Characterization of Fracture System for Comprehensive Safety Evaluation of Radioactive Waste Disposal Site in Subsurface Rockmass (방사성 폐기물 처분부지의 안정성 평가검증을 위한 균열암반 특성화 연구)

  • The purpose of this study is the simulation of discontinuous rockmass and identification of characteristics of discontinuity network as a branch of the study on characteristics of groundwater system in discontinuous rockmass for evaluation of safety on disposal site of radioactive waste. In this study the site for LPG underground storage was selected for the similarities of the conditions which were required for disposal site of radioactive waste. Through the identification of hydraulic properties. characteristics of discontinuities and selection of discontinuity model around LPG underground storage facility. the applications of discrete fracture network model were evaluated for the analysis of pathway. The orientation and spatial density of discontinuities are primarily important elements for the simulation of groundwater and solute transportation in discrete fracture network model. In this study three fracture sets identified and the spatial intensity (P$_{32}$) of discontinuities is revealed as 0.85 $m^2$/㎥. The conductive fracture intensity (P$_{32c}$) estimated for the simulation area around propane cavern (200${\times}$200${\times}$200) is 0.536 $m^2$/㎥. Truncated conductive fracture intensity (T-P$_{32c}$) is calculated as 0.26 $m^2$/㎥ by eliminating the fracture with the iowest transmissivity and based on this value the pathway from the water curtain to PC 2.   • Youngju multipurpose dam is planned to minimizing the damage by flood and obtaining the water for industrial use in Nakdong river region. Faults in rock mass have strong influences on the behaviors of dam structure. Thus, it is very important to analyse for the characteristics of fault rocks in dam design. However, due to the limitation of geotechnical investigation in design stages, engineers have to carry out the additional geological survey including directional boring to find the distribution of faults and the engineering properties of faults for stability of dam. Especially, the selection of location of dam and type of dam considering fault zone must be analyzed through various experimental and numerical analysis. In this study, various geological survey and field tests were carried out to analyse the characteristics of the large fault zone through the complex dam is designed in foundation region. Also, the distribution of structural geology, the shape of faults and the mechanical properties of fault rock were studied for the reasonable design of the location and type of dam for long-term stability of the complex dam.

Development of Bialaphos-Resistant Transgenic Rice Using Agrobacterium tumefaciens (Agrobacterium tumefaciens를 이용한 bialaphos 저항성 형질전환 벼의 개발)

  • The bialaphos is a potent inhibitor of glutamine synthease in higher plants and is used as a non-selective herbicide. We have used the bialaphos resistant gene(Bar) encoding for an acetyltransferase isolated from Streptomyces hygroscopicus SF1293. Callus derived from mature seeds of rice(Oryza sativa L. cv. Dong Jin) were co-cultivated with Agrobacterium tumefaciens EHA101 carring a plasmid pGPTV-HB containing genes for hygromycin resistance (HygR) and Bar. Transgenic plants showing in vitro resistance to 50 mg/L hygromycin and 10 mg/L bialaphos were obtained by using a two-step selection/regeneration procedure. Transformation efficiency of rice was about 30% which was as high as reported in other dicotyledons. Progenies ($\textrm{T}_{1}$ generation) derived from primary transformant of 17 lines were segregated with a 3 resistant : 1 sensitive ratio in medium containing hygromycin and bialaphos. Stable integration of Bar gene into chromosomal DNA was proven by Southern blot analysis of genomic DNA isolated from $\textrm{T}_{2}$ progenies. Transgenic plants ($\textrm{T}_{3}$) grown in the field were resistant to bialaphos (Basta) at a dosage lethal to wild type plants.

Quantitative analysis of water-soluble vitamins and polyphenolic compounds in tomato varieties (Solanum lycopersicum L.) (토마토(Solanum lycopersicum L.) 품종 간 수용성 비타민과 폴리페놀계 성분 함량 변이 분석)

  • Tomato fruit quality is determined by the contents of various functional metabolites in addition to fruit appearance. To develop tomato cultivars with higher amounts of functional compounds, an efficient quantification method is required to identify the natural variations in the compounds in the tomato germplasm. In this study, we investigated tomato varieties, which included 23 inbred lines and 12 commercial F1 cultivars, for their contents of seven watersoluble vitamins (vitamin C, vitamins B1, B2, B3, B5, B6, and B9) and five polyphenolic compounds (quercetin, rutin, kaempferol, myricetin, and naringenin chalcone). The results of high performance liquid chromatography and liquid chromatography-mass spectrometry showed that vitamin C and naringenin chalcone were the major water-soluble vitamins and polyphenolic compounds, respectively, and their abundance was highly variable depending on the cultivar. By contrast, the contents of vitamin B1, quercetin, and kaempferol were lowest among the cultivars. With regard to the relationship between metabolic compounds and fruit characteristics, a significant association was found in fruit size, indicating that cherry tomato varieties contain higher amounts of the compounds compared to large fresh-type varieties. However, no direct association was detected in fruit color, except for naringenin chalcone. The results of this study provide new insights on the quantification of metabolic compounds and the selection of breeding materials, which are prerequisites for the development of functional tomato varieties.

  • Children's literature has a significant impact on children's intelligence and emotional development. The interest in children's literature is increasingly growing with the expectation of the award winning books and the educational values of children's books. Although the interest has brought about a quantitative increase in the domestic children's books, it has difficulty selecting high-quality children's books among various books. In such an environment, the Children's Literature Award will provide an authoritative source of choice, which will help guide the selection of quality books to those who want to read children's books. In addition to this trend, the library should check the status of winning works and incorporate them into new collection development policies. The purpose of this study is to present the direction of the library book development policy of the public library. For this purpose, 10 public libraries in Seoul, Korea were selected and analyzed quantitatively and qualitatively through the collection status of the children's literature winning books, loan status, turnover, Bonn's use Factor, and Trueswell's 80/20 rule. Based on the results of this analysis, we would like to suggest some issues to be reflected in library development policy of the future.

Analysis of Quantitative Trait Loci for Yield Component Traits in Soybean Using Recombinant Inbred Lines (콩에서 수량구성요인과 관련된 양적형질유전자좌의 분석)

  • Seed and pod numbers are the main yield components in soybean. Selection for increased yield potential is main goal of plant breeding. The objective of this study was to identify quantitative trait loci(QTLs) that control pod number per plant, seed number per plant and pod in soybean. The 117 $F_{2:10}$ recombinant inbred lines(RILs) developed from a cross of 'Keunolkong' and 'Shinpaldalkong' were used. Two independent QTLs for pod number per plant were identified from linkage group(LG) F and L. Two QTLs for seed number per plant were located on LG F and L. Seed number per pod was related with three QTLs located on LG D1a, D1b and F. Pod and seed number per plant have two common QTLs on LG F and L.

Vegetation Structure in Otter (Lutra lutra) Home Range of Hwacheon, Gangwon-do (강원도 화천군 수달(Lutra lutra) 서식지의 식생 구조)

  • In order to determine whether vegetation would be one of the factors for the selection of otter home range, vegetation structure and other potential factors were studied in Hwacheon, Korea. Thirteen sites, otter's activity found and not found, were investigated in North Han River and connected tributary streams of Hwacheon-gun. Three types of vegetation were classified by cluster analysis, which is short grass, tall grass and shrub type. Vegetation zone of each channel is composed of either one type, or mosaic of tall grass and shrub type. Short grass type is common in Lake Paro and upper North Han-river where water level is highly variable throughout a year. Therefore, annual species such as Persicaria nodosa, Fimbristylis dichotomam and Chenopodium ficifolium are the most dominant. Shrub type is common at the downstream sites of Jichon stream and along mainstream of North Han River down Lake Paro. A shrub species, Salix koreensis, is the most common. Tall grass type is dominant occupying the most vegetation zone of the tributary channels. Phragmites japonica is absolutely dominant. Due to its dense cover, a few plant species are co-existed. Otter activity was found in all three vegetation types and no marked activity was found at some sites of tall grass type. There is no difference in species composition and physiognomy between tall grass sites with and without otter activity, while it shows significant difference in fish availability between two groups. Overall we found that home range of otters in the region is along the mainstream and downstream of tributary streams with high fish availability in all vegetation types and in various human activity levels.

Selection of Promising 'Ssam' Vegetable for Summer Production in Highland (고랭지 여름출하용 유망 쌈채소 선발)

  • Field investigations were initiated to determine possible new 'ssam' vegetable that could be grown during the summer season in highland. Korean 'ssam' means cooked rice in vegetables. The 'ssam' vegetable is called that those leaves has been used for wrapping at meal. Seventeen leafy vegetables were studied, including chard (Beta vulgaris L. var. flavescens DC.), leaf broccoli (Brassica oleracea L. var. italica), Chinese leek (Allium tuberosum Rottl.), Japanese hornwort (Cryptotaenia japonica Hassk), red leaf mustard (Brassica juncea L.), green leaf mustard (Brassica juncea L.), leaf parsley (Petroselinum crispum Nijim), pakchoi (Brassica campestris L. ssp. chinensis Jusl.) tatsoi (Brassica campestris var. narisoma), kale (Brassica oleracea var. acephala), collard (Brassica oleracea var. acephala), Korean mint (Agastache rugosa O. Kuntze), romaine lettuce (Lactuca sativa L. var. longifolia Lam.), red leaf chicory (Cichorium intybus L. var. folisum), red chicory (Cichorium intybus L. var. folisum), green leaf chicory (Cichorium intybus L. var. folisum), and sprouting broccoli (Brassica oleracea L. var. italica). Date of the first harvest in 1999 was July 10th in chard, leaf broccoli, red mustard, mustard, leaf parsley, pakchoi, tatsoi, kale, collard, Korean mint, red leaf chicory, red chicory, green leaf chicory, and sprouting broccoli. Date of the first harvest for Chinese leek, Japanese hornwort, and romaine lettuce was from the middle of June to the late of August. Based on ratings on marketable yield, we found good leaf shape, taste and yield in chard, red leaf mustard, green leaf mustard, pakchoi, tatsoi, romaine lettuce, red leaf chicory, red chicory, and green leaf chicory. Even though the most of all these vegetables were obtained high scores by test panels in shape, taste and quality, but some of them revealed inadequate leaf size as a 'ssam' vegetable. That problem will be solved by control of harvesting time. On the basis of this observation, chard, red leaf mustard, green leaf mustard, pakchoi, tatsoi, romaine lettuce, red leaf chicory, red chicory, and green leaf chicory were rated as promising 'ssam' vegetable by analysis of profits, adaptation, and quality.

Inheritance of Fruit Ripening Time in Oriental Pear (Pyrus pyrifolia var. culta Nakai) (동양배 과실 숙기형질의 유전분석)

  • To improve the breeding efficiency of oriental pear, heritability, correlation and frequency distribution of fruit ripening date were analyzed using 4,035 seedlings obtained from 15 families between 13 parental cultivars. Although variation of fruit ripening time was higher in most early-ripening parental cultivars than in late-ripening parental cultivars, according to analysis of average, standard deviation, and coefficient of ripening variation for ten years, fruit ripening time obtained from all parental cultivars was inherited narrower and more stable variation than others fruit trait, with 0.92-3.41 in coefficient of variation. The heritability of fruit ripening time was calculated to be over 0.8 in almost all crosses and average fruit ripening time of seedlings from cross combinations could be predicted based on that of the parental cultivars due to its superior heritability relative to other fruit traits. The average ripening time was earlier than the mid-parental value in families obtained from cross combinations using at least one late-ripening cultivar as parent, indicating that the early-ripening trait was more likely to be dominant compared to the late-ripening trait. By contrast, average ripening time was clustered in families of crosses not only between mid-season and early-season cultivars, but also between mid-season and mid-season cultivars. There was highly significant relationship (at 0.68) between mid-parental and progeny mean fruit ripening time. The correlation between fruit ripening time and fruit weight was also highly positive and thus, the mid-parental fruit ripening time could be a potent criterion for indirect selection of fruit weight.

A Comparison Study of Foreign Nation's Risk Management Programs for Controlling Foodborne Pathogens (제외국 식중독균 위해관리 정책 비교 연구)

  • This study was performed to acquire the information on the foodborne pathogen risk management programs in a couple of developed countries by the expert meeting and searching the information on the web. The backgrounds, strategies and effects related to microbial hazards of the foodborne pathogen reduction programs in fresh produce (US), Escherichia coli O157:H7 in ground beef (US), Salmonella in chicken, pork and eggs (Denmark), and Vibrio parahaemolyticus in seafood (Japan) were investigated for case study. A comparison among the pathogen reduction programs was conducted to find advantages and disadvantages and implications of the policies to bring out implications of the programs. A model for foodborne pathogen reduction program was developed based on both the CODEX risk management scheme and the case studies as follows; 1) preliminary risk management activities, 2) planing a foodborne pathogen reduction program, 3) option identification and selection, 4) implementation (conducting the each stake-holders role and applying the intervention methods), 5) monitoring activities, 6) interim review, 7) continuation or amendment of implementation method by the interim review before achieving the goal, and 8) final review and additional cost-benefit analysis if necessary. This proposed model according to the role of the stake-holders can be used to conduct microbial risk management programs in Korea in the near future.
