Staphylokinase production mediated by lysogenic conversion in Staphylococcus aureus isolated from bovine mastitis (젖소 유방염유래 Staphylococcus aureus에서 용원변환에 의한 staphylokinase 산생)

  • Staphylokinase is seldom formed by animal Staphylococcus aureus strains. In this study, 76(72.4%) of 105 Staph aureus strains isolated from bovine mastitis were found to produce a staphylokinase. These staphylokinase producing strains were tested for lysogenic conversion by means of delysogenization procedure and isolation of serotype B and F converting phages. By the application of delysogenization method comprised of UV irradiation and acriflavine treatment to 76 staphylokinase-positive strains, the delysogenized cells could be observed in 29(38.2%) of the strains and delysogenization rates in 16(55.2%) of 29 delysogenized strains were 0.9% or less. A total of 7 serological group F phages were isolated from 76 staphylokinase-positive strains, and these phages could be again divided into three groups by the immunity reaction. Of 7 serotype F phages, 2 were isolated from the original lysogenic strains producing colonies of delysogenized cells after delysogenizing treatments and 5 were isolated from strains in which delysogenized cells were not observed after delysogenizing treatments. Difference of sensitivities to serological group F phages between original lysogenic strains and strains from which delysogenized cells were not isolated after delysogenizing treatments was not observed These data suggest that staphylokinase production of the remaining 42 strains might be also mediated by lysogenic conversion.

Serotaxonomical Analyses of Some Streptomyces Strains Using Antibodies against Cell Envelope (균사 외피 항체를 이용한 Streptomyces 속 균주들의 혈청학적 유사성 분석)

  • The similarities among five strains of Streptomyces were measured by the serological methods. Antigens were prepared by dissolving cell envelope fractions in Tween 20 buffer and antisera were produced from the immunized rabbits. Immunodiffusion studies and ELISA results showed that the degree of antigen-antibody reaction was not exactly matched to the taxonomic distance; i. e. the strains of the cluster group A exhibited low level of cross-reactions each other, while relatively strong cross-reactions were observed between Streptomyces lavendulae of cluster group F and Streptomyces viridochromogenes of cluster group A.

  • The diagnosis of human hydatidosis is primarily made using radiological and serological methods. Radiological methods are generally of low specificity and serological methods lack sensitivity, especially for pulmonary disease. In this study the capabilities of a new rapid test, the hydatid antigen dot immunobinding assay (HA-DIA), which was developed for the diagnosis of pulmonary hydatidosis, were studied and compared with another immunodiagnostic method, indirect hemagglutination (IHA). The study subjects included 18 patients, 9 women, 9 men; range 7 to 63 years; mean 30 years, with surgically proven pulmonary hydatidosis, a control group comprised of 14 patients; viral respiratory infections (1), cirrhosis (2), connective tissue disease (2), taeniasis (3), and 6 healthy donors. We found that the HA-DIA test had a sensitivity of $67\%$ and specificity of $100\%$, and that the IHA test had a sensitivity of $50\%$ and specificity of $100\%$. We conclude that HA-DIA is a simple, rapid, low cost assay that does not require instrumentation and has a higher sensitivity than IHA for the diagnosis of pulmonary hydatidosis.

A Serological Study on Toxoplasma gondii Infection Among People in South of Tehran, Iran

  • Although Toxoplasma gondii infection generally shows subclinical courses in adults, it may have severe pathologic effects on the fetus. For a survey of toxoplasmosis in Shahr-e-Rey area, south of Tehran, Iran, we conducted a serological study on 1,187 general populations using indirect fluorescent antibody (IFA) test. In this cross sectional study, cases were selected randomly by a local primary health care (PHC) system. Blood samples were collected in microcapillary tubes and sera were examined by IFA after centrifugation. It has been shown that 68.3% of cases in all age groups had IFA titer of 1:10 and higher, which we consider as positive cases, and the rate for 1:400 or over was 30.2%. There was meaningful correlation between anti-Toxoplasma antibody titers and age, sex, and occupation of subjected people (P < 0.05%). The incidence was higher in females and highest in the children age group (10-14 years old). The present study demonstrates high prevalence of Toxoplasma infection among the people in south of Tehran, Iran.

  • Myocarditis refers to inflammation, necrosis, or myocytolysis that may be due to many infectious, connective tissue and many other causes affecting the myocardium or involvement of the endocardium or pericardium. The most common manifestation is congestive heart failure, although arrhythmias and sudden death may be the first sign of myocarditis. Viral myocarditis is typically a sporadic but occasionally epidemic illness, noted as an acute potentially fulminant disease of 1-to 4-wk-old infants, as an acute but more benign myopericarditis of toddlers and young children. The most common casuative agent in viral myocarditis is Coxsackievirus and the outcome of the biopsy-proven chronic dilated cardiomyopathy associated with Coxsackievirus is poor without therapy. Myocarditis may be confirmed by percutaneous endomyocardial biopsy and the viral myocarditis may be diagnosed by the serological viral study with the clinical manifestations. He was admitted for the management of tachyarrhythmias occurred suddenly without prodromal symptoms and signs and diagnosed as viral pancarditis by serological Coxsackievirus study, echocardiogram, chest x-ray, EKG and other clinical manifestations.

Rearing managements of pig farms and survey on pneumonia of slaughtered pigs (양돈장 사양관리와 도축돈 폐 병변조사)

  • This studies were carried out to investigate the rearing managements of pig farms and survey on pneumonia of the slaughtered pigs from 5 selected herds located in Jangsu, Jeonbuk. Isolated aerobic microorganisms from pneumonic lung were examined antibiotic susceptibility and tested serological antibody titers of the herd base. Prevalence rate of pneumonia were examined according to rearing and health management conditions of pig farms. Prevalence rate of pneumonia were detected in 78.8% and enzotic pneumonia. In 47.7%, pleuropneurnonia in 31.1%. In serological antibody titers showed the positive reaction with 54.4% in Actinobacillus pleuropneumonia serotype 5, 44.8% in Pasteurella multocida, 36.8% in A pleuropneurnonia serotype 2, 13.6% in Mycoplasmal pneumonia. Isolated aerobic microorganisms were examined antibiotic susceptibility and showed the high activity in gentamicin (58.3%), enrofloxacin (53.3%), norfloxacin (51.6%), cephalothin (41.7%) and low activity in amoxycillin (98.3%), oxytetracycline (98.3%), penicillin G (90.0 %), tetracycline (88.4%), ampicillin (88.3%). Farm managements were deficient effect of humidity in swine house but ammonia gas all appeared the 10 ppm that were recommendation density, below.

Prevalence of fowl adenovirus and chicken anemia virus in Jeonbuk, Korea (전북지역 조류아데노바이러스 및 닭전염성빈혈 감염률 조사)

  • Fowl adenovirus (FAdV) and chicken anemia virus (CAV) have gained much importance as an immunosuppressive and economically important emerging pathogen of poultry. This study was carried out to investigate the prevalence of FAdV and CAV infection in chickens. The groups were divided into Korean native chickens, broiler, layer hens and broiler breeder and set up groups according to age. As results, 12.5% of the native chicken, 2.5% of broiler and 6.7% of layer chicken were positive, respectively by PCR for FAdV. Serological test showed that 84.8%, 79.0%, 97.7% and 96.1% of chickens were positive for antibody to FAdV in native chickens, broiler, layer hens and broiler breeder. The prevalence of CAV infection were 20.0%, 7.5%, 16.7% and 10.0%, based on CAV gene detection by PCR. In serological test of CAV, 40.6%, 35.9%, 84.8% and 73.9% of chickens were positive in that groups.

  • Porcine reproductive and respiratory syndrome (PRRS) is an economically important disease of swine that occurs all over the swine industry worldwide. It was first observed in the Unite States in 1987 then in Europe in 1990. It has been described in Japan and in Korea in 1993. PRRS virus is divided into two distinct types, North American and European, genetically. Based on our limited knowledge there has been no report on the existence of European PRRSV. But according to the government's Korea Customs Service there has been many importations of breeding pigs from Europe. These seem to make an estimate that European PRRSV could be introduced in Korea by inflow of European breeding pigs. We first detected the European PRRSV could be introduced in Korean pig farms by using polymerase chain reaction (PCR). Further, it is also identified that there are not only North American PRRSV antibody but also a European PRRSV antibody. According to the genetical and serological experiment results, the presence of established North American PRRSV in Korea is due to the use of live vaccines made of North American PRRSV strain as well field virus infection, and the European PRRSV is possibly introduced from imported breeding stock.

  • To establish a statistically reliable sampling strategy for serological surveillance of classical swinefever (CSF) in Korea, antibody test data from CSF surveillance conducted during year 2005 were analyzed.The most appropriate sampling method was determined to be stratified multi-stage random sampling strategy,in which the primary sampling unit is a pig farm and the secondary are the pigs by the strata of breedersand finishers in the selected farm. The optimum sample size was 5 to 19 including 1 to 2 breeders accordingto the number of pigs in the farm. The optimum sampling strategy demonstrated in this study was veryFindings of our study provide practical guidelines for surveillance of herd immunity level to CSF in Korea.

  • Serological investigations for the leptospirosis on hospitalized patients in Choonchun Sungsim Hospital during the periods from August to November 1985 and 841 inhabitants of Kangwondo area including Choonchun, Choonsung, Inje, Chulwon, Hwachun, Gosung, Taibaik, Samchuk and Yangju area were carried out. 1. Among 58 hospitalized patients who were suspected as leptospirosis, 10 patients were detected to have antibody against Leptospira. All of positive sera had the highest antibody titer against serogroup Icterohemorrhagiae and most positive sera were also reactive to serogroup Australis and Canicola. Antibody titer of positive sera detected by microscopic agglutination(MA) test were ranging from 1 : 40 to 1 : 2,560. Antibody titer detected by ELISA method were higher than those detected by MAT(ELISA 1 : 400$\sim$1 : 25,600) and IgM titer of positive sera were generally higher than IgG titer. 2, Of 841 inhabitants in 8 area of Kangwondo, 17 persons (2,02%) possessing antibody against Leptospira were detected by ELISA method, IgG titer in positive sera were generally higher than IgM titer. Persons possessing antibody to Leptospira were distributed in both sex and in various age group, and no significant regional and occupational fluctuations were obserbed.
