New Characterization Methods for Block Copolymers and their Phase Behaviors

  • In this feature article, we briefly review the new methods we have utilized recently in the investigation of morphology and phase behavior of block copolymers. We first describe the chromatographic fractionation method to purify block copolymers from their side products of mainly homopolymers or block copolymer precursors inadvertently terminated upon addition of the next monomer in the sequential anionic polymerization. The chromatographic method is extended to the fractionation of the individual block of diblock copolymers which can yield the diblock copolymer fractions of different composition and molecular weight, which also have narrower distributions in both molecular weight and composition. A more detailed phase diagram could be constructed from the set of block copolymer fractions without the need of acquiring many block copolymers each prepared by anionic polymerization. The fractions with narrow distribution in both molecular weight and composition exhibit better long-range ordering and sharper phase transition. Next, epitaxial relationships between two ordered structures in block copolymer thin film is discussed. We employed the direct visualization method, transmission electron microtomography(TEMT) to scrutinize the grain boundary structure.

  • Beryllium (Be), cobalt (Co), thallium (Tl) and vanadium (V) are candidates of 21 priority soil pollutants in Korea. The distribution of their concentration in soils from three contamination sources including industrial, roadside and mining areas was investigated. Concentrations of the metals were evaluated quantitatively using pollution indices and the fractionation of metals was conducted using modified SM&T (Standards Measurements and Testing programme) sequential extraction. Concentrations of the metals for all samples from industrial and roadside soils were within the range of natural background levels, while some of Be in soils from abandoned mines exceeded that the range. Enrichment Factor (EF) and Nemerow Integrated Pollution Index (NIPI) for Be, Co, Tl and V showed that there are effects or possibilities of anthropogenic activities. Pollution Load Index (PLI) analyses indicated all investigated sites needed further monitoring. The results of sequential extractions indicated mobile fractions (F1+F2) of Be, Tl and V were below 30% except some of Co in soil, which implies their low mobility to neighboring environment media. Variable tools like sequential extraction, comparison with background/actual concentration and pollution indices, as well as aqua regia extraction should be considered when evaluating Be, Co, Tl, V in soil.

Comparison of concurrent chemoradiotherapy versus sequential radiochemotherapy in patients with completely resected non-small cell lung cancer

  • Purpose: Our institution has implemented two different adjuvant protocols in treating patients with non-small cell lung cancer (NSCLC): chemotherapy followed by concurrent chemoradiotherapy (CT-CCRT) and sequential postoperative radiotherapy (PORT) followed by postoperative chemotherapy (POCT). We aimed to compare the clinical outcomes between the two adjuvant protocols. Materials and Methods: From March 1997 to October 2012, 68 patients were treated with CT-CCRT (n = 25) and sequential PORT followed by POCT (RT-CT; n = 43). The CT-CCRT protocol consisted of 2 cycles of cisplatin-based POCT followed by PORT concurrently with 2 cycles of POCT. The RT-CT protocol consisted of PORT followed by 4 cycles of cisplatin-based POCT. PORT was administered using conventional fractionation with a dose of 50.4-60 Gy. We compared the outcomes between the two adjuvant protocols and analyzed the clinical factors affecting survivals. Results: Median follow-up time was 43.9 months (range, 3.2 to 74.0 months), and the 5-year overall survival (OS), locoregional recurrence-free survival (LRFS), and distant metastasis-free survival (DMFS) were 53.9%, 68.2%, and 51.0%, respectively. There were no significant differences in OS (p = 0.074), LRFS (p = 0.094), and DMFS (p = 0.490) between the two protocols. In multivariable analyses, adjuvant protocol remained as a significant prognostic factor for LRFS, favouring CT-CCRT (hazard ratio [HR] = 3.506, p = 0.046) over RT-CT, not for OS (HR = 0.647, p = 0.229). Conclusion: CT-CCRT protocol increased LRFS more than RT-CT protocol in patients with completely resected NSCLC, but not in OS. Further studies are warranted to evaluate the benefit of CCRT strategy compared with sequential strategy.

Size Determination of Pollens Using Gravitational and Sedimentation Field-Flow Fractionation

  • Pollens are known to be an allergen. They penetrate human respiratory system, triggering a type of seasonal allergic rhinitis called pollen allergy (hey fever). The purpose of this study is to test two field-flow fractionation (FFF) techniques, gravitational FFF (GrFFF) and sedimentation FFF (SdFFF), for their applicability to sizecharacterization of micron-sized pollens. Both GrFFF and SdFFF are elution techniques, providing sequential elution of particles based on size. They allow the size distribution as well as the mean size of the sample to be determined from the elution time. In this study, GrFFF and SdFFF were used to determine the size distribution of Paper Mulberry and Bermuda Grass pollens. For the Paper Mulberry pollen, the mean size obtained by GrFFF is 12.7 μm, and agrees rather well with the OM data with the relative error of 8.0%. For the Bermuda Grass pollen, the mean size obtained by GrFFF is 32.6 μm with the relative error of 12.3%. The mean sizes determined by SdFFF are 12.4 (relative error = 10.1%) and 27.1 μm (relative error = 5.2%) for the Paper Mulberry and the Bermuda Grass pollen, respectively. Although SdFFF tends to yield more accurate size distribution due to lower band broadening under the field strength higher than 1 G, the sizes determined by GrFFF were not significantly different from those by SdFFF.

  • As the size of semiconductors becomes smaller, it is necessary to perform high precision polishing of nanoscale. Ceria, which is generally used as an abrasive, is widely used because of its uniform quality, but its stability is not high because it has a high molecular weight and causes agglomeration and rapid precipitation. Such agglomeration and precipitation causes scratches in the polishing process. Therefore, it is important to accurately analyze the size distribution of ceria particles. In this study, a study was conducted to select dispersants useful for preventing coagulation and sedimentation of ceria. First, a dispersant was synthesized and a ceria slurry was prepared. The defoamer selection experiment was performed in order to remove the air bubbles which may occur in the production of ceria slurry. Dynamic light scattering (DLS) and asymmetrical flow field-flow fractionation (AsFlFFF) were used to determine the size distribution of ceria particles in the slurry. AsFlFFF is a technique for separating nanoparticles based on sequential elution of samples as in chromatography, and is a useful technique for determining the particle size distribution of nanoparticle samples. AsFlFFF was able to confirm the presence of a little quantities of large particles in the vicinity of 300 nm, which DLS can not detect, besides the main distribution in the range of 60-80 nm. AsFlFFF showed better accuracy and precision than DLS for particle size analysis of a little quantities of large particles such as ceria slurry treated in this study.

Distribution of Arsenic Fraction in Soil Around Abandoned Mining Area and Uptake by Rice

  • Arsenic (As) contamination of agricultural soils resulting from mining activity has caused major concern due to the potential health risk. Therefore the current study was carried out to investigate the relationship between fractionation of As in soil and rice uptake and to provide a basic information for adequate management of As contaminated agricultural soil. Twenty agricultural soils and rice affected by the abandoned mining sites were collected. Soil chemical properties and As concentrations (total and sequential extracted) in soils were determined and As concentrations in polished rice were analyzed. The average concentration of As in non-specifically adsorbed (F1), specifically adsorbed (F2), amorphous hydrous oxides of Fe and Al (F3), crystalline hydrous oxides of Fe and Al (F4) and residual phase (F5) were 0.08, 1.38, 10.34, 3.26 and $10.98mgkg^{-1}$, respectively. Both soil pH and available phosphorus were positively correlated with the concentrations of As in F1 and F2. These results indicate that increasing the soil pH and available phosphorus can significantly increase the easily mobile fractions of As (F1 and F2). The average concentration of As in polished rice was $0.09mgkg^{-1}$. The concentrations of As in F1 and F2 showed a positive correlation with the concentrations of As in polished rice. Therefore soil pH and available phosphorus affect the distribution of As fractionation in soils and thus affect As bioavailability.

Development of TPA-induced Ornithine Decarboxylase (ODC) Inhibitors from Plants as Cancer Chemopreventive Agents

  • Chemical carcinogenesis is associated with the increase of intracellular polyamine levels, and 12-O-tetradecanoylphorbol-13-acetate (TPA)-induced mouse epidermal ODC activity are essential to skin tumor promotion by TPA. Therefore, for the discovery of new cancer chemopreventive agents, we have evaluated about 73 kinds of natural products to study inhibitory effects against ODC activity induced by TPA in T24 cell culture system. The total methanol extracts of plants fractionated into three layers (hexane, ethyl acetate and water layer) were tested and the hexane fraction of Angelica gigas $(root\;bark,\;IC_{50}:\;7.4\;{\mu}g/ml)$ and the ethyl acetate fraction of Corydalis ternata $(root,\;IC_{50}:\;7.5\;{\mu}g/ml)$ were the most effective on the inhibition of TPA-induced ODC activity, These active fractions are under investigation with further sequential fractionation using column chromatography.

Antimicrobial Effect of Roselle (Hibiscus sabdariffa L.) Petal Extracts on Food-Borne Microorganisms

  • In this study, we investigated the antimicrobial effect of 14 different herbal petal extracts on various foodborne and food spoilage bacteria. Herbal petal extracts were prepared with 70% ethanol followed by sequential hexane, chloroform, ethyl acetate, n-butanol, and water fractionation. Antimicrobial activity was highest in the ethanol fraction from roselle (Hibiscus sabdariffa L.) petals as determined by the paper disc method. The roselle ethanol extract retarded the growth of food spoilage bacteria in kimbap (rice rolled in dried laver). Foodborne microorganisms (e.g. Bacillus cereus and Clostridium perfringens), on the other hand, were most efficiently inhibited by the ethyl acetate fraction of the roselle petal extract as determined by growth inhibition curves. Our study shows that roselle petals harbor antimicrobial activity against foodborne and food spoilage microorganisms. The critical ingredient is highly enriched in the ethyl acetate fraction of the extract.

  • Chemical studies of nitrogen compounds of Panax ginseng seem relatively rare, Probably due to the isolation difficulties, subsequently the investigations of biological activities are little. The experimental conditions were established for highly complete extraction of peptides (basic, acidic and neutral) from Panax ginseng. This task was achieved by applying the follow isolation procedure: 1 , the sequential extraction with water, 0.1% TFA in 20% acetonitril and buffer pH 6.5 (water-pyridine-acetic acid 100:3:900) : 2, fractionation by ultrafiltration : 3, n-butanol extraction 4, cation- and anion-exchange chromatography : 5, chromato-electrophoresis. The comparison of red ginseng (xylem Sl pith part) and red ginseng inside white (xylem Sc pith part) was also provided. To analyze the peptide mixture the chromato-electrophoresis method of separation was applied. Optimal conditions for peptides mapping of sample were explored. Our experiments revealed the quantitative difference of peptide between xylem & pith and phloem & cortex part. We have also found the qualitative difference in the composition of polypeptides between normal red ginseng (xylem Sc pith part) and red ginseng inside-white (xylem St pith part)

  • Background: Combination chemotherapy is now considered to be the cornerstone of small cell lung cancer (SCLC). management but the optimal management of limited SCLC is not well defined. The role of thoracic radiotherapy (TRT) is less well established. Recent meta-analyses reports revealed that TRT combined with chemotherapy produce "good" local control and prolonged survival. But other reports that survival was not changed. The liming, dose, volume and fractionation for TRT with the combined chemotherapy of SCLC remains unsettled. In this study, we analyzed the effects according to the timing of thoracic radiotherapy in limited SCLC. Method: All fifty one patients received cytoxan, adriamycin and vincristine(CAV) alternating with etoposide and cisplatin(VPP) every 3 weeks for 6 cycles were randomized prospectively into two groups: concurrent and sequential. 27 patients received 4500cGy in 30 fractions(twice daily 150cGy fractional dose) over 3 weeks 10 the primary site concurrent with the first cycle of VPP(concurrent gorup). 24 patients received 4000 to 5000cGy over 5 or 6 weeks after completion of sixth cycles of chemotherapy(sequential group). Results: 1. Response rates and response duration : Response rates were not significantly different between two groups(p=0.13). But response duration was superior in the concurrent group(p=0.03). 2. Survival duration was nor different between two groups(p=0.33). 3. Local control rate was superior in the concurrent group(p=0.00). 4. Side effects and toxicities: Hematologic toxicities, especially leukopenia, infection and frequency of radiation esophagitis were higher in the concurrent group (p=0.00, 0.03, 0.03). Conclusion: The concurrent use of TRT with chemotherapy failed to improve the survival of limited stage SCLC patients compared with the sequential use of TRT but response duration and local control rate were superior in the concurrent group. Frequency of radiation esophagitis, life threatening hematologic toxicities and infection were more frequent in the concurrent group than sequential group. So, the selection of an optimal schedule of chemotherapy combined with TRT that would lead to a major increase in survival with minimal toxicity is remained to be validated in large scale study in the future.
