Anti-Helicobacter pylori Compounds from Polygonum cuspidatum

  • Anti-Helicobacter pylori activity guided fractionation led to the isolation of five anthraquinones, two stilbenes and one naphthoquinone from the EtOAc fraction of Polygonum cuspidatum, using silica gel column chromatography, Sephadex-LH20, MPLC and recrystallization. The chemical structures were identified to be physcion (1), emodin (2), anthraglycoside B (3), trans-resveratrol (4), anthraglycoside A (5), polydatin (6), 2-methoxy-6-acetyl-7-methyljuglone (7) and citreorosein (8) by UV, $^1H$-NMR, $^{13}C$-NMR and mass spectrometry. Anti-Helicobacter pylori activity including MIC values of each compound was evaluated. All of the isolates exhibited anti-H. pylori activity of which MIC values were lower than that of a positive control, quercetin. Compounds 2 and 7 showed potent growth inhibitory activity. Especially, a naphthoquinone, compound 7 displayed most potent antibacterial activity with $MIC_{50}$ value of $0.30{\mu}M$ and $MIC_{90}$ value of $0.39{\mu}M$. Although anti-H. pylori activity of this plant was previously reported, this is the first report on that of compounds isolated from this species. From these findings, P. cuspidatum roots or its isolates may be useful for H. pylori infection and further study is needed to elucidate mechanism of action.

Triterpenoids from the Fruits of Cornus kousa Burg. as Human Acyl-CoA: Cholesterol Acyltransferase Inhibitors

  • The fruits of Cornus kousa Burg. were extracted with 80% aqueous methanol (MeOH) and the concentrated extract was partitioned with ethyl acetate (EtOAc), n-butanol (n-BuOH), and $H_2O$. From the EtOAc traction, 5 triterpenoids were isolated through repeated silica gel ($SiO_2$), octadecyl silica gel (ODS), and Sephadex LH-20 column chromatography (c.c.). These compounds were determined to be ursolic acid (1), corosolic acid (2), taraxasterol (3), betulinic acid (4), and betulinic aldehyde (5) on the basis of their spectroscopic data including electronic ionization mass spectrometry, ultraviolet spectroscopy, infrared spectroscopy, and nuclear magnetic resonance. This is the first reported isolation of these compounds from this plant. Also, compounds 1, 3, 4, and 5 show a relatively high inhibitory activity against human acyl-CoA: cholesterol acyltransferase-1 (hACAT-1) with inhibition values of $52.8{\pm}0.7$, $91.1{\pm}0.4$, $93.0{\pm}0.7$, and $96.2{\pm}0.2%$ at a concentration of $100{\mu}M$, respectively.

복분자 열매에서 항산화활성을 지닌 quercetin의 분리 및 동정 (Isolation and Identification of Quercetin with Antioxidative Activity from the Fruits of Rubus coreanum Miquel)

  • 복분자 열매에 함유된 항산화활성 물질을 탐색하기 위하여 MeOH로 추출하고, 이 추출물을 용매분획하여 EtOAc가용 중성획분을 얻었다. 이 획분에 함유된 항산화활성 물질을 흡착성과 분자체 효과를 이요한 Sephadex LH-20 column chromatography로 정제하고, HPLC에 의해 1종의 항산화활성물질을 단리하였다. 분리된 활성물질의 구조 해석을 위해 EI-MS와 $^1H-NMR$ 분석을 실시하여, 활성물질은 구조식이 $C_{15}H_{10}O_7$이며 분자량이 302인 quercetin으로 동정되었다. 또한 항산화활성 물질 quercetin은 HPLC에 의해 복분자 열매 100g 중에 $0.25{\pm}0.02\;mg$이 함유되어 있는 것으로 분석되었다.

도화(桃花, Pruni persicae Flos) 분획물의 항산화 및 항염증 효과 (Antioxidant and Anti-inflammatory Effects of Fractions from Pruni persicae Flos)

  • Objectives : The solvent extracts of Pruni persicae Flos were investigated for the activities of anti-oxidant and anti-inflammatory effects to apply as a functional ingredient for cosmetic products. Methods : In this study, the fractions of P. persicae Flos were extracted with 70.0% acetone and purified using Sephadex LH-20 column chromatography. As a result, eight fractions were isolated. We performed MTT assay, total polyphenol contents, DPPH free radical scavenging assay, SOD-like activity, xanthine oxidase inhibition assay, astringent activity assay, hyaluronidase inhibition assay and the production of nitric oxide. Results : For anti-oxidant effects, the electron donating ability of fraction (Fr.) 2-5, Fr.-8 isolated from P. persicae Flos was above 90.0% at 100 ppm respectively. The superoxide dismutase (SOD) - like activity of Fr.-5 isolated from P. persicae Flos was 92.1% at 1,000 ppm. The xanthine oxidase inhibitory effect of Fr.-6 isolated from P. persicae Flos was about 83.3% at 1,000 ppm. Hyaluronidase inhibition activity related to the anti-inflammation effect was 94.0% for Fr.-4 isolated from P. persicae Flos at 500 ppm. In the anti-inflammation effect, the Fr.-4 isolated from P. persicae Flos inhibited the generation of nitric oxide. Conclusions : All these findings suggested that the fractions of P. persicae Flos has a great potential as a cosmeceutical ingredient with a anti-oxidant and anti-inflammatory effects.

Neuraminidase Inhibitors from the Fruiting Body of Phellinus igniarius

  • During our ongoing investigation of neuraminidase inhibitors from medicinal fungi, we found that the fruiting bodies of Phellinus igniarius exhibited significant inhibitory activity against neuraminidase from recombinant H3N2 influenza viruses. Two active compounds were isolated from the methanolic extract of P. igniarius through solvent partitioning and Sephadex LH-20 column chromatography. The active compounds were identified as phelligridins E and G on proton nuclear magnetic resonance ($^1H$ NMR) and electrospray ionization mass measurements. These compounds inhibited neuraminidases from recombinant rvH1N1, H3N2, and H5N1 influenza viruses, with $IC_{50}$ values in the range of $0.7{\sim}8.1{\mu}M$.

홍화적색소 Carthamin의 효과적인 분리 및 화학구조 분석 (Efficient Purification and Chemical Structure ldentification of Carthamin from Carthamus tinctorius)

  • 우리나라에서 오랫동안 적색 및 황색색소원으로 널리 사용하여 왔던 홍화(Carthamus tinctorius)로부터 전통적인 추출방법을 응용한 새로운 방법을 사용하여 적색소를 효과적으로 분리정제하였다. 홍화꽃잎을 물 및 메탄올로 처리하여 황색소를 제거한 다음 건조파쇄하여 0.5 M $Na_2CO_3$로 홍색소를 추출하고 0.5 M citrate 수용액으로 침전시킨 후 cellulose 흡착, Sephadex LH-20 관크로마토그라피로 분리정제하였다. 분리정제된 적색소는 $300^{\circ}C$에서 분해되었고 silica gel TLC 상에서 BAW(n-BuOH : HOAc : $H_2O$=4 : 1 : 5)로 전개하였을 때 $R_f$값이 0.56이었다. 에탄올 용액에 녹인 적색소의 UV/Vis 흡수스펙트럼은 519, 372, 311, 244 nm에서 최대 흡수피크를 나타내었고, IR 스펙트럼은 특히 $3400\;cm^1$ 넓은 영역에서 hydroxyl기에 의한 강한 흡수띠를 보여주었다. $^{1}H$$^{13}C$ NMR data로 부터 enolized ${\beta}-triketone$, p-hydroxycinnamoyl, methine 및 glucosyl moieties를 확인하였고 그 값을 제시하였다. 이상의 data를 문헌과 비교한 결과 분리한 홍화적색소의 화학구조는 $6-{\beta}-D-glucopyranosyl-2-[[3-{\beta}-D-glucopyranosyl-2,3,4-trihydroxy-5-[3#-(4@-hydroxyphenyl)-1#-oxo-2#-propenyl]-6-oxo-1,4-cyclohexadien-1-yl]methylene]-5,6-dihydroxy-4-[3#-(4@-hydroxyphenyl)-1#-oxo-2#-propenyl]-4-cyclohexene-1,3-dione$인 carthamin으로 확인하였다.

Pyridoxatin, an Inhibitor of Gelatinase A with Cytotoxic Activity

  • Gelatinase A is a member of the matrix metalloproteinases that play an important role in cancer invasion and metastasis. In the course of screening gelatinase A inhibitors from microbial sources, a fungal strain PT-262 showed a strong inhibitory activity. The strain was identified as Chaunopycnis alba on the basis of its morphological characteristics. The inhibitor was isolated from acetone extract of mycelial cake by sequential chromatographies on MCI-gel, Sephadex LH-20, and a reverse-phase HPLC column. The purified inhibitor was identified as pyridoxatin by its physico-chemical properties and spectroscopic analysis. Pyridoxatin is not a peptide analog and has cyclic hydroxamic acid moiety. It inhibited activated gelatinase A with an $IC_{50}$ value of 15.2 ${\mu}M$ using fluorescent synthetic peptide. It also had a strong cytotoxicity against human cancer cell lines in vitro. Furthermore, this compound inhibited DNA synthesis with an $IC_{50}$ value of 2.92 ${\mu}M$ in PC-3 prostate cancer cells by [$^3H$]thymidine incorporation assay.

The constituents of taraxacum hallaisanensis roots

  • Three sesquiterpene lactone compounds, two novel(1.betha.,3.betha.-dihydroxy-6.betha.,11.betha.,4.alpha.,5.alpha.,7.alpha.H -eudesm-12, 6-olide-1-O-.betha.-D-glucopyranoside, 1.betha.,3.betha.-dihydroxy-6.betha.,11.betha.,4.alpha.,5.alpha.,7.alpha.H-eudes m-12,6-olide-1-O-.betha.-D-glucopyranoside) and 1.betha.,3.betha.-dihydroxy-6.betha.,11.betha.,4.alpha.,5.alpha., 7.alpha.H-eudesm-12,6-olide were isolated from the aqueous fraction of MeOH extract of the roots from Taraxacum hallaisanensis (Compositae) employing Amberlite XAD-2, ODS-gel, silica gel and Sephadex LH-20 column chromatographics. Another known compound, (-)-epicatechin, was isolated from the aqueous fraction of the MeOH extract. The total MeOH extract also contained phytosterol and a mixture of .betha.-amyrin acetate, .alpha.-amyrin acetate and lupeol acetate. Structures of isolated compounds were elucidated by spectroscopic parameters of IR, Mass, /sup 13/C-NMR, /sup 1/H-NMR, /sup 1/H-/sup 1/H COSY, /sup 13/C-/sup 1/H COSY and HMBC.

[ $\beta$ ]-Secretase (BACE1) Inhibitors from Sanguisorbae Radix

  • In the course of screening anti-dementia agents from natural products, two $\beta$-secretase (BACE1) inhibitors were isolated from the ethyl acetate soluble fraction of Sanguisorbae Radix by the activity-guided purification using silica gel, Sephadex LH-20, and RP-HPLC. They were identified as 1,2,3-trigalloyl-4,6-hexahydroxydiphenoyl-$\beta$-D-glucopyranoside (Tellimagrandin II, 1) and 1,2,3,4,6-pentagalloyl-$\beta$-D-glucopyranoside (2) and were shown to non-competitively inhibit $\beta$-secretase (BACE1) with the $IC_{50}$ values of $3.10{\times}10^{-6}M\;and\;3.76{\times}10^{-6}M$, respectively. The Ki values of 1 and 2 were $6.84{\times}10^{-6}M\;and\;5.13{\times}10^{-6}M$. They were less inhibitory to asecretase (TACE) and other serine proteases such as chymotrypsin, trypsin, and elastase, suggesting that they were relatively specific inhibitors of BACE1.

Analysis of Functional Constituents in Mulberry (Morus alba L.) Twigs by Different Cultivars, Producing Areas, and Heat Processings

  • Four functional constituents, oxyresveratrol 3'-O-${\beta}$-D-glucoside (ORTG), oxyresveratrol (ORT), t-resveratrol (RT), and moracin (MC) were isolated from the ethanolic extract of mulberry (Morus alba L.) twigs by a series of isolation procedures, including solvent fractionation, and silica-gel, ODS-A, and Sephadex LH-20 column chromatographies. Their chemical structures were identified by NMR and FABMS spectral analysis. Quantitative changes of four phytochemicals in mulberry twigs were determined by HPLC according to cultivar, producing area, and heat processing. ORTG was a major abundant compound in the mulberry twigs, and its levels ranged from 23.7 to 105.5 mg% in six different mulberry cultivars. Three other compounds were present in trace amounts (<1 mg/100 g) or were not detected. Among mulberry cultivars examined, "Yongcheon" showed the highest level of ORTG, whereas "Somok" had the least ORTG content. Levels of four phytochemicals in the mulberry twigs harvested in early September were higher than those harvested in early July. Levels of ORTG and ORT in the "Cheongil" mulberry twigs produced in the Uljin area were higher than those produced in other areas. Generally, levels of ORTG and ORT in mulberry twigs decreased with heat processing, such as steaming, and microwaving except roasting, whereas those of RT and MC did not considerably vary according to heat processing. These results suggest that the roasted mulberry twigs may be useful as potential sources of functional ingredients and foods.
