Human papillomavirus (HPV) 16, E7 proteins derived from the prototype (Bac73) and natural variant (Bac101) E7 open reading frame were produced in Sf9 insect cells. The variant E7 gene occurred naturally by substitution mutation at the position of 88 nucleotide, resulting serine instead of asparagine. Using E7 specific monoclonal antibody (VD6), both E7 proteins were identified in recombinant baculovirus infected SF9 cells. Radiolabelling and immunoprecipitation analysis revealed that both E7 proteins were phosphoproteins. Immunostaining result showed that E7 proteins were mainly localized in the cytoplasm. Nuclear form of E7 proteins was also detected after a sequential fractionation procedure for removing chromatin structure. Considering that the VD6 recognition site in E7 protein is located within 10 amino acid at the N-terminus, this region appears to be blocked by the nuclear component. Western blot analysis revealed that nuclear form was more abundant than cytoplasmic E7 proteins. Time course immunostaining showed that the primary location of E7 protein was the nucleus and exported to the cytoplasm as proteins were accumulated. These events occurred similarly in both Bac73 and Bac101 infected Sf9 cells, suggesting that these two proteins may have similar biological functions.

Data spread over 11 years (1986-1996) pertaining to a synthetic population developed by inter se mating of half-breds of Corriedale and Russian Merino with Nali maintained at CCS Haryana Agricultural University, Hisar were utilized for the estimation of genetic parameters for growth traits. The means for birth weight (BWT), weaning weight (WWT), six month body weight (SWT), yearling weight (YWT), preweaning daily gain (PRW) and post weaning daily gain (POW) were 3.35 kg, 10.79 kg, 13.28 kg, 18.96 kg, 82.6 gm and 27.6 gm, respectively. The effects of year and season of birth and sex of lamb were significant for all the traits under study except the effect of season of birth for WWT, SWT and PRW. No definite trend was observed over the years for the averages of body weight and gain. Lambs born during the spring season performed better for BWT, WWT and PRW while the performance of lambs born during autumn was better for the other traits included in the study. The male lambs were heavier than the females for body weight at all stages and gain in weight. The heritability estimates for WWT and PRW were low; for BWT and SWT were moderate and for YWT and POW were high. Birth weight had high heritability and high genetic correlations with subsequent body weights and gains but due to the presence of a maternal effect on BWT and WWT, a sequential selection procedure is recommended for the improvement of growth rate in sheep.
This study investigates effects of an aging period on arsenic (As) chemical forms in soils and phytotoxicity using artificially As-contaminated soils with a range of As concentrations (0-300 mg/kg) and aging periods (0 and 3 months). A sequential extraction procedure showed that the increasing As concentration in soils increased the ratio of non-specifically and specifically bound As, which are known to be bioavailable. This resulted in increasing As uptake by tomatoes with increasing As concentration (R2=0.87 for exponential fitting); however, the seed germination was not sensitive to the As concentrations of the soil samples. The seed germination was also statistically similar in the soils with 75 and 150 mg-As/kg regardless of the aging period. The time taken until the seed germination (i.e., lag phase), on the other hand, decreased from 10 d to 3 d with aging for 3 months. This can be attributed to the decreased amount of bioavailable As with aging. Overall, this study shows that when the toxic effects of the As-contaminated soils are assessed using tomato plants, it is better to use more sensitive methods than seed germination such as the As accumulation or the lag phase for seed germination.
This paper is aimed at source tracking of soil heavy metal contamination at a site established by reusing construction wastes. The soil heavy metal concentration at the study site peaked at a depth range of 5-10 m. Column studies were conducted to investigate the possibility of the contamination scenario of infiltration of stormwater carrying heavy metals of ground origin followed by selective heavy metal accumulation at the 5-10 m depth range. Almost all amount of lead, zinc, cadmium, and nickel introduced to the columns each packed with 0-5 m or 5-10 m field soil were accumulated in the column. The very poor heavy metal mobility in spite of the weak association of the heavy metals with the soil (characterized by a sequential extraction procedure) can be attributed to the high pH (10-11) of the construction wastes. From the results, the heavy metal contamination of the subsurface soil by an external heavy metal source was determined to be very unlikely at the study site. The column study applied in the current study is expected to be a useful methodology to present direct evidence of the contaminant source tracking at soil contamination sites.
Isotope amount ratios of lead in a bronze sample have been successfully determined using multicollector inductively coupled plasma mass spectrometry (MC-ICP-MS). Matrix separation conditions were tested and optimized using ion exchange chromatography with anion-exchange resin, AG1-X8, and sequential elution of the 0.5 M HBr and 7 M $HNO_3$ to separate lead from very high contents of copper and tin in bronze matrix. Mercury was also removed efficiently in the optimized separation condition. The instrumental isotope fractionation of lead in the MC-ICP-MS measurement was corrected by the external standard sample bracketing method using an external standard, NIST SRM 981 lead common isotope ratio standard followed by correction of procedure blank to obtain reliable isotope ratios of lead. The isotope ratios, $^{206}Pb/^{204}Pb$, $^{207}Pb/^{204}Pb$, $^{208}Pb/^{204}Pb$, and $^{208}Pb/^{206}Pb$, of lead were determined as $18.0802{\pm}0.0114$, $15.5799{\pm}0.0099$, $38.0853{\pm}0.0241$, and $2.1065{\pm}0.0004$, respectively, and the determined isotope ratios showed good agreement with the reference values of an international comparison for the same sample within the stated uncertainties
Substructuring methods are often used in finite element structural analyses. In this study a multi-level substructuring(MLSS) algorithm is developed and proposed as a possible candidate for finite element fluid solvers. The present algorithm consists of four stages such as a gathering, a condensing, a solving and a scattering stage. At each level, a predetermined number of elements are gathered and condensed to form an element of higher level. At the highest level, each sub-domain consists of only one super-element. Thus, the inversion process of a stiffness matrix associated with internal degrees of freedom of each sub-domain has been replaced by a sequential static condensation of gathered element matrices. The global algebraic system arising from the assembly of each sub-domain matrices is solved using a well-known iterative solver such as the conjugare gradient(CG) or the conjugate gradient squared(CGS) method. A time comparison with CG has been performed on a 2-D Poisson problem. With one domain the computing time by MLSS is comparable with that by CG up to about 260,000 d.o.f. For 263,169 d.o.f using 8 x 8 sub-domains, the time by MLSS is reduced to a value less than $30\%$ of that by CG. The lid-driven cavity problem has been solved for Re = 3200 using the element interpolation degree(Deg.) up to cubic. in this case, preconditioning techniques usually accompanied by iterative solvers are not needed. Finite element formulation for the incompressible flow has been stabilized by a modified residual procedure proposed by Ilinca et al.[9].

It is essential to embed product quality in the design process to win the global competition. Many components found in many products including automobiles and electronic devices are fabricated using sheet metal forming processes. Wrinkle and fracture are two types of defects frequently found in the sheet metal forming process. Reducing such defects is a hard problem as they are affected by many uncontrollable factors. Attempts to solve the problem based on traditional deterministic optimization theories are often led to failures. Furthermore, the wrinkle and fracture are conflicting defects in such a way that reducing one defect leads to increasing the other. Hence, it is a difficult task to reduce both of them at the same time. In this research, a new design method for reducing the rates of conflicting defects under uncontrollable factors is presented by using operating window and a sequential search procedure. A new SN ratio is proposed to overcome the problems of a traditional SN ratio used in the operating window technique. The method is applied to optimizing the robust design of a sheet metal forming process. To show the effectiveness of the proposed method, a comparison is made between the traditional and the proposed methods using simulation software, applied to a design of particular sheet metal forming process problem. The results show that the proposed method always gives a more robust design that is less sensitive to noises than the traditional method.

This study presents a reliability-based optimum design of reinforced concrete frames, in which the AFOSM and SOSM methods are applied for the evaluation of the failure probabilities, and the sequential linear programming method is used as a practical approach to the system optimization. One-story two-bay reinforced concrete frame is chosen for the numerical illustration of the proposed reliability-based optimum design. As a result, it is found that the proposed procedure for the reliability-based optimization of RC frames could provide the accurate estimation of the optimal level of safety, and appears applicable to real structures with reasonable complexity. It is shown in the paper that the probability distributions of the basic random variables and the uncertainties of the applied loadings and material strengths may have great effect on the optimum design, but the AFOSM and SOSM methods do not show significant discrepancy in the optimum design results, but the former appears more realistic and time saving than the latter for this specific study.
Purpose: Given the several proper models for given mixture components-process variables experimental data, we propose a strategy to find the optimal condition in which the performance of the responses is well-behaved under those models. Methods: Given the mixture experimental data with process variables, first we choose the reasonable starting models among the class of admissible product models based on the model selection criteria and then, search for the candidate models that are the subset models of the starting model by the sequential variable selection method or all possible regressions procedure. Good candidate models are screened by the evaluation of model selection criteria and checking the residual plots for the validity of the model assumption. Results: We propose a strategy to find the optimal condition in which the performance of the responses is well-behaved under those good candidate models by adopting the optimization methods developed in multiple responses surface methodology. Conclusion: A strategy is proposed to find the optimal condition in which the performance of the responses is well-behaved under those proper combined models. This strategy to find the optimal condition is illustrated with the example in this paper.

Objective: The purpose of this study was to propose a sequential procedure for the simultaneous analysis of soluble and insoluble metal compounds. Methods: Methods for sampling and analyzing metal compounds such as ISO standards, NIOSH methods, HSE methods, and OSHA methods were reviewed. Results: Some metals have different OELs depending on the solubility of the compound. Therefore, we should take into account these characteristics and perform an exposure assessment. Soluble metal compounds are first extracted from the filter, and then the filter is digested by acids to analyze residual insoluble components. The extraction of soluble compounds can be completed by agitation for about 60 minutes with a leach solution (water) in a water bath at 37℃. For the analysis of insoluble compounds, the sample filter and the filtration filter remaining after the extraction of the soluble compounds are analyzed. This allows simultaneous determination of soluble and insoluble metal compounds. For hexavalent chromium compounds, soluble hexavalent chromium can first be extracted from the filter by using sulfate buffer. The insoluble hexavalent chromium remaining in the filter can then be extracted using carbonate buffer. Conclusion: Workers are often exposed to many hazardous substances with different exposure limits at industrial sites. The OELs for compounds of the same metal can be set differently depending on solubility. This study can help evaluate a worker's exposure to metal compounds by suggesting methods for the simultaneous determination of soluble and insoluble metal compounds.
