Multi-cycle chromatographic separation of Iysozyme from egg white was investigated. Multi-cycle chromatography was performed by repeated cycling(one cycle: resin equilibration, sample loading, washing, elution). Two types of cation exchange resins, Cellufine CM C-200 and Bio-rex 70, were used to determine the optimum condition for the separation of Iysozyme by multi-cycle chromatography. The resin was equilibrated in 20 mM Na-phosphate buffer(pH 7.0). Chromatograms of UV absorbance levels of every cycle were compared to confirm the eluting ability of Iysozyme in the two types of gel. Collected samples from eluting regions in every cycle were assayed by 15% SDS-PAGE.
We synthesized HPP-g-GMA copolymer using pre-irradiation method by E-beam and aminated HPP-g-GMA using amination reaction. Degree of grafting increased with increasing GMA monomer concentration and showed the maximum value of 130% at 1.46 M of GMA. The degree of amination increased with increasing the degree of grafting. When the degree of grafting was 100%, degree of amination showed the maximum value of 37.4%. The ion exchange capacity of aminated HPP-g-GMA was about 3.78 meq/g, and it showed remarkable adsorption ability of hollow fiber ion exchanger. Through the BET analysis, the surface area of aminated HPP-g-GMA was 54.83 $\m^2/g$ and the mean pore size was $26\AA$. These showed the decrease of surface area and the slight increase of the mean pore size. SEM results show that the thickness of fiber increased after the step of reaction and there pore blocking phenomena was not observed. The aminated HPP-g-GMA was synthesized successfully and found to be suitable for the adsorption and separation of anion.
Independent component analysis is a multivariate approach to separate mixed signals into original signals. It is the most widely used method of blind source separation technique. ICA uses linear transformations such as principal component analysis and factor analysis, but differs in that ICA requires statistical independence and non-Gaussian assumptions of original signals. PCA have a natural ordering based on cumulative proportion of explained variance; howerver, ICA algorithms cannot identify the unique optimal ordering of the components. It is meaningful to set order because major components can be used for further analysis such as clustering and low-dimensional graphs. In this paper, we compare the performance of several criteria to determine the order of the components. Kurtosis, absolute value of kurtosis, negentropy, Kolmogorov-Smirnov statistic and sum of squared coefficients are considered. The criteria are evaluated by their ability to classify known groups. Two types of data are analyzed for illustration.
Background: The Pediatric Balance Scale (PBS) was developed to assess of balance ability in children with balance problem. The PBS was translated into Korean and its reliability had been studied. However, it had need to be verified using psychometric characteristics including item fit and rating scale. Objects: The purpose of this study was to investigate the item fit, item difficulty, and rating scale of the Korean version of PBS using Rasch analysis. Methods: In total, 40 children with cerebral palsy (CP) (boy=17, girl=23) who were diagnosed with level 1 or 2 according to the Gross Motor Function Classification System participated in this study. The PBS was performed, and was verified regarding the item fit, item difficulty, rating scale, and separation index and reliability using Rasch analysis. Results: In this study, the 'transfer', and 'turning to look behind left and right shoulders while standing still' item showed misfit statistics. in total 40 children with CP. Also, 'transfer', 'standing unsupported with feet together' and 'standing with one foot in front' items showed misfit statistics in diplegia CP group. Regardless of the classification of CP, the most difficult item was 'standing on one foot', whereas the easiest item was 'sitting with back unsupported and feet supported on the floor'. The 4 rating scale categories of PBS were acceptable with all criteria. Both item and person separation indices and reliability showed acceptable values. Conclusion: The PBS has been proven reliable, valid and is an appropriate tool, but it needs to modify the items of PBS according to CP classification.
A sulfonated PP-g-styrene ion-exchange hollow fiber membrane was prepared by pre-irradiation method with E-beam followed by sulfonation reaction. Degree of grafting increased with the increase of styrene monomer concentration and showed the maximum value of 128% at 80% of styrene monomer composition. Sulfonation yield increased with the degree of grafting. At 100% degree of grafting, sulfonation yield showed the maximum value of 13.4%. Ion exchange capacity of sulfonated HPP-g-styrene of 3.42 meq/g was attained, resulting in the remarkable increase of adsorption ability BET analysis proved that the surface area of sulfonated HPP-g-styrene was 62.54 $m^2/g$ and the mean pore size was 25 $\AA$. From the BSA adsorption experiments, the adsorption amount of BSA was increased with sulfonation. At 13.4% sulfonation yield the adsorption amount of BSA was maximum as 3.8 mg/g. Sulfonated HPP-g-styrene was synthesized successfully and suitable for the adsorption and separation of BSA.
Bacillus subtilis XF-1, a strain with demonstrated ability to control clubroot disease caused by Plasmodiophora brassicae, was studied to elucidate its mechanism of antifungal activity against P. brassicae. Fengycin-type cyclopeptides (FTCPs), a well-known class of compounds with strong fungitoxic activity, were purified by acid precipitation, methanol extraction, and chromatographic separation. Eight homologs of fengycin, seven homologs of dehydroxyfengycin, and six unknown FTCPs were characterized with LC/ESI-MS, LC/ESI-MS/MS, and NMR. FTCPs (250 ${\mu}g/ml$) were used to treat the resting spores of P. brassicae ($10^7/ml$) by detecting leakage of the cytoplasm components and cell destruction. After 12 h treatment, the absorbencies at 260 nm ($A_{260}$) and at 280 nm ($A_{280}$) increased gradually to approaching the maximum of absorbance, accompanying the collapse of P. brassicae resting spores, and nearly no complete cells were observed at 24 h treatment. The results suggested that the cells could be cleaved by the FTCPs of B. subtilis XF-1, and the diversity of FTCPs was mainly attributed to a mechanism of clubroot disease biocontrol.
As an effective method for extraction of succinic acid, aqueous two-phase systems based on piperidinium ionic liquids were used in this study. Effects of the alkyl chain length of cation in piperinidium ionic liquids on phase diagram and extraction efficiencies were investigated. Experimental results show that aqueous two phase systems can be formed by adding appropriate amount of piperidinium ionic liquids to aqueous $K_2HPO_4$ solutions. It can be found that the ability of piperidinium ionic liquids for phase separation followed the order [OMPip][Br]>[HMPip][Br]>[BMPip][Br]>[EMPip][Br]. The biphase-forming ability of piperidinium ionic liquids was higher than that of imidazolium and pyrrolidinium ionic liquids in the presence of $K_2HPO_4$. 75~95% of the succinic acid could be extracted into the ionic liquid-rich phase in a single-step extraction. There was little difference in the extraction efficiency of succinic acid by piperidinium ionic liquids comparing to other ionic liquids such as imidazolium and pyrrolonidium ionic liquids. This aqueous two phase system by piperidinium ionic liquid is suggested to have effective application for the separation of succinic acid.
Natural anti-oxidative compounds have important disease prevention and food preservation properties, in addition to anti-bacterial, anti-inflammation, anti-cancer, and skin whitening effects. High-performance liquid chromatography (HPLC), with an ultra vilolet (UV) detector coupled to a reverse phase C18 column and an online measurement system for 1, 1-diphenyl-2-picryl hydrazyl (DPPH) radicals, was used to search for potent antioxidative compounds in crude extracts. The online HPLC-DPPH assay was then applied to confirm antioxidative compounds in water extracts from Radix of Pueraria lobata, Rhizoma of Zingiber officinale, Fructus of Chaenomeles sinensis, Cortex of Ulmus pumila, and Radix of Astragalus membranaceus. To determine the yields of the extracts, the Brix% of each extract solution was measured using a refractometer. When the relative DPPH radical scavenging ability values of the water extracts were compared with those of a positive control (ascorbic acid), the water extracts of P. lobata, C. sinensis, and U. pumila were 7.77%, 4.71%, and 4.19%, respectively. The results suggest that this method provides a useful assay for rapid measurement of DPPH radical scavenging abilities and conformation of antioxidative compounds in natural products. Moreover, it can reduce the time spent on the separation of active compounds from natural materials, such as medicinal plants, in addition to the use of reagents for separation.
The composite fiber adsorbents containing amidoxime group were prepared and separation properties of uranium ion from seawater were investigated. The amount of uranium adsorption was increased with an increase in adsorption time. When the mole ratio of monomer and comonomer, such as acrylonitrile (AN), tetraethyleneglycol dimethacrylate(TEGMA), and divinylbenzene (DVB), were 1 :0. 1 :0.003, this resin showed the maximum adsorption ability for uranium at a level of pH 8. The amount of uranium adsorption was also increased linearly to one hour with an increase in the content of adsorbent which was added in the composite fiber adsorbents(CFA). The maximum adsorption for uranium of CF A showed at $25^{\circ}C$. Hence, the adsorption ability of CF A for calcium and magnecium ions were increased gradually by the recycling of adsorption and disorption, the adsorption content of their on were 0.3, 0.9mmole/g-adsorbents, respectly. It also showed that the adsorption contents of Ca and \1g ions were much lower than them of uranium. The desorption of uranium on the CF A was carried out , bout 100% within 30min, and the desorption rate of various CF A were equalled.
Background: Rasch analysis estimates the probability that a respondent will endorse an item and select a particular rating for that item. It has the advantage of placing both the items and the person along a single ration scale and calibrates person ability and item difficulty onto an interval scale by logits. In addition, Rasch analysis is a useful tool for exploring the validity of questionnaires that have been developed using traditional methods. Therefore, it has been recommended as a method for developing and evaluating functional outcome measures. Objects: The purpose of this study was to investigate the item fit, item difficulty, and rating scale of the Korean version of the Fullerton Advanced Balance Scale (KFAB) using Rasch analysis. Methods: Total 97 subjects (39 males and 58 females) with dwelling elderly adults were participated, but 3 people were excluded for misfit persons. Rasch analysis was then done by means of the Winsteps program to determine the item fit, item difficulty, rating scale, and reliability of the KFAB. Results: In this study, the 'standing with feet together and eyes closed', 'two-footed jump', 'walk with head turns', and 'stand on foam, eyes closed' items shown misfit statistics. The most difficult item was 'stand on one leg', whereas the easiest item was 'turn in full circle'. The rating scale was acceptable with all criteria. Both item and person separation indices and reliability showed acceptable values. This would indicated that each domain covers a useful range of item difficulty that is appropriate for measuring a person with a wide range of functional ability. Conclusion: The KFAB has been proven reliable, valid and an appropriate tool with which to evaluate the balance of the elderly people.
