An Eigen Analysis with Out-of-Plane Deformable Ring Element (면외변형 링 요소를 이용한 고유해석)

  • This paper presents the theoretical natural frequencies of out-of-plane deformable ring based on the variables such as out-of-plane deflection, torsional rotation and shear rotation. Based on the same variables, a finite element eigen analysis is carried out by using the $C^0$-continuous, isoparametric element which has three nodes per element and three degrees-of-freedom at each node. Numerical experiments are peformed to find the integration scheme which produces accurate natural frequencies, natural modes and correct rigid body motion. The uniformly reduced integration and the selective reduced integration give more accurate numerical frequencies than the uniformly full integration, but the uniformly reduced integration produces incorrect rigid body motion while selective reduced integration does correct one. Therefore, the ring element based on the three variables which employes selective reduced integration is recommended to avoid spurious modes, to alleviate the error due to shear locking and to produce correct rigid body motion, simultaneously.

System for Repeated Integration of Various Gene Expression Cassettes in the Yeast Chromosome (효모염색체내에 다양한 유전자발현 cassette의 반복적 integration을 위한 system 구축)

  • In this study, a repeated yeast integrative plasmid (R-YIp) harboring Cre/loxP system was constructed to integrate various gene expression cassettes into the yeast chromosome. The R-YIp system contains a reusable selective marker (CgTRP1), loxP sequence, and target sequence for integration. Therefore, many gene expression cassettes can be integrated into the same position of the same yeast chromosome. In the present study, several model enzymes involving xylan/xylose metabolism were examined, including endoxylanase (XYLP), ${\beta}$-xylosidase (XYLB), xylose reductase (GRE3) and xylitol dehydrogenase (XYL2). Efficient expression of these genes was obtained using two promoters (GAL10p and ADH1p) and various plasmids (pGMF-GENE and pAMF-GENE plasmids) were constructed. The XYLP, XYLB, GRE3, and XYL2 genes were efficiently expressed under the control of the GAL10 promoter. Subsequently, R-YIps containing the GAL10p-GENE-GAL7t cassette were constructed, resulting in pRS-XylP, pRS-XylB, pRS-Gre3, and pRS-Xyl2 plasmids. These plasmids were sequentially integrated into chromosome VII of a Saccharomyces cerevisiae strain by repeated gene integration and selective marker rescue. These genes were integrated by the R-YIp system and were stably expressed in the yeast transformants to produce active recombinant enzymes. Therefore, we expect that the R-YIp system will be able to overcome current limitations of the host cells and allow selective marker selection for the integration of various genes into the yeast chromosome.

Development of a Multicopy Integration Vector in Yarrowia lipolytica (Yarrowia lipolytica의 Multicopy Integration Vector 개발)

  • Multicopy integration vector is a very useful vector system in that they can be integrated into chromosomal DNA in several copies and stably maintained under non-selective conditions. To develop a multicopy integration vector system in the yeast Yarrowia lipolytica, P-type ribosomal DNA was cloned from Y lipolytica. A HindIII-BglII fragment of the cloned rDNA and a promoterless URA3 gene were inserted into pGEM1, generating multicopy integration vectors, pMIYL-1 and pMIYL-2. The rDNA fragment is for targeted homologous recombination between the vector and the chromosomal DNA of Y. lipolytica, and the promoterless URA3 gene is a defective selection marker for inducing multicopy integration. pMIYL-1 and pMIYL-2 have an unique restriction enzyme site, KpnI, and two unique restriction enzyme sites, KpnI and EcoRI, repectively, which can be used for targeting of the vectors into the rDNA of Y. lipolytica chromosomal DNA. After transformation of the vectors into Y. lipolytica, copy number and stability were analyzed by Southern hybridization. The vectors were found to be present in less than 5 copies per cell and were stably maintained during growth in non-selective media.

Performance analysis of TR, DTR and PRM UWB systems in frequency selective channel (주파수 선택적 채널에서의 TR, DTR과 PRM UWB 통신 시스템 성능분석)

  • The UWB signal distortion in frequency selective channel has made it difficult to implement the channel estimator and the synchronizer of the UWB receiver. In this paper, we examine the performances of TR(Transmitted Reference) and DTR(Differential TR) UWB which use either reference pulse or differential method to estimate the channels. we also propose a FSK-based PRM(Pulse Repetition Modulation) UWB system as an another form of UWB system which is advantageous in frequency selective channel. Finally, resorting to statistical analysis, we examine the noise effect due to noisy reference at an auto-correlation(AcR) receiver. moreover, SNR dependance of the integration length in the AcR receiver is also investigated.

Quality Variable Prediction for Dynamic Process Based on Adaptive Principal Component Regression with Selective Integration of Multiple Local Models

  • The measurement of the key product quality index plays an important role in improving the production efficiency and ensuring the safety of the enterprise. Since the actual working conditions and parameters will inevitably change to some extent with time, such as drift of working point, wear of equipment and temperature change, etc., these will lead to the degradation of the quality variable prediction model. To deal with this problem, the selective integrated moving windows based principal component regression (SIMV-PCR) is proposed in this study. In the algorithm of traditional moving window, only the latest local process information is used, and the global process information will not be enough. In order to make full use of the process information contained in the past windows, a set of local models with differences are selected through hypothesis testing theory. The significance levels of both T - test and χ2 - test are used to judge whether there is identity between two local models. Then the models are integrated by Bayesian quality estimation to improve the accuracy of quality variable prediction. The effectiveness of the proposed adaptive soft measurement method is verified by a numerical example and a practical industrial process.

  • Rapid prototyping is becoming an increasingly importnat techniuqe involved in the design cycles of modern industry. The majority of the rapid prototyping systems currently available use photo-reactive resins and waxes as the raw materials. The models produced by these systems often have relatively poor mechanical and physical properties and as such have a limited application to the production of advance prototypes but are excellently suited to the manufacture of engineering prototyes. This work identifies the need to produed near production grade advance prototypes from a variety of metals and a novel prototyping process based on the techniques of selective laser sintering and conventional machining is proposed. The integration of a carbon dioxide laser and a conventional machine tool to create the opto-mechanical by multi-layer sintering and some of the problems involved are also discussed.

  • Some studies on electrochemical additive manufacturing of metals were summarized in this technical report, and development status of selective electrochemical 3D printing technology was introduced. In order to apply it to the PCB mass production process, essential considerations how to overcome the fundamental problems, such as the sizing, process sequence and PCB process design have been described.

  • A less simple approach developed for generation of marker-free transgenic plants is to select transformants without the use of selective marker genes. Some results about development of marker-free transgenic plants were obtained using a non-selective approach in several crops such as rice, potato and tobacco. However, the study did not provide evidence on detailed characterization of introduced gene on genome, a critical step for confirming the stable integration and transmission of a foreign gene. In this study, we evaluated structure and integration sites of transgene (mCry1Ac) in the transgenic Bt rice plants which were made via conventional Agrobacterium-mediated transformation by non-selective method. Structure and integration sites of transgene in these transgenic plants had similar fashion as those recovered under selection.

A Motion-driven Selective Visual Attention System (모션 기반 선택적 주의 시스템)

  • In this paper, a selective visual attention module based on motion stimuli is introduced for the purpose of detecting ROI(region of interest) or FOA(focus of attention) in motion pictures. Analysis of motion fields in our approach is in direct contrast to some of the previous studies of selective visual attention module. Motion that presents temporal visual saliency in an aspect between two successive frames is analyzed based on psychological studies in 'DORF(double opponent receptive fields)' and 'NF(noise filtration)' in MT(middle temporal cortex). Analyzed results are integrated based on the theory of 'motion integration' in MT to obtain a single conspicuous region. Experiments through a human subjective evaluation showed generally accepted results.

A Case Study on Sandplay Therapy for a Girl Suffering from Selective Mutism (선택적 함묵증 여아의 모래놀이치료 사례연구)

  • This study explored the case of sandplay therapy for a 4th grade girl suffering from selective mutism. Her selective mutism apparently began following an extremely embarrassing experience in kindergarten. Her symptoms were a combination of symbiotic, reactive and passive- aggressive type behaviors. The goal of the therapy undertaken with this child was to enable her to express her repression and suppression, within a free and protective space during sandplay therapy. There were a total of 60 sessions of sandplay therapy. The client described the situations she had experienced in the first sandtray, by placing babies absent caring adults and food on the sandbox. She also placed baby fish away from their mother. In the mid-point of the sessions, she repeated her regressive behaviors by babbling like a baby and fought with snakes and monsters. In the final sessions, she showed integration and adaptation by engaging in snow play, expressing the union of opposites, placing blue and red mig and making a cross on the sand. This study showed the effectiveness of sandplay therapy since her selective mutism was lessened.
