산조인(酸棗仁)과 Lithium Salt가 가토(家兎) 혈청(血淸) Ceruloplasmin에 미치는 영향(影響) (Effects of Zizyphi spinosi Semen and Lithium Salt on the Serum Ceruloplasmin in Rabbits)

  • The effects of the ether extract of Zizyphi spinosi semen and lithium chloride on the serum ceruloplasmin level in blood of rabbits were examined. An oral dose 1gm/kg/day of the ether extract and oral dose of lithium chloride 1 Meq/kg/day with 1gm/kg/day of the ether extract were administered respectively to rabbits for eight days, The results are: 1)Serum ceruloplasmin level in blood of the rabbits treated with the ether extract was higher than that of the control group. 2)The level of the rabbits treated with both the ether extract and lithium chloride was higher than that of the control. 3)The administration of lithium chloride alone also increased ceruloplasmin blood level.

Comparison of the deleterious effects of yaji and cadmium chloride on testicular physiomorphological and oxidative stress status: The gonadoprotective effects of an omega-3 fatty acid

  • Objective: This study investigated testicular oxidative stress status and physiomorphological function in Wistar rats fed with yaji and cadmium chloride (CdCl2). Methods: Sixty male albino Wistar rats (12 per group) were randomly assigned to five groups: group I (control), group II (300 mg/kg.bw of yaji), group III (500 mg/kg.bw of yaji), group IV (2.5 mg/kg.bw of CdCl2), and group V (2.5 mg/kg.bw of yaji+4 mg/kg.bw omega-3). Each group was evenly subdivided into two subgroups and treatment was administered for 14 days and 42 days, respectively. Semen quality (sperm count, progressive motility, normal morphology, and gonadosomatic index), hormones (testosterone, follicle-stimulating hormone, and luteinizing hormone), testicular oxidative stress markers (superoxide dismutase, catalase, glutathione peroxidase, and malonaldehyde) and testicular histomorphological features were examined. Results: Yaji caused significant (p< 0.05) dose- and duration-dependent reductions in semen quality, the gonadosomatic index, testosterone, follicle-stimulating hormone, and luteinizing hormone. Yaji also caused significant (p< 0.05) dose- and duration-dependent decreases in superoxide dismutase, catalase, and glutathione peroxidase activity, as well as increased testicular malonaldehyde levels. Yaji induced distortions in the testicular histological architecture. CdCl2 damaged testicular function by significantly (p< 0.05) reducing semen quality, reproductive hormone levels, and oxidative stress markers in albino Wistar rats. CdCl2 also altered the histology of the testis. Conclusion: This study shows that yaji sauce has similar anti-fertility effects to those of CdCl2, as it adversely interferes with male reproduction by impairing oxidative stress markers and the function and morphological features of the testis.

  • This study was undertaken to determine Juglandis Semen extraction (JS) has a protective effect against the cell injury caused by oxidants, t-butylhydroperoxide (t-BHP) and $H_{2}O_2$ in rabbit lung slices. Cell injury was estimated by measuring tissue water content and peroxidation of membrane lipids was assessed by measuring malondialdehyde (MDA), an end-product of lipid peroxidation. t-BHP significantly increased water content in lung tissues over concentrations of 2-10 mM, and such effects were prevented by 5% JS. JS exerted the beneficial effect in a dose-dependent manner. $H_{2}O_2$ (100 mM) also increased water content in tissues, which was almost completely prevented by 5% JS. t-BHP induced lipid peroxidation in a dose-dependent fashion in lung tissues over concentrations of 0.5-10 mM. JS significantly reduced t-BHP induced lipid peroxidation and oxidant-independent endogenous lipid peroxidation, and such effects were dose-dependent at concentration of 0.5-10%. JS prevented $H_{2}O_2$ (100 mM)-dependent lipid peroxidation. These results suggest that JS prevents ceil injury induced by oxidants in the lung, and such effects may be attributed to inhibition of lipid peroxidation. The precise mechanisms remains to be explored.

  • Purpose: This study was conducted to investigate effects of Allii tuberosi Semen, Ginseng Radix Alba and Morindae officinalis Radix extract on reproductive capacities in mice. Methods: The 8-week-old ICR rats are used in the study, and we administered the extract solution of Allii tuberosi Semen, Ginseng Radix Alba and Morindae officinalis Radix concentration of 1, 10, 100mg/0.3ml to each rats and water to another rats(control group) once a day for each 30, 60, 90 and 120 days. we evaluated the total sperm count and morphology. Results: For 30 days, the total sperm count and normal sperm most increased in the group administered by 100mg/0.3ml Allii tuberosi Semen. For 60 days, the total sperm count and normal sperm most increased in the group administered by 100mg/0.3ml Morindae officinalis Radix solutions. For 90 days, the total sperm count most increased in the group asministered by 10mg/0.3ml Morindae officinalis Radix and 100mg/0.3ml Allii tuberosi Semen solutions. For 120 days, the total sperm count and normal sperm most increased in the group administered by l0mg/0.3ml Ginseng Radix Alba. Conclusion: This study shows that there were dose and duration effects of Allii tuberosi Semen, Ginseng Radix Alba and Morindae officinalis Radix extract solution on the total sperm count and morphology.

Effects of intravenous multiple busulfan injection on suppression of endogenous spermatogenesis in recipient stallion testes

  • Preparation of recipient stallions is critical step to produce donor spermatogonial stem cell (SSC) derived sperm using transplantation technique. This study was conducted to evaluate the effects of intravenous busulfan infusion on germ cell depletion, semen production, and libido in stallions. Six Thoroughbred stallions were separated into two treatment groups: 1) a multiple low-dose (2.5 mg/kg bw for the first 4 weeks and 5 mg/kg bw for the 5th week); and 2) control group treated with PBS. Testicular samples were obtained at 11 weeks and classified into three different patterns of spermatogenesis, such as normal, Sertoli cell only, and destroyed. Semen collection and libido experiments were performed 1 week before treatment, and 4 and 8 weeks after treatment. For the sperm analysis, total spermatozoa and motility were measured using a light microscope with a motility analyzing system. In the multiple low-dose group, the numbers of tubules categorized as Sertoli cell only were significantly higher than those in the control as well as the total population and total/progressive motility of sperm were significantly decreased 8 weeks after the start of the treatment. The sperm production and motility in the multiple low-dose group appears to be reduced, while libido was maintained. In conclusion, multiple administration of 2.5 mg/kg bw busulfan depletes endogenous germ cells in the stallion recipients for SSC transplantation.

활성산소에 의한 심근독성 및 동과의 보호효과에 관한 연구 (Cardiotoxicity and Effect of Benincasae Semen on the Reactive Oxygen Species)

  • To examine the cardiotoxicity of glucose oxidase(GO) in cultured myocardial cells, cardiotoxicity was measured by MTT assay. Myocardial cells were treated with 1~50 mU/ml GO for 5 hours. The cardioprotective effect of Benincasae Semen(BS) was measured by MTT assay in these cultrures. Cell viability was significantly decreased in dose- and time-dependently after myocardial cells were exposed to 30mU/ml GO for 5 hours. In the cardioprotective effect of BS on the cardiotoxicity induced by GO, BS prevented the cardiotoxicity induced by GO in these cultures. From these results, it suggests that GO had cytotoxic effect in cultured myocardial cells and herb extract, BS showed protective effect on GO-induced cardiotoxicity.

메틸수은으로 손상된 배양 심근세포에 대한 동과자의 영향 (Effect of Benincasae Semen on Methylmercury-Induced Myotoxicity in Cultured Myocardial Cells)

  • To clerify the toxic effect of methylmercuric chloride(MMC) in cultured mouse myocardial cells, cytotoxic effect was measured by MTT assay after cultured myocardial cells were incubated for 48 hours in the media containing 1~30 uM concentrations of MMC. And also, the protective effect of Benincasae Semen (BS) was assessed in these cultrures. Cell viability was significantly decreased in a dose-dependent manner after cultured myocardial cells were exposed to 30 uM MMC for 48 hours. In the neuroprotective effect of BS on MMC-induced cytotoxicity, BS blocked the MMC-induced myotoxicity in these cultures. From these results, it suggests that MMC is toxic on cultured mouse myocardial cells and BS is effective in blocking the neurotoxicity induced by MMC.

속수자 추출물의 HT-29 대장암세포 증식에 대한 억제효과 (Inhibitory effects of Euphorbiae lathyridis Semen extract on cell growth in HT-29 human colon cancer cells)

  • Objectives. In this study, we investigate that methanol extract of Euphorbiae lathyridis Semen contributes to growth inhibitory effect on the HT-29 human colon cancer cells. Methods. Euphorbiae lathyridis Semen (ELS) was extracted with 80% methanol. HT-29 cells were treated with different concentrations of ELS extract for 24-72 hrs. Growth inhibitory effect was determined by MTT assay. Cell apoptosis was determined by surveying caspases cascades activation using Western blot. Cell cycle arrest was analyzed by flow cytometry with PI staining. Results. Exposure to ELS extract showed in inhibitory effects on HT-29 cell growth as a dose-dependent manner. Cell growth inhibition by ELS extract was related with induction of cell apoptosis with DNA fragmentation through the activation of caspases-3, caspase-9 and PARP cleavage. Conclusion. ELS extract significantly inhibited cell growth and induced cell apoptosis in HT-29 human colon cancer cells, therefore, These results suggest that ELS extract can be used as chemoprevention agent of colon cancers.

토사자 추출물의 ketoconazole로 유도된 고환 독성 흰쥐의 산화적 스트레스 저해효과 (Protective Effects of Water Extract from Cuscutae Semen on Ketoconazole-Induced Oxidative Stress in Testicular Damage Male Rats)

  • Ketoconazole (KET) is an antifungal drug with a broad spectrum of activity that also induces reproductive toxicity in humans and animals. KET inhibits C17-20 lyase which blocks the conversion of 17 ${\alpha}$-hydroxyprogesterone to androstenedione. The effect of Cuscutae semen(CS) extract against KET-induced testicular damage was evaluated in male rats. CS extract was administered orally (100 and 200 mg/kg) for 26 days. Three weeks after CS extract administration, KET was CS-administered intraperitoneally at a dose of 100 mg/kg once a day for 5 days. KET-induced reproductive toxicity was associated with clear reductions of the weights of testes and epididymides, sperm indices and serum testosterone levels. In addition, marked oxidative damage to testicular lipids and alterations of natural antioxidant enzymes were reported in association with KET toxicity. Most of the KET-induced effects were greatly decreased with the concomitant application of CS extract. This study suggests a protective role of Cuscutae semen extract that could be attributed to its antioxidant properties.

Single Oral Dose Toxicity Study of Prebrewed Armeniacae Semen in Rats

  • Armeniacae semen (AS) has been considered a toxic herb in the Korean medicine as it contains hydrogen cyanide and amygdalin, especially in its endocarp. Therefore, prebrewed AS that is devoid of endocarp has been traditionally used. In the present study, amygdalin content of the prebrewed AS was significantly lower ($2.73{\pm}0.32{\mu}g/ml$; p<0.01) than the content in the extract that contained the endocarps ($28.50{\pm}6.71{\mu}g/ml$); amygdalin content corresponded to 10% of the extract in the present study. Because of single oral dose toxicity of prebrewed AS according to the recommendation of Korea Food and Drug Administration Guidelines (2009-116, 2009), which was based on single oral dose toxicity study of prebrewed AS, mortality due to toxic principles was significantly reduced. In this study, 2,000 mg/kg of prebrewed AS led to death of 1 female rat and 1 male rat at the end of 2 hr of administration. Based on these results, the 50% lethal dose in both male and female rats was determined to be 9279.5 mg/kg. Seizure, loss of locomotion, and increases in respiration and heart rate were observed as prebrewed AS treatment-related toxicological signs; these signs were restrictedly manifested in the prebrewed AS (2,000 mg/kg)-treated rats. In addition, no changes were observed in body weight, organ weight, gross features, and histopathological parameters with 2,000 mg/kg of AS in both male and female rats. These findings serve as direct evidence that amygdalin in AS is the toxic principle, which can be reduced by the traditional prebrewing method involving the exclusion of endocarp.
