Effects of Dietary Protein Levels on the Serum Lipids and Enzyme Activities of Ethanol-Administered Rats (에탄올을 투여한 흰쥐에서 식이 단백질 섭취 수준이 혈청중 지방 및 효소활성에 미치는 영향)

  • This study was undertaken to investigate effects of alcohol and dietary protein levels on serum lipids and enzyme activities in 15 week-old male rats given a normal diet. Rats were divided into 8 groups : control group (16% protein 16PC) and 8%(8PE) 16%(16PE) and 24% protein groups(24PE) to which was given 5% ethanol mixed into their drinking water after 4 weeks and 10 weeks. Body weight organ weight and various blood components were determined at 4 and 10 weeks. Body weight gain organ weight hemoglobin concentration and hematocrit value were not influenced by ethanol and dietary protein levels. The levels of total cholesterol HDL-cholesterol and phospholipid in serum were not affected by ethanol consumption. Serum triglyceride concen-trations after 10 weeks were significnatly increased ethanol-treated group compared with that of control group and the effect was greater in low protein group than control group. Serum ALP activity was significantly higher in 8PE group than other group but there was no influence by ethanol consumption.

  • Parathyroid hormone(PTH) is known to stimulate bone resorption and to inhibit bone collagen synthesis. In contrast, as the evidence of stimulation of bone formation by PTH has recently been observed, the study on the role of PTH involved in osteoporosis draws remarkable attention. This study has dealt with the role of alkaline phoshatase(AP), a marker enzyme for bone formation and osteoblast action, Animals(BALS/cmice) were divided into three dietary groups(high and medium Ca and Ca-free) and hormones including PTH, calcitonin(CT), cholecalciferol(citamin D) were i.p. injected. AP in the serum and liver was measured using Sigma 221 alkaline buffer solutions containing 9mM of p-nitrophenyl phoshate. Enzume was reacted at 37$^{\circ}C$ for 10 minutes and the reaction was stopped by 1.8ml of 0.1N NaOH and measured at 410nm. We found that serum and liver AP activity was increased by low dietary Ac. Compared to the control, and serum Ap activity was enhanced by PTH and vitamin D regardless of the dietary Ca. On the other hand, liver AP activity was inhibited by OTH and vitamin D at all levels of dietary Ca. CT inhibited the action of PTH and vitamin D in the serum. But, the inhibition of PTH and vitamin D action by CT was not observed in the liver, unlike in the case of serum.

  • This study was performed to determine the changes of serum enzyme activities in rats with hepatic injury induced by the administration of AAPH. Minor behavioral change, brittleness of skin hair and decreased water and fled intake were observed in rats administered intraperitoneally with AAPH. Serum AST and ALT activities pre-treatment were $65{\pm} 13.8 and 32{\pm}$ 12.6 IU/L, respectively and increased sharply from 2 hours of administration and reached $1248{\pm} 77.6 and 946{\pm}$ 45.6 IU/L, respectively at 48 hours of administration. Serum ALP and $\gamma -GTP activities pretreatment were 221 {\pm} 75.6 and 2.2{\pm}$ 0.35 IU/L respectively and increased sharply from 8 hours of administration and reached $767{\pm} 44.9 IU/L and 8.0{\pm} 1.23 IU/L,$ respectively at 48 hours of administration.

  • To understand the activity of non-specific defence factors in cultured Sebastes, the antibacterial effect of the serum, skin mucus and homogenate of various organs from cultured oblong rockfish (Sebastes oblongus) and rockfish(Sebastes schlegeli) against pathogenic bacteria, Aeromonas hydrophila, Edwardsiella tarda, Vibrio anguillarum, and Streptococcus sp. was compared with that of flounder(Paralichthys olivaceus) and seabass(Leteolabrax japonicus). And the activities of proteolytic enzyme, chitinolytic enzyme and haemolycin as non-specific defence factor were investigated on the oblong rockfish and rockfish. Samples from oblong rockfish showed the highest antibacterial activity by lysoplate assay on agar plate mixed with pathogens, followed in descending order by rockfish, seabass, and flounder. Turbidimetric assay was carried to evaluate the lysozyme activity of fish samples against lyophilized cells of Micrococcus lysodeiktikus. The serum, kidney, liver, stomach, intestine and eyeball of oblong rockfish and the mucus and gill of rockfish appeared to have the highest lysozyme activity among the fish strains investigated. All samples except skin mucus, liver, and eyeball of oblong rockfish and rockfish showed proteolytic enzyme activity. Chitinolytic enzyme activity was showed in random sampling and haemolytic activity was remarkable in oblong rockfish. Therefore, Sebastes strain was proved to have effective defense mechanisms based on the antibacterial activities, and lysozyme, proteolytic enzyme, chitinolytic enzyme, and haemolycin were considered to act as the non-specific defence factor of Sebastes.

Effect of Fed Chlorella on the Change of Lipid Components and Enzyme Activity in Serum of Rat by Lead Exposure (납에 노출된 흰쥐에서 Chlorella 섭취가 혈청내 지방성분 및 효소활성에 미치는 영향)

  • This study was performed to investigate the influence of Chlorella which effected the change of Pb contents, enzyme activity and lipid compounds on the rats fed the beverage involved Pb and the different contented Chlorella added-diets for 18weeks. The rat's weight of 200ppm Pb group was decreased 6.04% and the cause of that was Pb intake. But the rat's weight of Chlorella added-diets +200ppm Pb group was increased 4.02% (p<0.01). When feeding the different(0%, 2%, 5%, 10% ) Chlorella added-diets with the Pb contented beverage to the rats, we could know that the Pb contents accumulated on tissue were decreased to 20.70(0%), 12.88(2%), 14.83 (5%) and 19.56(10%), compared with the quantity of Pb taken in. Total-cholesterol, triglyceride, glucose content and AST, ALT, ALP, LDH enzyme activity in serum were the highest on 200ppm Pb group and those were decreased by the order of different(2%∼5% >10%) Chlorella contents + 200ppmPb group. A significance was recognized on the level of 1%. Therefore, when rats were exposed to Pb, it was thought that the amount of Chlorella intake was adequate on content 2∼5% for reducing the lead toxicity.

Serum Levels of Xanthine Oxidase Activities in Cyclohexanone-Treated Rats Pretreated with Carbon Tetrachloride

  • To investigate an effect of cyclohexanone (CHO) treatment on the serum levels of xanthine oxidase (XO) in liver damaged animals, the rats were intraperitoneally pretreated with 50% carbon tetrachloride ($CCl_4$) in olive oil (0.1 mL/ 100 g body weight) 14 times every other day. To the $CCl_4$-pretreated rats, CHO (1.56 g/kg body weight) was injected once and then the animals were sacrificed at 4 hours after CHO treatment. The increasing rate of serum and liver XO activities to the control was higher in CHO-treated animals pretreated with $CCl_4$ than the $CCl_4$-pretreated those. Concomitantly CHO injection to the $CCl_4$-pretreated animals showed somewhat higher Vmax and lower Km value in the kinetics of liver XO enzyme. Furthermore, increasing rate of hepatic malonedialdehyde content to the control was also higher in CHO-treated animals pretreated with $CCl_4$ than $CCl_4$-pretreated those. On the other hand, the injection of CHO to the $CCl_4$-pretreated animals showed the more enhanced liver damage on the basis of liver function finding; liver weight per body weight (%), serum levels of alanine aminotransferase activity and hepatic glucose-6-phosphatase activity. In conclusion, injection of CHO to the $CCl_4$-pretreated rats led to more increased activity of serum XO and it may be caused by acceleration of hepatocyte membrane permeability and induction of enzyme protein.

  • This paper was intented to charily the cause of an increase of serum guanase activity in rats following injection of $CCI_4$. The cause of increasing serum guanase was focused on the change of guanase activities in both serum and liver, and additionally, these results were compared with the previously known alanine aminotransferase (ALT). Concomitantly the microscopic investigation on the histologic changes, and the determination of lipid peroxides of liver were combined in this experiment for a correlation to observe that the activity of guanase would be effected by the various degree of hepatic injury induced by $CCI_4$. The serum levels of guanase were increased about 2 fold in the fatty change stage (3-12 days), 5.2 fold representing the peak value in necrosis stage (21days), 4.5 fold in early cirrhosis stage (48 days), and 2 fold in severe cirrhosis stage (92 days). These changes of serum guanase activity showed similar patterns to those of ALT activity and lipid peroxides in liver cell. The changes of liver guanase activities showed an increase, whereas ALT activities in liver were markedly decreased. It is likely that the increase of serum guanase activity is based on the excess leaking of guanase into blood by the result of accelerated enzyme synthesis in liver cell of $CCI_4$ intoxicated rats. In addition, the possibility could not be ruled out, however, that the increase of serum guanase activity would be caused by the alteration of membrane permeability.

  • Progesterone catabolizing enzyme, the enzyme $20{\alpha}$-hydroxysteroid dehydrgenase($20{\alpha}$-HSD) is pivotal in the regulation of ovarian function in rodents, which catabolizes progesterone into biologically inactive $20{\alpha}$-hydroxypregn-4-en-3-one($20{\alpha}$-OHP). In this study was carried out the influence of $20{\alpha}$-HSD activity of ovarian function, we investigated changes in ovarian cytosol $20{\alpha}$-HSD activity and serume progesterone concentration during the estrous cycles and pregnancy in rat. During the estrous cycles, the $20{\alpha}$-HSD activities were highest on the progestrous, but serum progesterone concentration was lowest on this phase. During the pregnancy, the $20{\alpha}$-HSD activities were relatively higher early pregnancy(day-1-3 gestation) and late pregnancy(day 21 to parturition), serum progesterone concentration was maintained significantly high to day 19 of gestation. The $20{\alpha}$-HSD activities were lower during the middle pregnancy. From these results, ovarian $20{\alpha}$-HSD activities may possibly act as physiologically very important in the control and maintenance of estrous cycles in rat.

  • The purpose of this study was to probe the influences of krill (Euphausia superba) meal with NaF oral administration on a dose-effect relationship between fluoride levels of krill meal and serum enzyme activity such as alkaline phosphatase (ALP), aspartate aminotransferase (AST), alanine aminotransferase (ALT), and lactate dehydrogenase (LDH) in rats fed experimental diets for 5 weeks. There were no significant decreases in the activities of ALP, AST, ALT, and LDH in sera among krill meal diet groups (KF10, KF20, KF30). However, these groups were significantly (p<0.05) lower enzyme activities than normal diet (ND) plus NaF 10 mg group (NF). The fluoride levels of serum and organ tissues (liver, brain, heart, lung, kidney) in NaF 10 mg groups (NF, KF10, KF20, KF30) were significantly increased by adding krill meal in comparison with normal diet group. The results indicate that a difficult to found toxicity to the tissues from krill meal diet groups.

The Effect of Benzene on the Activity of Adenosine Deaminase in Tissues of Rats

  • Swiss Albino (Rat rattus norvegicus) rats were intraperitoneally injected with a 100 mg $kg^{-1}$ dosage of benzene, a toxic and carcinogenic agent widely used for industrial purposes. Changes in the adenosine deaminase (ADA) activity in the liver, kidney and serum of rats were investigated at 0, 2, 4, 8, 16, 32 and 64 h following injection. Serum physiological was administered to each control group. Enzyme activities were measured spectrophotometrically. Our purpose was to further investigations of some diseases caused by benzene, and present evidence of variations in the activity of ADA enzyme effected by benzene. While benzene caused significant inhibitions in ADA activity in the liver at 16 and 32 h and at 0.05 probability level, no significant inhibition or activation occurred at other test periods (hours). ADA activity did not present any significant variation in the kidneys. It was observed that ADA activity displayed similar patterns in the control groups. Comparisons of ADA activities in the two groups showed a statistically significant decrease between $4^{th}$ and $64^{th}$ hours (p< 0.05), demonstrating a direct correlation between benzene and its effects on ADA enzymes.
