  • The studies were carried out to investigate the changes in the activities of glucose, ALT, ALP, GGT, cholesterol, $T_3$, and cortisol among the serum when the dogs were administered the prednisolone acetate. The experiments were designed to 3 groups and twenty-four dogs were randomly assigned to 3 groups of 8 dogs each. In groups I, each dog was injected 1mg of prednisolone acetate/kg/day for 14 days, intramuscularly. In groups II, 10mg of prednisolone acetate/kg/day for 14 days. And in the control group, 0.1ml of saline daily for 14 days. The results were summarized as follows: 1. In group I, II and control, mean serum glucose activities remained in the normal range during the study. 2. In group I, mean serum alanine aminotransferase(ALT) activities slowly increased until day 3, than reached the maximum(317IU/l) until day 21, and then decreased in 32.3IU/l until day 40(p<0.005). In group II, reached the maximum(184IU/l) until day 3, and gradually decreased in 140IU/l until day 54(p<0.001). 3. In group I, mean serum alkaline phosphatase(ALP) activities reached the maximum (786IU/l) until day 12 and gradually decreased in 153IU/l until day 47(p<0.001). In group II, reached the maximum(381IU/l) until day 24 and rapidly decreased in the range of control until day 33. 4. In group I, mean serum GGT activities reached the maximum(29.3IU/l) until day 12, and slowly decreased in the range of control until day 47(p<0.005). In group II, reached the maximun(102IU/l) until day 54(p<0.005). 5. Mean serum cholesterol activities of group I decreased than those of control(p<0.05). In group II, decreased than those of control(p<0.05). 6. In group I, mean serum $T_3$ activities reached the maximum(32.8ng/dl) until day 12 and increased in the range of control until day 21(p<0.05). In group II, gradually decreased in 25.1ng/dl until day 12, then decreased under the level of 25ng/dl from day 15 to day 40, and then gradually increased but remained under the range of control(p<0.001). 7. In group I, mean serum cortisol activities slowly decreased and reached the minimum($0.4{\mu}g/dl$) until day 18(p<0.001), and then increased in the range of control until day 47. In group II, rapidly increased and reached the maximum($22.1{\mu}g/dl$) until day 12(p<0.05), but decreased to the minimum($0.64{\mu}g/dl$) until day 40, and then increased in the range of control until day 47.

  • Diets consist of two different pork samples: pork of a Jeju native pig ( 260 days old, $101{\sim}103kg$ ) not fed tangerine byproduct during finishing period ($T_0$), and pork fed 8% and 15% tangerine byproduct during growing and finishing period ($T_1$), respectively. The effects of the diet on the physiological activities of rats were studied by feeding 17-week old rats with the two diets for 4 weeks. There was no significant difference between $T_0$ and $T_1$ in the rat's feed intake, feed efficiency ratio, and weight gain. Furthermore, there was no significant difference between $T_0$ and $T_1$ in the rat's weight of liver, kidney, spleen, epididymal fat pad, triglyceride and cholesterol of liver. Both $T_0$ and $T_1$ showed similar trends in terms of total lipid, phospholipid, triglyceride, total cholesterol, atherogenic index, protein, glucose, hemoglobin level, mineral level, and ${\gamma}$-GTP, ALT, AST and ALP activities. However, $T_1$ showed the trend of increasing amount of the serum's HDL and LDL cholesterol level, compared with $T_0$.

  • The purpose of this study was to investigate the effect of Bojungikgitang(補中益氣湯), Soeumin bojungikgitang(少陰人 補中益氣湯) and its component groups on diabetes in alloxan induced diabetic rats. Sprague-Dawley male rats weighing $200{\pm}10\;gm$ were randomly assigned to one normal, one control and five experimental groups. The experimental groups was classified into to Bojungikgitang(B1 group), Soeumin bojungikgitang(B2 group), Bogihyulyak(B0 group), Siho-sengma(B1+ group) and Gwakhyang- soyeup(B2+ group). Diabetes was experimentally induced by abdomenal injection of 100 mg/kg of body weight of alloxan. Animals were sacrificed at the 6th day and 11th day of diabetic states. The results were obtained as follows: In carbohydrate metabolism, the level of glucose in control group was three-fold higher after alloxan injection than that of normal group. In B1, B2 groups the level of glucose was decreased by 39% and 43% compared with that of control group, and in component groups it was decreased by 73%, 81%, and 80% respectively. In fat metabolism, the level of triglyceride and total cholesterol in serum were lower in B1 and B2 groups with high significant difference and had similar tendency in components groups compared with that of control group. The level of HDL cholesterol on serum were increased in B1 and B2 group with significant difference. And in component group it had similar tendency compared with that of control group. In protein metabolism, the level of total protein and albumin were higher in B1 and B2 group with significant difference, and in component group also increased but with no significancy compared with control group. In kidney function, the level of creatinine group with significant difference, and other compared with control group. In liver function, the level of GOT, GPT, ${\gamma}$-GTP. ALP and bilirubin were lower in B1 and B2 group with significant difference, and in component had similar tendency compared with control group. But in the level of LDH, it was decreased with no significancy. These results indicate that Bojungikgitang(補中益氣湯), Soeumin bojungikgitang(少陰人 補中益氣湯) has strong effect on diabetes and it is useful to prevent diabetes, and in component groups the difference between B1+ and B2+ group was not significant.

  • This study examined the effects of baked garlic powder on the lipid metabolism in rats fed a high-fat/highcholesterol diet for 4 weeks to induce hyperlipidemia. Male Sprague-Dawley rats were assigned to four groups according to the dietary fat, cholesterol and baked garlic powder levels. The experimental groups were normal diet group (N), a high-fat/high-cholesterol diet group (C), a high-fat/high-cholesterol diet with 1.5% baked garlic powder group (GPL) and a high-fat/high-cholesterol diet with 3% baked garlic powder group (GPH). The body weight gain, food intake and food efficiency ratio were similar in the experimental groups. The epididymal adipose tissues weight of the C group was higher than that of the N group, whereas those of the groups fed baked garlic powder were decreased gradually. The ALT and ALP activities were similar in the C groups, but the serum AST and LDH activities elevated by a high-fat/high-cholesterol diet were decreased significantly by feeding a 3% baked garlic powder diet. The serum triglyceride, total cholesterol and LDL-cholesterol levels as well as the atherogenic index and cardiac risk factor tended to decrease in the groups fed baked garlic powder than the C group, whereas the serum HDL-cholesterol level was lower in the C group and remarkably in groups fed baked garlic powder than the control group. The total cholesterol level in the liver and mesenteric adipose tissue and the triglyceride level in epididymal tissue were lower in the groups fed baked garlic powder than the C group. These results suggest that baked garlic powder reduces the serum lipid components and improves the lipid metabolism in hyperlipidemic rats induced with a high-fat/high-cholesterol diet.

  • BACKGROUND/OBJECTIVE: There is intense interest in soy isoflavone as a hormone replacement therapy for the prevention of postmenopausal osteoporosis. A new kind of isoflavone-enriched whole soy milk powder (I-WSM) containing more isoflavones than conventional whole soy milk powder was recently developed. The aim of this study was to investigate the effects of I-WSM on bone metabolism in ovariectomized mice. MATERIALS/METHODS: Sixty female ICR mice individually underwent ovariectomy (OVX) or a sham operation, and were randomized into six groups of 10 animals each as follows: Sham, OVX, OVX with 2% I-WSM diet, OVX with 10% I-WSM diet, OVX with 20% I-WSM diet, and OVX with 20% WSM diet. After an 8-week treatment period, bone mineral density (BMD), calcium, alkaline phosphatase (ALP), tartrate-resistant acid phosphatase (TRAP) 5b, osteocalcin (OC), procollagen 1 N-terminal propeptide (P1NP), and osteoprotegenin (OPG) were analyzed. RESULTS: BMD was significantly lower in the OVX group compared to the Sham group but was significantly higher in OVX + 10% I-WSM and OVX + 20% I-WSM groups compared to the OVX group (P < 0.05). Serum calcium concentration significantly increased in the OVX + 10% and 20% I-WSM groups. Serum ALP levels were significantly lower in the OVX + 10% and 20% I-WSM groups compared to the other experimental groups (P < 0.05). OC was significantly reduced in the OVX group compared to the Sham group (P < 0.05), but a dose-dependent increase was observed in the OVX groups supplemented with I-WSM. P1NP and OPG levels were significantly reduced, while TRAP 5b level was significantly elevated in the OVX group compared with the Sham group, which was not affected by I-WSM (P < 0.05). CONCLUSIONS: This study suggests that I-WSM supplementation in OVX mice has the effect of preventing BMD reduction and promoting bone formation. Therefore, I-WSM can be used as an effective alternative to postmenopausal osteoporosis prevention.

  • Objective: This study was to assess liver involvement in multiple myeloma with the aid of liver function tests. Materials and Methods: A hospital based retrospective study was undertaken using data retrieved of multiple myeloma from the register maintained in the Department of Biochemistry of the Manipal Teaching Hospital, Pokhara, Nepal between $1^{st}$ January, 2007 and $28^{th}$ February, 2012. We collected biomarkers of liver profiles including bilirubin (Total, Direct and Indirect), total protein, albumin, AG ratio, SGOT, SGPT, ALP, ${\gamma}GT$, LDH, ferritin, renal profile and hematological profile. Descriptive statistics and testing of hypothesis were used for the analysis using EPI INFO and SPSS 16 software. Results: Out of 37 cases of multiple myeloma, serum level of AST, ALT, ALP, ${\gamma}GT$ and LDH were increased above the cut-off point in 22 (59.5%), 24 (64.86%), 13 (35.13%), 9 (24.3%) and 11 (29.7%) respectively. The mean values of AST ($65.5{\pm}28.18$ U/L), ALT ($68.37{\pm}29.74$ U/L), ALP ($328.0{\pm}148.4$ U/L), ${\gamma}GT$ ($44.5{\pm}29.6$ U/L) and LDH ($361.7{\pm}116.5$ U/L), total protein ($9.79{\pm}1.03$ gm/dl) were significantly increased when compared with controls. In contrast, albumin ($3.68{\pm}0.43$ gm/dl) and the AG ratio ($0.62{\pm}0.15$) were significantly decreased. Similarly, anemia, hyperuricemia, azotemia, hypercalcaemia and Bence Jones proteinuria were found in 30 (78.9%), 27 (71.1%), 19 (51.5%), 15 (39.5%) and 16 (42.1%) respectively, in cases of multiple myeloma. Conclusions: While clinical manifestation of liver disease among the multiple myeloma was not common, abnormalities in liver function were characteristic.

  • This study was designed to investigate the effect of Codonopsis lanceolata extracts on carbon tetrachloride (CCl₄)induced hepatotoxicity in rats fed high, medium or low fat diet. The liver weight was increased not by the level of fat in diet but by administration of CCl₄. Activities of serum aspartate aminotransferase (AST) and alkaline phosphatase (ALP) were significantly increased by CCl₄, the activities of AST and alanine aminotransferase (ALT) were significantly decreased in case of CCl₄ treatment with C. lanceolata extracts. In high fat diet with CCl₄ administration, the content of total lipid in liver was significantly increased, which was significantly decreased by the administration of C. lanceolata extracts. Administration of C. lanceolata water extracts remarkably prevented the tissue injury such as lipid degeneration, infiltration of inflanunatory cells and necrosis caused by CCl₄, but those methanol extracts did not affect histopathological changes of the liver as water extract. The data suggest that intake of C. lanceolata extracts would prevent lipid accumulation and hepatotoxicity induced by high fat diet and CCl₄ abministratim, and the water extract of C. lanceolata is supposed to be more effective than those methanol extract.

  • [ ${\alpha}-Naphthylisothiocyanate$ ] (ANIT) induces intrahepatic cholestasis, involving damage to biliary epitheial cells. This study investigates hepatic gene expression and histopathological alterations in response to ANIT treatment in order to elucidate early time response of ANIT-induced hepatotoxicity. ANIT was treated with single dose (3, 6, and 60 mg/kg) in corn oil by oral gavage. Serum biochemical and histopathological observation were performed for evaluation of hepatotoxicity level. Affymetrix oligo DNA chips were used for gene expression profile by ANIT-induced hetpatoxicity. Hepatic enzyme levels (ALT, AST, and ALP) were increased in 24 hr high dose group. In microscopic observations, moderate hepatocellular necrosis, were confirmed 24 hr high dose groups. We found that gene expression patterns were dependent on time and dose. Our selected genes were related inflammation and immunomodulation. In this study, ANIT-induced hepatotoxicity was involved in acute phase responses and provides evidence for role of neutrophil could be mechanism associated with ANIT-mediated hepatotoxicity.

  • The effects of mycelium of Cordyceps militaris on the growth, the lipid metabolism, the serum protein levels and the enzyme activities in male rats were studied. Sprague-Dawley rats were given four different types of diets for a succeeding period of five weeks: either a control diet, a control diet supplemented with 2%, 3% or 4% mycelium of Cordyceps militaris (CM) powder. The body weight gain, hepatic weight, feed efficiency ratio and the feed intake of the rats given diets with 2%, 3% or 4% CM were similar to those in rats given the control diet. The concentrations of hepatic total lipid and triglyceride of rats fed the 3% or 4% CM diets were significantly lower than those of rats fed the control diet. But the concentrations of hepatic total cholesterol and phospholipid of rats fed the all CM diets were similar to those of rats fed the control diet. The concentrations of total lipid, total cholesterol, triglyceride phospholipid, and the atherogenic index, and the activities of glutamic oxaloacetic transaminase and lactic dehydrogenase in serum of rats fed the all CM diets were significantly lower than those of rats fed the control diet. No differences were noted in the concentrations of HDL-cholesterol, LDL-cholesterol, total protein, albumin and creatinine, and the activities of glutamic pyruvic transaminase, ${\gamma}$ -glutamyltranspeptidase and alkaline phosphatase in the serum among the rats with on all the experimental diets. These results showed that the all CM diets feeding decreased the total cholesterol, the triglyceride, the phopholipid, and the atherogenic index in serum of rats.

  • Background: In chronic hepatitis B (CHB), the presence of hepatic steatosis (HS) seems to be associated with known host and viral factors which may influence the long-term prognosis of chronic hepatitis B (CHB), probably leading to cirrhosis and hepatocellular carcinoma (HCC). Different from chronic hepatitis C (CHC), factors associated with HS in CHB are not clearly explored. Materials and Methods: 160 CHB patients were divided into two groups depending on the results of liver biopsy. Group I consisted of 71 patients with confirmed steatosis. Group II comprised 89 patients without steatosis. The groups were compared in terms of basal characteristics, body mass index (BMI), liver enzymes (ALT, AST, ALP), serum fasting blood sugar (FBS) and lipids, hepatitis B e antigen (HBeAg), viral load, and histological findings. Results: In terms of host factors, male gender, older age, BMI, high serum FBS and lipid levels were associated with HS. On the other hand, ALT levels, the HAI scores of necroinflammation and stage of fibrosis did not associate with HS. On multivariate analysis, parameters of sex, BMI, cholesterol and FBS levels were independently associated with HS. Regarding viral factors, HBeAg negativity was significantly associated with HS (81.7%, p value 0.006), but not HBV DNA level (p value 0.520). Conclusions: HS in CHB appears to be unrelated to the status of HBV replication. However, fibrosis progression in CHB is related to variable host factors. HS may be enhanced through these factors in HBV chronic patients.
