쥐 Sertoli 세포돌기의 주사전자현미경적 연구 (Scanning Electron Microscopic Study of the Sertoli Cell Processes in the Rat)

  • The three-dimensional structure of the Sertoli cell in the rat was investigated by scanning electron microscopy. Morphologically, seven types of Sertoli cell processes were evident : Shrot, flat and ramified processes are projected from the lateral side of the basal portion of Sertoli cell. Leaf-like processes are attached to the surface of spermatocytes and spermatids. Slender cord-like processes, flat and irregular shaped processes, sucker-like processes and club-like processes are observated in the middle and apical portion of seminiferous epithelium. The sheet-like processes rest upon more than one-thirds of the surface of each spermatogonium, spermatocyes and spermatids located in the proximity of the Sertoli cell. All Sertoli processes are originated from Sertoli cell column. Just before spermiation, the processes which are attached to the head of maturation spermatid are eliminated. Though the mechanism for elimination of residual body is not known, these observations segget that the Sertoli cell process are thought to have a reciprocity with the germ cells.

한우 Sertoli 세포의 주사전자현미경적 연구 (Scanning Electron Microscopic Study of the Sertoli Cell in the Korean Native Bull)

  • The three-dimensional structure of the Sertoli cell in the Korean native bull was investigated by scanning electron microscopy. Morphologically, four types of Sertoli cell processes were evident: 1) sheet-like processes, 2) sleeve-like processes, 3) bough-like processes and 4) finger-like processes. The sheet-like processes rested upon more than half of the surface of each spermatogonia, spermatocyte and spermatid. Sleeve-like processes, bough-like processes and finger-like processes are observed in the middle and apical portion of seminiferous tubule. All Sertoli cell processes are originated from Sertoli cell column. Just before spermiation, the apical sheet-like processes are shifted from their position at the spermatid head, and bough-like processes covered the disengaged residual body, after which the residual body was no longer evident in the tubule. Though the mechanism for this elimination is not known, the process suggests a reciprocity between the Sertoli and germ cells.

세르톨리세포 관찰을 위한 PA-TCH-SP-PD 염색의 적용 (Application of Periodic Acid Thiocarbohydrazide Silver Proteinate Physical Development ( PA-TCH-SP-PD) Stain to Observation of Sertoli Cell)

  • 본 연구는 Sertoli 세포 column 과 Sertoli 세포돌기 미세구조의 연구를 위해 당질검출을 위한 PA-TCH-SP, -PD 투과전자현미경용 특수염색을 정세관에 적용한 후 각 정세포와 Sertoli 세포를 관찰하고 비교하여 이 분야 연구에 본염색법의 적용 가능성을 검토하였다. PA-TCH-SP, -PD 특수염색을 개의 정세관에 적용하여 투과전자현미경으로 관찰하였던 바, Sertoli 세포 column 과 돌기 부위에 수많은 반응과립이 관찰되었으며 정세관 기저부의 정조세포질에서도 약간의 반응과립이 관찰되었다. 이와 같은 반응과립은 각 정세포의 핵, 정모세포질, 정자세포질 및 잔류체에는 존재하지 않아 반응과립이 존재하는 Sertoli 세포 colunm 과 돌기 및 정자세포질에 침투된 돌기들을 명확히 구분할 수 있었다. 따라서 PA-TCH-SP, -PD 염색법은 투과전자현미경하에서 Sertoli 세포 colunm 과 돌기의 형태학적 및 기능학적 연구에 대단히 유용한 염색법으로 사료된다.

진도견 정세관의 정세포와 Sertoli 세포내 glycogen의 분포 (Glycogen distribution of germ cells and Sertoli cells of seminiferous tubules in Jindo dog)

  • In an effort to obtain basic data of carbohydrate metabolism during spermiogenesis of the sexually-matured Jindo dog, the glycogen distribution in the testis was investigated by light and transmission electron microscopy. Periodic acid thiocarbohydrazide silver proteinate physical development(PA-TCH-SP-PD) staining method provided better results in the detection of glycogen granules from Sertoli cells and germ cells than the periodic acid schiff(PAS) staining method did. Pre-treatment of the tissue sections with ${\alpha}$-amylase elicited a significant decrease in PA-TCH-SP-PD stained granules, which suggested that the stained granules were of glycogen origin. High concentration of the glycogen granules were observed in the Sertoli cells, especially in its column, sheet-like processes, club-like processes, and tubular processes. The glycogen granules were unevenly distributed in some Sertoli cell columns. These results strongly indicated that the Sertoli cells of Jindo dogs showed vigorous activity of carbohydrate metabolism.

진도견 정자형성계 세포들의 Lectin-binding patterns (Lectine-binding patterns of spermatogenic cells in the Jindo dog)

  • The lectin-binding patterns in the testis of the sexually matured Jindo dog were investigated to study the distribution of glycoconjugates in the seminiferous tubule under light and transmission electron microscopy. Positive reactions to Wheat germ agglutinin(WGA) and Dolichos biflorus agglutinin (DBA) were observed in the Sertoli cell and in the residual body of spermatid with a stronger reaction in the Sertoli cell to the lectins than in the residual body. Strong reactions to Soybean agglutinin(SBA) and Peanut agglutinin(PNA) were observed in the acrosome vesicles of the Golgi- and cap-phase spermatid, while a moderate reaction was observed in the acrosome-phase, maturation-phase spermatid and the residual body. The acrosome area of the spermatid reacted intensively to Griffonia simplicifolia agglutinin( GS-I) when the cell was in the acrosome-phase and maturation-phase, and the same reaction to the GS-I was observed in the residual body. However, the seminiferous tubule did not react to Ulex europeus agglutinin I(UEA-I). The gold-labelling of the Sertoli cells with DBA resulted in positive reactions of the Sertoli cell column and processes when observed under the electron microscopy, while the Golgi-, cap- and acrosome-phase spermatids reacted positively to SBA in the peripheral low-dense area of the acrosome vesicle of spermatid. Based on these results, we concluded that differences in the lectin-binding pattern of the seminiferous tubules were recognized in the Jindo dog compared to other animals.

Functions of somatic cells for spermatogenesis in stallions

  • Spermatogenesis and testis development are highly structured physiological processes responsible for post-pubertal fertility in stallions. Spermatogenesis comprises spermatocytogenesis, meiosis, and spermiogenesis. Although germ cell degeneration is a continuous process, its effects are more pronounced during spermatocytogenesis and meiosis. The productivity and efficiency of spermatogenesis are directly linked to pubertal development, degenerated germ cell populations, aging, nutrition, and season of the year in stallions. The multiplex interplay of germ cells with somatic cells, endocrine and paracrine factors, growth factors, and signaling molecules contributes to the regulation of spermatogenesis. A cell-tocell communication within the testes of these factors is a fundamental requirement of normal spermatogenesis. A noteworthy development has been made recently on discovering the effects of different somatic cells including Leydig, Sertoli, and peritubular myoid cells on manipulation the fate of spermatogonial stem cells. In this review, we discuss the self-renewal, differentiation, and apoptotic roles of somatic cells and the relationship between somatic and germ cells during normal spermatogenesis. We also summarize the roles of different growth factors, their paracrine/endocrine/autocrine pathways, and the different cytokines associated with spermatogenesis. Furthermore, we highlight important matters for further studies on the regulation of spermatogenesis. This review presents an insight into the mechanism of spermatogenesis, and helpful in developing better understanding of the functions of somatic cells, particularly in stallions and would offer new research goals for developing curative techniques to address infertility/subfertility in stallions.

A Role of Plasminogen Activators in Animal Reproductive Cells and Organs

  • Plasminogen activators (PAs) are serine proteases that convert plasminogen to plasmin. Two type of PAs are urokinase-type PA (uPA) and tissue-type PA (tPA). Plasminogen is present in most extracellular fluids. PAs play in various reproductive processes including implantation, ovulation and fertilization. In the spermatozoa, PAs and PAIs play a role in sperm motility and fertilization. PAs in the sertoli cell are stimulated spermatozoa maturation and sperm activation through the phospholipase A2. The oocyte maturation is the process for fertilization and implantation. PAs in cumulus-oocyte complexes (COCs) are related to oocyte maturation by protein kinase A and C. In the ovulatory process, PAs activity are changed and it are related to reducing the tensile strength of ovarian follicle wall. The uterine environment is important for reproduction and the uterus undergo tissue remodeling. In the uterus and oviduct of mammals, expression and activity of PAs are changed during estrous cycle. Thus, expression and activity of PAs are concerned to many reproductive functions. Therefore, PAs seem to important factor of regulator in reproductive events.
