Sample Preparation and Stability of Human Serum and Urine Based on HPLC-DAD for Metabonomics Studies

  • Many literatures focus on the biological relevance and the identification of biomarkers for disease activity assessment while less attention has been paid to the development of standard procedures for sample preparation and storage based on liquid chromatography technique. The influencing factors including protein precipitation, storage temperature, storage time, and reconstitution by ultra pure water were analyzed employing HPLC-DAD. The effects were investigated from five participants over three months by principal components analysis (PCA) and the values of percent changes (PC). The samples with protein precipitation might slow the rate of bacterial enzymatic conversion. After protein precipitation, the average PC of urine samples ($0.136{\pm}0.013$, n = 5) is relatively less than that of the serum samples ($0.173{\pm}0.026$, n = 5) for three months. Minimal effects on metabolic profiles of serum and urine (PC < 0.15) are reasonable for metabolomic studies after protein precipitation and storage at $-20^{\circ}C$ for two months.

효소적 갈변 단백질이 흰쥐 혈청콜레스테롤 농도에 미치는 영향 (Effect of Proteins Modified by Enzymically Oxidized Caffeic Acid on the Concentration of Serum Cholesterol of Rats)

  • 카제인, 대두 단백질에 caffeic acid와 tyrosinase를 첨가한 후 온도 $30{\sim}35^{\circ}C$, pH 6.8에서 5시간 반응시켜 갈변단백질을 조제하였다. 이러한 갈변단백질 및 무처리 카제인, 대두단백질을 단백질 수준 20.0% 되도록 콜레스테롤 무첨가 사료에 혼합, 7주령의 Wistar계 흰쥐 수컷에 14일간 급여하였다. 갈변단백질의 아미노산 cystine의 손실이 눈에 띄는 것 이외 아미노산 손실은 인정되지 않았다. 단백질 소화율은 갈변에 의하여 저하되고, 갈변 카제인의 경우는 현저한 맹장의 비대가 인정되었다. 혈청 총콜레스테롤, LDL-콜레스테롤, 트리글리세라이드 농도는 갈변에 의해 유의차는 인정되지 않았으나, 상승하는 경향이었다. 단백질은 효소적 갈변에 의해 단백질 소화율이 저하되고, 그로 인한 분량(糞量)도 증가하지만, 혈청지질의 저하작용은 없었다.

Zearalenone Affects Immune-Related Parameters in Lymphoid Organs and Serum of Rats Vaccinated with Porcine Parvovirus Vaccine

  • Rats were administered zearalenone (ZEA) via gavage at dosages of 0, 1, 5, and 30 mg/kg for 36 days. On treatment day 8, inactivated porcine parvovirus vaccine (Vac) was injected intraperitoneally. Antibody production against porcine parvovirus was then measured as a function of ZEA treatment. Compared to the vaccine alone, ZEA treatment, with or without Vac, decreased the serum level of IgG. The level of IgM decreased in all ZEA groups at day 22, but the decrease was sustained only in the medium-dose ZEA group at day 36. The level of IgA was unchanged in the Vac only and ZEA groups at day 22, but was decreased in the 5 mg/kg ZEA plus Vac group compared to the Vac only group at day 36. The level of IgE was decreased by all doses of ZEA at day 22, but was unaffected in ZEA plus Vac groups compared to the Vac only group. The levels of IL-1 in the thymus and spleen; INF-${\gamma}$ in serum; IL-2, IL-6, and IL-10 in the thymus; and IL-10 and IFN-${\gamma}$ in the spleen decreased after ZEA administration. Furthermore, the levels of IL-$1{\beta}$ in the spleen and mesenteric lymph node, IL-$1{\beta}$ in the thymus, IL-2 in the thymus and spleen, IL-6 in the thymus, IL-10 and IFN-${\gamma}$ in the spleen, and GM-CSF and TNF-${\alpha}$ in the thymus decreased after vaccination in rats exposed to ZEA. In conclusion, these results suggest that ZEA exposure via drinking water can cause an immunosuppressive effect by decreasing immunoglobulins in serum and cytokines in lymphoid organs.

Human Serum Amyloid A-1 단백질 농도 분석을 위한 CdSe/ZnS 양자점 기반의 Lateral Flow Immunoassay 방법 개발 (Analysis of Human Serum Amyloid A-1 Concentrations Using a Lateral Flow Immunoassay with CdSe/ZnS Quantum Dots)

  • 본 논문에서는 수용성의 CdSe/ZnS 양자점을 합성하고 이에 항체기능성을 도입하여 lateral flow immunoassay (LFIA) 플랫폼에 융합하여 폐암 질병진단에 활용 가능한 단백질 바이오마커[예: 인간 혈청 아밀로이드 A-1 (hSAA1)]의 농도 분석에 적용하고자 한다. 면역분석법 센서 스트립은 니트로셀룰로오즈 막에 테스트라인과 대조라인으로 각각 항hSAA1 단일클론항체(10G1)(anti-hSAA1)와 항chicken IgY (anti-chicken IgY)를 스프레이하여 제작하였다. 이와 함께, 유기상에서 합성된 CdSe/ZnS 양자점은 카르복실기로 변형된 알케인티올기를 이용한 리간드 교환방법으로 수용성으로 전환하였으며, 이에 타겟 단백질인 hSAA1에 특이적으로 결합 가능한 항체인 항hSAA1 단일클론항체(14F8)로 컨쥬게이션하여 형광검출용 입자[QDs-anti hSAA1 (14F8)]로 사용하였다. 제작된 LFIA 스트립 위에 순차적으로 다른 농도의 hSAA1과 QDs-anti hSAA1 (14F8)의 복합체를 흘려주면, 테스트라인에 anti hSAA1 (10G1)/hSAA1/QDs-anti hSAA1 (14F8) 샌드위치 복합체가 형성되어 양자점에 의한 발광신호가 검출됨을 측정하였다. 최적화된 측방흐름이 가능한 완충용액 조건에서 100 nM 농도의 hSAA1 단백질의 유무를 5 min 안에 눈으로 확인 가능하였다.

Anti-hepatotoxic activity of Fruit pulp of Momordica dioica Roxb. (Cucurbitaceae)

  • The Hexane Extract (HE) and Ethyl Acetate Soluble Fraction of the Methanolic Extract (EASFME) of the fruit pulp of Momordica dioica Roxb. (Cucurbitaceae) was evaluated for its anti-hepatotoxic activity in rats. Acute hepatotoxicity was induced by administering paracetamol (2 g/kg, p.o.) for 3 days. The extracts, at a dose of 400 mg/kg (p.o.) administered for 7 days exhibited a significant therapeutic effect by lowering Serum Glutamate Oxaloacetate Transaminase (SGOT), Serum Glutamate Pyruvate Transaminase (SGPT), Serum Alkaline Phosphatase (ALP) and Serum bilirubin and increasing the serum protein levels. These biochemical observations were supplemented by histopathological examination of the liver sections. The activity of extract was also comparable to the standard drug Silymarin, which is a well-known natural anti-hepatotoxic drug.

Aequorin을 표지물질로 사용한 신경전달물질, 세로토닌에 대한 생물발광면역분석법 (Bioluminescence immunoassay for neurotransmitter, serotonin using aequorin as a Label)

  • 본 연구는 생물발광단백질인 aequorin을 표지물질로 사용해 신경전달물질인 serotonin에 대한 생물 발광면역분석법을 처음으로 개발하였다: serotonin-avidin 접합체는 Mannich 법으로 합성하였으며, 최적화된 avidin: formaldehyde: serotonin의 초기 몰 비는 1:12,000:25 이었다. 개발된 serotonin에 대한 생물발광면 역분석법은 좋은 감도(LOD of 0.68 ng/mL)와 넓은 검출 범위($5.0{\times}10^{-10}\;M\sim5.0{\times}10^{-7}\;M$)를 나타내었다(참고, 혈청 내 serotonin의 양은 $151{\pm}45\;ng$/mL 이다.). 교차반응성에 대한 연구 결과, 5-methoxytryptamine은 28.0% 의 교차반응성을 보인 반면, 3-methylindole, melatonin 그리고 5-hydroxylindole-3-acetic acid 은 교차반응성을 보이지 않았다. 또한, 혈청에서의 회수율은 만족할만한 값이 얻어졌다. 그러므로, 본 연구에서 제안된 생물발광면역분석법은 혈청 내의 serotonin 을 모니터링 하는데 좋은 분석법으로 사용될 수 있다.

병원자료에 근거한 혈액 및 혈액화학 검사항목의 참고구간 설정 (Reference Intervals from Hospital-Based Data for Hematologic and Serum Chemistry Values in Dogs)

  • Reference interval is critical for interpreting laboratory results, monitoring response to therapy and predicting the prognosis of the patients in clinical settings. The aim of the present study was to update established reference intervals for routine hematologic and serum chemistry values for a population of clinically healthy dogs (range, 1-8 years) seen in an animal hospital. Blood was obtained by venipuncture while animals were physically restrained, and samples were analyzed for 9 chemistries on MS9-5H (Melot Schloesing Lab, France) and 6 hematology on Vet Test 8008 (IDEXX, USA). Data from 105 dogs (52 males and 53 females) for hematology and 113 dogs (37 males and 76 females) for chemistry were used to determine reference intervals using the parametric, nonparametric and bootstrap methods. Prior to analysis, all parameters were tested for normal distribution using Anderson-Darling criterion. Of the 9 biochemical analytes, alkaline phosphatase, alanine aminotransferase, aspartate aminotransferase, creatinine, total protein, and glucose concentrations did not fit normal distribution for both original and transformed data. All but eosinophil count satisfied normal distribution for either original or transformed data. Parametric method can be used for original cholesterol concentrations, RBC, WBC, and neutrophil counts. This technique can also be used for power-transformed values of blood urea nitrogen concentrations and for logarithm of lymphocyte and monocyte counts. Non-parametric or bootstrap method was the preferred choice for the remaining 7 biochemical parameters and eosinophil count as they did not follow normal distributions. All three statistical techniques performed in similar reference intervals. When establishing reference intervals for clinical laboratory data, it is essential to assess the distribution of the original data to increase the accuracy of the interval, and non-parametric or bootstrap methods are of alternative for the data that do not fit normal distribution.

  • The interaction of a polyoxometal (POM), K6SiW11Co(H2O)O39.10H2O (K6) as a Keggin, with human serum albumin (HSA) was studied by different methods and techniques. Binding studies show two sets of binding sites for interaction of POM to HSA. Binding analysis and isothermal calorimetery revealed that, the first set of binding site has lower number of bound ligand per mole of protein (ν), lower Hill constant (n), higher binding constant (K), more negative entropy (ΔS) and more electrostatic interaction in comparison to the second set of binding site. In addition, differential scanning calorimetery (DSC) and spectrophotometery data showed that, there are two energetic domains. The first domain is less stable (lower Tm and Cp) which corresponds to the tail segment of HSA and another with more stability is related to the head segment of HSA. Polyoxometal also decreases the stability of protein as Tm, secondary and tertiary structure as well as quenching of the fluorescence decrease. On other hand, perturbations in tertiary structure are more than secondary structure.

  • Holstein cows being raised in the Kangweon district were examined on the values of blood chemistry in infertile cows. The results obtained were as follows; 1. Total serum protein was 7.52$\pm$0.45g/100ml 2. Serum albumin was 3.06$\pm$0.28g/100ml 3. In values of serum globulin, $\alpha$1-globulin, $\alpha$2- globulin, $\beta$-globulin and ${\gamma}$-globulin were 0.79$\pm$0.14, 0.60$\pm$0.10, 0.86$\pm$0.13 and 2.21$\pm$0.35ng/100ml, respectively. 4. A/G ration was 0.69$\pm$0.09 5. Total cholesterol in serum was 119.73$\pm$34.52mg/100ml 6. Blood glucose was 55.24$\pm$6.46mg/100ml 7. Serum calcium was 9.27$\pm$0.87mg/100ml 8.
