계 골격근에서 순수분리한 Serine Protease Inhibitor의 특성과 작용기구 (A Potent Inhibitor of Pancreatic Serine Proteases from Chick Skeletal Muscle)

  • Trypsin을 비롯한 여러 serine protease를 저해하는 단백질을 계골격근으로부터 여러 chromatography 방법을 이용하여 순수하게 분리하였다. 이 저해제의 분자량은 젤 여과법을 이용하여 측정한 결과 66,000 달톤이었으며 sodium dodecyl sulfate 존지하에서 전기영동하였을 때 66,000과 64,000 달톤의 두 단백질로서 나타났다. 이 중 64,000 달톤의 단백질은 순수분리과정 혹은 생체 내에서 일어난 부분 절제현상에 기인한 66,000 달톤 단백질의 부산물인 것으로 사료된다. 이 저해제는 약 7.4의 등전점을 가진 당 단백질임을 알 수 있었으며, 다량의 cysteine잔기를 포함하고 있다.

Inhibition of Various Proteases by MAPI and Inactivation fo MAPI by Trypsin

  • MAPI (microbial alkaline protease inhibitor) was isolated from cultrue broth of Streptomyces chromofuscus SMF28. The Ki values of MAPI for the representative serine proteases such as chymotrypsin and proteinase K were 0.28 and $0.63{\;}\mu\textrm{M}$, respectively, and for the cysteine proteases cathepsin B and papain were 0.66 and $0.28{\;}\mu\textrm{M}$, respectively. These data indicate that MAPI is not a potent selective inhibitor of serine or cysteine proteases. Progress curves for the inhibition of three proteases by MAPI exhibithe characteristic patterns; MAPI exhibited slow-binding inhibition of cathepsin B. It was rapidly associated with chymotrypsin before the addition of substrate and then reactivation of MAPI-inhibited enzyme was investigated in the presence of substrate. On the other hand, MAPI-proteinase K interaction was typical for those classical inhibitors. When MAPI was incubated with trypsin, there was an extensive reduction in the ingibitory activities of MAPI corresponding to 66.5% inactivation of MAPI, indicating that trypsin-like protease may play a role in the decrease of the inhibitory activity during cultivation.

Recovery of serine protease inhibitor from fish roes by polyethylene glycol precipitation

  • The fractionation of serine protease inhibitor (SPI) from fish roe extracts was carried out using polyethylene glycol-4000 (PEG4000) precipitation. The protease inhibitory activity of extracts and PEG fractions from Alaska pollock (AP), bastard halibut (BH), skipjack tuna (ST), and yellowfin tuna (YT) roes were determined against target proteases. All of the roe extracts showed inhibitory activity toward bromelain (BR), chymotrypsin (CH), trypsin (TR), papain-EDTA (PED), and alcalase (AL) as target proteases. PEG fractions, which have positive inhibitory activity and high recovery (%), were the PEG1 fraction (0-5 %, w/v) against cysteine proteases (BR and PA) and the PEG4 fraction (20-40 %, w/v) against serine proteases (CH and TR). The strongest specific inhibitory activity toward CH and TR of PEG4 fractions was AP (9278 and 1170 U/mg) followed by ST (6687 and 2064 U/mg), YT (3951 and 1536 U/mg), and BH (538 and 98 U/mg). The inhibitory activity of serine protease in extracts and PEG fractions from fish roe was stronger than that of cysteine protease toward common casein substrate. Therefore, SPI is mainly distributed in fish roe and PEG fractionation effectively isolated the SPI from fish roes.

Alterations in Membrane Transport Function and Cell Viability Induced by ATP Depletion in Primary Cultured Rabbit Renal Proximal Tubular Cells

  • This study was undertaken to elucidate the underlying mechanisms of ATP depletion-induced membrane transport dysfunction and cell death in renal proximal tubular cells. ATP depletion was induced by incubating cells with 2.5 mM potassium cyanide(KCN)/0.1 mM iodoacetic acid(IAA), and membrane transport function and cell viability were evaluated by measuring $Na^+$-dependent phosphate uptake and trypan blue exclusion, respectively. ATP depletion resulted in a decrease in $Na^+$-dependent phosphate uptake and cell viability in a time-dependent manner. ATP depletion inhibited $Na^+$-dependent phosphate uptake in cells, when treated with 2 mM ouabain, a $Na^+$ pump-specific inhibitor, suggesting that ATP depletion impairs membrane transport functional integrity. Alterations in $Na^+$-dependent phosphate uptake and cell viability induced by ATP depletion were prevented by the hydrogen peroxide scavenger such as catalase and the hydroxyl radical scavengers(dimethylthiourea and thiourea), and amino acids(glycine and alanine). ATP depletion caused arachidonic acid release and increased mRNA levels of cytosolic phospholipase $A_2(cPLA_2)$. The ATP depletion-dependent arachidonic acid release was inhibited by $cPLA_2$ specific inhibitor $AACOCF_3$. ATP depletion-induced alterations in $Na^+$-dependent phosphate uptake and cell viability were prevented by $AACOCF_3$. Inhibition of $Na^+$-dependent phosphate uptake by ATP depletion was prevented by antipain and leupetin, serine/cysteine protease inhibitors, whereas ATP depletion-induced cell death was not altered by these agents. These results indicate that ATP depletion-induced alterations in membrane transport function and cell viability are due to reactive oxygen species generation and $cPLA_2$ activation in renal proximal tubular cells. In addition, the present data suggest that serine/cysteine proteases play an important role in membrane transport dysfunction, but not cell death, induced by ATP depletion.

Cloning and Characterization of TMPRSS6, a Novel Type 2 Transmembrane Serine Protease

  • We have identified TMPRSS6, a novel type 2 transmembrane serine protease. TMPRSS6 possesses all the signature motifs of the family of transmembrane serine proteases (TMPRSSs), including a transmembrane domain, an LDL receptor class A (LDLRA) domain, a scavenger receptor cysteine-rich (SRCR) domain, and a serine protease domain. The substrate specificity of TMPRSS6 is slightly different from those of other TMPRSS family members. Combined with the finding that TMPRSS6 is expressed strongly in the thyroid and weakly in the trachea, this may indicate that TMPRSS6 has a specialized role.

  • A 29kDa cysteine protease of Taenia solium metacestodes was purified by Mono Q anion-exchanger and Superose 6 HR gel filtration chromatography. The enzyme was effectively inhibited by cysteine protease inhibitors, such as iodoacetic acid (IAA) and trans-epoxy-succinyl-L-leucyl-amido (4-guanidino) butane (E-64) while inhibitors acting on serine- or metallo-proteases did not affect the enzyme activity. The purified enzyme degraded human immunoglobulin G (IgG), collagen and bovine serum albumin (BSA), but human IgG was more susceptible for proteolysis by the enzyme. To define the precise biological roles of the enzyme, more detailed biochemical and functional studies would be required.

남북극 유래 저온성 박테리아 Culture Collection에서 저온활성 프로테아제 생산균주의 스크리닝과 효소 특성 (Screening for Cold-Active Protease-Producing Bacteria from the Culture Collection of Polar Microorganisms and Characterization of Proteolytic Activities)

  • 극지연구소(KOPRI)는 국내외적으로 유일하게 남북극 지역에서 분리한 저온적응성 박테리아 균주를 대상으로 culture collection(약 6,300균주)을 구축하여 운영하고 있다. 보유 중인 프로테아제(protease) 생산 균주들(총 874균주) 중에서 활성이 높은 프로테아제를 생산하는 78개의 균주들을 1차 선발한 후, 1% skim milk가 포함된 0.1${\times}$ ZoBell 고체배지에 접종하고 다양한 온도($5-30^{\circ}C$)에서 배양하면서 세포외분비성 프로테아제의 활성을 비교하였다. 위의 신속하고 직접적인 균주 스크리닝 방법을 통해서, 최종적으로 저온활성 프로테아제를 생산하는 15개의 저온적응성 균주들을 선발하였다. 최종 선발된 균주들은 16S rRNA 유전자의 분석결과 Pseudoalteromonas (13균주)와 Flavobacterium (2균주) 속(genus)으로 분류되었고, $5-15^{\circ}C$ 저온에서도 활성을 나타내는 저온성 프로테아제를 생산하였다. 15개 균주들이 생산하는 각각의 프로테아제는 특이적 화합물에 의한 효소활성 억제 정도에 따라 5개의 그룹(serine protease, aspartic protease, cysteine protease, metalloprotease, 그리고 미분류 프로테아제)으로 분류되었다. 본 실험을 통해서 선발한 남북극 유래 박테리아 균주들은 새로운 저온활성 프로테아제를 발굴하기 위한 유용한 생물자원으로서의 가치를 가지고 있다.

Minor Thermostable Alkaline Protease Produced by Thermoactinomyces sp. E79

  • Thermoactinomyces sp. E79 produced two types of thermostable alkaline proteases extracellularly. A minor protease was separated from a major protease by using DEAE-column chromatography. This enzyme was purified to homogeneity by ammonium sulfate and DEAE-Sepharose ion-exchange chromatography. The purified minor protease showed different biochemical properties compared to the major protease. The molecular mass of the purified enzyme was estimated by SDS-PAGE to be 36 kDa. Its optimum temperature and pH for proteolytic activity against Hammarsten casein were $70^{\circ}C$ and 9.0, respectively. The enzyme was stable up to$75^{\circ}C$ and in an alkaline pH range of 9.0-11.0. The enzyme was inhibited by phenylmethylsulfonyl fluoride (PMSF) and $Hg^{2+}, indicating that the enzyme may be a cysteine-dependent serine protease. In addition, the enzyme cleaved the endoproteinase substrate, succinyl-Ala-Ala-Pro-Phe-p- nitroanilide, and the $K_m$ value for the substrate was 1.2 mM.

류마티스 관절염 실험용쥐의 활액에서 단백분해효소의 활성 및 항산화에 대한 녹용약침의 효과 (Effect of Cervi Pantotrichum Cornu Herbal acupuncture on protease activities, antioxidant in Rheumatoid arthritis rats)

  • 류마티스 관절염의 쥐의 활액에서 단백분해효소, 산화제와 유리기에 대한 녹용약침의 비특이적 면역억제효과를 연구하였다. 일련의 실험표본으로서 여러가지 세포질, 리소좀, 기질 백분해효소의 제 활성을 RA대조군과 녹용약침군의 활액에서 카르보닐기 유도로 생성되는 유리기-유발 단백질손상과 항산화를 비교하였다. 전반적으로 단백분해효소활성이 정상군과 비교하여 RA대조군에서 유의성 있게 증가하였다. 세포질 단백분해효소들은 정상군과 RA군의 차이에서는 유의성이 없었다. 녹용약침처리($100{\mu}g/kg$)결과 세포질, 리소좀, 기질 단백분해효소생성을 억제하였으며, RA군과 녹용약침군 또는 정산군 사이에 활액 또는 세포질 항산화에서 유의성 있는 차이가 없음에도 불구하고, RA군 활액의 단백질손상을 유발하는 유리기는 녹용약침군과 정산군에 비교하여 약 2배 정도 높았다. 이상의 결과에서 단백분해효소와 유리기는 RA유발시 단백질손상을 유도하는 물질로 밝혀졌으며, 따라서 단백분해효소 저해와 유리기소거능을 갖는 치료법개발이 새로운 RA예방치료법으로 제시되었다. 나아가서 여러가지 기질특이성을 갖는 활액내 단백분해효소류(cysteine, serine, metallo proteinases와 peptidases)에 대한 효과적인 저해제개발이 필요한 것으로 보인다. 따라서 본 녹용약침은 이와 같은 새로운 개념의 2가지(유리기제거, 단백분해활성) 관절염치료 요소를 충족하는 약리활성을 포함하는 훌륭한 제제로 평가된다.

어류 알의 Protease Inhibitor 활성 분포 (Distribution of Protease Inhibitors from Fish Eggs as Seafood Processing Byproducts)

  • To identify and examine the distribution of proteolytic inhibitory activity in crude extracts from fish eggs, and to determine the applicability of these protease inhibitors as anti-degradation agents in surimi-based products and fish meat, we compared the inhibitory activities of various extracts from fish eggs to those of commercial proteases, such as trypsin and papain. We used the optimal conditions for the screening of trypsin activity: 30 ug/uL of 0.1% trypsin and 0.6 mM Na-benzoyl-L-arginine-p-nitroanilide (BAPNA) with a pH of 8.0 at $40^{\circ}C$ for 60 min. The activities of papain and four commercial proteases were investigated after mixing with 100 ug/uL enzymes and 0.3% casein with a pH of 8.0 at $40^{\circ}C$ for 60 min. We performed a screening assay to detect the inhibitory activity (%) of crude extracts from eight species of fish eggs against the target proteases trypsin and papain. The assay revealed a wide distribution of trypsin and papain inhibitors in fish eggs. The specific inhibitory activities (11.6.28.6 U/mg) of crude extracts from fish eggs against trypsin and BAPNA substrate were higher than that (0.64 U/mg) of egg whites, used as a commercial inhibitor. The inhibitory activities of crude extracts from fish eggs against trypsin, and of egg whites against casein substrate (1.94.4.51 U/mg), were higher than those of papain (0.24.1.57 U/mg) and commercial protease (0.04.0.32 U/mg). The extracts from fish eggs were rich in protease inhibitors that exhibited strong inhibitory activity against trypsin, a serine protease, and papain, a cysteine protease.
