새로운 Tetracycline 유도적 유전자 발현 System의 In Vitro 검정 (Examination of Improved Tetracycline Inducible Gene Expression System In Vitro)

  • Until recently the most popular tetracycline-inducible gene expression system has been the one developed by Gossen and Bujard. In this study, we tested the latest version of same system and the results are summarized as follows: Compared with previous one, the difference of new system are minor changes of nucleotide sequences in transactivator and tetracycline response element (TRE) regions. Sensitivity to the doxycycline (a tetracycline derivative) was improved. Leakiness of GFP marker gene expression in non-inducible condition was significantly decreased. Higher expression of the marker gene was observed when the cells were fed with doxycycline-containing medium. Optimal insertion site of woodchuck posttranscriptional regulatory element (WPRE) sequence which was known to increase gene expression was different depending on the origin of cells. In chicken embryonic fibroblast, location of WPRE sequence at 3' end of TRE resulted in the highest GFP expression. In bovine embryonic fibroblasts, 3' end of transactivator was the best site for the GFP expression.

탄성콘크리트 댐의 모양최적설계 (Shape Optimal Design of Elastic Concrete Dam)

  • 본 연구에서는 평면 변형도상태 하에서 정수압을 받는 2차원 탄성 콘크리트 댐의 단면 모양을 최적화함으로써 댐의 질량을 최소화하였다. 최적화 문제의 목적함수로는 댐의 단면적이, 제약조건으로는 주응력 제약조건과 두께 제약조건들이, 설계변수로는 모델 경계의 모양이 채택되었다. 모델 영역의 변화에 따른 설계감도해석을 위해 최적화 문제를 범함수 형태로 변환한 후 연속체 역학의 물질미분 개념과 Adjoint Variable Technique 을 활용하였고, 최적화를 위해서는 Gradient Projection Method 를 사용하였다. 연구 결과 본 연구에 적용된 이론이 효율적이고 실제 탄성구조물 설계에 광범위하게 응용될 수 있음이 밝혀졌다.

Neuro-Fuzzy Control of Interior Permanent Magnet Synchronous Motors: Stability Analysis and Implementation

  • This paper investigates a robust neuro-fuzzy control (NFC) method which can accurately follow the speed reference of an interior permanent magnet synchronous motor (IPMSM) in the existence of nonlinearities and system uncertainties. A neuro-fuzzy control term is proposed to estimate these nonlinear and uncertain factors, therefore, this difficulty is completely solved. To make the global stability analysis simple and systematic, the time derivative of the quadratic Lyapunov function is selected as the cost function to be minimized. Moreover, the design procedure of the online self-tuning algorithm is comparatively simplified to reduce a computational burden of the NFC. Next, a rotor angular acceleration is obtained through the disturbance observer. The proposed observer-based NFC strategy can achieve better control performance (i.e., less steady-state error, less sensitivity) than the feedback linearization control method even when there exist some uncertainties in the electrical and mechanical parameters. Finally, the validity of the proposed neuro-fuzzy speed controller is confirmed through simulation and experimental studies on a prototype IPMSM drive system with a TMS320F28335 DSP.

Rhodamine derivative synthesis: dual-detectable chemosensor

  • Rhodamine chromophore/fluorophore have been attracted to many researchers due to its excellent photophysical properties. In this study, we have designed and synthesized a strong emissive fluorescent dye chemosensor for toxic elements. A rhodamine-based sensor was prepared by incorporation the rhodamine fluorophore and several functional host groups with high affinity to hazardous metal and anion. This sensor shows a high selectivity and an excellent sensitivity and is a dual-responsive colorimetric and fluorescent metal/anion-specific sensor. In addition, the 1:1 binding mode was proposed based on Job's plot method. Finally, computational calculation was simulated and calculated to approach for HOMO/LUMO of this dye chemosensor.

A New Rhodamine B Derivative As a Colorimetric Chemosensor for Recognition of Copper(II) Ion

  • A new rhodamine-based sensor 1 was designed and synthesized by incorporating rhodamine B and benzimidazole moieties. Sensor 1 exhibits high selectivity and sensitivity to $Cu^{2+}$ in $CH_3CN$-water solution (HEPES buffer, pH = 7.0) with an obvious color change from colorless to pink. Other metal ions such as $Hg^{2+}$, $Ag^+$, $Pb^{2+}$, $Sr^{2+}$, $Ba^{2+}$, $Cd^{2+}$, $Ni^{2+}$, $Co^{2+}$, $Fe^{2+}$, $Mn^{2+}$, $Cu^{2+}$, $Zn^{2+}$, $Ce^{3+}$, $Mg^{2+}$, $K^+$ and $Na^+$ had no such color change and have no significant influence on $Cu^{2+}$ recognition process. The interaction of $Cu^{2+}$ and sensor 1 was proven to adopt a 1:1 binding stoichiometry and the recognition process is reversible.

만성중이염에 대한 Pipemidic Acid의 치료효과 (Therapeutic Effect of Pipemidic Acid on OMPC)

  • 지난 약 반세기에 걸쳐서 Penicillin을 비롯하여 많은 항생제 및 화학요법제의 등장으로 급성 혹은 만성중이염의 치료에 많은 도움을 주었다. 그런데, 근래에 이같은 약제의 무절제한 사용 및 오용으로 인한 내성균의 출현과 균교대현상 등으로 치료에 곤란을 겪고 있는 실정이다. 연자 등은 만성적인 이루를 주소로 하여 내원한 만성중이염 환자 50례를 대상으로 무균상태하에서 이루를 채취하여 균을 배양 및 동정하였으며 pipemidicacid를 비롯한 여러 항생제에 대한 감수성검사를 시행하였다. 그결과 중이염환자 50례에서 staphylococcus aureus 및 epidermidis, pseudomonas aeruginosa 및 putrefasciens, proteus mirabilis, $\alpha$-hemolytic streptococcus, klebsiella pneumonia, unproved gramnegative rod, serratia marcescens, corynebacterium이 검출되었으며, pipemidic acid를 투여하여 pseudomonas와 proteus에 의한 중이염에 좋은 치료효과를 보였으며 staphylococcus를 비롯한 다른 감염균에도 괄목할만한 치료효과를 나타냈다. Pipemidic acid는 piromidic acid의 새로운 유도체로서 gram양성균 뿐만 아니라 pseudomonas를 포함한 gram음성체에도 저명한 항균효과가 있음을 알 수 있었다.

Strategy for the Seamless Mode Transfer of an Inverter in a Master-Slave Control Independent Microgrid

  • To enable a master-slave control independent microgrid system (MSCIMGS) to supply electricity continuously, the microgrid inverter should perform mode transfer between grid-connected and islanding operations. Transient oscillations should be reduced during transfer to effectively conduct a seamless mode transfer. This study uses a typical MSCIMGS as an example and improves the mode transfer strategy in three aspects: (1) adopts a status-tracking algorithm to improve the switching strategy of the outer loop, (2) uses the voltage magnitude and phase pre-synchronization algorithm to reduce transient shock at the time of grid connection, and (3) applies the hybrid-sensitivity $H_{\infty}$ robust controller instead of the current inner loop to improve the robustness of the controller. Simulations and experiments show that the proposed strategy is more practical than the traditional proportional-derivative control mode transfer and effective in reducing voltage and current oscillations during the transfer period.

인쇄 제판용 Photoresist의 잔막수율법에 의한 용해도 비교 (Solubility Comparison by Yield Method of Residual of Photoresist for Printing Plate)

  • Photoresist is defined as substance that makes chemical changes in its solubility, colouring and hardening by light energy. In this study, photosensitive photoresists of the positive type for a printing plate were studied. PF, o-, m-, p-CF resins as a matrix resin were synthesized at an identical condition. Photoresists were prepared by mixing NDS derivatives with a matrix resin at various mixing ratios. Characteristics of photoresists were studied by yield method of residual using solubility and Optical microscope was also analyzed. Prepared photoresist using NDS derivatives shows excellent photosensitivity and solubility compared with commercial product. The mixing ratio of 1:4(by mass) of NDS derivative[III] and m-CF resin shows the highest dissoultion rate among others. In addition, photoresist was obtained at this condition resulted in the superior sensitivity and contrast.

고질소 추진물질 합성 연구 (The Study on the Synthesis of Propellant with High Nitrogen Content)

  • 기존에 사용되고 있는 대부분의 추진물질들은 연소 시 이산화탄소, 염산가스 등의 환경유해 물질을 다량 발생시킨다. 본 연구에서는 이러한 문제점을 개선하기 위한 테트라진 계열의 저탄소 고질소 화합물인 DAAT의 합성공정을 확립하였다. 또한, 문헌에 빠져있는 구체적인 공정법 및 특성 분석 결과를 서술하였다. 그리고 분광분석(NMR, IR)을 통한 DAAT의 구조분석과 열, 충격, 마찰 안정성을 측정하였고, Gaussian 09와 EXPLO5를 이용하여 생성열과 폭발 특성(폭압, 폭속) 등을 계산하였다.

  • Lead, a heavy metal widely employed in various industries, continues to pose a threat to both human health and the environment. Therefore, the development of a sensor capable of rapidly and accurately detecting lead(II) ions in real-time at contaminated sites is crucial. In this study, we have engineered a fluorescent sensor with the ability to efficiently detect lead(II) ions under actual environmental conditions, including tap water and freshwater. The compound, tetraphenylethylene carboxylic acid derivative (TPE-COOH), exhibits high selectivity and sensitivity toward lead(II) ions in aqueous solution, where the interaction between TPE-COOH and lead(II) ions leads to its aggregation, thus triggering a fluorescence "turn-on" based on the aggregation-induced emission (AIE) mechanism. Impressively, compound TPE-COOH proficiently detects lead(II) ions within a range of 30 to 100 𝜇M in tap water and freshwater, even in the presence of various interfering substances.
