Purpose: When deciding a treatment plan in diabetic foot ulcer patients, predicting a possibility of healing wounds is important since not a few patients have poor general condition to get successful wound healing. This study was planned to find out if a serum collagen level can be used as a predictor for healing wounds in diabetic foot patients. Methods: Fifty-seven patients, who visited our clinic from January to June, 2007 for treatment of diabetic foot ulcers, were included in this study. Serum levels of type I collagen were checked using carboxy terminal type I propeptide kits. Simultaneously serum levels of vitamin C and iron, cofactors of collagen synthesis, were checked. The patients were divided into two groups; a group of successfully healed wounds and the other of unhealed wounds. Serum levels of the parameters were compared between the 2 groups. Results: The serum level of collagen was $197.65{\pm}86.26ng/ml$ in a healed group and $87.91{\pm}28.76ng/ml$ in the unhealed group(p<0.05). The serum iron and vitamin C levels were did not show significant differences. Conclusion: The serum collagen level may predict healing or nonhealing wounds in diabetic foot ulcers.
The induction of proliferation of adult rat islets was investigated under various culture conditions. The islets were isolated from male Sprague-Dawley rats and subsequently embedded in collagen gels, which mimic the in vivo three-dimensional surroundings. During the culture period, the effects of heterologous serum (fetal bovine serum, FBS), epidermal growth factor (EGF), and retinoic acid (RA) on islet growth were examined with respect to the morphological and total DNA content changes. To investigate these changes at the cellular level, whole mount immunocytochemistry using specific antibodies for insulin and glucagon was performed. The results showed that (i) collagen gels as an extracellular matrix can maintain islets in a similar way to that in vivo, (ii) heterologous serum (FBS) had stimulatory effects on islet proliferation in a dose-dependent manner, (iii) RA had inhibitory effects on islet proliferation induced by the serum in a dose-dependent manner, (iv) EGF had weak inhibitory effects on islet proliferation induced by the serum except at the concentration of 10 nM where its effect was not significant, and (v) whole mount immunocytochemistry revealed that newly proliferated islet cells were mainly $\beta$-and $\alpha$-cells.

The aim of this study was to evaluate the effect of Lycii fructus (LF) on serum lipid and the collagen content of the connective tissues in ovariectomized rats. From day 2 until day 37 after the ovariectomy, Sprague-Dawley female rats were randomly assigned to the following groups : sham-operated rat (Sham), ovariectomized control rat (OVX-control), and ovariectomized rats supplemented with the LF 50mg/kg bw/day (OVX-LF). The LF ethanol extract were orally administrated 1ml per day. Body weight gain was not significantly different in groups. Although total-cholesterol and triglyceride were increased in the ovariectomized control, supplementation with the LF extract decreased the levels. Moreover, the serum HDL-cholesterol levels were significantly increased after supplementation with the LF extract(p<0.05). Supplementation with the LF extract prevented a decrease in the collagen level in bone and cartilage tissues. These results are consistent with the conclusions based on the estrogenic activities of LF. Therefore, it may be used to possibly improve the quality of life in menopausal women.
The aim of this study was to evaluate the effects of Artemisia Princeps Var. Orientalis (AP) on serum lipids and the collagen content of the connective tissues in ovariectomized rats. The OVX rats were significantly heavier than the sham-operated rats at all time points, but supplementation with the AP extracts tended to gain weight less than OVX-control. Although total-cholesterol was increased at OVX-control, supplementation with the AP extracts tended to result in less than OVX-control. Triglyceride was significantly decreased after supplemented with the AP extracts (p<0.05). HDL-cholesterol is appeared higher AP extracts group than OVX-control. According to the results, we could know the fact that AP extracts were effective on serum lipids content throughout decreasing total-cholesterol, triglyceride and increasing HDL-cholesterol in ovariectomized rats. Supplementation with the AP extracts prevented a decrease in the collagen level in bone and cartilage tissues. These results are consistent with the conclusions based on the estrogenic activities of AP. Therefore, it may be used to possibly improve the quality of life in menopausal women.

This study was carried out to know the immunological effect of GDGT on CIA(collagen induced arthritis) mice, a model of rheumatoid arthritis. For this purpose, GDGT was orally administerd to mice with arthritis induced by collagen II and then value of cytotoxicity on hFLSs and liver, the arthritis index, immunocyte in paw joint and DNL, rheumatoid factor (IgG and IgM), collagen II specific antibody in the serum were measured. The cytotoxicity were not shown on hFLSs and liver. The arthritis index decreased significantly after 3 week. In total cell counts of DLN and paw joint, there was a significant increase in DLN and significant decrease in paw joint. In DNL, $CD19^+$, $CD3^+$, $CD4^+$, $CD3^+/CD69^+$, $CD8^+$, $CD4^+/CD25^+$, $ CD3^+/CD49b^+$ cells increased significantly. In Paw joints, $CD3^+$, $ CD4^+$, $CD4^+/CD25^+$ cells decreased significantly. The level of serum IgG and IgM decreased significantly. The level of collagen II in the serum was decreased significantly. Marginal erosion, necrotic chodrocytes, cartilage and bone degradation were improved in histological section of paw joints. The results present significant immunological effect of GDGT on rats with arthritis induced by collgen II. So we expect that GDGT should be used as a effective drugs for not only rheumatoid arthritis but also another auto-immune disease.
Objectives : This study was carried out to investigate the anti-inflammation, anti-development and curative effects of Oyaksunki-sangamibang (OSKM) on collagen-induced arthritis in Wistar rats and ICR mice. Materials & Methods : D experiment part II, the inhibitory effects of nitric oxide synthesis, pro-inflammatory cytokines, and cyclooxygenase were studied. In experiment part II, paw eduma volume and thickness of ankle joint were measured at 0, 10, 15, and 20 days after immunization. The incidence and arthritis score were evaluated 14 days after immunization, At 15 days after immunization, serum $TNF-\alpha$ was analyzed. In experiment part III paw edema volume and thickness of ankle joint were measured at 0, 10, and 15days after treatment. At 15 days after treatment, serum $TNF-\alpha$ was analyzed. Results : In experiment part I: 1. Nitric oxide synthesis ·md pro-inflammatory cytokines were inhibited significantly by OSKM extract. 2. Cyclooxygenase 2 (COX-2) was inhibited by OSKM extract. In experiment part II: Paw edema volume, thickness of ankle joint and serum $TNF-\alpha$ level of the teated group were significantly decreased compared with the control group at 20 days after immunization. In experiment part III: Incidence of arthritis was $70\%$. OSKM-treated group had no significant change on paw edema volume, thickness of ankle joint and serum $TNF-\alpha$ level. Conclusions : These results indicated that OSKM has anti-inflammation effects on the ICR mouse, and higher inhibitory effects on the onset but lower inhibitory effects on the progression of collagen-induced arthritis in rats.

Objectives & Methods : The purpose of this study is to observe the effects of Angelicae Radix Pharmacopuncture(AR-P) at Joksamni(ST36) on collagen II induced arthritis in DBA/1J mice. The authors evaluated arthritis index, arthritis incidence and joint edema, and measured body weight, spleen size and stenosis rate, serum cytokine level, serum antibody level, immune cell populations In spleen, lymph node, and knee joint, and performed histological analysis of CIA mouse joint. Results : In the AR-P group, arthritis index, arthritis incidence and joint edema were decreased, and the enlargement, malformation and stenosis of spleen and the malformation of joint appeared milder than the control group. In AR-P group, the levels of $IL-1{\beta}$, IL-6, $TNF-{\alpha}$, $IFN-{\gamma}$ in serum were significantly decreased. And the level of anti-collagen II in serum was maintained lower in AR-P group than in the control group. In AR-P group, the level of $IFN-{\gamma}$ and IL-10, and $IFN-{\gamma}/IL-4$ ratio were significantly decreased, and the ratios of $CD3e^+$ cells to $CD45^+$ cells, $CD4^+$ cells to $CD8^+$ cells in spleen were similarly maintained as those of the normal group. In the AR-P group, the populations of $CD4^+/CD25^+$ cells in spleen and lymph nodes, $CD45R^+/CD69^+$ cells in lymph nodes, $CD3^+/CD69^+$ cells and $CD11b^+/Gr-1^+$ cells in knee joint were decreased. In the histological analysis, the cartilage destruction, synovial cell proliferation and the collagen fiber destruction were decreased in the AR-P group Conclusions : The results suggest that AR-P at right ST36 has a therapeutic effect on collagen-induced arthritis in mice.

Total cell number of paw joint in CIA mice(ESWTT extract administration group) was decreased significantly. 4. The absolute number of CD3+, CD3+/CD69+, CD4+, CD4+/CD25+, CD49b+, CD3+/CD49b+ cells in CIA mice(ESWTT extract administration group) were decreased significantly. 5. The levels of IL-6 and $TNF-{\alpha}$ in the serum of CIA mice(ESWTT extract administration group) were decreased significantly. 6. The levels of total IgG and IgM in the serum of CIA mice(ESWTT extract administration group) were decreased significantly. 7. The level of collagen II specific antibody in the serum of CIA mice(ESWTT extract administration group) was decreased significantly. Conclusions : Comparison of the results for this study showed that ESWTT had immunomodulatory effects of suppressing. So we expect that ESWTT could be used as an effective drugs for not only rheumatoid arthritis but also auto-immune disease.

In order to examine the effect of ${\beta}-estradiol$ on the cell growth, using a primary rabbit kidney poximal tubule cell culture system. We investigated the effect of ${\beta}-estradiol$ on alpha 1 (IV) collagen and ${\beta}-actin$ mRNA levels from primary rabbit kidney cell cultures, and also the effects of 3 growth factors and ${\beta}-estradiol$ supplementation on the growth of primary rabbit kidney proximal tubule cells in the serum-free medium. 1 nM of ${\beta}-estradiol$ showed a sizable potentiation effect on the growth of the proximal tubule cell in serum-free medium, but higher concentration (> 10 nM) of estradiol indeed inhibited the growth. In the absence of hydrocortisone as a growth supplement in serum-free medium, ${\beta}-estradiol$ caused to potentiate the growth of the cell. In the presence of hydrocortisone, ${\beta}-estradiol$ also potentiated the growth of the proximal tubule cells. According to the Northern analysis, ${\beta}-estradiol$ increased the level of ${\beta}-actin$ mRNA, although mRNA level of the alpha I(IV) collagen was not changed significantly.

The objective of this study was to investigate the effects of ingestion of low-molecular-weight collagen peptides on lipid composition, blood glucose level, and enzyme activities in the serum of hyperlipidemic rats fed experimental diets for 5 weeks. Concentrations of total cholesterol, low-density lipoprotein (LDL)-cholesterol, free cholesterol, triglyceride (TG), phospholipid (PL), and blood glucose, the atherosclerotic index, and the cholesteryl ester ratio were higher in serum of the hyperlipidemic group (CW group), and the cholesterol-plus-collagen peptides extract group (CCP group) than in the control group (BG group basal diet plus water). However, the concentrations of total cholesterol, LDL-cholesterol, free cholesterol, TG, PL, and blood glucose, the atherosclerotic index, and the cholesteryl ester ratio in serum were lower in the CCP group than in the CW group. By contrast, the ratio of HDL-cholesterol to total cholesterol and the absolute HDL-cholesterol level in the CCP group were higher than in the CW group. The activities of alkaline phosphatase (ALP) and aminotransferases (AST, ALT) in serum were lower in the CCP group than in the hyperlipidemic CW group. The results indicate that an extract of low-molecular-weight collagen peptides effectively inhibited increases in lipid elevation, blood glucose level, and enzyme activities, in the serum of hyperlipidemic rats.
