골격근 손상에 대한 '사향서활정'(麝香舒活精)치료의 혈청 CK, LDH활성도 및 골격근 ${\alpha}-actin$ mRNA 발현 변화의 관찰 (Effect of 'Sexiang Shuhuo Jing' for CPK, LDH Activities and Skeletal Muscle ${\alpha}-actin$ mRNA Expression after Skeletal Muscle in Rats)

  • The study examined clinical effect of the 'Sexiang Shuhuo Jing' on serum CK and LDH activities and skeletal muscle ${\alpha}-actin$ mRNA expression concentration 140days after skeletal muscle injury in rats. The clinical research consisted of observing and measuring the serum CK, LDH activities and skeletal muscle ${\alpha}-actin$ mRNA expression, at the time of injury and during recovery. All experimental data were analyzed by repeated measurement with ANOVA on of SPSS(11.5v), accepting level for all significances was above ${\alpha}\;=.05.$ The results were as follows: That skeletal muscle injury in rats there existed a substantial increase serum CK, LDH activities and expression of skeletal muscle ${\alpha}-actin$ mRNA And Sexiang Shuhuo Jing treatment group's serum CK, LDH activities lower and faster recovery than control group. The 1 st day after skeletal muscle injury, Sexiang Shuhuo Jing treatment group's skeletal muscle ${\alpha}-actin$ mRNA expression was much more higher than control group, after 2 day's faster recovery normal level than control group. There existed a substantial increase again serum CK, LDH activities and skeletal muscle ${\alpha}-actin$ mRNA expression 3rd days after injury in control group. But in Sexiang Shuhuo Jing treatment group's can't be found that.

국내 동물원에서 사육 중인 일본원숭이의 혈액상 (Hematological analyses of Japanese macaque (Macaca fuscata) housed in Everland Zoological Gardens)

  • Hematological and serum biochemical values were assessed from 20 clinically healthy Japanese Macaques raised in Everland Zoological Gardens and compared to the International Species Information System (ISIS) reference range that is used internationally as standard for wildlife animals. Taking our standard on sexual maturation at age 4, tRBC values in Macaques under age 4 were significantly lower than those over age 4, but the Hb and PCV values were significantly higher. Compared to the ISIS standard, the tRBC values in Macaques under age 4 were significantly lower whereas the Hb and MCHC values were significantly higher. Moreover, in the samples of Macaques over age 4, the PCV and MCV values were significantly lower than the ISIS standard. On serum biochemistry values the creatinine and amylase values in the Macaques under age 4 were significantly lower than those over age 4. In comparison with the ISIS standard, the values of ALT, ALP, BUN, IP, $Ca^{2+}$ and $K^+$ in the Macaques under age 4 did have no significant difference. The values of TP, GGT, tBil, amylase, TG and UA were significantly higher than the ISIS standard, but the values of albumin, AST, glucose, creatinine, cholesterol, CPK, LDH, $Na^+$ and Clwere significantly lower. In contrast, the values of TP, albumin, ALT, ALP, creatinine, cholesterol, amylase, TG, IP and $Na^+$ in the Macaques over age 4 did have no significant difference, but the values of GGT, BUN, tBil, UA and $Ca^{2+}$ were significantly higher, while the values of AST, glucose, CPK, LDH, $K^+$ and $Cl^-$ were significantly lower. On the other hand, there was no significant difference in hematological and serum biochemical values between the groups of male and female.

신경근전기자극이 흰쥐 골격근의 미세구조에 미치는 영향 (Effect of Neuromuscular Electrical Stimulation(NMES) on the Ultrastructure of Skeletal Muscle in Rats)

  • This study conducts electrical stimulation to male white rat of Spargue-Dawley which is 7 weeks, has the weight of 240 g and is seemingly healthy for one or two weeks by means of neuromuscular electrical stimulator in order to examine the effects of neuromuscular electrical stimulation on its gastrocnemius, measures change of weight of gastrocnemius, serum and enzyme activity and then obtains the following conclusions. There is little difference in AST and CPK of weight and serum of gastrocnemius after one or two weeks of conducting neuromuscular electrical stimulation in all experimental groups. On the one hand, as a result of histochemical observation, NMES I group showed hypertrophy of perimysium and increase of sectional diameter of muscle fiber compared to comparison group, but NMES II group showed a similar result to comparison group. When ultrasubstructure was observed under electron microscope, I-type muscle fiber of NMES I group showed well-arranged mitochondria and it was similar to comparison group. II-type muscle fiber showed a large quantity of glycogen granules within sarcoplasmatic and the extension of luminal of T-tubule. I-type muscle fiber of NMES II group had small mitochondria and showed the vacuolar degeneration of mitochondria and extended T-tubule. II-type muscle fiber showed the extension of agranule cytoplasma reticulum with T-tubule and the reduction of amount of glycogen granule within partial sarcoplasmatic.

죽력(죽초액)이 유영운동으로 유발된 피로에 미치는 영향 (The Effect of BCL (Bambusae Caulis in Liquamen) on Fatigue Induced by Swimming Exercise)

  • Objectives : Bambusae Caulis in Liquamen (BCL) has been used for stamina fortification in Oriental Medicine for thousand years. The goal of the present study was to investigate the muscle antifatigue effect of BCL. Methods : The mice were divided into three groups. The control group was allowed to swim for 10 minutes without BCL infusion. For the preventive effect of BCL, another group referred to BCL-1 was fed with BCL an hour before the swimming exercise. BCL-2 group for the reversion effect was forced to swim for 10 minutes at first, then fed with BCL followed by half an hour rest and another 10 minute swimming exercise. All the serum samples collected by heart puncture were used for the measuring biochemical factors such as LDH, CPK, glucose, lactate, triglycerides and total cholesterol. Results : In the BCL-2 group, lactate and LDH were significantly reduced compared to the control. BCL infusion during the exercise was effective in metabolizing LDH, reducing the conversion ratio of pyruvate into lactate. In the BCL-1 group, it was not effective in antifatigue function. The level of glucose was significantly reduced in BCL-1 group compared to the control. When the BCL was infused before the exercise, it is assumed that glycogenolysis seen during the exercise was prevented. When the BCL was infused during the exercise, the triglycerides and total cholesterol were increased significantly compared to the control. BCL seems to cause the increase of lipolysis to utilize the fat as an energy source. Unlike other parameters, CPK did not show any changes from BCL infusion. Conclusions : Based on the above results, BCL is found to be involved in energy metabolism. Further studies are needed to find out the underlying mechanism of BCL for its effect on stamina fortification.

운동 후 지연된 근통종과 근손상의 지표에 관한 연구 (A study on the Delayed Muscle Soreness after Exercise and the Indicators of Muscle Damage)

  • The purpose of this study was to analyze the delayed muscle soreness after exercise and the indicators of muscle damage. Subjects of this study were 9 male students. Some muscle enzymes and others(GOT, LDH, ALD, CPK in serum, WBC, ESR) that known as the indicators of muscle damage were measured before exercise. After measurements they run on a treadmill ($incline:25\%$, speed;3.5miles/h) for 7 min. on an average. The intensity of this kind of exercise was $90.5\%$ of maximal exercise intensity. After exercise, same indicators were measured at the just after exercise, 24hr after, 48hr after, 72hr after. Also muscle soreness level was evaluated at same intervals by make use of Modified Abraham's scale. The result was as follow: There was no relationships between muscle soreness and the indicators of muscle damage but ALD was the most important indicator that can be explains the muscle soreness very well.

강아지에서의 골막섬유육종 증례 (A Periosteal Fibrosarcoma in a Puppy Dog)

  • A 4 months old Golden retriever male dog was referred. By the history taking, it was kwon that the mass has been formed at the parietal region of the patient after fourth prevention inoculation. The appetite, urination and excretion were normal. The mass confirmation which was limited in the parietal region, did not seem to be enlargement of lymph node from physical examination palpation. Complete blood count and serum biochemical profiles were consistent with mild lymphocytosis, mild monocytosis, anisocytosis, regenerative anemia and elevated ALP, CPK. Survey radiographs indicated a mixed density in parietal region. We performed surgical resection and chemotherapy for cure and biopsy. Histological finding was periosteal fibrosarcoma. The application of cisplatin was topically used in operation site. Up to now the indication of recurrence is not visible.

개에서 외부고정법을 이용한 양측성 하악골절의 치유 일례 (A Case of External Skeletal Fixation for Treatment of Bilateral Mandibular Fracture in a Dog)

  • A 5-year-old, male mixed breed dog was presented to Gyeongsang National University Animal Hospital for bilateral mandibular open fracture by bite. The dog had loss of the teeth and injury of the oral mucosa. Radio-graphic and blood examinations were conducted. Fracture lines were observed between first premolar and second premolar, but temporomandibular luxation was not observed on radiographs. In serum analysis, elevated levels of CPK and LDH were found. The dog was anesthetized and prepared for aseptic surgery. Acrylic external skeletal fixator with Kirschiner wire was applied. Pin loosening and malalignment of the right fracture line was observed at 7 weeks after surgery. We applied Kirschner-Ehmer fixator and half pins during the second surgical procedure. At 4 weeks after a second surgical procedure, the fixator and pins were removed from mandible. The fractures were healed with no complications.

화학검사 결과의 검증을 위한 용혈 지수 일치화 도구 개발 (Harmonization of Hemolysis Index in Clinical Chemistry Laboratory and Its Application as a Result Verification Tool)

  • 용혈 지수(hemolysis index)는 생화학 장비에서의 용혈을 위한 반정량 지표이다. 용혈 지수의 특성이 상업 플랫폼마다 다르기 때문에 용혈 지수의 표준화 또는 일치화는 현재 되어 있지 않다. 진단검사의학에서 일치화(harmonization)는 측정 절차와 상관없이 동일한 결과와 동일한 해석을 할 수 있는 능력을 말한다. 임상 환자의 혈액 중에서 무작위로 추출한 40개의 검체를 인위적으로 체외 용혈(in vitro hemolysis)시킨다. 혈액 자동화 장비인 XE-2100 (Sysmex, Japan)에서 측정된 혈청 헤모글로빈을 기준으로 검체의 혈청을 0~300 mg/dL 사이에서 20 mg/dL 차이로 각각을 희석시킨다. 희석된 혈청 헤모글로빈을 생화학 자동화 장비 Hitachi-7600 (Hitachi, Japan)으로 측정하여 용혈 지수와 혈청 헤모글로빈을 평가하였다. 용혈 지수와 혈청 헤모글로빈은 선형성을 보였으며 용혈 지수 1은 대략 혈청 헤모글로빈 20 mg/dL 정도였다. 용혈 거절 기준을 결정하기 위해 용혈 지수를 세 그룹으로 나눈다: 용혈 지수 0~1, 용혈지수 4~6, 용혈 지수 9~15. 또 다른 임상 검체 40개의 검체를 Hitachi-7600 (Hitachi, Japan)로 각각의 임상 검사 종목을 측정한 후, 각각의 검체를 주사기의 피스톤을 앞 뒤로 움직여 인위적인 체외 용혈을 시키고 Hitachi-7600 (Hitachi, Japan)로 측정하였다. 세 그룹 사이의 임상 검사 종목의 용혈로 인한 백분율 차이(percentage difference)를 ANOVA 또는 Kruskal-Wallis test 분석하였다. 사후 검정에서 용혈 지수 0~1인 그룹과 용혈지수 5~6인 그룹 사이에 유의한 차이가 나는 종목들은 Glucose, creatinine, total protein, AST, direct bilirubin, uric acid, phosphorus, triglyceride, LDH, CPK, magnesium, potassium이였다. 많은 임상 검사 종목들이 용혈지수 4~6인 그룹과 참조 그룹인 용혈지수 0~1인 그룹 간에 통계적으로 유의미한 차이를 보이기 때문에 용혈 기준은 대략 용혈지수 5 (혈청 헤모글로빈 100 mg/dL)가 적합하다 판단된다.

녹용 에탄올 추출물이 2,3,7,8 Tetrachlorodibenzo-p-dioxin에 노출된 랫드의 일반 혈액 화학 지수 및 조직 병변에 미치는 효과 (Effect of Antler Velvet Ethanol Extract on Common Serum Chemistry Panels and Histopathological Change in Rats Exposed to 2,3,7,8-Tetrachlorodibenzo-p-dioxin)

  • 본 연구에서는 TCDD에 노출된 랫드에 있어 녹용의 에탄올 추출물(EAV)이 혈액상(complete blood count) 및 임상화학지수(common serum chemistry panels) 및 장기별 조직 병변에 미치는 효과를 구명하기 위하여 수행되었다. 본 연구 결과 TCDD에 노출된 랫드의 혈액상 및 혈액화학적 지수 중 혈소판 수 감소(p<0.01), AST(p<0.01) 및 ALT (p<0.05) 활성 증가, CPK (p<0.01) 및 LDH(p<0.05) 활성 감소, BUN 활성 증가(p<0.05), glucose 함량 감소(p<0.05), lipase 활성 증가(p<0.05), TG(p<0.01) 및 LDH-cholesterol(p<0.05) 함량 증가가 대조군에 비하여 유의하게 변화한 것으로 관찰되었다. 특히 혈소판 수 감소, serum lipase 활성 증가, abdominal lipoprotein lipase(LPL) 활성 저하 및 male에서 serum estrogen 함량 증가는 동물 종과 관계없이 매우 재현성이 있게 나타나는 독성 반응으로 본 실험에서도 대조군 대비 현저한 차이를 보였다. 본 실험에서 TCDD-노출 랫드의 체중 감소는 복부 지방 조직내 LPL 활성 저화와 serum에 존재하는 HSL의 활성 증가와 관련이 있을 것으로 사료된다. 또한 장기별 무게에 있어서도 TCDD는 고환의 무게를 현저하게 감소시켰다(p<0.01). 한편, 녹용의 에탄올 추출물(EAV)은 TCDD에 노출된 랫드에 있어 LDH의 활성 감소(p<0.05), glucose 함량 감소(p<0.05), lipase 활성 증가(p<0.05), 고환 무게 감소(p<0.01)에 대하여 유의하게 억제하는 것으로 관찰되었다. TA군의 흉선은 거의 퇴화되어 CO 흉선 무게의 3.8%에 불과하였다. TCDD에 노출된 랫드에 EAV를 전처리하면 TA군과 비교하여 비록 무게 감소는 유의하게 억제되나 CO군과 비교하면 현저하게 작은 값을 나타냈다(p<0.01). 이상의 결과로 보아 녹용은 랫드에 있어 다이옥신에 의하여 야기되는 일반 혈액화학지수 및 조직 병변에 미치는 효과를 개선시킬 수 있다고 판단한다.

Streptozotocin 유도 당뇨성 흰쥐의 혈당, 전해질 및 지질대사에 삼백초 추출액이 미치는 영향 (The Effects of Saururus chinensis Baill Extract Administration on the Blood Glucose, Electrolyte and Lipid Metabolism in STZ-Induced Hyperglycemic Rats)

  • Streptozotocin(STZ 55 mg/kg BW., I.P. injection)으로 유발된 당뇨성 Sprague Dawley계 수컷 흰쥐에 있어서, 삼백초 생리활성물질 추출액(3.5 g% extract)의 섭취에 의한 혈당, 전해질, 혈청 지질개선효과 및 당질대사 이상 등에 관여하는 효소의 활성 변동을 생리생화학적 측면에서 검토하기 위하여 본 실험을 수행하였다. 기본식이만을 급여한 대조군인 Normal군을 비롯한 STZ 당뇨 유발군(NSW군), 당뇨 유발에 삼백초 잎 추출액을 섭취시킨 군(NSSL군) 및 당뇨 유발에 삼백초 뿌리 추출액을 섭취시킨 군(NSSR군)을 5주간 실험 사육하였다. 혈당 농도는 당뇨 유발군에 삼백초 추출액을 급여하므로서 유의적으로 저하됨을 관찰할 수가 있었다. 또한 혈청 총 콜레스테롤, 동맥경화지수, LDL, LDL- 콜레스테롤, 유리 콜레스테롤, 콜레스테롤 에스테르 비, 중성지방 및 인지질 농도 등은 삼백초 추출액을 섭취시킨 군(NSSL군, NSSR군)에서 농도가 유의성 있게 감소되는 것으로 나타났다. 한편, HDL-콜레스테롤 및 총 콜레스테롤에 대한 HDL-콜레스테롤 비 등은 삼백초 추출액 섭취(NSSL군, NSSR군)에 의해서 유의성 있게 증가 되는 것으로 나타났다. 혈청 중의 전해질(Na, K, Cl) 및 creatinine 농도는 STZ으로 당뇨 유발군에 삼백초 추출액을 섭취시킴으로서 유의성 있는 농도 저하 효과를 보였다. 또한, 혈청 중 aminotransferase(AST, ALT), creatine phosphokinase(CPK), lactate dehydrogenase(LDH) 및 alkaline phosphatase(ALP)의 활성은 STZ으로 당뇨 유발 시킨 후 삼백초 추출액을 섭취시킴으로서 저하되는 것으로 나타났다. 한편, 삼백초 잎(NSSL군)과 뿌리(NSSR군) 추출액 섭취에 따른 혈당, 각 종 지질 성분 및 효소 활성치의 두 군 간의 비교는 삼백초 잎이 뿌리 추출액에 비하여 다소 효과적인 차이를 보였으나 유의성 있는 변화는 보이지 않았다. 이상과 같은 결과들을 미루어 볼 때, 삼백초 중의 생리활성물질이 STZ으로 유발된 당뇨성 흰쥐의 혈당 조절 기능이상 및 혈청 지질대사 이상 등에서 오는 각종 질환의 예방 및 개선 작용에 효과가 있을 것으로 생각된다.
