Effect of Placental Serum to Cultures of the Healthy Human Peripheral Blood Lymphocytes (태반혈청이 사람의 임파구 배양에 미치는 영향)

  • To investigate the effects of placental serum to in vitro culture of the normal human lymphocytes the peripheral blood lymphocytes were isolated by ficoll-hypaque separation method in 20 healthy human adults. The blast transformation respone of lymphocytes to mitogens were observed by stimulation with PHA($25{\mu}g/ml$) and Con A($25{\mu}g/ml$) using RPMI 1640 media containing 20% placental serum(PS), tetal calf serum(FCS), or humans AB serum(AB). And one-way mixed lymphocyte culture test was performed between these unrelated person compounded into stimulators and responders to investigate the effect of placental serum. The following results were obtained. 1. In 20 experimental cases, these were no significant diffenence between FCS, AB, and PS in untreated control cultures. 2. In PHA-treated cultures, whereas the blast transformation rate of the FCS groups and AB groups were $40.8{\pm}4.3%$ and $44.6{\pm}4.3%$ respectively, that of PS groups were $21.7{\pm}3.4%$, Similar results were obtained in Con A-meated cultures. Therefore, placental serum inhibited the mitogenic response of lymphocytes significantly. 3. In MLC tests, stimulation index of the FCS groups and AB groups were $18.5{\pm}3.5$ and $20.1{\pm}3.3$ respectively. But placental serum inhibited MLC response of lymphocytes significantly($7.4{\pm}1.9$).

Characterization of Polysaccharides Obtained from Purslane (Portulaca olerace L.) Using Different Solvents and Enzymes

  • Physiochemical properties, such as yield and molecular weight distribution of polysaccharide fractions, of polysaccharides in the enzymatic hydrolysates of purslane were investigated and characterized. A higher amount of micro nutrients, such as potassium (9,413 mg/100 g), phosphorus acid (539 mg/100 g), leucine, alanine, lysine, valine, glycine, and isoleucine, was present in whole purslane. The yield of water soluble polysaccharides (WSP) was 0.29, 7.01, and 7.94% when extracted using room temperature water (RTW), hot-water (HW), and hot temperature/high pressure-water (HTPW), respectively, indicating that HW or HTPW extraction may be effective to obtain WSP from purslane. The average ratio of L-arabinose:D-galactose in the WSP was 37:49, 34:37, and 27:29, when extracted using RTW, HW, and HTPW, respectively. These results indicate that water was a suitable extraction solvent for preparation of the arabinogalactan component of whole purslane. A higher yield and total carbohydrate content was obtained by using Viscozyme L instead of Pectinex 5XL during extraction of the WSP, which indicates that enzymatic treatment of purslane may be an effective method to control the Mw of polysaccharides. Finally, it was confirmed that Viscozyme L is a suitable enzyme for the hydrolysis and separation of polysaccharides obtained from purslane.

Analysis of Volatile Compounds and Enantiomeric Separation of Chiral Compounds of Dried Sancho (Zanthoxylum schinifolium Siebold & Zucc)

  • The volatile compounds of dried sancho (Zanthoxylum schinifolium), an aromatic plant were extracted by simultaneous distillation and extraction (SDE) method and identified by gas chromatograph-mass spectrometry (GC-MS). Selected chiral constituents of sancho oil were characterized by enantiodifferentiation using multidimensional gas chromatograph (MDGC)-MS. A total of 57 compounds were identified and quantified, and the major compounds were identified estragole, nonanoic acid, octanoic acid, $\beta$-phellandrenene, and limonene. Among them, estragol (63.9%) was found as the predominantly abundant component of sancho. $\alpha$-pinene and nerolidol, and $\beta$-pinene and linalool were determined to be enantiomerically pure (100%) for their (S)-form and (R)-form, respectively. The enantiomeric composition of limonene in sancho revealed 83.9% purity for the (S)-enantiomer, whereas (E)- and (Z)-rose oxides showed mixtures of both enantiomers. The enantiomeric excess (%) for citronellal was 22.6% with the (R)-enantiomer as major enantiomer. The enantiomeric composition of these compounds can be used as parameter for authenticity control of sancho.

Real-time monitoring of net setting and hauling process in fishing operations of Danish seine vessel using ECDIS (ECDIS에 의한 외끌이 기선저인망 어선의 투양망 조업 과정의 실시간 모니터링)

  • This paper describes on the real-time monitoring of net setting and hauling process for fishing operations of Danish seine vessels in the southern waters of Korea as an application of a PC based ECDIS system. Tracking of fishing process was performed for the large scale Danish seine vessel of G/T 90 and 350 PS class using the fishing gear which the length of net, ground rope, head rope and sweep line including warp in both sides were 86m, 104m, 118m and 3,200m, respectively. Tracking information for net setting and hauling process was continuously recorded for 23 fishing operations performed on November and December, 2003. All measurement data, such as trawl position, heading, towing course and past track which was individually time stamped during data acquisition, was processed in real time on the ECDIS and displayed simultaneously on the ENC chart. The results indicated that after the separation of a marker buoy from Danish seiner, the averaged running speed of vessel and the averaged setting period while shooting the seine on the course of diamond shape to surround the fish school in the 23 fishing operations were 8.3 knots and 13.1 minutes, respectively. And with the maker buoy taken on board, the averaged running speed of vessel and the averaged towing period while closing the seine on the straight route was 1.0 knots and 47.0 minutes, respectively. After the closing stage of hand rope, the hand rope was towed by the averaged speed of 2.2 knots during the 13.0 minutes. The average area for route of diamond shape swept by sweep lines of the seine in 23 fishing grounds was $709,951.6m^2$. Further investigation is also planed to provide more quantitative tracking information and to achieve more effective surveillance and control of Danish seine vessels in EEZ fishing grounds.

Antifungal Mechanism and Properties of Antibiotic Substances produced by Bacillus subtilis YB-70 as a Biological Control Agent

  • Antibiotic substances were produced by Bacillus subtilis YB-70, a potential biocontrol agent found to suppress root-rot of eggplant (Solanum melonggena L) caused by Fusarium solani, in a dextrose glutamate medium and isolated by isoelectric precipitation. Partial purification was performed by column chromatography on silica gel with two solvent systems: chloroform-methanol and methanol-chloroform-water as eluting solvents, This active fraction YBS-1 s contained antifungal activity were soluble in ethanol, methanol, and water, but were not soluble in other solvents including acetone, butanol, ethyl ether, dimethylformamide, propanol, and etc. High performance liquid chromatography and thin layer chromatographic separation of YBS-1s showed that they have been composed of three biological active bands that were named YBS-1A, -1B, and -1C. The substances were stable to heat and resistant to protease. YBS-1s were active against a wide range of plant pathogenic fungi but did not inhibit the growth of bacteria and yeasts. They were not only fungicidal but also fungistatic against chlamydospores of F. solani. The $ED_{50}$ values for the chlamydospore germination and the germ-tube growth of F. solani were $O.725\mu\textrm{m}/ml\;and\;O.562\mu\textrm{m}/ml$, respectively. Microscopic observations proved the substances restricted the growth of phytopathogenic fungus F. solani by spore burst followed by dissolving of its germ-tube, and caused abnormal hyphal swelling after application to chlamydospores or growing hyphae. Cultural filtrate of B; subtilis YB-70 also suppressed the development of root-rot of eggplant in pot tests.

  • Hybrid process of thermally-induced phase separation and stretching was developed for the preparation of microporous polypropylene hollow fiber membranes. Precursor for stretching was prepared by using soybean oil as a diluent and benzoic acid as a nucleating agent far the sphenlite control and it was stretched far the micrporous hollow fiber membrane. The effects of stretching ratio and deformation rate for stretching process were investigated. Increase of stretching ratio resulted in the greater pore size with nonuniform size distribution. Higher deformation rate also increaser the pore size with uniform size distribution. Stretching ratio was closely related with the orientation of polymer chain and increased the mechanical strength of the fiber. Increase of deformation rate had little effects on the orientation of crystalline phase, and decreased the orientation of amorphous phase which caused the decrease of tensile strength of the fiber and broke the micro-fibrils connecting spherulites to form a circular pore shape.

Sphingolipid Metabolic Changes during Chiral C2-Ceramides Induced Apoptosis in Human Leukemia Cells

  • N-acetylsphingosine (C2-ceramide) is a synthetic water-soluble ceramide mimicking the activity of natural ceramides. By fixing chiral conformation on carbon numbers 2 and 3 in the ceramide structure, four chiral C2-ceramides naming d-erythro-, I-erythro-, d-threo-and 1-three C2-ceramide were synthesized. We have investigated the chiral effects of these C2-ceramides on the sphingolipid metabolism, particularly on both the sphingolipid bio- synthetic pathway and on the degradation pathway. In both HL-60 and U937 cells, the chiral C2-ceramide ($10{\mu}\textrm{m}$) showed sphingosine accumulation monitored fluoromatrically by a high performance liquid chromatographic separation of the sphingoid bases. Most importantly, in HL-60 cells, l-erythro C2-ceramide induced a 50 fold increase in sphingosine as compared to the control, while l-threo C2-ceramide exhibited a minimal 7-fold in-crease. In contrast, sphinganine, another sphingoid base, showed less accumulation by any chiral C2-ceramide tested under the same conditions. These results suggested that chiral C2-ceramide primarilyacts on the sphingolipid degradation pathway rather than on the sphingolipid biosynthetic route. The strong $C_0/G_1$ phase arrest in the cell cycle by treatment of I-erythro C2-ceramide indicates that the blockade of the sphingolipid degradation pathway might be concomitantly involved in the dysfunction of the cell cycle. On the other hand, the fact that all chiral C2-ceramides tested failed to inhibit the activity of sphingosine kinase acting on the removal of sphingosine by producing sphingosine-1 -phosphate demonstrates that chiral C2- ceramides may increase sphingosine by activating various ceramidases by which natural ceramides are divided into sphingosine and free fatty acids. However, the precise steps involved in this interaction are still unknown.

A Study on the Final Evaluation Criteria of Allocation Exceedance Regional in Total Maximum Daily Load (오염총량관리 할당부하량 초과지역의 최종 평가기준에 관한 연구)

  • The Total Maximum Daily Load (TMDL) is a watershed management system that involves the establishment of the target water quality, the calculation of permission loading (allocation loading), and the control of total pollutants for each unit watershed. Allocation loading is assessed through the comprehensive implementation assessment of the previous year's plan. Assessment results are used for follow-up management measures such as the limit of development and updating of TMDL Management Implementation Plans for the next planning period. Although detailed assessment criteria are important, they are not currently available. Therefore, we suggested assessment criteria by comparing two methods('integration method' and 'separation method') using combination point and non-point discharge loading. We also examined the penalty criteria considering controllable load local government and updating methods of the TMDL Management Implementation Plan for the next planning period.

Analytical Method for Triazine Herbicide Cyanazine Residues in Major Medicinal Crops (주요 약용작물에 대한 Cyanazine 제초제의 잔류 분석법)

  • Background: Cyanazine is used as a pre-emergent herbicide once during the growing season to control weeds of many upland crops worldwide. This study aimed to establish a method to determined cyanazine residue levels in major medicinal crops by using high performance liquid chromatography-UV detection/mass spectometry (HPLC-UVD/MS). Methods and Results: Cyanazine residue was extracted with acetone from the raw products of four representative medicinal plants - Scutellaria baicalensis, Paeonia lactiflora, Platycodon grandiflorum and Angelica gigas. The extract was diluted with a large volume of saline water and directly partitioned into dichloromethane to remove polar co-extractives in the aqueous phase. It was then purifined using optimized Florisil column chromatography. HPLC analysis conducted using an octadecylsilyl column allowed the successful separation of cyanazine from co-extractives of the samples, and the amount was sensitively quantified by ultraviolet absorption at 225 nm with no interference. The accuracy and precision of the proposed method were validated by conducting recovery experiments on each medicinal crop sample fortified with cyanazine at two concentration levels per crop in triplicate. Conclusions: The mean recoveries ranged from 91.2% to 105.3% for the four representative medicinal crops. The coefficients of variation were less than 10%, irrespective of the sample types and fortification levels. The limit of quantification of cyanazine was 0.02 mg/kg as verified by the recovery experiment. A confirmatory method was performed by liquid chromatography/MS using selected-ion monitoring technique to clearly identify the suspected residue.

Development of the On-Site Vermicomposting Unit for Sewage Sludge (하수슬러지 지렁이 퇴비화 장치개발에 관한 연구)

  • The on-site sewage sludge vermicomposting equipment was designed and evaluated on the batch and continuous tests. The vermicomposting equipment was designed to consider the mechanization as well as automation. In terms of performance evaluation of equipment with continuous experiments, the highest treatment rate was found in the condition of 5mm in particle size and 10mm in e feeding height. Also, 20~25kg of earthworm per 50kg of sewage sludge was the most appropriate density of earthworm. In addition, the most effective separation of cast was found when the earthworm breeding box height was 80cm. The performance evaluation on equipment allowed the establishment of earthworm composting capacity of 2ton per a day with a size of $5{\times}3{\times}1m$ and the automatic control devices.
