Protective Effects of Moschus Against Carbon Tetrachloride-Induced Acute Hepatotoxicity in ICR Mice (사향이 CCl4로 유발된 mouse의 간 손상에 미치는 영향)

  • Objectives : This study was aimed at investigating liver protection mechanism of Moschus by inducing liver toxicity through $CCl_4$ in mice and evaluated histological and serological findings. Methods : Experiment groups was categorized into untreated normal group, $CCl_4$ treated control group, and orally administered Moschus experiment group. At the termination of experiment, gross examination of the liver as well as histological findings, and Total protein, Total bilirubin, Direct bilirubin SGOT, SGPT, and ALP contents in the serum were evaluated. Results : 1. For gross examination and histological findings, $CCl_4$ treated control group showed destroyed lobular structure, increased fibrosis, as well as hepatic cirrhosis. For the group treated with Moschus, the lobular structure suffered less damage, and showed lower level of fibrosis and liver cirrhosis compared to the control group. 2. For serum analysis, Total protein were significantly increased in the Moschus experiment group than the control group. 3. Total bilirubin didn't show significant differences between the two groups. but direct bilirubin was significantly increased in the Moschus experiment group than the control group. 4. SGOT, SGPT, were significantly decreased in the normal and Moschus experiment groups compared to the control group. 5. ALP was significantly decreased in the normal group compared to the control group, but Moschus experiment group didn't show significant differences compared to the control group. Conclusion : Taken together, Moschus can be effectively used for recovering the liver functions and further researches must be conducted to verify the efficacies of Moschus bile juice.

Effects of Angelicae gigantis Radix Aqua-acupuncture at Ganshu(BL18) and Qimen(LR14) on Liver Damage induced by Acetaminophen in Rats (간수, 기문혈의 당귀 약침자극이 acetaminophen으로 유발된 흰쥐의 손상간에 미치는 영향)

  • This study was done in order to investigate the protective effects of A.G.R.(Angelicae gigantis Radix) aqua-acupuncture on acetaminophen induced liver damage in rats. The liver damage was induced by acetaminophen (500mg/kg) injection into the peritoneum. The A.G.R. aqua-acupuncture solution was injected into the corresponding loci to Ganshu($BL_{18}$) and Qimen($LR_{14}$) of human body and a blank locus of the root of tail on four consecutive times at 0, 3, 6, and 12 hours after acetaminophen injection. And the serum GOT, GPT, LDH, ALP activities, total bilirubin, direct bilirubin levels were measured in the rats. The serum GOT, GPT, LDH ALP activities and bilirubin level were decreased comparing with that of a control group in case of A.G.R. aqua-acupuncture treated group, specially Ganshu and Qimen aqua-acupuncture treated groups showed an obvious significant decrease.

Effects of the Herbal-Acupuncture with Corni Fructus Extract at Eumgok(KI10) on Osteoporosis in Ovariectomized Mice (음곡(陰谷)(KI10) 산수유 약침이 난소적출 생쥐의 골다공증에 미치는 영향)

  • Objective & Method: The purpose of this study is to investigate the effects of herbal-acupuncture with Corni Fructus extract (CF-HA) at Eumgok(KI10) on osteoporosis in ovariectomized (OVX) ddy mice. I carried out several experimental items to analyze the changes in body weight, uterine weight, uterus index, tibial length, the ash bone weight, tibial BMD, the levels of serum ALP, osteocalcin, Ca, P and the levels of tibial Ca, P, Ca/P ratio, and we performed histological and histomorphological analysis as well. Result: 1. CF-HA at Eumgok(KI10) significantly did not increase the level of bone mineral density in overiectomized mice. 2. CF-HA at Eumgok(KI10) significantly decreased the level of serum phosphorus in ovariectomized mice and sinificantly increased the level of serum calcium in ovariectomized mice. 3. CF-HA at Eumgok(KI10) significantly increased the levels of tibial calcium and phosphorus in ovariectomized mice. 4. CF-HA at Eumgok(KI10) significantly decreased the number of tibial osteoclast like cell in ovariectomized mice. 5. In the histomorphometric analysis of tibia, GPL(growth plate length) was significantly decreased by CF-HA at Eumgok(KI10) in ovariectomized mice. Conclusions: These results suggest that CF-HA at Eumgok(KI10) may have useful therapy effects on osteoporosis in ovariectomized mice.

Effect of Diet with Meat of Crossbred Black Pig Fed with Tangerine Peel on Lipid Metabolism, Protein Level and Enzyme Activities in Rats (감귤부산물을 급여한 제주 개량흑돼지 고기가 흰쥐의 지질대사, 단백질 농도 및 효소활성에 미치는 영향)

  • Diets consist of two different pork samples: pork of a Jeju crossbred black pig not fed with tangerine peel during finishing period $(T_0)$, and pork fed with $6\%\;and\;10\%$ tangerine peel during growing and finishing period $(T_1)$, respectively. The effects of the diet on physiological activities of rats were studied by feeding 17 weeks old rats with the two diets for 4 weeks. The feed intake, weight gain, feed efficiency ratio, and weight of liver, kidney, spleen and epididymal fat pad for the rats were similar among the diets. The total lipid level and triglyceride of liver were similar among $T_0$ and $T_1$. All of the diet groups showed similar trends in terms of the serum total lipid, phospholipid, triglyceride, total cholesterol and HDL-cholesterol level, and atherogenic index, hemoglobin level, and $\gamma-GTP$, ALT, AST and ALP activities. However, it was found that the cholesterol level of liver and the LDL-cholesterol of serum in $T_1$, was significantly lower than those in $T_0(p<0.05)$.

Effects of Guisinhwan on the Ovariectomized Rat Model of Postmenopausal Osteoporosis (귀신환이 난소적출 백서의 골다공증에 미치는 영향)

  • Aging is characterized by an age-dependent reduction in bone density. Pronounced bone loss by postmenopausal estrogen deficiency induces a bad effect on dynamic balance of bone metabolism. When the bone density is reduced to the level below the fracture threshold, the risk for fracture is greatly increased. The purpose of this study is to examine what are the effects of the Guisinhwan(GSH : 歸腎丸) on the ovariectomized rat model of postmenopausal osteoporosis. The results of the experiment are as follows: Body weight in control group showed significant increase in comparison with sham, but that in GSH-treated showed no change in comparison with control. The level of serum albumin in control group showed significant decrease in comparison with sham. That in GSH-treated was slightly decreased in comparison with control. The level of serum ALP activity in control group showed significant increase in comparison with sham, but that in GSH-treated was significantly decreased in comparison with control. The level of serum phosphorous, calcium, GOT, GPT, T3 did not significant change among the three groups. The level of serum estrogen in control group showed slightly decreased in comparison with sham, but that in GSH-treated showed no change in comparison with control. The level of serum T4 in control group showed significant increase in comparison with sham, but that in GSH-treated was significantly decreased in comparison with control. Trabecular bone area as well as trabecular thickness in control group showed significant decrease In comparison with sham. Those in GSH-treated showed significant increase in comparison with control. Trabecular number did not significant change among the three groups. Trabecular separation only in GSH-treated showed significant decrease in comparison with control. Osteoclast number in control group showed significant increase in comparison with sham. That in GSH-treated showed significant decrease in comparison with control. Oseoblast surface in control group showed significant increase in comparison with sham, but that in GSH-treated showed no change in comparison with control. According to the above these results, GSH has shown to be capable of preventing and curing osteoporosis caused by old-aged and postmenopause.

Effect of Dietary Addition of Turkish Propolis on the Growth Performance, Carcass Characteristics and Serum Variables of Quail (Coturnix coturnix japonica)

  • We examined the effect of dietary Turkish propolis and flavomycin on growth performance, carcass characteristics, internal organ weights and some serum variables in quail (Coturnix coturnix japonica) birds. One hundred and fifty day-old quails were randomly divided into five groups, with ten replicate pens per treatment and three birds per pen. One group received the basal diet (antibiotic-free), the control. The flavomycin at 10 mg/kg diet and propolis at 0.5, 1 and 1.5 g/kg diet were added to the basal diet. Body weight gain, feed consumption and feed efficiency were determinated weekly. Carcass characteristics, internal organ weights and serum variables were determinated at the end of the study (35 day). The results showed that body weight gain, feed efficiency and carcass weight were improved significantly (p<0.01) when compared to control group for birds fed diets containing propolis and flavomycin between 14 to 35 days. The addition of 1 g/kg propolis to the diet resulted in significantly (p<0.01) better-feed efficiency as compared to control and other treatment groups. There were no significant differences in carcass yield, abdominal fat, liver gizzard, proventriculus and intestinal weight and intestinal pH among the groups. In addition, serum ALP, total protein, uric acid, cholesterol and triglyceride were not influenced by the any supplementation. However, birds fed with propolis tended to have higher serum HDL and lower level than birds fed the control diet. In conclusion, supplementation of propolis and flavomycin during the growth period showed similar effects on growth performance in quail. Therefore, it can serve as a natiral substitute for antibiotics in poultry diets.

Effect of Powder of Small Water Dropwort (Oenanthe javanica DC) and Brewer's Yeast(Saccharomyces cerevisiae) on the Liver Function and Serum Lipid Metabolism in Alcohol-Consumed Rats (돌미나리 가루와 맥주 효모가 알코올을 섭취한 흰쥐의 간 기능 및 혈청 지질대사에 미치는 영향)

  • This study investigated the effect of the powder of the small water dropwort(PSWD) and brewer's yeast(BY) on the liver function and serum lipid metabolism in alcohol-consuming rats. Male Sprague-Dawley rats were fed an AIN-93 diet(control), ethanol plus control(A), ethanol plus PSWD(AS), ethanol plus BY(AB), ethanol plus PSWD and BY(ABS) diet for seven weeks. The feed intake, body weight gain and feed efficiency ratio were the lowest and in the alcohol-consuming groups. The weight of the liver, kidney, spleen and epididymis of the anatomized rats showed positive correlation with the body weight gain. Compared with group A, the content of the lipid in the serum was significantly low in the AS and AB groups. In particular, it was the most effective in the AB group. The GOT, GPT and ALP level in the serum showed a significant difference in the alcohol-consuming groups. Compared with the A group, they showed a significantly low difference in the AS and AB groups. The total cholesterol and triglyceride levels in the liver were similar in all groups. The weight of the feces was significantly different but there was no significant difference in the content of total cholesterol and triglyceride levels in the AS and AB groups. These results suggest that BY and PSWD improve the liver function, and had an effect on reducing the lipid content of the serum and feces of alcohol-consumed rats. In particular, the effect of BY which contained protein, dietary fiber and vitamin B was higher than that of PSWD.

The Experimental Study on the Quinaldine(2-Methylquinoline) Toxicity for a Long Anaeathetization Period (장기 마취시 어류에 미치는 quinaldine 독성에 대한 기초 연구)

  • This study was taken to induce either anaesthesia or sedation for the purpose of applying to transport of live fish safely. 7 species of fish in addition to Tilapia mossambica were exposed to 250 ppm concentration of the anaesthetic quinaldine to determine the safe level for handling and transportation of these species. The results obtained are as follows ; 1. The time taken to lose balance increased with a decrease on the concentration of the anaesthetic. 2. Anaesthetization must be carried out under temperature lower or higher rather than optimum temperature. 3. The longer the length of the fish, the longer the anaesthetization time and recovery time of fish. 4. Coefficient of recovery period and body length is 0.78. 5. At 10-15 min. after anaesthetization, the serum levels of glucose, ALP and SGOT were at peat. 6. LDH of the anaesthetized fish is much more increased than that of the unanaesthetized. 7. In the more 250 ppm treatment, the pyknosis of the brain and spleen tissue appeared.

Effects of Low Level Laser Treatment at LR2 and LR8 acupoint on the liver damage induced in D-GalN in rats (간경(肝經)의 형화혈(滎火穴)과 합수혈(合水穴)에 시술한 레이저침이 D-GalN 간손상 유발 흰쥐에 미치는 영향)

  • Objectives : This study was performed to investigate the effect of invasive laser acupuncture treatment at Liver Brook (LR2) acupoint and Liver Sea (LR8) acupoint on liver damage induced by D-galactosamine (D-GalN) in rats Methods : Liver damage was induced by D-GalN. The experimental rats were divided into two groups (control group, Low Level Laser Treatment (LLLT) group). Control groups were classified into small groups. Intact group had no liver damage and no treatment. D-GalN group was induced liver damage induced by D-GalN and not treated. LLLT group were induced liver damage induced by D-GalN and then treated at the LR2 or LR8 acupoint with 532, 658, 904 nm invasive laser acupuncture. The treatment was carried out three days at a time for 15days at both acupoints. To examine mechanism of the effect of invasive laser acupuncture, we measured the contents of ASP, ALT, ALP, TBIL in serum, CBC in blood and SOD in liver tissue. Results : The change of body weight increased in all groups. That change was AST and ALP, the AST activity decreased significantly compared with the control groups and decreased by 532 nm and 904 nm both LLLT groups. But ALP increased at LR8 acupoint by 658 nm. TBIL level significantly decreased in all LLLT groups. The SOD of LLLT groups increased in the liver tissue of rats compared to the control groups. SOD activity indicated that LLLT can help cellular defense mechanism by preventing scavenging hydrogen peroxide. In the change of WBC, it was increased in D-GalN Control group compared to intact group and LLLT groups. Conclusions : These results suggested that invasive laser acupuncture treatment at LR2 or LR8 acupoint reduced activation of hepatic enzyme and damage of liver tissue. Thus, the effect of invasive laser acupuncture was nearly identical to the way of the traditional acupuncture for the treatment of hepatocytotoxicity.

Hepatoprotective effect of electroacupuncture at GB34 (Yangreungcheon) in $CCl_4$-intoxicated rats (양릉천 저주파 전침자극이 $CCl_4$로 유발된 흰쥐의 간 손상에 미치는 영향)

  • Objectives : To investigate the effect of electro-acupuncture (EA) at GB34 on hepatotoxicity in $CCl_4$-intoxicated rats. Methods : Rats were injected with $CCl_4$ and treated with acupuncture or 2 Hz electro-acupuncture (EA) at left GB34 three times a week for 10 weeks. A non-acupoint in left gluteal area was selected as a sham point. To estimate the effects of EA on hepatotoxicity in rats, body weight, liver weight and liver index were measured, and biochemical assays for serum ALT, AST, ALP and total cholesterol, and hematological analysis for RBC, WBC, PLT, hemoglobin, lymphocytes, neutrophils and monocytes, and histology analysis of liver tissue were performed. Results : 1. Lymphocyte level in blood was significantly decreased by $CCl_4$-intoxication and significantly increased by acupuncture and 2 Hz EA at left GB34. 2. Neutrophill and monocyte level in blood was increased by $CCl_4$-intoxication and significantly reduced by acupuncture and 2 Hz EA at left GB34. 3. Acupuncture and 2 Hz EA at left GB34 significantly reduced serum ALT and AST which were increased by $CCl_4$-intoxication. 4. EA at GB34 significantly reduced serum ALT and AST as compared with EA at sham point in $CCl_4$-intoxicated rat. 5. No significant difference was found between the effects of acupuncture and that of 2 Hz EA on $CCl_4$-induced liver damage in rats. Conclusions : 2 Hz EA at GB34 has hepatoprotective effects on $CCl_4$-induced liver damage in rats and the point-specificity of GB34 may be involved in these effects.
