Serological Survey on Bovine Babesiosis in Korea (한우(韓牛)의 바베시아병(病)에 대(對)한 혈청학적조사시험(血淸學的調査試驗))

  • In this study, the serological survey on bovine babesiosis was conducted using indirect fluorescent antibody technique. A total of 472 serum samples were taken and tested with the following results. 1. 63.3% or 299 out of 472 serum samples were positive against bovine babesiosis. 2. The positive rate in Seoul, Jeon-buk and Jeju areas were 63.8, 30.6 and 91.4 per cent, respectively. 3. In spring, the rate of positive reaction was 37.5 to 54.0 per cent, in summer it was 69.1 to 81.8 per cent and in autumn 65.3 to 68.5 per cent.

Serological activity of fractions of mycobacterial antigens (항산성균 항원분획의 혈청학적 활성)

  • A study on the production of mycobacterial antogens has been made in order to improve immunological reactivity and specificity, which have long been explored for the better use of immunological diagnosis of mycobacterial infections. Instead of culture filtrate cell extract was used as a starting material for the production of antigens in this study. Cell extract was fractionated though several steps such as salting out, gel filtration and ion exchange column chromatography and reactivity and specificity of the fractions so produced were enaluated by the various serological methods. The result showed that the species-specific antigenic components distributed mostly in the fractions, Tc of M. tuberculosis, Kc of M. kansasii, Sa of M. scrofulaceum, Aa of M. avium and Fa, Fb, Fc (FF1) of M. fortuitum, which were fractionated by ion exchange column prior to concentrating by salting out and molecular sieving.

  • In order to know the current status of sensitivity of Corynebacterium renale to some antimicrobial agents of current use, the sensitivity of 37 strains isolated from urine and vaginal cavity of healthy Korean native cattle and dairy cattle was studied by agar plate dilution method, and their sensitivity according to serological types was compared. The antimicrobial agents tested were streptomycin (SM), erythromycin (EM), oxytetracycline (OT), penicillin G (PC), gentamycin (GM), kanamycin (KM), ampicillin (AP), leucomycin (LM), chloramphenicol (CM), colistin (CL), sulfadimethoxine (SDM) and sulfadiazine (SDZ). Most strains were highly sensitive to PC, GM, AP, LM and KM, and moderate to CM and OT, but resistant to SM, EM, CL, SDM and SDZ. Serological type II was more sensitive to EM, SDM and SDZ than type I and II, but more resistant to SM.

  • Infectious coryza is one of the important acute respiratory diseases causing a significant egg drop and retarded growth in chicken. An attempt for the isolation of etiologic agent was made by utilizing SPF sentinel birds housed in commercial farms and the results obtained are as follows. Fifteen isolates of Haemophilus gallinarum were tested for their biological and serological characteristics with reference strains, 221 and Modesto and subsequently classified into two serotypes. Of them, isolates immunolocally identical to the standard strains were also selected as vaccine strains for future studies.

  • Lymphocystis is a viral disease of fish primarily in marine and brackishwaters. Here we report the cloning, expression, and the serological applications of the lymphocystis disease virus (LCDV) major capsid protein (MCP). The MCP gene was amplified by PCR from the genomic DNA of LCDV isolated from Schlegel's black rockfish, Sebastes schlegeli, and expressed in E. coli. Mouse antisera raised against the purified recombinant MCP (rMCP) reacted with the viral MCP in an immunofluorescence assay, indicating that this rMCP would be useful for serological studies of field samples.

Isolation of adenovirus from porcine alveolar macrophages (돼지 폐포탐식세포로부터 adenovirus의 국내분리주 작성)

  • One cytopathogenic virus was isolated in alveolar macrophages from pig without any apparent respiratory clinical signs. Biophysical properties and electron microscopy of the isolate showed the characteristics of adenovirus. Intracytoplasmic inclusion bodies were seen in virus-inoculated cells. The genetic analysis indicated the presence of DNA with the size of >20Kb. In a serological survey of 40 serum samples collected from two different farms in slaughter house, 9 sera were positive for neutralizing antibody against the isolate. The potential implications of the isolate as the causative agent in respiratory disorder were discussed.

Identification of a Strain of Babesia Isolated from Korean Cattle (한우(韓牛)로부터 분리(分離)한 바베시아 원충(原蟲)의 동정(同定))

  • In order to identify unknown Babesia spp. which was isolated from Korean cattle, the morphology of Korean strain was compared with that of Babesia bigemina (Kochinda strain) and Babesia spp. (Miyake strain). Immunofluorescent technique was used to identify the serological character of the parasites. The results obtained were summarized as follows: 1. Korean strain was morphologically very similar to Babesia spp.(Miyake strain) which mostly showed parallel-bigeminate forms, while B. bigemina (Kochinda strain) was mostly round and oval forms. 2. By the indirect fluorescent antibody technique: a) Anti-Babesia spp. and Korean Babesia spp. sera showed a higher antibody titers with Babesia spp. (Miyake strain) antigen (1:500) than with B. bigemina (Kochinda strain) antigen (1:50). b) Anti-Babesia bigemina sera showed a lower titer with Babesia spp. antigen (1:50) than with B. bigemina antigen (1:250). 3. On the basis of morphological and serological confirmantions, a Babesia strain isolated from a Korean cattle was very similar, if not identical, to Miyake strain of Babesia spp.

Studies on Properties of Avian Reovirus Isolated in Korea (국내에서 분리한 닭 레오바이러스의 성상에 관한 연구)

  • Avian reoviruses have been implicated in respiratory disease enteric conditions including Cloacal pasting in young thicks, pericarditis, hydropericardium, anaemia with swollen spleen and liver and petechiation of skeletal muscle and viral arthritis. This study was conducted to examine properties of reovirus field 3 strains isolated from affected broiler from several farms. An infectious agent was isolated from leg tendons and intestine of broiler with clinical tenosynovitis. The agent grew well on the chicken embryo kideny cells(CEK). One of them produced cytopathic effects(CPE) of round type and formed intranuclear inclusions, and the other was characterized by CPE of syncytical type and cytoplasmic inclusion. The properties and serological classification of field strains were examined by hemagglutin test, virus neutralization test, agar gel precipitin, electropherotype. They showed no hemagglutination reactions and not well neutralization and to possess common antigens detectable by AGP test. RNA electropherotype presented 10 segment band as the previous report. These data suggest that the field strains and standard strains (1133, 1733) may be the same group.

A Serological Test for Syphilis among Prostitutes by VDRL Slide Test (VDRL Slide Test에 의한 접대부의 매독혈청검사 성적)

  • A Serological test for Syphilis by VDRL Slide tests were made to find the degree of treponemal infection. For this study sera were collected from 5, 312 Prostitutes of Seoul, Inchun, Euijungbu, Yangzoo, Pajoo and Pyungtaig from November 27, to December 7, 1972 and were examined on the following items. 1. VDRL Slide Qualitative test. 2. VDRL Slide Quantitative test. As the results of this study the following Conclusions were obtained. 1) This test were obtained average reactive 5.92% for 5, 312 prostitutes in Seoul 7.87% for 254 prostitutes, in Iuchun 6.77% for 354 prostitutes, in Euijungbu 6.62% for 740 prostitutes, in Yangjoo 5.82% for 1, 958 prostitutes, in Pajoo 6.08% for 937 prostitutes and in Pyungtaig 4.82% for 1, 069 prostitutes respectively 2) The highest Reagin titer was 1: 32 serum dilution.

이종의 식특성 "바이러스"의 합성기작에 관하여

  • The mechanism of synthesis of the toacco mosaic virus(TMV) and the potato virus X(PVX) was investigated using the methods of ultraviolet light irradiation and serological analysis. In vitro irradiation of UV on the infected tobacco juice for 10 minutes caused the infectivity of TMV and PVX to decrease markedly on their respective local lesion indicator hosts, Nicotiana glutinosa L. and Gomphrena globosa L., indicating that UV destroys directly the infectivity of the virus particles. Ten minutes after the UV was irradiated on the leaves of the two indicator hosts before inoculation, the infectivity of TMV decreased as it was irradiated in vitro, whereas that of PVX increased by 26% as compared with the unirradiated control. When the two viruses were mix-inoculated in the common host of tobacco and the synthetic products were analyzed by serological methods for a two week infection period, it was found that both viruses were multiplying more rapidly and abundantly than they were singly inoculated into the same host species. Titers from mixed series were often two times as high as those of singly inoculated series. A mechanism of competition in the synthesis between the mixed viruses in the common host is postulated.
