제주지역 한우의 요네병 감염실태 조사 (Prevalence of Johne's disease of Korean native cattle in Jeju Province, Korea)

  • The bacterium causing Johne's disease (JD), Mycobacterium avium subspecies paratuberculosis (MAP), is distributed worldwide and causes progressive intestinal disease, also known as paratuberculosis, in domestic and exotic ruminants, including cattle. The subclinical form of this disease results in progressive weight loss, reduced milk production, lower slaughter value, and premature culling, with possible impacts on fertility and udder health. Eventually, infection can lead to the clinical form that manifests chronic diarrhea, emaciation, debilitation, and eventual death. JD is a major cause of economic loss in cattle industry. In this study, serological testing was conducted by ELISA to determine the prevalence of MAP in Korean native cattle of Jeju during the duration of 2010 to 2013. A total of 5,150 blood sera were collected in 111 farms and the samples were classified into four different age groups for MAP; group 1 (2 years or less), group 2 (2~4 years), group 3 (5~7 years), and group 4 (over 8 years). Overall seroprevalence of MAP was 5.2% (269/5,150), and annually from 2010 to 2013, they were 11.6% (106/915), 2.1% (10/474), 5.5% (89/1,627), and 3.0% (64/2,134), respectively. Also, the prevalence rates was determined by age groups: 48 of group 1 (17.8%), 154 of group 2 (57.3%), 48 of group 3 (17.8%), and 19 of group 4 (7.1%) were seropositive for MAP. Although seropositive samples were found in all age groups, the highest seroprevalence was found in group 2. This study showed that during the investigation the seroprevalence of MAP tended to decrease.

  • The present study investigated serological and molecular prevalence of Mycoplasma gallisepticum (MG) and Mycoplasma synoviae (MS) infection in unvaccinated broiler breeder farms in Jeonbuk providence. An enzyme-linked immunosorbent assay (ELISA) and polymerase chain reaction (PCR) had been used to determine antibody titers against MG and MS, and genome of these pathogens, respectively. Seventy five percent of farms were seropositive for MG and 94% of farms were seropositive for MS. In addition, the rate of antibody positive flocks against MG were 65.3% (32/49), while the rate of positive flocks against MS were 84.2% (80/95). The geometric mean antibody titers were $802.2{\pm}626$ and $27,726.7{\pm}2426$ against MG and MS, respectively. Interestingly, none of samples was positive for MG genome by PCR, while 94% (farms), 82% (flocks) and 62.6% (broiler breeder) were positive for MS genome by PCR. These findings suggest that the prevalence of MG or MS infection could be higher than expected. Thus, strict prevention program including vaccination and environmental sanitation should be implemented to avoid disease transmission from breeder to broilers as well as transmission among broilers.

돼지 생식기호흡증후군 바이러스의 항체분포 및 역학조사 (Seroprevalence and epidemiological analysis of porcine reproductive and respiratory syndrome virus in Korea)

  • A nation wide sero-epidemiological survey of porcine reproductive and respiratory syndrome(PRRS) was carried out to analyze the current status of the PRRS virus infections in the field using the indirect immunofluorescent antibody assay(IFA) with the field isolate PL96-1. Since the first report of the antibody detection to PRRSV in 1993, the prevalence of seropositive pigs has increased dramatically and the data indicate that over 21% of the pigs and around 60% of the farms showed seropositives to the PRRS virus. A slightly higher positive rate was recognized in breeders than fattenings and it might be due to the higher age at the time of testings. No significant regional differences were detected in the sero-epidemiological survey. Higher sero-positive rate in growers indicates that PRRSV infection in the field was common after weaning(around 40 days). However, the number of seropositive pigs were declined in fattening pigs. Sows showed around 26% of sero-positive rate that there is a higher chance of continuous virus circulation in the infected farms. Low rate of sero-positivity in boars(9.8%) implies that there is high demand in proper control measures to prevent virus spreading through breeding procedures such as natural or artificial insemination. Therefore it was concluded that PRRSV infection in domestic swine herds is endemic and the positive rate and economic loses will be increased by spontaneous infections in naive farms.

경남 지역내 소 Neospora caninum에 대한 감염률 조사 (Seroprevalence of antibodies to Neospora caninum in Korean indigenous cattle in Gyeongnam central area)

  • This survey was carried out to investigate the seroprevalence of antibodies to Neospora caninum in Korean indigenous cattle that was raised in central area province Gyeongnam, Korea. A total of 719 sera were tested for N. caninum antibodies using ELISA (Herdcheck anti-Neospora, IDEXX Laboratories Inc., Westbrook, Maine USA). Seroprevalence of individual and farm were 29.8%(214/719) and 53.2%(50/94). Regional seropositive rates of the samples were 61%(47/77), 23.3%(7/30), 13.8%(49/355), 37.6%(77/205), 65.4%(34/52) at Changwon, Jinhae, Gimhae, Miryang, Yangsan, respectively. It showed difference at the age and on the herd size of farms. A herd of cattle above the age of 5 was more infective than under 4 years. And in seroprevalence by herd size farms having under 30 heads was top(35.7%). Seropositive 214 herds of N. caninum antibidies were tested for brucellosis by test tube. Positive rate of double infection was 16.4%(35/214).

  • The prevalence of Coxiella burnetii in native Korean goats in Jeonbuk province was investigated using ELISA (sera) and PCR (vaginal mucus). A total of 798 blood samples from 189 farms were collected in 2013 and 2015. Seroprevalence was 13.6% in 2013 and 15.7% in 2015. Tracking survey of six seropositive farms on the prevalence of C. burnetii was conducted. 137 (41.4%) out of 331 goats were seropositive and 47 (17.6%) out of 267 goats were positive in PCR. The higher seropositivity observed in adult goats and female goats significantly, the seroprevalence of C. burnetii was higher as age increased (<1 y=13.2%~${\geq}5$ y=100%)(P<0.05) and female goats (44.8%) was higher than male goats (24.1%) on the seroprevalence (P<0.05). 21 (7.9%) goats positive in PCR was seronegative. The prevalence of C. burnetii in native Korean goats in slaughter house was 30.0% in ELISA and 11.5% in PCR. Significantly, female goats (62.8%) was higher than male goats (12.3%) on the seroprevalence (P<0.05). Based on these data, C. burnetii was easily exposed in domestic animals: native Korean goats-related areas such as farms and slaughter house.

Incidence and sero-surveillance of feline viruses in Korean cats residing in Gyeonggi-do

  • Incidences of major feline viral diseases provide basic information for preventing viral disease in cats. Despite the growing interest in feline viral diseases, sero-surveillances have been lacking. In this study, we analyzed the diagnoses of feline viral diseases and conducted a sero surveillance of feline panleukopenia virus (FPV), feline calicivirus (FCV), feline herpesvirus-1 (FHV-1), and feline infectious peritonitis virus (FIPV) in Korean cats. Of the 204 confirmed cases since 2015, the numbers of diagnoses for FPV, FIPV, FCV, feline influenza virus, and FHV-1 were 156, 32, 12, 3, and 1 case, respectively. In total, 200 sera, collected between 2019 and 2021, were screened for the presence of antibodies against FPV, 2 FCVs, FHV-1, and FIPV using a hemagglutination inhibition test and a virus-neutralizing assay (VNA). The overall seropositive rates in cats tested for FPV, the 2 FCVs, FHV-1, and FIPV were 92.5%. 42.0%, 37.0%, 52.0%, and 14.0%, respectively. A low correlation (r = 0.466) was detected between the VNA titers of 2 FCV strains. The highest incidence and seropositive rate of FPV reveal that FPV is circulating in Korean cats. The low r-value between 2 FCVs suggests that a new feline vaccine containing the 2 kinds of FCVs is required.

SARS-CoV-2 IgG Antibody Seroprevalence in Children from the Amritsar District of Punjab

  • The majority of the children experience milder coronavirus disease 2019 (COVID-19) symptoms. Children represent a significant source of community transmission. Children under 18 years of age account for an estimated 4.8% of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) infections globally. However, no conclusive statements pertaining to the multi-fold aspects of the virus in children could be drawn due to the lower prevalence of pediatric cases. The present study was conducted to identify the indirect impact of SARS-CoV-2 infections on developing herd immunity among children in the age group 3 to 18 years by investigating their antibody levels. In the study, 240 children aged 3~18 years were recruited by the Department of Pediatrics, Government Medical College and Hospital, Amritsar, India, and quantification of the antibodies was performed at the Viral Research and Diagnostic Laboratory (VRDL), Government Medical College (GMC), Amritsar, India. Out of the 240 serum samples, 197 (82.08%) showed seropositivity, while 43 (17.92%) were seronegative. When stratified, it was observed that in the age group 3~6 years, 22.33% of children were found to have anti-SARS-CoV-2 antibodies while in the age groups 7~10 years, 11~14 years, and 15~18 years, respectively, 37.06%, 30.46%, and 10.15% were seropositive. Although there was seroconversion among children which was useful for predicting the next wave, no differences in seropositivity were observed between adults and children.

Comparison of blood parameters according to fecal detection of Mycobacterium avium subspecies paratuberculosis in subclinically infected Holstein cattle

  • Background: Mycobacterium avium subspecies paratuberculosis (MAP) causes a chronic and progressive granulomatous enteritis and economic losses in dairy cattle in subclinical stages. Subclinical infection in cattle can be detected using serum MAP antibody enzyme-linked immunosorbent assay (ELISA) and fecal polymerase chain reaction (PCR) tests. Objectives: To investigate the differences in blood parameters, according to the detection of MAP using serum antibody ELISA and fecal PCR tests. Methods: We divided 33 subclinically infected adult cattle into three groups: seronegative and fecal-positive (SNFP, n = 5), seropositive and fecal-negative (SPFN, n = 10), and seropositive and fecal-positive (SPFP, n = 18). Hematological and serum biochemical analyses were performed. Results: Although the cows were clinically healthy without any manifestations, the SNFP and SPFP groups had higher platelet counts, mean platelet volumes, plateletcrit, lactate dehydrogenase levels, lactate levels, and calcium levels but lower mean corpuscular volume concentration than the SPFN group (p < 0.017). The red blood cell count, hematocrit, monocyte count, glucose level, and calprotectin level were different according to the detection method (p < 0.05). The SNFP and SPFP groups had higher red blood cell counts, hematocrit and calprotectin levels, but lower monocyte counts and glucose levels than the SPFN group, although there were no significant differences (p > 0.017). Conclusions: The cows with fecal-positive MAP status had different blood parameters from those with fecal-negative MAP status, although they were subclinically infected. These findings provide new insights into understanding the mechanism of MAP infection in subclinically infected cattle.

만성 B형 간염 바이러스 감염 환자에서 혈청 Cytokine에 관한 연구 (A Study of Serum Cytokines in the Patients with Chronic Hepatitis B Virus Infection)

  • 목 적: 혈청 B형 간염 바이러스 표면 항원(HBsAg)이 양성인 만성 보유자 35명에서 혈청 Th1계 싸이토카인(IL-2, TNF-${\alpha}$ 및 IFN-${\gamma}$)과 Th2계 싸이토카인(IL-4와 IL-10)을 측정하여 만성 B형 간염 바이러스 감염 환자의 병태에 따른 싸이토카인의 변화를 구명하고자 하였다. 방 법: 대조군은 항 HBs 항체 양성인 정상 소아 7명이었고 환자군 35례중 HBeAg 양성군은 20례, 항 HBe 항체 양성군은 15례이었으며 간 조직검사를 시행한 19례중 만성 지속성 간염은 10례, 만성활동성 간염은 9례이었다. 말초 정맥혈로부터 혈청을 분리하여 aminotransferase, B형 간염 바이러스 표지자 검사 및 ELISA로 싸이토카인(IL-2, TNF-${\alpha}$, IFN-${\gamma}$, IL-4 및 IL-10)을 측정하였다. 결 과: 1) HBe 항원 양성인 환자에서 항 HBe 항체 양성인 환자에 비해 그리고 만성 활동성 간염 환자에서 지속성 간염 환자에 비해 의의 있게 혈청 AST와 ALT치가 높았다. 2) HBe 항원 양성인 환자에서 항 HBe 항체 양성인 환자에 비해 IL-10이 의의 있게 높았다. 3) 만성 활동성 간염 환자에서 지속성 환자에 비해 통계학적인 의의는 없었으나 측정한 모든 싸이토카인이 높은 경향을 보였다. 4) 혈청 아미노전이 효소치가 100 U/L 이상인 환자에서 IL-2, TNF-${\alpha}$, IFN-${\gamma}$ 및 IL-10이 높은 경향을 보였고 혈청 아미노전이 효소치가 정상인 환자에서 효소치가 높은 환자에 비해 IL-4가 높은 경향을 보였다. 결 론: Th2계 싸이토카인인 IL-10의 증가는 간염의 활성도보다는 HBe 항원의 지속성과 관련이 있고 Th1계의 싸이토카인인 IL-2, TNF-${\alpha}$ 및 IFN-${\gamma}$는 간염의 활성도 증가와 관련이 있음을 시사하였다.

광주, 전남 지역 소아의 A형 간염 항체 보유율에 대한 연구 (An Epidemiologic Study on the Seropositive Rate of Hepatitis A Virus in Children of Gwangju and Jeonnam)

  • 목 적: 소아 특히 영유아는 A형 간염에 감염되어도 증상이 대부분 없기 때문에 다른 사람들에게 전파시키는 원인이 된다. 위생상태의 개선으로 A형 간염바이러스에 노출될 기회가 적어지면서 항체가 형성되지 않아 20~30대 젊은 세대에서 A형 간염이 급격히 증가하고 있으며 최근 대유행의 조짐을 보이고 있다. 저자들은 광주. 전남 지역 소아의 A형 간염 항체 보유율을 조사하여 역학적 특성을 이해하고 예방대책 기준을 수립하기 위한 기초자료를 얻고자 본 연구를 시행하였다. 방 법: 2009년 1월 1일부터 2009년 12월 31일까지 조선대학교병원 소아청소년과에 입원한 18세 이하의 환자 중 수혈이나 면역억제요법을 받은 적이 없는 광주, 전남지역에 거주하는 소아 1,435명(남아 788명, 여아 647명)을 대상으로 IgG anti-HAV 검사하였다. 연령대는 1세 미만, 1세~5세 미만, 5세~10세 미만, 10세~15세 미만, 15세 이상으로 구분하였으며 1세 미만은 개월 수로 세분화하여 비교하였다. 결 과: IgG anti-HAV 양성은 40.8%였으며, 연령에 따른 항체 양성률은 1세 미만은 41% (107/261), 1세~5세미만 49.9% (264/236), 5세~10세 미만 51.1% (180/352), 10세~15세 미만 12.9% (30/232), 15세 이상 8.2% (5/61)로 10세 이상에서 현저하게 감소하였다. 1세 미만을 세분화 하여 조사한 결과 양성률이 1개월 미만은 47.6%였으며 이후 30% 이상의 양성률을 보이다가 7개월경에 감소하기 시작하여 9개월에 14.3%로 감소하였다. 성별에 따른 양성률은 남아 40.6% (320/788), 여아 41.1% (266/647)로 였고 성별 간의 의미 있는 차이는 없었다. 거주지에 따른 비교는 광주 42.5% (415/976), 전남 37.3% (171/459)로 광주지역이 높았다. 결 론: 소아에서 IgG anti-HAV 양성률은 40.8%였으나 10세 이상에서 12.9%로 현저히 감소하여 이들이 성인이 되는 경우 현증 A형 간염의 집단 발생 가능성이 높아질 것으로 생각되며 A형 간염에 대한 적극적인 예방접종이 필요할 것으로 판단된다.
