Segregation Mode of Plant Height in Crosses of Rice Cultivars Ⅸ. Crosses between Semi-dwarf Japonicas and Semi-dwarf(d-t) gene Testers (수도 품종간 교잡에 있어서 간장의 유전분리 Ⅸ. 단간 Japonica 품종과 Semi-dwarf (d-t) gene 검정친과의 조합)

  • In order to search for the semi-dwarf japonica varieties allelic to the semi-dwarf rice cultivar which is controlled by d-t gene, seven dwarf japonica varieties. Reimei, Hoyoku. Shiranui, Kokumasari, M 7. S.224 and S.295 were crossed to the semi-dwarf cultivar, wx 817. wx 817 is known to have semi-dwarf gene d-t. Their F$_1$, F$_2$ and F$_3$ were grown in 1984 and 1985 and culm lengths were measured at harvest. The results are summarized as follows. 1. The F$_2$s of all 7 cross combinations showed normal distribution and no segregation. 2. The range of culm length variation in the F$_3$ was variable depending on the cross combination, but the general pattern was similar in the all 7 crosses. 3. The mean of F$_3$ and parental F$_2$ mean which were selected into short, medium and tall groups were similar and showed no segregation, implying the selection efficiency in F$_2$. 4. From the results of F$_2$ and F$_3$ segregations, it is concluded that the culm length of the 7 semi-dwarf japonicas tested here are controlled by the same major gene d-t although they are modified by different minor genes.

  • The sorghum (Sorghum bicolor (L.) Moench) variety 'Sodamchal' was developed and registered by Department of Sothern Area Crop Science, National Institute of Crop Science (NICS) in 2013. This variety was developed from the cross between 'Hwanggeumchal' with brown grain and 'Jungmo4001' with semi-dwarf trait. 'Sodamchal' is the first semi-dwarf stature with waxy endosperm ever developed in Korea. The yield performance of 'Sodamchal' was 2.95 MT/ha in local adaptability test for 2 years. It had 91 cm culm length, which was 58 cm shorter than that of 'Hwanggeumchal', 26.3 cm panicle length, 22.0 mm thickness of culm, 21.1 g in 1,000 grain weight. It could be reduce the lodging problem and also adapted to mechanized cultivation. 'Sodamchal' produces high quantities of polyphenol and radical scavenging activitiy with diphenyl-picrylhydrazyl (DPPH), 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) in pigmented testa. Above all 'Sodamchal' is suitable for use as a parental source for the development of improved pollinator parent lines for developing semi-dwarf sorghum with waxy endosperm and antioxidant activity. Analysis of genome variants in dwarf soybean lines obtained in F6 derived from cross of normal parents (cultivated and wild soybean)

  • Plant height is an important component of plant architecture and significantly affects crop breeding practices and yield. We studied DNA variations derived from F5 recombinant inbred lines (RILs) with 96.8% homozygous genotypes. Here, we report DNA variations between the normal and dwarf members of four lines harvested from a single seed parent in an F6 RIL population derived from a cross between Glycine max var. Peking and Glycine soja IT182936. Whole genome sequencing was carried out, and the DNA variations in the whole genome were compared between the normal and dwarf samples. We found a large number of DNA variations in both the dwarf and semi-dwarf lines, with one single nucleotide polymorphism (SNP) per at least 3.68 kb in the dwarf lines and 1 SNP per 11.13 kb of the whole genome. This value is 2.18 times higher than the expected DNA variation in the F6 population. A total of 186 SNPs and 241 SNPs were discovered in the coding regions of the dwarf lines 1282 and 1303, respectively, and we discovered 33 homogeneous nonsynonymous SNPs that occurred at the same loci in each set of dwarf and normal soybean. Of them, five SNPs were in the same positions between lines 1282 and 1303. Our results provide important information for improving our understanding of the genetics of soybean plant height and crop breeding. These polymorphisms could be useful genetic resources for plant breeders, geneticists, and biologists for future molecular biology and breeding projects.

Possibility of breeding super rice cultivars using gene linkage

  • The rapid growth of world population, reduction of cultivated areas for crop production, and detrimental effects of pests, diseases, and climate changes have required to breed new rice cultivars with high yield, accepted quality, but strong resistance to abiotic and biotic stresses. However, traditional breeding needs much time to breed a new cultivar, whereas the successful use of molecular breeding is still questionable. We have developed a novel mutation which allow to cross many rice cultivars together with low segregation, that allow to breed a new cultivar in only several cropping. The mechanism has been unknown, but we suggest that gene linkage may play a crucial role, of which the semi dwarf gene might be the center gene for gene linkage occurrence. The phenomenon of this possible gene linkage is contrary to Mendel rules, but it is promising to breed new rice cultivars, of which, the most elite genes in rice might be able to gather in a targeted rice variety.

  • In order to investigate the causes of epidemic out break of rice blast disease in 1978, investigations were undertaken in respect of climatic conditions, variety, cultural practice and plant pathology. During 1978, especially in August at heading time, it was higher temperature and humidity, higher frequency and amount of rainfall, lower amount of sunshine and solar radiation than less blast infested years. Nitrogen content in rice plant was higher than previous years. Acreage increase of semi-dwarf varieties brought about a result of proportional increase of new blast races which are able to infect the semi-dwarf varieties. It was concluded that those conditions mentioned above might have caused the result of severe neck blast disease in rice varieties in Korea, 1978.

Varietal Difference in Changing Aspect of Daily Sink Filling During the Grain Filling Period in the Rice Plants (수도증열기간중 Sink충전의 경시적 변화와 그 품종간 차이)

  • The experiment was conducted to know the varietal difference in changing aspect of daily filling sink during of rice using three cultivars; ‘Suweon 295’ (japonica), ‘Suwoen 264’ and ‘IR 1317-70-1’ (semi-dwarf indica) cultivated in the field and green house of the Crop Experiment Station in 1983. There was not significant varietal difference in flowering habit and duration of flowering period each panicle. Semi-dwarf indica cultivars showed shorter days to physiological maturity of rice kernel than the japonica one. The former also displayed earlier decrease in water content of rice grain during ripening than the latter. Japonica variety revealed larger difference in ripening advancement between vigorous florets and inferior one than semi-dwarf indica varieties. The semi-dwarf indica rice cultivars expressed significantly shrewder parabola of daily filling of sink per panicle during ripening than the japonica one. The time showing maximum capacity of daily filling of sink per panicle during ripening was 10-12 days after anthesis. The filling capacity of daily sink filling per panicle for 'IR 1317-70-1', 'Suweon 264' and 'Suweon 265' at this time were about 240 mg, 165 mg and 145 mg, respectively. Pattern of sink filling per panicle during ripening was slightly changed by the cultural environment. ‘Suweon 295’ showed slightly sharper parabola and 2-3 days earlier time showing maximum capcity of daily sink demand per panicle during ripening in the field than those in the green house.

Varietal Differences in Agronomic Characters under Different Altitudinal Locations with Equal Latitude in Paddy Rice (동일위도상 표고의 차이에 따른 수도품종의 수량형질변이)

  • The present study was conducted to investigate the variations in morphological traits, yield and yield-related components of rice cultivars exposed to different weather conditions by growing at the locations with different altitude but with equal latitude. Three Japonica varieties (Daegoldo, Jinheung, Shin #2) and three Tongil type semi-dwarf varieties(Josaengtongil, Suweon 287, wx 817-65-2-3) were seeded at Suweon on April 21 and transplanted at Suweon ($127^{\circ}$ N37 20', altitude 37m) and Jechun ($128.2^{\circ}$, N37 10', altitude 280-300m) on June 1 with the spacing of 30 $\times$ 15 em. The morphological traits, yield and yield-related components were measured. Culm length, paincle length and 1000-grain weight showed a little increase in Japonica varieties in Jechun as compared to those in Suweon and vice versa in semi-dwarf varieties, showing no significant differences. Number of panicles, per hill and number of spikelets per panicle among yield-related components showed remarkable increase in all varieties in Jechun as compared to those in 'Suweon but such inter-locational differences differed with varieties within varietal group and between varietal groups. On the contrary, grain fertility was higher in Suweon than that in Jechun and in Japonica varieties than that in semi-dwarf varieites , showing greater inter-locational difference in semi-dwarf varieties. Among Japonica varieties Jinheung and Shin #2 showed remarkable increase in yield in Jechun as compared to that in Suweon but in semidwarf varieties it was just the opposite. The importance of each of the yield-related component contributing to yield showed similar tendency 'regardless of locations in Japonica varieties. However, it was evidently different between locations in semidwarf varieties, where number of spikelets per panicle showed greatest direct effect on yield in Suweon but in Jechun so did grain fertility followed by the number of spikelets per panicle.

Evaluation of Varietal Difference and Environmental Variation for Some Characters Related to Source and Sink in the Rice Plants (벼의 Source 및 Sink형질의 품종간차이와 환경변이의 평가)

  • Experiments were carried out to evaluate the standard gravity in determining potential kernel size and to determine the effective sampling way by analyzing intra - and inter - plant variations for some source and sink characters using eleven semi-dwarf indica and three japonica cultivars including four semi-dwarf indica nearisogenic lines. Also, additional experiments were conducted to understand yearly variation and variety x year interaction effects for ten characters related to source and sink and to characterize the varietal difference of pre- and post-heading self-competition employing three parental varieties and their F$\sub$5/ progenies in 1982 and 1983. It is desirable to determine the potential kernel size by average kernel wight of rice grains showing above 1.15 specific gravity. There was significant difference in leaf area per tiller, spikelets and sink capacity per panicle among vigorous, intermediate and inferior tillers classified by differentiated order and vigorousness. Although it was difficult to find out any significant difference in grain-fill ratio, ratio of perfectly ripened grain, potential kernel size and sink/source ratio between vigorous and intermediate tillers, there was big difference between them and inferior one. The coefficients of variation within each tiller-group for some characters related to source and sink were larger with the order of vigorous tillers < intermediate one '||'&'||'lt; inferior one, and the average heritability of all characters, evaluated by the ratio of varietal variance (equation omitted) to total variance (equation omitted), were higher with the order of inferior tillers '||'&'||'lt; intemediate one '||'&'||'lt; superior one. Therefore, it is desirable to sample the vigorous tillers to represent the varietal difference of these traits. '82-'83 year variations of three parental cultivars were significant for all traits except for leaf area/tiller, panicles/hill, leaf area index and rough rice yield. The characters showing highly significant variance of variety x year interaction were growth duration from transplanting to heading, leaf area/tiller, sink/source ratio, sink capacity/panicle and grain yield. Generalized yearly response of three parental varieties (Suweon 264, Raegyeong, IR1317-70-l) and their F$\sub$5/ progenies on the 1st and 2nd principal components extracted from ten source and sink characters generally exhibited reduction in both source and sink. However, there were diverse variety x year interactions such as progenies showing similar reaction with their parents and intermediate or recombinational yearly response with little or considerable yearly movement on the four-dimensional planes of the two upper principal components between 1982 and 1983. Sink characters revealing highly significant border effect were grain-fill ratio, spikelets and sink capacity per panicle. Among them the latter two especially showed significant variety x border effect interaction. Self-competition characterized by relative weakness of inside plant's sink characters compared to the border one was more severe during the reproductive stage before heading than maturing stage. Though the larger sink capacity per panicle generally disclosed the severer self-competition, some lines (like Suweon 264) revealed severe self-competition with small sink capacity while a few others showed tender self-competition in spite of big sink capacity per panicle.

Morphological and Progeny Variations in Somaclonal Mutants of 'Ilpum' (Oryza sativa L.) ('일품'벼 체세포변이체의 표현형과 후대변이)

  • A total of 424 plants was regenerated from the seed-culture of a rice cultivar, 'Ilpum'. The regenerated plants were grown in a greenhouse. The 297 plants with high fertility were selected among 424 plants. The harvested seeds from each plant were planted to each line at experiment field in 2008 and 2009. The each line was evaluated for the agronomic and morphological traits, also. The 64 lines (21.5%) showed significant differences in agronomic and morphological traits from donor cultivar 'Ilpum' among 297 lines. The heading date different from donor cultivar 'Ilpum' showed highest frequency in 297 lines, and accounts for 9.1% (29 lines). The phenotype of opaque endosperm and rolling leaf account for 1.7% and 1.3% in 297 lines, respectively. The genetic segregation was observed in dwarf/semi-dwarf, rolling leaf and opaque endosperm at $S_1$ generation, but not in $S_2$ generation. These results suggest that the mutant derived from a tissue-culture will be one of the promising genetic resources, due to its wide variation and high frequency of mutation, comparatively.
