The Effect of Hair Essence (HHRHG0202-80) Containing Five Herbal Extracts on Hair Growth and the Prevention of Alopecia in vitro & vivo (5종의 한약 추출물이 함유된 헤어 에센스제품(HHRHG0202-80)이 in vitro 및 in vivo에서 육모 및 탈모 방지에 미치는 효과)

  • Backgrounds : Androgenetic alopecia is a relatively common disorder, but its precise mechanism is not elucidated. There are two commercial drugs approved by FDA. One(finasteride) has an inhibition activity of 5$\alpha$-reductase(type 2) and the other(minoxidil) has a vasodilation activity. Objectives : A verified herbal remedy for baldness is needed for medicinal treatment or preventing alopecia, which could be demonstrated by alopecia-related in vitro & in vivo tests Methods : On the basis of oriental pharmacognosy, we classified many herbal medicines into four groups (malnutrition, aging, alopecia and gray hair) according to its effect. The mitosis induction of hairy dermal papillae cell and the metabolic inhibition for type 2 $5{\alpha}-reductase$ were tested with five herbal extracts. Also, five herbal extracts were added to the normal essence formulation (HHRHG0202-80) in ranges of 0.1~0.3%, which was applied two-mouse models to validate each hair growing activity in vivo. Results : Stimulation of follicular papillae cell proliferation was observed in treatment of three herbal extracts (Glycyrrhizae radix:159.7%, Corni fructus : 144.7%, and Coicis semen : 136.6%) at a dose of $10\mu\textrm{g}/ml$. Three herbal extracts (Biotae semen. Glycyrrhizae radix and Coicis semen) showed inhibitory activity for $5{\alpha}-reductase$(type 2) at 93.18%, 73.36% and 47.6%, respectively at the same dose. We observed the enhancement of hair growth activity in C57b1/6 mouse and the inhibition of alopecia in AGA mouse after topical administration of the hair essence. Conclusions : Hair essence product, which contains five medicinal plants, would be used for the remedy for male pattern baldness (MPB) and the other alopecia diseases.

Protective Effects of Thujae Semen against Neurotoxicity Induced by 6-hydroxydopamine in PC12 Cells (백자인의 6-하이드록시도파민으로 유도된 뇌세포독성에 대한 보호효과)

  • Objectives : This study was performed to evaluate the neuroprotective effect of water extracts from Thujae Semen(TSW) in PC12 cells. Methods : We performed 2,2-diphenyl-2-picrylhydrazyl(DPPH) radical scavenging assay, 2,2-azinobis- (3-ethyl-benzothiazoline-6-sulfonic acid(ABTS) cation scavenging assay, and determination of total polyphenolic content to examine the antioxidant effects of TSW. We also carried out 3-(4,5- dimethylthiazol-2-yl)-2,5-diphenyl-tetrazolium bromide assay(MTT), reactve oxygen species(ROS) assay, and nitric oxide(NO) assay to examine neuroprotective effects against 6-hydroxydopamine(6-OHDA) in PC12 cells. Results : TSW showed $IC_{50}$ values of 404.3 and 219.9 ${\mu}g/mL$ in DPPH and in ABTS assays, respectively. TSW showed 9.74 ${\mu}g/mL$ of total polyphenol contents. TSW incresed cell viability in a dose dependent manner and it showed protective effect against 6-OHDA neurotoxicity at the concentration of 25-200 ${\mu}g/mL$. Moreover, it recovered 6-OHDA induced cell death at the same concentrations. The extract showed a dose dependent reduction of ROS and NO generation by 6-OHDA. Conclusions : We concluded that TSW has neuroprotective effect against 6-OHDA-induced toxicity in PC12 cells through ROS and NO inhibition.

The Effect of Juglandis Semen Extract Solution on Oxidant-Induced Alteration of Glutamate Uptake in Rabbit Brain Synaptosome (호도약침액(胡桃藥鍼液)이 가토(家兎) 뇌(腦)의 Synaptosome에서 Oxidant에 의한 물질이동계(物質移動系)의 장애(障碍)에 미치는 영향(影響))

  • This study was undertaken to determine whether Juglandis semen extract solution (JLS solution) exerts protective effect against oxidant-induced inhibition of glutamate uptake by synaptosomes. Synaptosome was prepared from rabbit brain cortex. Glutamate uptake increased by incubation time during 10 minutes, which was significantly inhibited by 1mM t-buthylhydroperoxide(t-BHP). JLS solution prevented t-BHP-induced inhibition of glutamate uptake in a dose-dependent manner. t-BHP reduced glutamate uptake in dose-dependent fashion, which was significantly prevented by 2% JLS solution. t-BHP(1mM) and $ascorbate/Fe^{2+}(50/1{\mu}M)$ increased lipid peroxidation in synaptosomes by 5-fold, and it was significantly prevented by 2% JLS solution. $HgCl_2(0.1mM)$ inhibited glutamate uptake and increased lipid peroxidation. These changes were prevented by 2% JLS solution. Synaptosomal Na-K-ATPase activity was inhibited by t-BHP(1mM) and $H_2O_2(50mM)$, which was prevented by 2% JLS solution. The results indicate that JLS solution prevents oxidant-induced inhibition of glutamate by synaptosomes, and this may result from inhibition of lipid peroxidation induced by oxidants.

Assessing the Use of 5 ml Straws in the Cryopreservation of Boar Semen (돼지 정자 동결보존에 있어 5 ml straw의 한계성 극복)

  • The aim of this study was to overcome some of the limiting factors that the maxi cryopreservation straw of 5 ml presents in processing boar semen. Cryopreservation of semen samples was conducted in 0.5 ml and 5.0 ml straws at two freezing rates: -140℃ in 8 minutes and 30 seconds (FR-1) and -140℃ in 14 minutes (FR-2). The straws were then thawed and the semen parameters were compared by Computer Assisted Sperm Analysis, and sperm morphology and acrosome status were examined by Coomassie blue staining. The effects of different thawing temperatures and durations were also compared, namely 37℃ for 115 sec, 50℃ for 45 sec, or 70℃ for 25 sec. In general, the FR-1 group showed higher (p<0.05) sperm viability and motility than the FR-2 group in the 5.0 ml straws. Compared to other ranges, thawing at 50℃ for 45 sec showed the highest sperm viability and motility (68.4±3.6% and 69.5±2.2%, p<0.05), suggesting that thawing temperature should be adjusted concurrently with freezing rate. Sperm morphology and acrosome integrity did not significantly differ among the groups (p>0.05). The data obtained in this study suggest that improving the freezing-thawing protocol for one artificial insemination dose straws (5.0 ml) retains the sperm's parameters from 0.5 ml cryopreservation, and is more convenient to handle, which could result in enhanced reproductive performance.

Effects of Breeds, Insemination Time, Breeding Season, Sperm Concentration on Reproductive Performance of Sows Inseminated by Liquid Boar Semen (액상정액을 이용한 인공수정시 품종, 계절, 인공수정 횟수 및 정자농도가 번식성적에 미치는 영향)

  • This study was carried out to investigate the effects of liquid boar semen on reproductive performance in swine artificial insemination. Many factors, which were breeds, time of insemination, breeding season, sperm per dose etc, have been tried to improve reproductive efficiency. Boars were raised at Swine Artificial Insemination Center in National Livestock Research Institute, Sunghwan, Chungnam, Korea. This experiment was carried out from 1995 to 2000. There were no differences in the fertility results compared with 3 breeds (Landrace, Yorkshire and Duroc), frequencies of artificial insemination (double and triple) per estrus cycle and different seasons by using liquid boar semen. There were no significant differences in conception rate, farrowing rate and litter size using 4 trials of 3.0, 2.5, 2.0 and $1.5{\times}10^9/80ml$ in liquid boar semen with 70% of motile sperm cells. We confirmed that the sperm number per dose of $1.5{\times}10^9/80ml$ could be used for commercial artificial insemination.

  • This study was undertaken to determine if Juglandis Semen Aquacupuncture(Ja) has a protective effect against glycerol-induced acute renal failure in rats. Rats were dehydrated for 24hr and then injected with $5m{\ell}/kg$ of 50% glycerol, one-half of dose in each hindlimb muscle. In experiments for Ja effect, rats received $0.1m{\ell}$ of Ja extraction in both sides of corresponding $Sh{\grave{e}}ns{\bar{u}}$($BL_{23}$) of human body and non-acupuncture points(the root of tail) for 3 days after injection of glycerol. The experimental group were divided into the Normal group, the Control group, the Ja to $Sh{\grave{e}}ns{\bar{u}}$($BL_{23}$) group(Ja-AS), the Ja to non-acupuncture points group(Ja-AN). There were significant decrease of Urine vollume, total protein and phosphate level in Ja-AS as compared with the control group. There were not any significant change of Urine creatinine in Ja-AS as compared with the control group. There were significant decrease of Unine glucose in Ja-AS, Ja-AN as compared with the control group. This suggests that Ja-AS could be used in prevention and treatment of acute renal failure. However, the precise mechanisms of Ja protection remain to be determined.

Study on the Anti-allergic Effect of Armeniacae Semen Herbal Acupuncture Solution (행인약침(杏仁藥鍼)이 항알레르기에 미치는 영향)

  • Objectives : The purpose of this study was to examine the anti-allergic effect in vivo, and to observe single toxicity in mice of Armeniacae Semen herbal acupuncture solution (ASHA). Methods : We investigated anti DNP IgE-mediated passive cutaneous anaphylaxis in rodents and compound 48/80-induced active systemic anaphylatic shock in mice after treatment at both BL13 with ASHA of 25 ${\mu}{\ell}$(mice) or 50 ${\mu}{\ell}$(rats) 3 times for 5 days. To ascertain safety and toxicity of ASHA, we examined single toxicity test. In single test, three groups were treated with different dosages of ASHA (ASHA250, ASHA500 and ASHA1000) according to on Korean Food and Drug Administration, respectively. We observed attentively motality, abnormal clinical sign, body weight change, and organ weight of mice after ASHA treatment. Results : ASHA inhibited passive cutaneous anaphylaxis and active systemic anaphylatic shock by oral administration. During toxicity experiment period, there was no difference in body weight change, and organ weight among different dose groups. Death were not found in single test i.p. group. (ASHA250, ASHA500 and ASHA1000). Several individuals of single test i.p. group were observed yellow brown discharge around anus in early period after administration. Conclusions : These results indicate that ASHA have inhibition effects on passive cutaneous anaphylaxis and active systemic anaphylatic shock, and suggest that has some toxicity in high dosage.

Radical Scavenging and Antioxidant Effects of Juglandis Semen Extract(JSE) (호도약침액(胡桃藥鍼液)의 유리기(遊離基) 소거(消去)와 항산화(抗酸化) 효과(效果)에 대한 실험적(實驗的) 연구(硏究))

  • This study was performed to determine if Juglandis semen extract(JSE) has free radical scavenging and antioxidant activities. Superoxide anion generation by xanthine oxidase/xanthine and in neutrophils activated by phorbol-12, 13-dibutyrate was inhibited by JSE and its effect was dose-dependent. JSE also inhibited generation of $H_2O_2$ induced by glucose oxidase/glucose and in opossum kidney cells treated with antimycin A. JSE exerted a direct $H_2O_2$ scavenging effect. Exposure of opossum kidney cells to 1mM tBHP caused a significant increase in lipid peroxidation, which was prevented by JSE. JSE also prevented tBHP-induced LDH release. These data suggest that JSE has free radical scavenging and antioxidant activities. However, further studies should be carried out to find the active ingredient(s) of JSE that exerts radical scavenging action.

Study on the Effect of Benincasae Semen on Cultured Mouse Myocardial Cells Damaged by Adriamycin (Adiamycin에 의해 손상된 심근세포에 대한 동과의 영향에 관한 연구)

  • To examine the cardiotoxic effect of adriamycin on cultured rat myocardial cells, cytotoxicity was measured by MTT assay after cultured myocardial cells were grown with various concentrations of adriamycin(ADA) for 48 hours. The protective effect of Benincasae Semen(BS) on ADR-induced cardiotoxicity was also examined in these cultures. ADR decreased cell viability of cultured rat myocardial cells remarkably in a dose- and time-dependent manners. In protective effect of BS, it was very effective in blocking ADR-induced cytotoxicity. From these results, it is suggested that ADR shows cardioxicity, and the herba extract, as is very effective in preventing ADR-induced cytotoxicity on cultured rat myocardial cells.

Antifertility Effect of Neem (Azadrichta indica) Seed Kernel Meal in Chickens

  • The reproductive performance of forty two male broilers divided into three similar groups and fed on isocaloric and isonitrogneous diets containing 0, 10 or 20% water washed neem seed kernel meal (WWNSKM) was investigated from 20 to 32-wks of age. Results on semen characteristics revealed that feeding of WWNSKM led to significant (p < 0.05) reduction in semen volume, sperm concentration associated with increased incidences of morphological abnormalities in the spermatozoa when compared to that of the control birds. A drastic reduction in the fertilizing ability of spermatozoa was observed, the adverse effects being more at higher inclusion level of the cake. Hatchability of eggs also declined in the WWNSKM fed group. Histological examination of testes revealed a higher number of degenerating cells and poor spermatogenesis along with multinucleated giant cells in the seminiferous tubules of the testes of birds receiving the high dose of WWNSKM in diet. It may be concluded that the feeding of WWNSKM by incorporating in isocaloric and isonitrogneous diets to cockerels is associated with adverse effect on their fertility.
