Quantification of cholesterol in human serum by isotope dilution liquid chromatography/mass spectrometry (동위원소희석 액체크로마토그래피/질량분석법에 의한 혈청 내 콜레스테롤의 정량)

  • An ID LC/MS (isotope dilution liquid chromatography/mass spectrometry) was used as a primary method for the quantitative analysis of cholesterol in human serum. The separation of cholesterol was carried out by Thermo ODS hypersil $C^{18}$ column. The mobile phase was 100% methanol, and flow rate was $0.3m{\ell}/min$. Cholesterol and cholesterol-$3,4-13C_2$ were monitored at m/z 369.4 and 371.3, which correspond to $[M-H_2O+H]^+$ respectively. In order to verify the measurement method, NIST SRM 909b was analyzed. The results agreed well with certified values within uncertainty. The four kinds of serum certified reference material were prepared and certified. The repeatabilities of measurement were ranged from 0.1 to 0.8% (RSD), which were relatively good. The reproducibility between independent measurement run was below 0.24% (RSD). The expanded uncertainty was about 1.43% within the 95% confidence interval.

Analysis of tetracyclines in shrimp samples based on a two-step extraction approach prior to high-performance liquid chromatography

  • This study presents a sensitive and reliable method for determining tetracycline (TC), oxytetracycline (OTC), and chlortetracycline (CTC) residues in shrimp samples. A two-step process involving liquid-liquid extraction (LLE) followed by solid-phase extraction (SPE) was developed prior to HPLC analysis. The target analytes were effectively extracted using EDTA/McIlvaine buffer (pH 4.0): methanol (80:20, %v/v), with subsequent clean-up using a C18 SPE cartridge. HPLC separation was conducted on a C18 column (250 mm × 4.6 mm i.d., 5 ㎛) at 30 ℃, using 0.01 % trifluoroacetic acid (A) and acetonitrile (B) as the mobile phase. A gradient elution protocol was applied, transitioning from 85(A):15(B) %v/v to 70(A):30(B) %v/v at 7 min, with a 5 min hold, followed by adjustment to 85(A):15(B) %v/v for 13-14 min. The detection was performed using photodiode array (PDA) at 365 nm with a flow rate of 1.0 mL/min. The calibration curves exhibited good linearity within a concentration range of 0.4-6.0 ㎍/mL (R2 > 0.995). The limits of detection (LOD) for TC, OTC, and CTC in shrimp were 0.034, 0.029, and 0.021 ㎍/mL, respectively. The limits of quantitation (LOQ) for TC, OTC, and CTC were found to be 0.114, 0.097, and 0.071 ㎍/mL, respectively. Recoveries of TC, OTC, and CTC from spiked shrimp samples ranged from 91.0 % to 95.5 %, 92.4 % to 97.2 %, and 93.3 % to 96.6 %, respectively. This method was successfully applied to the determination of TC, OTC, and CTC residues in shrimp samples sourced from various local markets.

Simulation of D-limonene Separation from Mandarine Extract in Simulated Moving Bed (SMB) (감귤 추출물로부터 D-리모넨 분리를 위한 유사 이동층 크로마토그래피(SMB) 전산모사)

  • Limonene is orange flavored natural material that is mainly contained in mandarine and lemon peels. D-limonene was extracted from cold-storaged mandarine peel by using Soxhlet extractor at $120^{\circ}C$ for 2 hours with ethanol as solvent. Henry constants of d-limonene and impurity were calculated as $H_{Lim}=8.55$ and $H_{imp}=0.223$ from the result of HPLC analysis. 4-bed SMB of limonene simulation with $0.46{\times}25cm$ columns was conducted by using Aspen chromatography program. Then effective condition for purity was found by changing $m_2$ and $m_3$ values in triangle diagram. The highest purity was 98.59% at $m_2=2.57$, $m_3=9.55$. For this case, feed, desorbent, extract, and raffinate flow rates were 1 mL/min, 1.19 mL/min, 0.857 mL/min and 1.34 mL/min, respectively. Scale-up simulation was also conducted by increasing column diameter from 0.46 cm to 1.6 cm for getting the same efficiency. The increased flow rates were 12 mL/min, 14 mL/min, 10 mL/min, and 16 mL/min for feed, desorbent, extract, and raffinate. It was possible to scale-up with maintaining same limonene purity because linear isotherms of limonene and impurity were assumed.

Effects of Rice Straw Particle Size on Chewing Activity, Feed Intake, Rumen Fermentation and Digestion in Goats

  • Effects of particle size and physical effective fibre (peNDF) of rice straw in diets on chewing activities, feed intake, flow, site and extent of digestion and rumen fermentation in goats were investigated. A 4${\times}$4 Latin square design was employed using 4 mature Liuyang black goats fitted with permanent ruminal, duodenal, and terminal ileal fistulae. During each of the 4 periods, goats were offered 1 of 4 diets that were similar in nutritional content but varied in particle sizes and peNDF through alteration of the theoretical cut length of rice straw (10, 20, 40, and 80 mm, respectively). Dietary peNDF contents were determined using a sieve for particle separation above 8 mm, and were 17.4, 20.9, 22.5 and 25.4%, respectively. Results showed that increasing the particle size and peNDF significantly (p<0.05) increased the time spent on rumination and chewing activities, duodenal starch digestibility and ruminal pH, and decreased ruminal starch digestibility and $NH_{3}$-N concentration. Intake and total tract digestibility of nutrients (i.e. dry matter, organic matter, and starch) and ruminal fermentation were not affected by the dietary particle size and peNDF. Increased particle size and peNDF did not affect ruminal fibre digestibility, but had a great impact on the intestinal and total tract fibre digestibility. The study suggested that rice straw particle size or dietary peNDF was the important influential factor for chewing activity, intestinal fibre and starch digestibility, and ruminal pH, but had minimal impact on feed intake, duodenal and ileal flow, ruminal and total tract digestibility, and ruminal fermentation.

Characteristics of B cell proliferation by polysaccharide fraction of Paeonia japonica miyabe (백작약 조다당분획에 의한 B 세포 증식의 특성)

  • Background : Paeonia japonica Miyabe is a medicinal plant which has been widely used as a component of blood-building decoctions (Chinese medicinal concept : Bu-Xie). The immunopharmacological characteristics of the extract of Paeonia japonica (PJ) were investigated. Methods : The effects of fractions of PJ extract on lymphocyte proliferation were measured by $H^3$-thymidine incorporation assay. The proliferated lymphocyte subsets were analyzed in flow cytometry. The subset cell populations of spleen cells were separated by magnetic cell separation system, and their proliferation by the extract were investigated. The effect of the extract on antibody production was determined in mice challenged with sheep red blood cells (SRBC) using hemolytic plaque forming cell assay. Results : Spleen cells were proliferated by water extract of PJ. Polysaccharide fraction (PJ-P) of the extract was most active in the proliferation. It was found in flow cytometry that the lymphocyte subset proliferated by PJ-P was B cell population. Among the separated subset cell populations, T cell-depleted cell population and macrophage-depleted cell population were most proliferated by PJ-P. However, positively selected populations of B cells and T cells were not proliferated by PJ-P. These results indicate that B cell proliferation by PJ-P may require the assistance of macrophages or T cells. These results suggest that firstly PJ-P may stimulate macrophages or T cells, and then B cells are activated. The number of antibody-secreting cells was increased by administration of PJ-P in mice immunized with SRBC as a T-dependent antigen. Conclusion : These results suggest that macrophages and accessory cells are directly activated by PJ-P and then helper T cells and B cells are indirectly activated. As the results, immune responses might be coordinately improved. In conclusion, PJ-P, a polysaccharide of P. japonica, may be a characteristic immunostimulator, which is analogous to polysaccharides such as lentinan, PSK and ginsan.

Study on Material Segregation of Grout and Filling Characteristic of Grouting for Post-Tensioned Concrete Beam (PC 그라우트의 재료분리 및 PC 빔 그라우팅 충전성에 관한 연구)

  • This paper discusses a series of experiments including material improvement in order to ensure quality of grouting for the post-tensioned structure. In presstressed concrete, grouting refers to the construction procedure of filling empty space of duct enclosing the prestress tendons using cementitious material, To date, adequate quality control of the grouting has not been established in Korea because the relationship between the grouting and durability of post-tensioned structure is not well-recognized. The Korean standard does not consider the important material characteristic, wick effect, which is caused by tendons in the ducts, and furthermore current standard testing method does not quantify reasonable material segregation. As a result, the grout material, which satisfies the current material standards, may well exhibit excessive bleeding of water or shrinkage during construction. In this study, international codes and standards related to grouting were surveyed. The mix proportions of the constituents and novel admixtures were suggested to meet equivalently with these standards. Performance of this enhanced grout was compared with common domestic grouts using the international standard testing method. A series of mock-up specimens considering geometry of PC beam was constructed and grout flow pattern was observed as the grout was injected. It was observed that the grouting performance was highly influenced by material properties and filling characteristic can be varied depending on geometry of ducts.

Analysis of Two-Dimensional Pollutant Transport in Meandering Streams (사행하천에서 오염물질의 2차원 거동특성 해석)

  • In this study, RMA2 and RMA4, the 2-D depth-averaged models, were employed to simulate the two-dimensional mixing characteristics of the pollutants in the natural streams. The velocity and depth were first calculated using RMA2, 2-D hydrodynamic model, and then the resulting flow field was inputted to RMA4, 2-D water quality model, to compute the concentration field. RMA models were verified using the velocity and concentration data measured in S-curved meandering channel. The results showed that the RMA2 model simulated well the phenomenon that the maximum velocity line is located at the Inner bank of meandering channel, and the RMA4 model was well adapted to reproduce the general mixing behavior and the separation of tracer clouds. Comparing model simulations with measured data in the field experiments, RMA2 model simulated well general flow field and tendency that the maximum velocity line skewed toward the outer bank which were found in field experiments. The simulations of RMA4 model showed that the center of the tracer cloud tends to follow the path in which the maximum velocity occurs. In this study, the dispersion coefficients are fine-tuned based on the measured coefficients calculated using field concentration data, and the results show reasonable agreement with predictive equations.

A Method of Estimating the Volume of Exploitable Groundwater Considering Minimum Desirable Streamflow (최소하천유출량을 고려한 지하수 개발가능량 산정방안)

  • The concept of safe yield places an emphasis on balancing groundwater withdrawal with groundwater recharge but ignores naturally occurring groundwater discharge. Because streams and their alluvial aquifers are closely linked in terms of water supply and water quality, to be properly understood and managed they must be considered together. Therefore, some districts in Kansas have reevaluated their safe-yield policies to account for natural groundwater discharge and stream-aquifer interactions by amending their safe-yield regulations to include a portion of baseflow as the minimum desirable streamflow (MDS). This study proposes a modified safe-yield policy in which the drought flow is chosen as the MDS. Baseflow separation was conducted from streamflow hydrograph and the results are presented as a flow-duration curve. The exploitable groundwater can be determined by subtracting MDS from the cumulative baseflow. This method was tested in the Musimcheon watershed, which was validated for streamflow using the SWAT-K model. The annually averaged exploitable groundwater in the whole watershed was estimated to be 86 mm. The exploitable groundwater amounts were also estimated for each subwatershed in the Musimcheon watershed.

Near-wake Measurements of an Oscillating NACA 0012 Airfoil (진동하는 NACA 0012 에어포일의 근접후류 측정)

  • An experimental study was carried out in order to investigate the influence of Reynolds number on the near-wake of an oscillating airfoil. An NACA 0012 airfoil was sinusoidally pitched at the quarter chord point, and is oscillated over a range of instantaneous angles of attack of $\pm$6$^{\circ}$. An X-type hot-wire probe was employed to measure the near-wake of an oscillating airfoil, and the smoke-wire visualization technique was used to examine the flow properties of the boundary layer. The free-stream velocities were 1.98, 2.83 and 4.03 m/s and the corresponding chord Reynolds numbers were 2.3${\times}10^4$, 3.3$\times$104 and 4.8${\times}10^4$, respectively. The frequency of airfoil oscillation was adjusted to fix a reduced frequency of K=0.1. The results show that the properties of the boundary layer and the near-wake can dramatically be distinguished in the range of Reynolds numbers between 2.3${\times}10^4$ and 3.3${\times}10^4$, on the other hand, it is similar in the cases of Re=3.3$\times$104 and 4.8$\times$104. This is caused by that the unsteady separation point is dramatically delayed in case of Re= 2.3${\times}10^4$.

Effects of Wing Twist on Longitudinal Stability of BWB UCAV (날개의 비틀림이 동체-날개 융합익형 무인전투기의 종안정성에 미치는 영향에 대한 연구)

  • Lambda wing type Unmanned Combat Aerial Vehicle(UCAV) which adopts Blended Wing Body(BWB) has relatively less drag and more stealth performance than conventional aircraft. However, Pitching moment is rapidly increased at a specific angle of attack affected by leading edge vortex due to leading edge sweep angle. Wind tunnel testing and numerical analysis were carried out with UCAV 1303 configuration on condition of 50 m/s of flow velocity, $-4^{\circ}{\sim}28^{\circ}$ of the range of angle-of-attack. The effect of wing twist for longitudinal stability at the various angles of attack was verified in this study. When negative twist is applied on the wing, Pitch-break was onset at higher angle of attack due to delayed flow separation on outboard of the wing. On the other hand, pitch-break was onset at lower angle of attack and lift-to-drag ratio was increased when positive twist is applied on the wing.
