Biocontrol of plant pathogens provides an alternative means of reducing the incidence of plant diseases without the negative aspects of chemical pesticides. Nowdays, as the resistant fungi about the chemical fungicides have revealed and the concern of environment has increased, the biological control of phytopathogenic fungi by the antagonistic microorganisms is very much indispensable. For the selection of strong antagonistic bacterium for biological control agent of rice leafblast and cucumber gray mold rot, the antifungal strain KL-3 strain was selected among 120 strains isolated from the rhizosphere soils. And the strain was identified to be a species of Bacillus subtilis or closely related strain. In several biochemical and in vitro antibiosis tests, antifungal substances of Bacillus sp. KL-3 were presumed heat stable, micromolecular antibiotic substances. In vivo test and vinyl house field test, the antifungal substances of Bacillus sp. KL-3 represented excellent biocontrol ability aganist Alternaria mali, Phyricularia oryzae, and Alternaria kikuchiana as well as broad spectrum of other fungi. In particular, Bacillus sp. KL-3 strain showed more predominant activity than some chemical fungicides against fungi shown to resist chemcal fungicides.
Photosynthetic bacteria which can utilize ferulate as a sole carbon source for their metabolic activities were isolated from soils by liquid enrichment culture technique. The strain P-1 was selected by the highest capability of degrading ferulate in aerobic and anaerobic conditions. The strain P-1 was rod-shaped with its motility, strained gram negatively and could not utilize sulfur compounds. This strain has the bacteriochlorophyll a group I carotenoid and membrane structures like lamellae. As the results of physiological, morphological and cultural charactderistics, the isolate was identified as Rhodopseudomonas plaustris, one of the purple nonsulfer bacteria. The strain P-1 utilized 2mM/day in aerobic condition and 0.86 mM/day in anaerobic condition.
Quantification of Carbon stock has become in the contest of changing climate and mitigation potential of forests. Two different forest types, Dry Shiwalik Sal Forest and Moist Shiwalik Sal Forest in Barkot and Lachchiwala of Doon Valley, Western Himalaya are selected for the study. Volume equations, destructive sampling and laboratory analysis are done to estimate the carbon stock in different carbon pools like trees, shrubs, herbs and soils. Considerable variations are observed in terms of carbon stocks in different forest types. In Dry Shiwalik Sal Forest, carbon stock density varied between 129.81 and $136.00MgCha^{-1}$ while in Moist Shiwalik Sal Forest, carbon stock density ranged from 222.29 to $271.67MgCha^{-1}$. Tree species like Shorea robusta, Syzigium cumini, Miliusa velutina, Acacia catechu, and Mallotus philippensis had significant role in carbon sequestration. Shorea robusta had contributed highest in carbon stock due to highest density. Total of 2,338,280.165 Mg carbon stock was estimated in all the forest types.
This experiment was conducted to investigate the effects of turf grass growth to seedling rates and bed soil types. The results of this experiment were summarized as follows; the more increasing the seedling rates, the plant height and leaf length were longer, but the number of leaf and number of branch were fewer. Incase of transplanting of turf grass seedling by rice transplanting machine, it might be considered that the proper alternative bed soil was sandy loam soil with regarding to the economic aspects. The maximum seedling rate of turf grass in the seedling tray for rice was 1,000 of seedling amount due to the nutrient competition with intensive seedling. As the results indicated, it might be recommended that the proper alternative bed soil was sandy loam soil with 1,000 of seedling rates in case of transplanting of turf grass seedling by rice transplanting machine.
A promising biocontrol agent, A2l-4, against Phytophthora blight of pepper was selected from 351 bacterial isolates collected from rhizosphere soils and roots of onion (Allium fistulosum L.). The isolate A21-4 was identified as Serratia plymuthica based on its 16S rRNA sequence and key characteristics as compared with that of an authentic culture of S. plymuthica (ATCC No. 6109D01). The isolate readily colonized on roots of various crops including pepper when inoculated on seed and not. Strain A2l-4 showed narrow spectrum of antibiotic activity, as revealed in its strong inhibitory activity to the genera Pythium and Phytophthora, but not to Fuasrium and Rhizoctonia. In pot experiments, none of the pepper seedlings treated with A2l-4 were infected by Phytophthora capsici, while 86% of the control plants were killed by the pathogen.
This study dealt with the comparison of lateral baring capacity for vertical PHC pile between predicted and measured values driven in weathered granite soils to build world cup gymnasium in Kwangju area. Recently, the calculation of horizontal bearing capacity of piles foundation has been considered very important for earthquake or wind resistant design in Korea. During this study , Matlock & Reese, Broms and Chang's methods were selected in prediction of lateral resistant of PHC piles. As for case study, the prediction values were compared with 5 measured ones based on ASTM. The result showed that prediction values proposed by Matlock & Reese , Chang and Broms were smaller that real values. Three proposed methods by Matlock & Reese and Chang based on lateral deflection and Broms by ultimated lateral resistance turned out valid in view of engineering practice.
Soils contaminated by hazardous hydrophobic organic compounds, such as polycyclic aromatic hydrocarbons (PAHs), have become a major environmental issue due to toxic and carcinogenic properties of those compounds. In this work, we investigated effects of various metabolic inducers on phenanthrene biodegradation. Biodegradation tests were peformed with two different Pseudomonads: Pseudononrdia hydrocarboxydans (Gram positive) and Pseudomonas putida (Gram negative). Intermediates of phenanthrene metabolism (1-hydroxy-2-naphthoate, salicylate, catechol, phthalate and protocatechuate) were selected as inducers. The tests indicated that 1-hydroxy-2-naphthoate was the most effective inducer and enhanced the phenanthrene degradation rate up to 5.7 times, even though all the others also had induction ability to some extent. The effective induction could be achieved even at a low concentration of 1-hydroxy-2-naphthoate. Addition of metabolic inducers would be an attractive trick for the successful bioremediation of PAH-contaminated soil.
The EPS construction method-one kind of the load reducing methods-utilizes the EPS blocks, ultra-light materials whose unit weight is about 1/100 of soils and has been applied to many soft ground sites. It needed 3,000 days to get the 90% degree of the consolidation for the case of 12m high soil embankments on the 30m thick soft clayey foundations. The N value of SPT at this deposit was less than 5. The pack drain was installed to promote the radial consolidations. Although staged embankments were planned, designers failed to get a sufficient stability of the foundation ground. Therefore, the EPS fill method was selected to reduce the load and the construction period. EPS blocks(D-20 model) replaced the upper part of the soil embankments. These complex embankments reduced the ground settlement and the construction period. The possibility of lateral movements of the bridge abutments was checked and the design scheme was reviewed.
In this study, coarse-grained geomaterials were mixed with cementing binder. To do that, typical soils from road construction sites were selected to assess the strength and stiffness characteristics of cemented geomaterials mixed with cement and recycled fly ash. Mechanistic evaluation on these samples was performed depending on the various binder contents. Increasing cementing content tend to increase the resilient modulus under repeated loadings and unconfined strength respectively. In addition, the toughness of cemented geomaterials was also estimated in order to check the ability to resisting fatigue failure.
