Effects of Different Forms of Chromium Supplements on Serum Glucose, Insulin and Lipids in Rats

  • This study evaluated the effects of different forms of chromium supplements on serum glucose, insulin and lipid concentrations in rats. Sprague-Dawley male rats were randomly assigned to one of three dietary groups and fed AIN-76 semi-purified basal diets supplemented with 300 ppb Cr from Cr methionine (CrMet) and Cr chloride $(CrCI_3)$ or without Cr (control). By the end of the $4^{th}$ week, all rats were decapitated, blood collected, and serum glucose, insulin and lipid concentrations were determined. The CrMet and $CrCl_3$ supplementation did not affect weight gain and feed efficiency ratio. However, feed intake was significantly higher in CrMet groups than control (p < 0.05). CrMet-supplemented rats had markedly increased insulin levels (p < 0.05) compared with controls. Serum lipids were not significantly different between the control and the CrMet groups. $CrCl_3$ supplementation decreased total cholesterol and triglyceride, but the decreases were only significant for the control group. $CrCl_3$ supplementation was associated with significant decreases in total cholesterol compared with CrMet supplementation. These results indicate that CrMet supplementation is effective for increasing serum insulin, and $CrCl_32$ may improve lipid concentrations, because we observed decreased serum total cholesterol and an improved total cholesterol/HDL-cholesterol ratio (THR).

Effects of the Zizyphus jujuba Seed Extract on the Lipid Components in Hyperlipidemic Rats

  • The purpose of this study was designed to observe the effects of Zizyphus jujuba seed extract on the concentrations of the lipids and blood glucose in the S.D. rats fed the experimental diets for 4 weeks. Concentrations of total cholesterol, atherosclerotic index, LDL, LDL-cholesterol, free-cholesterol, cholesteryl ester, triglyceride (TG), phospholipid (PL) and blood glucose in serum were significantly higher in the cholesterol administration groups (group 2 (cholesterol+water), group 3 (cholesterol+Zizyphus jujuba seed extract)) than those in the control group (group 1, basal diet+water). But the concentrations of total cholesterol, atherosclerotic index, LDL, LDL-cholesterol, free-cholesterol, cholesteryl ester, TG, PL and blood glucose in serum ware remarkably lower in the group 3 than those in the group 2. In the ratio of HIDL-cholesterol concentration to total cholesterol and HDL-cholesterol concentration, Zizyphus jujuba seed extract administration group was higher percentage than in the group 2. The activities of aspartate aminotransferase (AST), alanine amino-transferase (ALT), lactate dehydrogenase (LDH) and alkaline phosphatase (ALP) in serum were rather lower in the Zizyphus jujuba seed extract administration group (group 3) than in the cholesterol diet group (group 2). From the above research, Zizyphus jujuba seeds were effective on the improvement of the blood glucose, lipid compositions in serum of dietary hyperlipidemic rats. And particularly, Zizyphus jujuba seeds were more effective as a therapeutic regimen for the control of metabolic derangements in adult disease.

Effects of Red-Yeast-Rice Supplementation on Blood Lipids and Relations among Cholesterol, Bone Markers, and Hormones in Overiectomized Rats (난소 절제 쥐에서 홍국의 첨가가 혈중 지질에 미치는 영향과 혈중 콜레스테롤과 골대사 지표 및 호르몬과의 관련성)

  • This study was performed to evaluate the effect of red-yeast-rice on blood lipids and also to investigated the relation of blood lipids, bone related hormones, and bone mineral density in overiectomized rats. Forty female rats (body weight $210\pm5$g) were divided into two groups. The rats in the first group were ovariectomized which the others received sham operation (SHAM). And then each rat group was further divided into control diet $(casein\;20\%)$ and red-yeast-rice powder supplemented $(0.1\%)$ diet group. All rats were fed on experimental diet and deionized water ad libitum for 9 weeks. Bone mineral density (BMD) and bone mineral content (BMC) were measured using PIXImus in spine and femur. Bone formation and bone resorption were measured by serum osteocalcin and urinary deoxypyridinoline (DPD) crosslinks value. Serum growth hormone, IGF-1 and calcitonin were analyzed using radioimmunoassay kits. The red-yeast-rice group had significantly lower the blood triglyceride concentration and had lower the blood total cholesterol concentration than casein group did in ovariectomized rats. And the red-yeast-rice group than casein group in ovariectomized rats. The red-yeast-rice group had significantly lower the atherogenic index than control group within the ovariectomized groups. In the experimental rats, serum total cholesterol concentration was negatively correlated with femur BMC and crosslink value. Crosslink value was negatively correlated with spine BMC and IGF-1. Osteocalcin concentration was negatively correlated with crosslink value. Growth hormone was negatively correlated with crosslink value and positively correlated with spine BMC. The findings of this study suggest that red-yeast-rices are beneficial for blood lipid in ovariectomized rats. And when considering a population-based public health approach to lowering cholesterol and bone loss, and preventing coronary heart disease and osteoporosis, the red-yeast-rice dietary supplement could provide a new approach for the maintenance of healthier cholesterol and bone.

Effects of Dietary Cu Levels and Kinds of Dietary Lipid on the Lipid Metabolism in Rats (식이내 Cu의 수준과 지방의 종류를 달리 하였을때 흰쥐의 체내 지방대사에 미치는 영향)

  • This study was performed to investigate the effects of different levels of Cu(0,6,60 ppm ) and kinds of lipid(corn oil, sesame oil or butter) in the diet upon lipids metabolism and Cu & Fe contents in weanling rats. The results obtained were summarized as following : 1) Food consumptions, total body weight gains and F.E.R.s showed no significant differences among groups with different dietary Cu levels and the kinds of lipid. 2) Weights of liver, hind limb muscle, kidney, spleen and epididymal fat pad showed no significant differences among groups. 3) The contents of total lipids and total cholesterols in serum, hind limb muscle were significantly higher in low Cu groups than in control and high Cu groups. But the contents of total lipid in liver were significantly lower in low Cu groups than in other groups. 4) The contents of total lipid and total cholesterol in serum, liver and total lipid in muscle were significantly higher in butter groups, but the contents of total cholesterol in muscle were significantly lower in butter groups. 5) The Cu concentrations in serum, liver, muscle and feces were significantly higher in high Cu groups than in control and low Cu groups. The Fe concentrations in serum tended to be lower and those in muscle were significantly higher in low Cu groups.

Effect of Chitosan-Ascorbate Containing Soyfiber Beni-Koji on Body Weight and Lipid Content of Obesity Rats Aid Induced from High Fat Diet (Chitosan-Ascorbate 함유 Soyfiber Beni-Koji의 식이가 고지방 식이로 유도된 비만 흰쥐의 체중과 지방 함량에 미치는 영향)

  • The dietary effects of soy fiber beni-koji(SBCA) containing chitosan-ascorbate on the body weight and serum lipids in obese rats induced by a high fat diet were investigated. The experimental plots(6 rats per each group) were divided into a normal group(NC), a high fat diet group(HF), a 1% SBCA diet group(SBCA1) and a 2% SBCA diet group(SBCA2), and fed for 6 weeks. Compared with the HF group, the mean body weight and the $R\'{o}hrer$ index of the SBCA1 and SBCA2 groups decreased by $3.4{\sim}7.4%$ and $8.1{\sim}13.9%$, respectively. In particular, the body lipids in the SBCA2 group decreased by 42.3%. The serum triglyceride content decreased by approximately $25.54{\sim}27.34%$ in the experimental diet groups. There was no significant difference in the serum total lipids between the HF and SBCA1 groups. On the other hand, the total lipids in SBCA2 group decreased by 19.12% compared with the HF group. In the SBCA2 group, the total serum cholesterol and LDL-cholesterol content decreased by 6.57% and 41.20% compared with the HF group, respectively, while the HDL-cholesterol increased by 10.0%. The risk factor index(RFI) in the SBCA2 group was decreased remarkably by 58.57%.

Effect of Tea Fungus/Kombucha Beverage on Serum and Liver Lipids Metabolism in Rats (발효홍차 음료가 흰쥐가 혈청과 간의 지질대사에 미치는 영향)

  • We investigated the effect of tea fungus/kombucha(TF) beverage on serum and liver lipid metabolism in growing male rats. The rats(Sprague-Dawley) were divided into five groups; control(TF free water), 20 or 40% TF groups(20 or 40% TF in water) and 20 or 40% TFS groups(20 or 40% TF autoclaved in water) according to the level of TF supplementation. The rats were fed ad libitum each of the experimental beverage for 7 weeks. The body weight gain of all the TF and TFS groups were similar to that of the control group. The total lipid and triglyceride concentrations of serum in the 40% TF group were increased more than those in the control group, but those in the 20% TF and 20% TFS groups were similar to that of the control group. The total cholesterol, HDL-cholesterol concentrations, HDL-cholesterol / total cholesterol ratio and atherogenic index of serum in all the TF and TFS groups were similar to those of the control group. The LDL-cholesterol concentration of serum was significantly lower in the 20% TF group than that in the control group. The LDL-cholesterol concentration of serum was significantly lower in the 20% TF group than that in the control group, but those of the 40% TF, 20% TFS and 40% TFS groups were similar to that of the control group. The liver weights of all the TF and TFS groups were heavier than that of the control group. The concentrations of hepatic triglyceride in the 20% TF and 20% TFS groups were significantly decreased, compared with that in the control group. The concentrations of hepatic total cholesterol in all the TF and TFS groups were significantly more decreased than that in the control group. These results indicated that the liver weights of tea fungus/kombucha beverage feeding rats were increased, also the serum triglyceride and total cholesterol concentrations of 40% TF feeding rats were increased. but the serum LDL-cholesterol, hepatic triglyceride and cholesterol levels of 20% TF feeding rats were decreased.

  • This study was done to investigate the effect of adolescent smoking on dietary intakes and nutritional status of serum lipids and antioxidant vitamins. Subjects were 82 somkers whose average pack-year was 0.73, and 85 nonsmokers of male technical high school students in Seoul. Anthropometric measurement was performed and % body fat was also analyzed by Bioelectrical Impedance Fatness Analyzer(GIF-891). Dietary intakes and habits were examined through questionnaires and nutrient intakes were analyzed by Computer Aided Nutritional (CAN) analysis programs. Serum TG and total cholesterol levels were measured by Spotchem sp-4410 and serum HDL-cholesterol levels were measured by test kit. serum vitamin C level was measured by 2,4-dinitrophenylhydrazine method and serum levels of vitamin A and vitamin E were measured by HPLC. All data were statistically analyzed by SAS PC package program. There was a significant difference in body fat percentage and systolic blood pressure while other anthropometric measurements were not different between smokers and monsmokers. Caloric intakes(2335㎉) in adolescent smokers tended to be higher than that of nonsmokers (2,175㎉) but the difference was not statistically significant. Intakes of protein(76.67g) and niacin(16.49㎎) in adolescent male smokers were significantly higher(P<0.05) than those of nonsmokers although other nutrient intakes were not significantly different. Analysis of serum lipids showed that serum HDL-cholesterol level of smokers was significantly different. Analysis of serum lipids showed that serum HDL-cholesterol level of smokers was significantly lower(p<0.05) than that of nonsmokers, whereas other lipid levels were not significantly different. Serum vitamin C level was also significantly lower(p<0.05) in adolescent smokers than in nonsmokers. In addition, serum vitamin E level, which was 7.85㎎/1 in smokers, was lower than that of 9.20㎎/1 in nonsmokers(p<0.05) while serum vitamin A level was not significantly different between the two groups. These results indicate that cigarette smoking in adolescence decreases serum levels of HDL-cholesterol, vitamin C and vitamin E even thoughth their smoking history is very short.   • This study was performed to investigate the effect of chitosan and NOCC from different sources and of different molecular weights on lipid metabolism. Sprague-Dawley rats were blocked into 26 groups according to body weight, and were raised for 4 weeks. Cadmium chloride was given at the level of 0 or 400 ppm in diet. Various chitosan and NOCC sources were given at the level of 0%(w/w) of diet. Total serum cholesterol and serum triglyceride concentrations were little affected by chitosan and NOCC supplements. However, HDL-cholesterol concentration and HDL : total cholesterol ratio were increased , and liver lipid, cholesterol , and triglyceride concentration were decreased by chitosan and NOCC supplements. The cholesterol and lipid lowering activity depends on fiber sources(crab and shrimp); type (chitosan and NOCC); and molecular weight(low , medium , and high). Among cadmium-free groups, chitosan-fed groups showed greater activity than NOCC -fed groups in lowering cholesterol and lipid levels, and greater fecal excretion of lipids and bile acids. Crab chitosans were more effective in cholesterol and lipid lowering activity than shrimp chitosans. The group fed high molecular weight crab chitosan showed the highest fecal excretion of lipids and bile acids. This indicated that high molecular weight crab chitosan was most effective in interfering with cholesterol and lipid absorption.

  • This study was undertaken to investigate effects of alcohol and dietary protein levels on serum lipids and enzyme activities in 15 week-old male rats given a normal diet. Rats were divided into 8 groups : control group (16% protein 16PC) and 8%(8PE) 16%(16PE) and 24% protein groups(24PE) to which was given 5% ethanol mixed into their drinking water after 4 weeks and 10 weeks. Body weight organ weight and various blood components were determined at 4 and 10 weeks. Body weight gain organ weight hemoglobin concentration and hematocrit value were not influenced by ethanol and dietary protein levels. The levels of total cholesterol HDL-cholesterol and phospholipid in serum were not affected by ethanol consumption. Serum triglyceride concen-trations after 10 weeks were significnatly increased ethanol-treated group compared with that of control group and the effect was greater in low protein group than control group. Serum ALP activity was significantly higher in 8PE group than other group but there was no influence by ethanol consumption.

A Study of Nutrient Intake, Serum Lipid and Leptin Levels of Elementary School Students with Different Obesity Index in Kyunggi Area (경기지역 일부 초등학생의 성별과 비만도에 따른 영양소 섭취상태와 혈청 Leptin 및 혈청 지질에 관한 연구)

  • The purpose of this study was to investigate the relationship among nutritional status, serum lipids and leptin of 134 elementary school students with different obesity index residing in Kyunggi-do, Korea. Subjects were assigned to under weight, normal weight or over weight group according to the their obesity index. Their nutritional status, serum lipid and leptin levels were evaluated based on 24-hr dietary recalls, anthropometric measurements and blood analysis. The mean weight, height and obesity index of total subjects was 150.8cm, 45.6kg and 5.4%. The mean % of body fat was higher while LBM and TBW (total body water) were lower in over weight subjects than those of other two groups. Total food, plant protein, carbohydrate, and vitamin C intakes of underweight male students were lower than those of overweight male students. The consumption of total fat was the highest in the female normal weight subjects and the lowest in female overweight subjects. Male students consumed more plant protein and vitamin C than female students. The mean daily intake of vitamin B$_2$, calcium and zinc of all subjects did not meet the Korean RDA. Also, iron intake of female students was lower than the Korean RDA. The mean level of serum leptin was the highest in male and female overweight subjects (p<0.001). The HDL-cholesterol level was the highest in the underweight group of male subjects (p<0.05) and in the normal weight group of female subjects (p<0.001) The serum level of leptin and atherogenic index (AI) of females were higher than those of males. Serum leptin of total subjects had significantly positive correlation with obesity index, weight, SBP, body fat, cholesterol intake, serum triglyceride, and AI. However, serum leptin was negatively correlated with LBM, TBW, intake of plant protein, vitamin B$_1$, vitamin C and serum HDL-cholesterol. Obesity index was also positively correlated with weight, SBP, body fat, serum triglyceride and AI. To summarize the results, the vitamin B$_2$, calcium, zinc, iron and fiber intakes should be supplied to elementary school students, especially those who were overweight. The average serum leptin level of students showed remarkably positive correlation with obesity index, body fat, serum lipids and AI. Also, it was suggested that the serum leptin might be related to the dietary factors like cholesterol, plant protein, vitamin B$_1$ and vitamin C of elementary school students.
