stimulators of proliferation and differentiation of muscle cells, respectively. The present study was performed to investigate the relationship of MSTN-induced growth inhibition to expression of the IGF system components and myogenin, a muscle cell-specific transcription factor, in rat L6 myoblasts. The L6 cells transfected with a green fluorescent protein-MSTN plasmid expression construct had a 47% less cell number than mock-transfected cells after 3-d serum-free culture, accompanied by delayed differentiation which was suggested by inhibited aggregation of cells. Moreover, cells transfected with the expression construct had decreased expression of IGF-II and myogenin genes, but not IGF-I or its receptor genes, as examined by reverse transcription-polymerase chain reaction. The reduced mitosis of the L6 cells transfected with the MSTN-expression construct increased following an addition of either IGF-I or IGF-II to the culture medium, but not to the level of mock-transfected cells. By contrast, myogenin gene expression in these cells increased after the addition of either IGF to the level of mock-transfected cells. Collectively, these results suggest that the inhibitory effect of MSTN on L6 cell proliferation and differentiation is likely to be partly mediated by serially suppressed expression of IGF-II and myogenin genes, not IGF-I gene.

Prognostic Value of Serum AFP, AFP-L3, and GP73 in Monitoring Short-term Treatment Response and Recurrence of Hepatocellular Carcinoma after Radiofrequency Ablation

  • Purpose: Alpha-fetoprotein (AFP), Lens culinaris agglutinin-reactive fraction of AFP (AFP-L3), and Golgi protein 73 (GP73) levels have been widely used as tumor markers for the diagnosis of hepatocellular carcinoma (HCC). The aim of this study was to investigate whether these tumor markers could be used to monitor short-term treatment response and recurrence of HCC in patients undergoing radiofrequency ablation (RFA). Methods: Between July 2012 and July 2013, 53 consecutive patients with newly diagnosed HCC were prospectively enrolled in this study. Among these, 32 patients underwent RFA, after which they were followed up prospectively at the First Hospital of Jilin University in China. Results: AFP, AFP-L3, and GP-73 values pre-RFA were not associated with tumor size, whereas AFP and GP-73 levels tended to be associated with tumor number, the presence of vascular invasion, deterioration of liver function, advanced-stage disease, and a poor performance status. GP-73 levels were dramatically elevated in the patients with hepatitis C-associated HCC. Neither pre-RFA nor 1-month post-RFA tumor marker values were associated with short-term outcome. The short-term recurrence rate of AFP-positive patients measured 1 month post-RFA was obviously higher than that of AFP-negative patients. Conclusions: AFP and GP-73 values were associated with clinical variables representing tumor growth and invasiveness, and the AFP value measured 1 month post-RFA was a strong predictor of short-term recurrence in patients with HCC.

Kupffer Cells Suppress Hepatocarcinogenesis and Metastasis in Tumor Orthotopic Implanted Kunming Mice

  • In this research, we used $GdCl_3$ (gadolinium chloride) to restrain the function of Kupffer cells and assessed effects on hepatocarcinogenesis and metastasis in the Kunming mouse. A 0.25% $GdCl_3$ solution (10 mg/kg b.w.) was infused via the vena caudalis of each mouse 1 week before inoculation of H22 cells and was continued once per three days. Then we observed the follow indexes 3 weeks after injection of H22 cells: tumor weight, histologic characteristics of tumor tissue by light microscopy, ultramicrostructure of Kupffer cells under the electron microscope, distribution and number of Kupffer cells by histochemical staining, and TNF-${\alpha}$ and IFN-${\gamma}$ levels in blood-serum and liver tissue by ELISA and RT-PCR. MMP-2 protein expression was tested by immunohistochemistry. The $GdCl_3$ pretreatment had no effect on the quantity of Kupffer cells, but clearly restrained their functions, with decrease of TNF-${\alpha}$ and IFN-${\gamma}$ levels and elevation of MMP2. Tumor immunity functions were markedly suppressed and tumor growth was accelerated with appearance of metastasis. Furthermore, survival time of trial mice was shortened.

A Study on Nutrients Intake , Physical and Biochemical Status of Elementary School Children in Taejon (대전지역 초등학교 아동의 체위 , 혈액성상 및 영양섭취에 관한 조사 연구)

  • The purpose of this study was to investigate nutrients intake, Rohrer index, serum cholesterol level, blood pressure and others for clinical and nutritional study on school children in Taejon. This survey was carried out in October, 1995. The subjects were 362 children, 6 to 11 years old. The results were summarized as follows. By Rohrer index, 4.6% of the children was lean, 69.6% was normal, 17.4% was overweight and 8.6% was obese group. Systolic blood pressure, diastolic blood pressure and skin fold thickness of all subjects were 111.7$\pm$15.7mmHg, 69.8$\pm$14.9mmHg and 14.5$\pm$7.3mm respectively. Systolic blood pressure and skin fold thickness were increased with Rohrer index. Total cholesterol, urea nitrogen and creatinine levels in blood were 160.8$\pm$23.9mm/dl, 11.5$\pm$3.5mm/dl and 0.71$\pm$0.12mm/dl respectively but not significantly different among groups. These levels have a slight tendency to increase in obese group but not significantly different among groups. Hematocrit and hemoglobin levels were 41.3$\pm$4.7%, 13.1$\pm$1.6g/dl. These levels of 11 years old girls were lower than same aged boys. Composition ratio of total energy intakes were 65% of CHO, 13% of protein and 22% of fat. Energy and most nutrients intakes were insufficient except for P and vitamin C. Especially the amount of vitamin A, Fe, Zn and folate intakes were lower than halves of RDA'S. Most nutrients intake were not significantly different among groups. Pearson' correlation in Rohrer index with nutrients intakes were all negative correlation significantly different of K, Na and Thiamin.

Optimum Dietary Lipid Level in Juvenile River Puffer Takifugu obscurus (치어기 황복(Takifugu obscurus) 사료 내 적정 지질 함량)

  • This study was conducted to evaluate the optimum dietary lipid level in juvenile river puffer. Five semi-purified diets were formulated with corn oil to contain graded levels of lipid levels of 6, 9, 12, 15 and 18%. Fish averaging $8.32{\pm}0.02$ g randomly were fed the experimental diets in triplicate groups for 8 weeks. After the 8-weeks feeding trial, weight gain and specific growth rate of fish fed the 9% diet were significantly higher than those of fish fed the 15 and 18% diets (P<0.05) but there was not significantly different from that of fish fed the 6, 9 and 12% diets. Feed efficiency and protein efficiency ratio of fish fed the 6, 9 and 12% diets were significantly higher than those of fish fed the 18% diet (P<0.05). Visceralsomatic index of fish fed 18% diet was significantly higher than that of fish fed the 6% diet (P <0.05) but there was not significantly different from that of fish fed the 9, 12, 15 and 18% diets. No significant differences were observed in condition factor, hepatosomatic index and whole body composition among all the fish groups. Serum cholesterol and triglyceride fish fed of 18% diet were significantly higher than that of fish fed the other diets (P<0.05). Optimum dietary lipid levels by using broken-line model and by using second order polynomial were estimated at 7.01% and 8.98% for the maximum growth of fish respectively. Therefore, these results suggested that the optimum dietary lipid level could be greater than 7.01% but less than 8.98% for the maximum growth in juvenile river puffer.

Korean Red Ginseng Saponin Fraction Downregulates Proinflammatory Mediators in LPS Stimulated RAW264.7 Cells and Protects Mice against Endotoxic Shock

  • Korean red ginseng has shown therapeutic effects for a number of disease conditions. However, little is known about the anti-inflammatory effect of Korean red ginseng saponin fraction (RGSF) in vitro and in vivo. Therefore, in this study, we showed that RGSF containing 20(S)-protopanaxadiol type saponins inhibited nitric oxide production and attenuated the release of tumor necrotic factor (TNF)-${\alpha}$, interleukin (IL)-6, granulocyte monocyte colony stimulating factor (GMCSF), and macrophage chemo-attractant protein-1 in lipopolysaccharide (LPS) stimulated murine macrophage RAW264.7 cells. Moreover, RGSF down-regulated the mRNA expressions of inducible nitric oxide synthase, cyclooxyginase-2, IL-$1{\beta}$, TNF-${\alpha}$, GMCSF, and IL-6. Furthermore, RGSF reduced the level of TNF-${\alpha}$ in the serum and protected mice against LPS mediated endotoxic shock. In conclusion, these results indicated that ginsenosides from RGSF and their metabolites could be potential sources of therapeutic agents against inflammation.

A Case of Secondary Renal Amyloidosis in Children with Juvenile Rheumatoid Arthritis (소아기 류마티스 관절염 환아에서 발생한 이차성 신유전분증 1 례)

  • Amyloidosis comprises a diverse group of systemic and local diseases characterized by organ involvement by the extracellular deposition of fibrils composed of subunits of a variety of normal serum proteins. Secondary amyloidosis is caused by the deposition of amyloid A(AA) protein in chronic inflammatory disease. Juvenile rheumatoid arthritis(JRA) has been known to be the most common cause of secondary amyloidosis. We experienced one case of secondary renal amyloidosis in a 12-year-old girl who had suffered from JRA for several years who had visited our renal clinic to evaluate the proteinuria with microscopic hematuria which was detected by chance at school urine screening examination. Apple green birefringence was observed under polarized light with Congo red stain at)d characteristic electron microscopic findings was also noted in renal tissues which was obtained by percutaneous renal biopsy. In our knowledge, this is the first case report of secondary renal amyloidosis developed in pediatric age in Korea.

Anti-obesity Effect of Aster Yomena Ethanol Extract in High Fat Diet-induced Obese Mice (고지방 식이로 유도된 비만 생쥐에서 쑥부쟁이 에탄올 추출물의 항비만 효과)

  • Aster yomena (AY) have been used as a traditional medicine to treat cough, bronchial asthma, and insect bites in Korea. In this study, we evaluated the inhibition of adipogenesis in 3T3-L1 cells and in high-fat diet (HFD)-induced obese mice by AY ethanol extract. Lipid accumulation measurement indicates that AY markedly inhibited adipogenesis in a dose-dependent manner. qRT-PCR results demonstrated that the mRNA expression of adipogenic transcription factors such as peroxisome proliferator-activated receptor-${\gamma}$ ($PPAR-{\gamma}$) in 3T3-L1 cells were significantly down-regulated by AY treatment. And inhibited the expression of FAS, a protein responsible for lipid synthesis, transport and storage. Oral administration of AY (100, 250, and 500 mg/kg, P.O/daily for 4 weeks) was conducted in high-fat diet induced obese mice and C57BL/6 mice. AY was orally administered for 4 weeks to extract liver and epididymal fat, and hematoxylin and eosin staining(H&E staining) was observed. Observation showed that the fat concentration of liver tissue tended to decrease dose-dependently and decreased significantly at 500 mg/kg concentration. The AY-administered group of HFD-induced mice had a lower body weight gain, along with decreased triglycerides and total cholesterol compared with the control mice, however, the HDL-cholesterol/total cholesterol ratio was increased. These results indicate that AY exhibits anti-obesity effects in obese mice by decreasing in serum lipid levels and lipogenesis related gene.
