To investigate antioxidative effects of phenolic compounds separated from persimmon leaves(PL)(Diospyros kaki Thunb.) on the ethanol-induced hepatotoxicity in rat, Sprague-Dawley rats weighing 100-150 g were divided into 5 groups; control group(CON), PL(70 mg/kg) administered group(PEl), ethanol(5 mL/kg, 25%) administered group(ETH), PL(70 mg/kg) and ethanol administered group (PE2), and PL(140 mg/kg) and ethanol administered group(PE3), respectively. The antioxidative activity of persimmon leaves decreased in order of ethylacetate>interphase materials>n-butanol>chloroform>n-hexane>water fraction. The growth rate and feed efficiency ratio decreased by ethanol were gradually increased to the adjacent level of CON by administering PL. The serum activities of ALT, alkaline phosphatase and lactic acid dehydrogenase elevated by ethanol were decreased significantly. It was also observed that the activities of SOD, catalase, and GSH-Px of rat liver increased by ethanol were markedly decreased in PL administered group as compared to ETH. The GSH content of liver was decreased by ethanol, but that was increased in PE1 and PE2 compared with ETH as a dose-dependant manner. These results suggested that phenolic compounds separated from persimmon leaves have a possible protective and relievable effect on the ethanol-induced hepatotoxicity in rats.

This study was performed to investigate the effect of Brassica rapa (BR) sprouts on weight reduction and cholesterol-lowering action in rats fed high fat diet for 4 weeks. Weight-matched male Sprague-Dawley rats were assigned to four groups according to dietary fat levels (10% or 20% of diet wt.). Experimental groups were normal diet group (N), high fat diet group (HF), high fat diet with 5% BR sprouts powder group (HF-BRL), and high fat diet with 10% BR sprouts powder group (HF-BRH). The body weight gain was increased in HF group, but gradually decreased to the corresponding level of the N group fed BR sprouts powder. The concentrations of serum LDL-cholesterol, atherogenic index and cardiac risk factor tended to decrease in the BR sprouts powder fed groups compared with the HF group. However, HDL-cholesterol concentration in serum decreased in the HF group and markedly increased in the BR sprouts powder fed groups. Concentrations of triglyceride and total cholesterol in liver were also markedly decreased in the BR sprouts powder fed groups. Triglyceride concentrations of epididymal and mesenteric adipose tissues in the BR sprouts powder fed groups were also decreased compared with the HF group. These results indicate that BR sprouts powder may reduce fat accumulation and body weight, and have cholesterol-lowering effect.
This study attempted to investigate if the soy isoflavone, genistein, influences bone metabolism in ovariectomized, 4-week-old female Wister rats. All the rats were divided into sham (SH) and ovariectomized (OVX) groups consisting of OVX-17${\beta}$-estradiol($10\;{\mu}g/kg$ b.w.), OVX-1mg or 5 mg or 10mg of genistein/kg b.w. The rates were allowed ad libitum access to foods and water for 8 weeks. The Results showed that body weight had significantly increased in the OVX group compared to the SH group (p<0.05) and was not different among the OVX-GEN and OVX-ES groups and the OVX group. The liver and uterus weights in the OVX groups were slighter than those in SH group (p<0.05). The kidney weight in the OVX groups was decreased compared to in that in the SH group but was not significantly different among all OVX groups. Femoral length in the OVX groups was longer than in the SH group and was not different. Rats in the OVX groups had higher creatinine levels than those in the SH group and hydroxyproline level did not differ significantly among any of the groups. Serum ALP activity and Ca in bone, feces, urine and serum did not change among the groups. Bone mineral density (BMD) decreased in the OVX groups compared to the SH group and was slightly increased by feeding genistein (p>0.05). Breaking force and stiffness did not change by ovariectomy and feeding genistein. Hence, these results suggested that estrogen may affect bone mineralization in growing rats and that genistein be may involved in the prevention of bone loss. However, more studies are needed to identify the proper mechanism of genistein and bone formation.

Among patients who are receiving treatments at an oriental medical hospital for various symptoms and diseases, patients whose main disease is accompanied by metabolic syndrome with abnormal liver function. This research was performed in order to observe the progression of changes in the liver function and serum lipid profile after the oriental medical treatments to patients who have been receiving oriental medical treatment for various other diseases and have been diagnosed as having metabolic syndrome at their first visit to the hospital based on NCEP ATP III diagnosis criteria and WHO Asia Pacific region criteria. Total number of subject patients were 39cases(mean age:55.58${\pm}$2.09 years) which had 20 male and 19 female. For the references for hepatic enzyme levels and blood lipid profile were measured in before treatment and four times after treatments(every 2.31${\pm}$06.17 weeks). Serum AST was 48.86${\pm}$7.46 IU/L before oriental medical treatment. After the treatment, 40.63${\pm}$4.69, 43.12${\pm}$5.46, 37.82${\pm}$4.52 IU/L were measured where although the level decreased to the normal level compared to pre-treatment, the value was not significant statistically(P>0.05). ALT was 66.26${\pm}$11.01 IU/L before oriental medical treatment. After the treatment 62.10${\pm}$8.20, 61.10${\pm}$8.76, 43.79${\pm}$5.68 were measured where although the level decreased, abnormally high level was maintained. The last result was significant statistically(P<0.05) compared to pre-treatment. ALP was 193.06${\pm}$14.20 IU/L before oriental medical treatment. After the treatment, 176.80${\pm}$6.48, 177.46${\pm}$11.81, 162.41${\pm}$9.06 where although compared to pre-treatment the last result was significant statistically(P<0.05), the change was within the normal range. ${\gamma}$-GGT was 87.83${\pm}$12.59 IU/L before oriental medical treatment. After the treatment, progressively near normal level was achieved with 118.73${\pm}$46.45, 85.03${\pm}$17.12, 70.64${\pm}$10.93 and the last result was statistically significant compared to pre-treatment (P<0.05). Blood triglyceride was 217.63${\pm}$32.18 mg/dL before oriental medical treatment. After treatment 215.09${\pm}$22.18, 189.93${\pm}$22.44, 191.22${\pm}$18.51 where abnormal values continued even after treatment although results was not statistically significant compared to pre-treatment(P>0.05). Total-cholesterol was 197.28${\pm}$9.24 mg/dL before oriental medical treatment, after treatment 201.55${\pm}$11.13, 186.87${\pm}$8.77 and 186.68${\pm}$7.61 were measured that results were not statistically significant compared to pre-treatment(P>0.05). HDL-cholesterol was 41.88${\pm}$2.38 mg/dL before oriental medical treatment, after treatment 48.75${\pm}$4.22, 44.10${\pm}$1.91, 48.00${\pm}$2.06 the results were not statistically significant compared to pre-treatment(P>0.05). LDL-cholesterol was 111.66${\pm}$13.08 mg/dL before oriental medical treatment, after treatment 109.94${\pm}$10.18, 101.79${\pm}$8.63, 104.00${\pm}$6.98 the results were not statistically significant compared to pre-treatment(P>0.05). With such results, even if common oriental medical treatments were given to metabolic syndrome patients with abnormal liver function, the liver function was confirmed not to be aggravated, and the concentration of lipids in the blood was confirmed not to be affected in most patients.

The objective of this study was to determine the effects of dietary Zn level on performance, serum Zn concentrations, alkaline phosphatase activity (ALP), and immune response of pigs inoculated with Porcine Reproductive and Respiratory Syndrome virus (PRRSv) and Mycoplasma hyopneumoniae. A $2{\times}4$ factorial arrangement of treatments was used in a randomized design. Factors included; 1) PRRSv and M. hyopneumoniae inoculation (n=36 pigs) or sham inoculation (n=36 pigs) with media when pigs entered the grower facility (d 0) at 9 weeks of age and 2) 10, 50, 150 ppm supplemental Zn sulfate (${ZnSO}_4$) from weaning until the completion of the study, or 2,000 ppm supplemental ${ZnSO}_4$for two weeks in the nursery and then supplementation with 150 ppm ${ZnSO}_4$for the remainder of the trial. The basal diet contained 34 ppm Zn. Pigs were weighed on d 0, 10, 17, 24 and 31 and blood samples were collected on d 0, 7, 14, 21 and 28. Pigs inoculated with PRRSv were serologically positive at d 28 and control pigs remained negative to PRRSv. In contrast, the M hyopneumoniae inoculation was inconsistent with 33.3% and 52.8% of pigs serologically positive at d 28 in the control and infected groups, respectively. A febrile response was observed for approximately one week after inoculation with PRRSv. Feed intake (p<0.01) and gain (p<0.1) were less in PRRSv infected pigs than control pigs for the 31 d study. However, performance did not differ among pigs in the four levels of ${ZnSO}_4$. Assessments of immune responses failed to provide unequivocal influence of either PRRSv inoculation or ${ZnSO}_4$level. These data suggest that PRRSv and M. hyopneumoniae act to produce some performance deficits and the influence of Zn supplementation of nursery age pigs does not have clear effect in grower pigs affected with disease.
Vanadium is an essential trace element but has not been identified with a specific biogical role. To study the direct effects of vanadium on osteoblast, we incubated murin osteoblast-like (MC3T3-El) cells with various corcentration of vanadium oxide & sodium orthovanadate. This study was designed to investigate the effect of vanadium on DNA synthesis, alkaline phosphatase (ALP) activity, cAMP formation responsive to parathormone(PTH) and type I $\alpha$ 2 collagen ribonucleic acid (mRNA) level in murin osteoblast-like (MC3T3-El) cells. The cells were cultured in $\alpha-minimal$ essential medium$(\alpha-MEM)$ supplemented with $10\%$ fetal bovine serum (FBS) and then changed to $0.1\%$ FBS with various concenoation of vanadium oxide & sodium orthovanadate. Quiescent cultured MC3T3-El cells incubated for 24 hours with 2,5,10,15,20 ${\mu}M$ vanadium oxide incorporated $[^3H]Thymidine;$ every concentration showed increases in $[^3H]Thymidine$ incorporations dose dependant manner, the greatest response occurred at $20{\mu}M$. Quiescent cultured MC3T3-E1 cells incubated for 3days with 2,5,10,15,20 ${\mu}M$ vanadium oxide, for 2days with sodium orthovanadate and alkaline phosphatase was assayed with disodium phenyl phosphate as substrate. Vanadium oxide increased the alkaline phosphatase content in MC3T3-El cells at $2{\mu}M\;&\;6{\mu}M$ ; the greatest response occurred at $2{\mu}M$. But decreased at other content sodium orthovanadate increased alkaline phosphatase content in MC3T3-El cells at all concenoation ; the greatest response occurred at $4{\mu}M$. Quiescent cultured MC3T3-El cells incubated for 3days with $5,10{\mu}M$ vanadium oxide , with $5,8{\mu}M$ sodium orthovanadate and cAMP formation was measured by Radioimmunoassay(RIA). Vanadium oxide & sodium orthovanadate showed the tendency of inhibitory effects on cAMP responsiveness to PTH in MC3T3-El cells. Quiescent cultured MC3T3-El cells incubated for 24hours with $10,20{\mu}M$ vanadium oxide, with $5,10{\mu}M$ sodium orthovanadate and Type I $\alpha$ 2 collagen ribonucleic acid (mRNA) expression was studied by Nothern blot analysis. Northern blot analysis of vanadium oxide treated cells showed decreasing effects 0& sodium orthovanadate revealed increasing effects in type I $\alpha$ 2 collagen ribonucleic acid (mRNA) level.

This study was conducted to examine the effect of calcium extracted from salted anchovy (Engraulis japonicus) on the calcium metabolism of rats. Sprague-Dawley male rats were fed low-calcium diets (0.15%) for 2 weeks after the adjustment period. Rats were divided into five groups and were fed experimental diet for four weeks. Experimental diets were low calcium (LC, 0.15% $CaCO_3$), 0.5% $CaCO_3$ (CC), seaweed calcium (SC), calcium lactate (LC), anchovy calcium (AC). The low-calcium diet group (LC) showed the lowest weight gain and had no differences among the groups with adequate calcium intake. Calcium retention was lowest in the LC group and higher in the CL, SC, AC groups than in SC groups. Serum alkaline phosphatase (ALP) level was highest in LC group, and significantly low in the CC and AC groups (p<0.05). Parathyroid hormone and osteocalcin levels showed no differences among experimental groups. The urine deoxypyridinoline (DPD) level was lower in AC and CC groups compared to the LC group (p<0.05). The dry weight of the femur showed no significant differences among normal calcium groups. The bone mineral density of the femur in AC and CC group were significantly higher than the LC group (p<0.05). From these results, calcium extracted from salted anchovy can be useful as a calcium supplement comparable with calcium carbonate.

Anti-obesity effects of ginseng and herbal plant mixtures were investigated to develop natural materials for anti-obesity. After inducing obesity with high fat diet for 8 weeks in male SD rats, ginseng and herbal plant mixtures DM1 (ginseng, puer tea, opuntia) and DM2 (ginseng, puer tea) were administrated orally to rats for another 8 weeks. During administration, food efficacy ratio and body weight of rat were measured twice weekly. After administration, body weight, body fat contents, and serum lipid level were estimated for anti-obesity effect and hematological analysis blood level of ALP and ASP was checked for safety. Body weight in rats fed high fat diet was significantly increased. Body weight in obese rats induced by high fat diet was significantly decreased by DM1 and DM2 feedings. The amount of body fat (epididymal, perirenal and visceral fat, brown adipose tissue) was significantly reduced by DM1 and DM2 treatments. The amount of TG, the concentration of leptin in blood plasma, and the concentration of insulin in blood plasma were significantly diminished by DM1 and DM2. Lipid accumulation on liver was reduced in DM2. There were no side effects among all groups according to blood analysis, hematological findings, and body weight. The findings of this study suggest that DM1 and DM2 may be effective materials for anti-obesity through reducing plasma triglyceride and body fats, and also decreasing body weight without side effects.

Crataegi fructus has been used as an oriental medicine and a folk remedy for the treatment of scurvy, constipation and stomach ailment. This study was aimed to investigate the effect of fermented Crataegi fructus vinegar (FCV) on the improvement of lipid metabolism in rats fed high fat diet (40% kcal% fat, fat source; beef tallow). Sprague-Dawley rats (n=32) were randomly divided into four groups [normal diet (ND), high fat diet (HD), and high fat diet supplemented with low (CFL; 1.5% wt/wt) and high (CFH; 3.0% wt/wt) doses of fermented Crataegi fructus vinegar] and were fed experimental diets for 6 weeks. At the end of the experiment, the body weight in high fat diet groups was higher than that of normal diet group, while the body weights of CFL and CFH group were significantly reduced by 7.2% and 10.0%, respectively, as compared with that of HD group. Moreover, liver and kidney weights in CFL and CFH group were significantly lower than that of HD group (p<0.05). The levels of serum triglyceride, total cholesterol, LDL-C, atherogenic index and hepatic triglyceride, total cholesterol in CFH group were significantly decreased as compared with HD group whereas it increased the serum level of HDL-C than HD group (p<0.05). CFL and CFH groups showed significantly decreased AST, ALT and ALP of serum as compared with HD group. Excretions of fecal saturated fatty acid in CFH group was significantly increased compared with ND and HD groups. These results imply that fermented Crataegi fructus vinegar can be used as possible food resources and functional food materials.
Objectives : Effects of Fructus Foeniculi extract on liver function were investigated in carbon tetrachloride(CCl4) intoxicated rats. Methods : Thirty two male Sprague-Dawley rats with mean weight of $227.28{\pm}7.92g$ were used in these experiments and housed with food and water ad libitum. Fructus Foeniculi extract was administerd at dose 100mg/kg/day and 200mg/kg/day p.o. for 2 weeks after that CCl4 was treated 3 times at dose of 2.5ml/kg, p.o. in alternate day basis. Then serum AFP(${\alpha}$-Fetoprotein), Total protein, Albumin, Triglyceride, Total cholesterol concentrations and ALP (Alkaline phosphatase), AST(Aspartate Aminotransferase), ALT(Alanine Aminotransferase), ${\gamma}$-GT( ${\gamma}$-Glutamyl transferase), LDH(Lactate Dehydrogenase) activities were determined with commercial kit by autoanalyzer. Results : Plasma ${\alpha}$-fetoprotein and total protein concentration showed a tendency to decrease in Fructus Foeniculi extract-treated groups. However, plasma albumin concentration showed no significant differences in all treatment groups. Activity of plasma aspartate aminotransferase and alanine aminotransferase in Fructus Foeniculi extract-treated groups showed a lower value than that of control group. Alkaline phosphatase and lactate dehydrogenase activities showed a tendency to decrease in Fructus Foeniculi treated groups. However, ${\gamma}$-glutamyl transferase activity showed no significant difference in all treated groups. Concentration of plasma triglyceride and total cholesterol showed a high level in CCl4 intoxicated rats but not in Fructus Foeniculi treated groups. Conclusion : Reviewing these experimental results, it appears that Fructus Foeniculi extract have recovering effect against liver injury.
